        비타민 C와 강황 에탄올 추출물이 인체 피부보습에 미치는 효과

        As the human body ages, various forms of physiological metabolic deterioration appear and skin aging is a typical phenomenon. Especially, occurrence of wrinkles due to cell senescence and reduction of moisture due to residual active oxygen are considered as representative factors of skin aging. In this study, a skin test formulation containing vitamin C that is effective for the removal of free radicals and Curcuma longa ethanol extract(CLEE) known to have antioxidant and skin moisturizing effect, was constituted and a skin clinical trial was conducted using it. We have produced a CLEE-containing formulation with the effect of moisturizing. And then we measured moisturizing effects each or together by Coneometer. After application of the vitamin C and CLEE each or together, the average levels in the applied parts of the skin significantly increased by 35.42%, respectively (p<0.05). These data indicate that coadministration of vitamin C and CLEE improve moisturizing effect in the human skin.

        배롱나무 잎 추출물 및 분획물의 항염 및 주름개선 효능평가

        To confirm the potential for application of Lagerstroemia indica leaves as functional cosmetic materials, samples were prepared by extraction with 70% ethanol and fraction with ethyl acetate, methylene chloride, and hexane. The result of evaluation of the anti-inflammatory activity at a concentration without cytotoxicity showed that the ability to inhibit NO production in the crude extracts was the highest at 24.44%. The result of evaluation of the mRNA expression levels of HAS-2 and AQP-3 showed that there was no significant effect (p<0.05). According to the result of confirming the mRNA expression level of MMP-1, treatment w ith the ethyl acetate fraction at a concentration of 60 μg/mL resulted in an expression inhibitory effect of 51.54% and significance was recognized (p<0.05). The results of this study confirmed the applicability of the L. indica leaves fraction as an anti-inflammatory and anti-wrinkle cosmetic.

        백화사설초(Hedyotis diffusa Willd) 에탄올 추출물의 Hyaluronic Acid 합성 효과

        Hedyotis diffusa Willd, a plant belonging to the Rubiaceae, is mainly used for medicinal purposes. It has traditionally been used in treatment of carcinoma and to neutralize snake venom. Currently, it is also widely used as a raw material for pharmaceuticals and cosmetics. Hyaluronic acid is an essential molecule for maintaining a moisturizing effect in human cells, including keratinocytes. We conducted an experiment to determine the effect of hyaluronic acid biosynthesis of Hedyotis diffusa Willd. on human skin. The results of the experiment confirmed that Hedyotis diffusa Willd. did not exhibit cytotoxicity in animal cells. The results showed that the expression of genes related to hyaluronic acid synthesis and the amount of hyaluronic acid produced on keratinocytes also increased. In particular, the biosynthetic effect of hyaluronic acid reached 106% compared to the positive control all trans retinoic acid (ATRA). Based on these results, it was determined that apples could exhibit a skin moisturizing effect by promoting production of hyaluronic acid in animal cells

        백선 추출물의 간세포 손상에 대한 연구

        Objectives : This study was carried out to evaluate whether the water extract from cause the cellular damage in HepG2 cell line. It was reported that Dictamnus dasycarpus Turcz(DDT) intake induce poisoning symptoms in human population. These symptoms was closely related to liver toxicity, however, mechanisms for liver toxicity caused by DDT have not been elucidated exactly. Here, hepatotoxicity caused by DDT was evaluated using HepG2 cell line. Methods : Water extract of DDT was treated into HepG2 cell with various doses such as 0, 0.1, 0.5, 1.0 and 5.0 ㎎/㎖. In order to cell viability, both MTT and LDH assay were carried out. Also, apoptosis array kit was used to identify whether cell death caused by DDT is due to apoptosis or not. In addition, reactive oxygen species (ROS) was measured after treatment of water extract. Results : We found out significant changes in the apoptosis related factors of hepatocyte. The cell viability of HepG2 treated with DDT water extract was decreased in dose-dependent. Also most of the apoptosis related factors were significantly increased. We found out that Caspase 3, Cytochrome C and ROS had increased in dose-dependent. In addition, other apoptosis related factors Bcl 2 and Bax, which were also constant changes. However, there was no significance. Conclusions : These results suggest that water soluble extract of DDT is expected to have oral toxicity, including hepatocellular damage Therefore, it is suggested that DDT could cause various side effects and toxicity of clinical conditions.

        냉증에 대한 산소챔버의 임상 효능 및 안전성 연구

        Cold hypersensitivity is regarded to be associated with blood circulation. This study is aims to evaluate the effects and safety of oxygen chamber therapy on cold hypersensitivity by comparing the temperature and Visual Analogue Scale. Methods: 42 outpatients who visited ○○ University Oriental Hospital from July 11th, 2013 to August 28th, 2013 were analyzed. Patients were subjected to thermometer, and those with thermal difference greater than 0.3℃ between upper arm and palm and also with more than VAS 4 of cold hypersensitivity were diagnosed with cold hypersensitivity. 42 outpatients diagnosed with cold hypersensitivity are divided into two groups, one is the experimental group consisted of 21 patients and other was control group consisted of 21 patients. The experimental group had oxygen chamber therapy 10 times for 4 weeks. Thereafter the effects of oxygen chamber therapy on cold hypersensitivity was analyzed with t-text using SPSS for Windows version 21. Results: After the oxygen chamber therapy, experimental group had considerable improvement on cold hypersensitivity, in consequence of decreasing rate of thermal difference and VAS of cold hypersensitivity. Ear deafness and hand numbness were reported as an adverse effects in experimental group, but there was no serious adverse effects. Conclusions: This clinical trial showed oxygen chamber therapy could be effective and safe to reduce cold hypersensitivity.

        탁리소독음 함유 크림의 피부탄력 개선 효과

        Dermal elasticity is closely related to generation of wrinkles. Especially, around face and eyes, wrinkles are particularly affected by skin elasticity. Taklisodokeum, a traditional oriental medicinal prescription, is typically used as a medicine for dermatological diseases. Recently, dermal damages are increasing due to environmental factors and pollution. Therefore, we investigated the dermal elasticity effect of a Taklisodokeum-containing cream. The dermal elasticity improvement was measured on eye side of face using a cutometer. Measurements were performed in triplicate before the product use (0 weeks) and after its use for 2 and 4 weeks. The cream increased the average dermal elasticity from 0.831 A.U. when the test was started to 0.853 A.U. (by 2.65%) after 2 weeks (p<0.005) and to 0.874 A.U. (by 5.17%) after 4 weeks (p<0.000). The above results indicate that Taklisodokeum cream has an obvious dermal elasticity improvement effect.

        죽력(竹瀝)의 인체 피부에 대한 안전성과 보습 효과

        Bamboo is a useful plant resource for mankind. Bambusae Caulis in Liquamen (BCL) is one of the processed products of bamboo, and is made by refining the sap of bamboo through heating. Although various uses of bamboo have been known, clinical study of moisturizing effect has never been conducted. In the present study, the clinical trial was conducted in accordance with the clinical trial regulations of the KFDA. Skin safety test results were determined to be non-stimulation without any irritation. Clinical trials were conducted on a non-treated group, control group, and test group, and significant results were obtained. The amount of moisture present on the skin was measured using corneometer. The measurement time was at 0 h, 0.5 h, 3 h, 6 h. The moisturizing effect significantly increased by 155% at 0.5 h compared to the non-treated group (p<0.001). Meanwhile, at 6 h, the gap between the two test groups reduced. This study is characterized by the clinical study using raw materials without making formulations. The test results were also confirmed as dose-dependent results. Based on the results of this clinical study, it is hoped that the raw materials can be used in various cosmetic formulations.

        인동등 에탄올 추출물의 HaCaT세포에서 Hyaluronic acid 생성 증가 효과

        Hyaluronic acid(HA) is composed of repeating disaccharide(D-glucuronic acid and N-acetyl-D-glucosamine). It has been known to play important roles in physiological and pathological events such as cell immigration and inflammation. Especially, HA is deeply associated with structural stiffness and hydration in skin. Production of HA by hyaluronic acid synthases(HASs), especially HAS2 in human and degradation of that by hyaluronidase are responsible for the HA homeostasis. Destruction of balance between production and degradation of HA could result in skin aging. Therefore, researches on molecules which induce the HAS up-regulation or inhibit the hyaluronidase activity have been carried out. In this study, we investigated the effect of Lonicera japonica ethanol extract(LJ) on HA production in HaCaT cells. LJ increased the 60% HA production compared to that of vehicle control, which could be mediated by HAS2 mRNA up-regulation not by hyaluronidase inhibition. Taken together, LJ could be developed for anti-aging and anti-wrinkle cosmetics. It still remains to be elucidated which compounds are responsible for HAS2 mRNA up-regulation and which signal pathways are involved in transcriptional regulation. 본 연구에서는 인동등의 에탄올 추출물이 HA 생성에 미치는 영향을 확인하고자 하였다. 인동등 에탄올 추출물 50, 100 및 200μg/mL 처리에서 각각 vehicle control 대비 39.26%, 50.14%, 60.01%의 HA 생성 증가를 보였다. HA량은 HA를 생성하는 HAS2와 분해하는 hyaluronidase 활성에 영향을 받기 때문에 인동등 에탄올 추출물이 hyaluronidase 활성을 저해하는지를 실험한 결과 인동 에탄올 추출물은 hyaluronidase 활성을 저해 효능을 보이지 않았다. HAS2 유전자 발현에 미치는 영향을 실험해 본 결과 50, 100, 200μg/mL 처리에서 vehicle control 대비 각각 1.99, 6.23, 7.36배의 증가를 보였다. 이를 통해 인동등 에탄올 추출물이 HAS2의 전사과정에 관여하여 유전자 발현을 유도함으로서 HA량을 늘릴 수 있음을 확인할 수 있었다. 따라서 인동 에탄올 추출물이 피부에서 보습, 항노화, 항주름 효과를 나타낼 수 있을 것으로 생각되며 기능성 화장품 원료로 개발이 가능할 것으로 보인다.

      • 원형탈모증의 임상양상 및 기술역학적 연구

        본 연구는 2004년 1월부터 2008년 10월까지 원광대학교 병원 피부과에 내원한 원형 탈모증 환자 126명중 치료 결과를 확인 할 수 있었던 95명을 대상으로 인구학적 특성 및 치료 방법에 따른 치료 효과의 차이를 알아보고자 빈도 분석 및 교차분석을 통하여 유의성을 검정하였다. 탈모반면적은 성별에서 남자와 여자가 각각 26-50%일 때 53.5%, 71.4%로 가장 많았으나 유의한 차이는 없었고, 연령별로는 20대 이하와 30대, 40대 이상이 각각 26-50%일 때 54.1%, 51.9%, 58.6%로 가장 많았으나 유의한 차이는 없었다. 치료방법은 성별에서 남자와 여자 각각 부신피질호르몬 국소방법을 94.3%, 85.7%로 가장 많았으나 유의한 차이는 없었고, 연령별로는 20대 이하, 30대, 40대 이상이 각각 스테로이드 국소 주사요법을 89.7%, 100%, 93.1%로 가장 많았으나 유의한 차이는 없었다. 치료효과는 성별에서 남자와 여자 각각 완치하였다가 64.8%, 71.4%로 가장 많았으나 유의한 차이가 없었고, 연령별로는 20대 이하와 30대의 경우 완치되는 경우가 각각 71.8%, 85.2%로 가장 많았고, 40대 이상은 부분반응 하는 경우가 58.6%로 가장 많았으며 통계적으로 유의한 차이를 보였다(p<0.05) 치료방법에 따른 치료효과에는 유의한 차이가 있었다(p=0.003). 스테로이드 pulse therapy를 사용한 경우 완치가 100%, DPCP 요법은 완치가 77.8%, minoxidil 국소요법은 완치가 73.8%, 스테로이드 pulse therapy의 경우 완치가 100%로 가장 많았다. Background: We embarked on this research in order to gain fundamental information and knowledge on the key differences in the effectiveness of various treatments and factors, such as key clinical conditions and demographic traits through the descriptive research-based procedures, on patients with alopecia areata. Methods: Based on the comprehensive data of 95 patients with verifiable treatment records out of 126 alopecia areata patients who have visited the department of dermatology of Wonkwang University Hospital from January 2004 to October 2008, We examined the significance tests through the analysis of frequency and range of the differences in the effectiveness of various treatments using demographic traits and medical treatments. Results: In terms of the treatment effectiveness, 64.8% of the male patients and 71.4% of the female patients showed the full recovery, with virtually no significant clinical data difference. In terms of the age group variations, 71.8% of the patients in 20's and under and 85.2% of the patients in 30's showed the full recovery, and 58.6% of 40's showed only partial recovery, rendering this difference quite significant.(p<0.05). There was a noticeable difference in effectiveness of various treatments.(p<0.01). Through the steroid pulse therapy, 100% of the patients fully recovered, 77.8% through the DPCP(Diphencyprone) method, 73.8% through the minoxidil application treatment, 65.2% through the steroid injection treatment, and finally 33.3% of the steroid application treatment fully recovering from the hair loss. Conclusion: In terms of the effectiveness, the steroid pulse therapy were most dominant and clear treatment, followed by DPCP treatment, the minoxidil application treatment, the steroid injection treatment, and the steroid application treatment in this order. In terms of the distribution of the gender factor of the observed patients, the number of female patients was far smaller than that of male patients, and tremendous barriers and challenges lie in the quest to make a conclusive finding through the use of the raw data of hospital patients. As such, a variety of rigorous and dynamic clinical research endeavors and the large sample of patients are necessary and strongly recommended at this point.

        뷰티소재 개발을 위한 머루포도 에탄올 추출물의 생리활성 평가

        . Grapes have been used as an important food resource and medicinal plant since the beginning of human civilization. Grapes are used as fresh fruits; dried grapes, which are easy to store, can be used to make alcoholic beverages through the process of fermentation. Scientific research on traditionally used natural raw materials is currently underway, and additional developments are anticipated. Muscat bailey A(MBA) is an improved variety from wild grapes and should be regarded as valuable as both a food ingredient and as a cosmetic ingredient. The purpose of this study was to examine the usefulness of grapes to the human body, the effects on antioxidant effects, cytotoxicity, and physiological factors involved in skin moisturization. Significant results were obtained in the evaluation of the antioxidant effect using the DPPH assay. The results from evaluation of HAS 2 gene expression in relation to the skin moisturizing effect confirmed an increase of approximately % compared to the control group. Hyaluronic acid (HA) is a polysaccharide involved in water retention in the human body, and hyaluronidase is an enzyme involved in decomposition of hyaluronic acid. Therefore, we examined the question of whether hyaluronidase can be inhibited by MBA; the results indicated approximately % greater effectiveness compared to the positive control. Based on these experimental results, recognition of the value of MBA as a raw material for maintaining skin health is anticipated.
