        Withaferin-A Inhibits Colon Cancer Cell Growth by Blocking STAT3 Transcriptional Activity

        Background: Withania somnifera (known as Ashwagandha) is a medicinal plant used in the ayurvedic medicines in India. Withaferin-A, a withanolide derived from the leaf extract of W. somnifera, has been reported to exhibit anti-tumor activity against various cancer cells, such as leukemia, breast cancer and colon cancer cells. Methods: We investigated the anti-cancer effects of withaferin-A on the proliferation and migration of human colorectal cancer (HCT116) cells. And we evaluated the effects of withaferin-A on the transcriptional activity of STAT3 and the growth of HCT116 cells in xenograft mouse tumor model. Results: In the present study, we found that withaferin-A inhibited the proliferation and migration of HCT116 cells in a concentration-dependent manner. Treatment of HCT116 cells with withaferin-A attenuated interleukin-6-induced activation of STAT3, which has been implicated in the development and progression of colon cancer. To examine the effect of withaferin-A on HCT116 cells proliferation in vivo, we generated HCT116 cells xenograft tumors in Balb/c nude mice and treated the tumor bearing mice with or without withaferin-A intraperitoneally. Treatment with withaferin-A exhibited significant decrease in the volume and weight of tumors as compared to untreated controls. Conclusions: The present study suggests that withaferin-A holds the potential to be developed as a small molecule inhibitor of STAT3 for the treatment of HCT116.

        청소년에서 구강건강관련 자기효능감과 사회적 지지가 구강건강관련 삶의 질에 미치는 영향

        Objectives: This study aimed to examine the effects of oral health-related self efficacy and social support on oral health-related quality of life, and analyze the related factors. Methods: A cross-sectional study was carried out on 979 high school students living in Iksan. Data were collected from January 15 to 30, 2014. Individual self-administered surveys were conducted through the sampling method. The contents of the survey were the characteristics of the subjects, oral health-related self efficacy (OHSE), social support related to oral health (OHSS), and quality of life related to oral health (QHRQoL). A survey tool designed by the author was used to measure OHSE and OHSS. QHRQoL used Child Oral Health Impact Profile-Short Form 19 (COHIP-SF 19) for examination. The statistical analysis methods used were mean difference, correlation, and multiple regression analyses. Results: QHRQol was correlated with OHSE, but not OHSS. Per the multivariate analysis, factors that affected QHRQoL were gender, mother’s education level, academic stress, and OHSE; OHSS did not have any effect. Conclusions: It was found that OHSE influenced QHRQoL. Therefore, it is necessary to develop education programs that can enhance OHSE.

        천연물 지표성분들의 세포독성 및 AP-1 활성 평가를 통한 암예방 기능성 탐색

        암을 유발하는 PMA는 세포를 자극하여 전사인자 C-Jun/C-fos의 발현을 증가시키며 핵 내의 AP-1의 활성 을 증가하게 되고 결과적으로 각종 암이 발생된다고 보고되고 있다. 본 연구에서는 천연물 지표성분의 안전성을 보 기위한 세포독성과 암예방 효과를 예측할 수 있는 Activator protein(AP-1) 활성억제를 관찰하였다. 천연물 지표성 분 38종을 대상으로 세포독성과 AP-1 활성억제를 관찰한 결과 섬수의 지표성분인 Bufogein 과 Cinobufagin에서는 세포독성을 보이며 AP-1 활성 억제와 비교 시 5배 정도 차이를 보임을 알 수가 있다. 반면에 Arctigenin, Manassantin A, B에서는 AP-1 활성 억제 농도는 2 μM 이하이면서 세포독성과의 비율은 15배 이상임을 알 수 있다. 본 실험의 결과를 통해 우방자와 삼백초의 지표성분인 Arctigenin, Manassantin A,B는 암 예방 연구의 가능성을 시사하였다. Cancer-inducing PMA stimulates cells to increase the expression of transcription factor c-Jun/c-fos and then increase the activity of AP-1 in the nucleus. The activity of AP-1 has been reported to cause cancer. In this study, We conducted cytotoxicity experiments to assess the safety of natural marker compounds and also observed inhibition of activator protein(AP-1) activity to predict cancer-preventing effects. The results of this experiment indicated that arctigenin, manassantin A, and B can predict the development of cancer prevention agents.

        해양심층수와 γ-Aminobutyric Acid (GABA) 함유 배양액의 결정화를 이용한 GABA함유 소금의 제조

        γ-Aminobutyric acid (GABA)-containing salt was prepared by crystallization of a mixture of salt water from deep sea and fermentation broth by lactic acid bacteria that contained GABA converted from glutamic acid. Salt from deep sea water has a lower sodium content but higher calcium, potassium and magnesium contents than commercial salt. Instead of monosodium glutamate (MSG), glutamic acid was used for solving the residual MSG problem. Fermentation by a lactic acid bacterium converted 90% of added glutamic acid (5%, w/v) to GABA, and continuous production of colorless fermentation broth containing more than 3% (w/v) GABA was achieved by using an activated carbon. Mixtures of salt water and fermentation broths with various GABA concentrations were co-crystallized and the GABA content was analyzed. This analysis showed that more than 90% of GABA from broth was adsorbed to salt. The appearance and surface of this prepared GABA-containing salt were examined with an image analyzer and scanning electron microscope. No difference was found with commercial sun-dried salt and no separated particles were detected, which indicates that the co-crystallization process used is suitable for the production of GABAcontaining salt.

        천연 생약에서 추출한 지표성분들의 모발 성장에 미치는 영향

        Many people around the world are stressed by hair loss, and drug-based therapies (minoxidil, finasteride) are available for the treatment of alopecia, but they have several side effects. Therefore, in order to reduce the side effects of anti-hair loss drugs, the use of new treatments using products developed from natural extracts has been emphasized. In this study, we focused on the mechanisms underlying the hair growth-promoting effects of marker compounds of natural herbal extracts. The 32 marker compounds of natural herbal (10 μM) were used to evaluate the growth of human hair follicle dermal papilla cells, the expression of mRNA genes, and the signaling transcription factor activities of Wnt and JAKs. Seven marker compounds were selected that induced equal or greater growth than minoxidil or tofacitinib in cell growth. For the seven marker compounds, we evaluated the expression of mRNA genes and the transcriptional activities of Wnt/β-catenin and IL-4/JAK3-STAT6, which are major signaling pathways that regulate hair growth. As a result of the evaluation, it was scientifically proven that the 4 marker compounds (honokiol, isoalantolactone, broussochalcone A, and 3-deoxysappanone B) induce hair growth. I have suggested the possibility of developing a natural product for improving hair loss with 4 marker compounds.

        여대생의 체형인식과 비만지식 및 비만스트레스 조사

        The purpose of this study is to know about real body shape and ideal body shape and to examine fatness knowledge and fatness stress. In order to examine general characteristics, frequency analysis was conducted. To know about gap between body shape recognition and ideal body shape, fatness knowledge and fatness stress index, t-test and One-Wey ANOVA. The subjects were 344 college woman being in K-woman`s university in Kwang-ju and their questionaire survey was the final analysis data. Study results were summarized as follows; The difference between recognition of real body shape and ideal body shape was meaningless. Fatness knowledge also showed no difference. Self cathexis express dissatisfaction. The fatter the body shape recognition, the higher the fatness stress index And also the higher the fatness knowledge, the higher the fatness stress.

        비만여대생의 등경락마사지 후 근체형 변화 연구

        The purpose of this study was to determine effects of back meridian massage on changes in the body type among obese female college students Subjects` BMI was over 23 and conducted a total of 24 sessions of back meridian massage, two sessions per week, for 12 weeks from May 19 to August 15, 2008, and analyzed changes in the muscular type. This study analysed by SPSSWIN 12.0 program. As for changes in the muscular type, there were significant differences in changes in horizontality of the upper scapula(X, Y, Z1) There were significant differences in changes in horizontality of the lower scapula(X, Y, Z2) among all the measurements twelve weeks after the (p<0.01 p<0.05). There were significant difference in changes in horizontality of the waist part(X, Y, Z3) in X and Z3 (p<0.001), Changes in Y4 for horizontality of the upper hips (X, Y, Z4), changes in X and Z4 according to the time of experiment was significant(p<0.001). The effects of back meridian massage on changes in the muscular body type were positive, therefore it is expected to be used as body type management programmed also to be used as basic data for marketing in the beauty industry or obesity and body type management programs.

        여고생의 통제의 소재에 따른 구강보건행태

        Objectives. The purpose of this study was to investigate the correlation between the oral health locus of control and the oral health behavior and attitude. Methods. 280 girls in W high school located in Ik-san city were chosen by the convenience sampling. A survey was made with the self-administration questionnaires. The questionnaires for the oral health locus of control were designed by the authors, and the internal locus of contol (ILOC) and the external locus of control (ELOC) were distinguished. Results. The oral health promotion behavior and attitude scale showed a significant positive correlation with ILOC, and the oral health promotion attitude scale showed a positive correlation with ILOC and a negative correlation with ELOC. The extraction of deciduous teeth in time among the oral health promotion behaviors showed the highest coefficient of correlation with the ILOC and ELOC. Such behaviors as toothbrushing after meal, toothbrushing before sleep, covering the multiple teeth with dental sealant, and taking the treatment after school oral examination showed the negative correlations with ELOC. The belief that early treatment is effective, taking good care of decayed teeth among the disease control is important, and preventing the oral diseases is economic showed a positive correlation with ILOC and a negative correlation with ELOC. The educational status of subject`s father was significantly correlated with ILOC. Conclusions. The oral health locus of control showed the statistically significant correlation with the oral health promotion behavior and attitude.

        녹차성분 EGCG의 CSK 단백질 조절을 통한 암예방 효과

        A great interest is emerging about green tea as a tool against human cancer proliferation or inflammation, as pointedout by recent reports describing the inhibitory action of epigallocatechin gallate (EGCG) on angiogenesis, urokinase, metalloproteinases,and induction of inducible nitric oxide synthase. We proposed that EGCG may regulate a multi target signalinghaving wider spectra of action than those actions of single enzymes. CSK (c-terminal Src kinase) protein is a non-receptortyrosine kinase involved in the cross-talk and mediation of many signaling pathways that promote cell proliferation, adhesion,invasion, migration, and tumorigenesis. Based on the knowledge that CSK activation is important for cancer proliferation wehypothesized that CSK could be a target of EGCG. Here we showed that EGCG effectively suppressed the growth of CSK MEFcell when compare with CSK knockout MEF cell growth. These results indicate that EGCG could be used as a chemopreventionto modulate CSK signal pathway in inflammatory processes and tumor formation.
