Background : A point mutation in the RET proto-oncogene, in medullary thyroid carcinoma (MTC) is well known, but no other genetic causes of MTC have been found. This study was performed to identify the most common DNA copy number changes in MTC by comparative genomic hybridization (CGH). Methods : Twenty-nine surgically resected MTC specimens were retrospectively selected from patients operated on between 1996 and 2004 at the Asan Medical Center. A review of the clinical data and pathological findings was performed. Congored staining and immunohistochemical stains (calcitonin, chromogranin A and CEA) were processed by tissue microarray. CGH analysis was performed. Results : The Congo-red stain was positive in only 12 cases. The immunohistochemical results were positive in 29 cases for chromogranin A, 26 cases for CEA and 25 cases for calcitonin. DNA copy number changes were found in 23 cases (79.3%). The most frequent change was a gain of 19q (65.5%); less frequent changes were gain of 22 (55.2%), 19p (51.7%), 16p (27.58%), 17q (17.24%), and loss of 4q (27.6%) and 3p (17.24%). Conclusions : DNA copy number changes of MTC were more common (79.3%) than reported in previous studies. The most frequent changes were gains in 19q, 22 and 19p. 노랑초파리의 GD STERILITY 에 관한 온도 및 AGE 의 영향에 관한 연구

An experiment was carried out to investigate the influence of temperature and aging on GD sterility from reciprocal crosses with π_2strain and Canton-S strain of Drosophila melanogaster. It was found that the GD sterility traits were observed display a characteristic reciprocal cross effect by low egg production and strongly depend on female aging effect and sensitive in various temperature. It was concluded that the GD sterility was induced by high and narrow range of temperature 25℃-29℃ and age a female produced high egg production.
쥐 유방암종에서 흉선세포 융합에 의한 전이능 증가 기작에 관한 연구

We have obtained three hybrid cell lines- FA4, FB4 and FB6 - through hybridization process between rat NMU mammary carcinoma cell line and allogeneic thymocytes. Those hybrid cell lines were inoculated with matrigel into the interscapular subcutaneous tissue in newborn Sprague-Dawley rats. About 5-14 weeks later, metastatic nodules were developed in the lungs and axillary lymph nodes among the animals, which were inoculated by FB6 or transplanted subsequently by those tumor masses. The expression of Thy 1.1 and T cell antigen was higher in FB6-12, one of the established cell lines which were gained from metastatic tumor lesions by inoculation of FB6, than in other cell lines. Also we have obtained FB6-HM subclone which was selected as a highly metastatic clone from FB6 by limiting dilution method. The evidence of hybridization between NMU and thymocytes was disclosed by the fact that FB6-HM clone showed a positive reaction against polyclonal rabbit sera raised by repeated immunization with S-D rat thymocytes. CD44 molecule, which has been known that the degree of its expression in the neoplastic epithelial cells was positively correlated with the increased metastatic potential, was also more expressed in FB6-12 than in any other cell lines. The results of this study strongly supported the hypothesis that epithelial tumor cells could gel metastatic potential through hybridization with lymphoreticular lineage cells. Additionally, by establishing the metastasizing mammary carcinoma in Sprague-Dawley rat model, we could perform proper experiments on the study of mechanism of hematogenous and lymphatogenous metastasis in cancer.

임신 13-14주에 해당하는 임산부 15명으로 대상으로 본원이 새롭게 establish한 in situ 방법 을 적용하여 조기 양수검사를 시행하였다. 통상의 검체 채취량보다 적은 10-15ml을 채취했 음에도 불구하고 모든 sample에서 성공적인 결과를 얻었고 banding 의 quality도 좋았다. 또 한 평균 9.2일 후에 harvest 할 수 있을 정도로 세포의 배양이 용이하였으며 coverglass위에 서 in situ로 염색체 분석을 하는 방법이므로 clonally analysis 가 가능하므로 mosaicism의 판 정을 용이하게 하는 잇점이 있다. 따라서 조기 양수검사는 좀 더 이른 시기에 유전적 산전 진단을 가능케 하는 임상적으로 매우 유용한 방법인 것으로 사료된다. Genetic Amniocentesis was performed at 13-14 weeks of gestation using in situ technique which was established by our laboratory. No problems were encountered. Culture failure was never observed although the extracted AF volume was 10-15ml and the banding quality of chromosome was good. Also the average cultrue duration of harvest was 9.2 days that had no connection with AF volume. In addition, we could observe 10-12 fields of metaphase per one coverglass for chromosome analysis. It is concluded that early amniocentesis is a clinically reliable procedure which permits prenatal diagnosis of genetic evaluation in early pregnancy.
