Immunostimulatory effects of Bordetella bronchiseptica antigen on mouse spleen cells

Bordetella bronchiseptica (B. bronchiseptica) is a Gram negative bacterium that causes a common respiratory disease in animals including pigs. It is a major etiological pathogen for atropic rhinitis, causing damage to the swine industry. Despite many efforts in development of an efficient vaccine against B. bronchiseptica, studies characterizing antigenic properties of B. bronchiseptica are lacking. In this study, we investigated the intact immunogenicity of B. bronchiseptica in the absence of vaccine adjuvant using mouse spleen cells. A preparation of formalin-treated intact B. bronchiseptica and mouse spleen cells was used for the experimental setup. The activation and viability of spleen cells treated with B. bronchiseptica was measured by MTT assay and Trypan blue exclusion test, respectively. A concentration range of B. bronchiseptica generated some clusters of spleen cells, indicating high proliferation. For functional analysis, production of TNFalpha was measured using specific ELISA after harvest of culture supernatants and expression of lymphocyte activation markers (CD25, CD69) was analyzed by flow cytometry. By itself, B. bronchiseptica showed significant proliferation and functional enhancement of spleen cells. Taken together, the intact immunogenecity of B. bronchiseptica may affect the development of its vaccine.

본 연구는 우리나라의 전통문화인 한지의 다양한 용도를 개발하고, 인피섬유를 주원료로 하는 한지의 특성상 폐기되어져 왔던 닥나무 목질부를 이용하여 기능성 재료인 세라믹의 제조 및 한지벽지에 이용되는데 기초 자료를 제공하고자 하였으며 다음과 같은 결론을 얻었다. 다양한 조건에서 제조된 닥나무 세라믹을 첨가한 수초지는 세라믹 제조 시의 수지함침율, 탄화온도 및 세라믹의 첨가량에 따라 전반적인 물리적 성질은 감소하였으나 인열지수는 거의 변화가 없었다. 또한 투기도의 경우에서는 세라믹이 닥섬유의 섬유결합을 저해하여 크게 증가하였다. 한편, 한지벽지로서 습윤인장강도, 일광견뢰도, 습윤마찰견뢰도, 은폐성 등은 KS 벽지 규격을 만족시켰으나 건조마찰견뢰도는 규격에 미달되어 이에 대한 보완이 요구되었다. 따라서 폐기성 닥나무 목질부를 활용하여 세라믹을 제조 및 한지벽지에 이용함으로서 닥나무 목질부의 다양한 활용도 개발과 양호한 품질의 한지벽지를 제조하는데 충분한 가능성을 확인하였다. This study utilized paper mulberry xylem to make functional ceramic materials and to manufacture Hanji wallpaper. Conclusions obtained from the results of this study were as follows. After manufacturing the ceramic with the paper mulberry xylem, it was added to the paper mulberry pulp and Hanji was produced with various conditions. When making the ceramic, overall physical properties of Hanji were decreased depending on resin impregnation rate, carbonization temperature and amount of ceramic content, but tearing index was not changed nearly. However, because ceramic hinders the fiber bonding of the paper mulberry pulp, air permeability was considerably increased. Hanji wallpaper which paper mulberry ceramic was added satisfied the wallpapers regulated in KS(Korean Standard) and the use of the wallpaper made of Hanji was expected as the environment friendly natural material.
핸드드립용 커피필터 제품을 위한 닥섬유 혼합 비율에 따른 한지의 물리적 특성 및 적용 가능성 평가

The objective of this study was to evaluate the possibility for the coffee filter paper of Hanji. In this study, the samples for Hanji filter papers were prepared with three different mulberry fiber contents of 10, 30 and 50%(w/w). Surface brightness and apparent density of hand-made Hanji decreased with increasing mulberry fiber contents. However, values of thickness, tensile strength, wet tensile strength, burst strength, and folding endurance for hand-made Hanji increased by increasing the contents of mulberry fibers due to fiber-to-fiber bonding. The results of the coffee extraction test using the developed filter papers showed no significant differences between commercial filter paper and Hanji filter paper. Therefore, Hanji filter paper containing more than 30%(w/w) mulberry fiber may have potential uses for the dripped coffee filter application from a physical property point of view.
