        Geranyllinalool에 의한 LLC-PK1 세포내 스핑고지질 생합성 억제

        Geranyllinalool, a polyisoprenoid compound, was found to block the early biosynthetic pathway of sphingolipids in LLC-PKl cells. Sphinganine, an intermediate in sphingolipid biosynthetic pathway, was abruptly accumulated in LLC-PKl cells at $2{\;}{\mu}M$ of fumonisin B1(FB1), a specific inhibitor of sphinganine N-acyltransferase, for 24 hr. Geranyllinalool lowered the $B_1(FB_1)$, a specific inhibitor of sphinganine N-acyltransferase, for 24 hr. Geranyllinalool lowered th FB1 and $50{\;}\mu$M geranyllinalool. l-Cy-closerine, an inhibitor of serine-palmitoyl transferase, was used as a positive control to evaluate the inhibitory effect of geranyllinalool. These results suggest that geranyllinalool may inhibit the serine-palmitoyl transferase, the first enzyme in de novo sphingolipid biosynthesis, resulting in the altered regulation of sphingolipid metabolism.

      • Effect of Dopamine on Propagation and Frequency of Slow Wave in Cat Isolated Stomach Muscle

        위(胃) 전기활동(서파)의 전파(傳播) 및 발생빈도에 미치는 dopamine의 영향을 주명하기 위하여 145마리의 고양이를 사용하며 다음의 실험을 실시하였다. 위의 복측부분을 적출하여 종주근의 주행 방향으로 대만쪽에서 길이 5cm, 폭 1.2cm인 근절편을 만들어, 95% $O_2$와 5% $CO_2$가 계속 공급되는 Krebs-Ringer용액 (PH 7.4, 온도 $36{\pm}0.5^{\circ}C$)내에 두고 가느다란 은선이 들어있는 모세관 전극 (Ag-AgCl)을 사음하여 단극성으로 서파를 기록하였다. dopamine의 첨가후 위서파의 전파방향은 첨가한 농도에 비례하여 전환이 많아졌으나, dopamine의 영향은 domperidone의 전처치로 유의하게 억제되었다. dopamine은 또한 농도가 증가함에 따라 불규칙한 위서파의 발생만도를 증가시켰으며 이 현상은 domperidone 및 Phentolamine에 의하여 억제되었으나 Propranolol, hexamethonium 및 tetrodotoxin에 의하여는 억제되지 않았다. 그러므로 dopamine은 고양이 위에서 dopamine receptor와 일부 ${\alpha}-adrenergic\;receptor$에 작용하며 이상적(異常的)서파를 발생시킨다고 사료된다.       • Plasma Secretin Concentrations in Fasting and Postprandial States of Normal Korean Subjects

        한국의 정상 성인 20 명에서 공복시 및 음식물 섭취후의 철장 secretin농도를 방사면역 측정법(radioimmunoassay)으로 다음과 같이 측정하였다. 20명중 12명은 단백질 음식물인 햄버거와 쌀밥을 일주일 간격으로 섭취하였다. 햄버거 섭취 추 평균 혈장 secretin농도$(12{\sim}16\;pg/ml)$는 공복시의 평균 혈장 secretin농도(10 pg/m1 이하) 보다 유의하게 증가하였으며 그 증가가 오랫동안 지속하였다. 쌀밥의 섭취 후에는 혈장 secretin농도$(9{\sim}13\;pg/ml)$가 공복시의 값보다 증가하는 경향이었으며, 단지 식후 30분의 값에서만 유의하게 증가하였다. 식후 혈장 secretin농도의 증가폭은 햄버거에서의 증가 값이 쌀밥에서의 값보다 더 컸다. 나머지 8명은 자당 용액과 생리 식염수를 마셨다. 자당 용액의 섭취후 평균 혈장 secretin농도$(10{\sim}14\;pg/ml)$는 공복시의 값보다 유의하게 증가하였으나 쌀밥의 값과 마찬가지로 일시적이었다. 그러나 생리 식염수를 섭취한 후에는 혈장 secretin농도에 이렇다할 변화가 없었다. 이상의 결과로 미루어 정상 한국 성인에서는 단백질 음식물은 물론 탄수화물 음식물의 섭취로도 혈장 secretin농도가 증가한다고 사료된다. This study was conducted to investigate fasting plasma secretin and postprandial secretin concentrations after ingestion of a protein meal or a sucrose solution in 20 healthy Korean subjects. In 12 subjects, ingestion of a protein meal, hamburger resulted in a significant and sustained increase in the mean plasma secretin concentrations, from mean fasting levels of less than 10 pg/ml to $12{\sim}16\;pg/ml$, and the mean plasma secretin concentrations, $9{\sim}13\;pg/ml$, after a rice meal increased significantly but transiently compared with mean fasting levels. The magnitude of postprandial increase in the Plasma secretin concentration after the hamburger was greater than that of the rice meal. In the remaining 8 subjects, drinking of a sucrose solution resulted also in a significant but transient increase in the mean Plasma secretin concentrations, from mean fasting levels of less than 10 pg/ml to $10{\sim}14\;pg/ml$ which were significantly greater than that after a physiological saline. Significant increase in the plasma secretin concentration was not observed during the postprandial period after the physiological saline. It is inferred from the above results that the Plasma secretin levels increase significantly after ingestions of a carbohydrate meal as well as a protein meal in the Korean race.

      • Prostaglandin $E_1$과 Acetazolamide가 흰쥐 전혈(全血)의 Carbonic Anhydrase 활성에 미치는 영향

        This study was undertaken to investigate the influence of prostaglandin $E_1(PGE_1)$ upon the activity of carbonic anhydrase and upon the inhibitory action of acetazolamide on carbonic anhydrase. The heparinized blood was sampled by cardiac puncture from Sprague-Dawley strain rats under ether anesthesia and was hemolysed by adding distilled water 1,000 times the amount of the blood. The activity of carbonic anhydrase of 0.1 ml of the hemolysate was measured by Maren's simplified micro-method. In the first experiment, the 7 rats were used, and the activity was measured by adding 0.1 ml of various concentrations of $PGE_1$(0.5, 1.25, 2.5, 5.0, 10 and $20\;{\mu}g/ml$). In the second experiment, the 6 rats were used and the activity was measured by adding 0.1 ml of $PGE_1(5\;{\mu}g/ml)$ and 0.1 ml of acetazolamide$(6{\times}10^{-7}M/l)$ respectively or simultaneously. Obtained results were as follows: 1) The activity of carbonic anhydrase was significantly inhibited by $PGE_1$ at doses of $0.5{\sim}10\;{\mu}g/ml$, maximally at a dose of $2.5\;{\mu}g/ml$, but inhibition was no more observed at a dose of $20\;{\mu}g/ml$. 2) The activity of the acetazolamide group was significantly less than that of the control group. 3) The activity of the $PGE_1+acetazolamide$ group was significantly less than those of the $PGE_1$ group and the acetazolamide group. It is inferred from the above results that the $PGE_1$ inhibits the activity of carbonic anhydrase dose-dependently and strengthens the inhibitory effect of acetazolamide on carbonic anhydrase.

        흰쥐 이자에서 Nitric Oxide ( NO )가 Insulin분비에 미치는 영향

        Background: NO (nitric oxide), derived from L-arginine through the action of nitric oxide synthase (NOS), is a short-lived free radical transmitting cellular signals for vasodilation, neurotransmission, and cytotoxicity. Recently, this molecule has been reported to be involved in the various glandular secretion. Although the relationship between NO and the pancreatic endocrine secretion has been widely investigated, the role of NO on insulin secretion has not been elucidated. Therefore, the present study was designed to reveal the precise action of NO on the secretion and synthesis of insulin following administration of NAME (L-NG -nitroarginine methyl ester) or L-arginine using immunocytochemistry and in situ hybridization techniques. Methods: NAME or L-arginine was administered into jugular vein of the male Sprague-Dawley rat (180~200 g, b,w.) exhibiting normoglycemia (80~120mg/dL). Blood glucose concentrations were measured at intervals of 30 minutes for 2 hours after drug treatment. The pancreatic tissues were taken out at 30 and 90 minutes following drugs administration for insulin immunocytochemistry and in situ hybridization. Results: Both NAME and L-arginine treatments diminished blood glucose levels. The decrease of blood glucose level was more prominent in NAME-treated rats than that of L-arginine. Insulin immunoreactivity in drugs-treated rat pancreas decreased compared to that in normal control, while the expression of insulin mRNA was significantly increased. Conclusion: On the basis of present study, it is concluded that the transient changes of NO con- centration, regardless of increase or decrease, in Langerhans islet might act as a potent stimulant in insulin secretion and its synthesis (J Kor Soc Endocrinol 14:719-72S, 1999).

      • 흰쥐의 위산분비 및 혈장 Gastrin농도에 미치는 대상회전 제거의 영향

        It has been recently reported that cingulate cortex mar facilitate gastric acid secretion, but its facilitatory mechanism on the gastric acid secretion is still unclear. This study was undertaken to investigate the facilitatory mechanism of the cingulate cortex upon gastric acid secretion in rats. Twenty·three male albino rats were divided into the cingulate(N= 13) and the operated control(N= 10) groups. The cingulate group in which cingulate cortex was removed by suction through a slit-shaped opening on each side of, and parallel to, the sagittal suture. In the operated control group, the surgical procedure was ended with the skull opening and the incision of dura mater. The gastric juice was collected via a chronic gastric cannula after 24 hours of fast, with water ad libitum. The juice was collected continuously for 6 hours, starting 3 hours prior to the injection of gastric secretagogue, pentagastrin$(12\;{\mu}g/kg)$ or histamine dihydrochloride $(320\;{\mu}g/kg)$. Three one·hour samples were obtained before ana after the administration of each secretagogue. The two agents were injected separately and subcutaneously at intervals of 1 week, the blood samples were drawn from the abdominal aorta for the radioimmunoassay of postprandial plasma gastrin concentration in response to the forced feeding of 10% cod liver oil. 1) After pentagastrin administration, the volume of gastric juice tended to decrease, but its acidity tended to increase in the cingulate group compared with those of the operated control group. However, there was no any difference in the acid output between the two groups. 2) Histamine-stimulated acid output and volume of the gastric juice of the cingulate group decreased significantly compared with those of the operated control group, while there was not significantly different in the acidity between the two groups. 3) Before pentagastrin or histamine administration, any change was not observed in the gastric acid secretion following the cingulate cortical ablation. 4) Postprandial plasma gastrin concentration in response to the forced feeding of 10% cod liver oil was insignificantly lower in the cingulate group than in the operated control group. It is inferred from the above results that the cingulate cortex exerts a facilitatory influence upon the histamine-stimulated gastric acid secretion in rats, and its influence may not be mediated by the stimulation of gastrin secretion.

      • 흰쥐의 위궤양 발생에 미치는 대상회전 제거 및 미주신경 절단의 영향

        This study was undertaken to investigate the effect of cingulate cortical ablation upon gastric ulceration, and the pathway through which cingulate cortex exerts the effect. 56 female rats were divided equally into cingulate (cingulate cortical ablation), cingulate-vagal (cingulate cortical ablation and vagotomy), normal control and vagal (vagotomy) groups. Cingulate cortex was ablated through a slit-shaped opening (1 mm in width, 13 mm in length) which was made symmetrically on both sides of, and parallel to, the sagittal suture by removing a bone flap from parietal and frontal bones on each side. Vagus nerves on both sides were transected around the distal end of the esophagus. In the normal control animals, surgical intervention ended with scalp incision. All rats were kept without restraint or food deprivation for 3 weeks after surgery. The stomach of each rat was inflated with 7 ml of physiological saline and then removed under deep anesthesia. The mucosal surface was examined under dissecting microscope for the location, shape and number of ulcers, and then enlarged photograph $(4{\times})$was taken. The incidence of ulcer in each group was counted and the number of ulceration as well as the total area of glandular mucosa were measured on the photograph. Results obtained were as follows: 1. The mean number of ulcer per stomach and the total area of ulcer exprssed as permillage of the total area of glandular mucosa were significantly higher in the cingulate group than the cingulate-vagal, the normal control and the vagal groups. There was no difference among the latter three groups. 2. The incidence of ulcer in the cingulate group was significantly higher than that in the .normal control group and was also higher, though not significantly, than those in the cingulate-vagal and the vagal groups. There was no difference among the normal control, the cingulate-vagal and the vagal groups. It is inferred from the above results that the cingulate cortex exerts an inhibitory influence upon gastric ulceration and that this influence is mediated by controlling the vagal activity.
