For a basic study of catecholamine metabolism and of the utilization of hydrogen peroxide in brain, rat brain peroxidase was solubilized and its properties were examined spectrophotometrically. The peroxidase was solubilized in 0.01 M phosphate buffer, pH 7.4 containing 0.05% Triton X-100 and 0.2 M KCI. The K_m values of brain peroxidase for 0-dianisidine and H₂O were 0.19 mM and 7.0 mM respectively. From these basic results, the standard assay condition of solubilized enzyme was established. The peroxidase was partially purified from solubilized protein by sephadex G-200 gel filtration and DEAE-cellulose ion exchange chromatography. The specific activity of brain peroxidase was increased 27-fold with 30% recovery. Substrate specificity of the enzyme was also investigated.

토양으로부터 분리한 1,300여 균주 중 50% 이상 ACAT 저해 활성을 나타내는 경우는 방선균 605균주로부터 4.0%, 곰팡이 500여 균주로부터 3.6%였으며, 세균 172균주로부터는 50% 이상 저해활성을 나타내는 것이 없어 좋은 대조를 이루었다 .이중 강한 ACAT 저해활성을 나타내는 생산균주 Streptomyces sp. A-3 배양액으로부터 XAD-7 column, silica-gel column, sephadex LH-20 column chromatography 및 역상 HPLC를 이용하여 활성저해물질(AI-3)을 분리·정제 하였다. AI-3를 UV, IR, FAB-MS, NMR을 통해 분석한 결과 분자량이 465이며, skin tumor promoter로 알려진 Olivoretin A와 거의 동일한 구조임을 알 수 있었다. 또한 rat liver microsome을 이용한 효소활성분석에서 AI-3의 IC_(50) value는 3.1 μM이었다. About 1,300 strains isolated from soil were evaluated for acyl-CoA:cholesterol acyltransferase (ACAT) inhibition activity. About 4.0% of actinomycetes and 3.6% of fungi showed greater than 50% inhibition activity, respectively. However, none of the isolated bacteria exhibited inhibition activity more than 50%. Among them, one Streptomyces sp. A-3 showed a higher ACAT inhibition activity in culture broth. Isolation of the ACAT inhibitor (AI-3) was achieved by Amberlite XAD-7 column, silica-gel column, Sephadex LH-20 gel-filtration and reverse phase HPLC. From the UV, IR, FAB-MS spectroscopy and NMR studies, the molecular weight of this compound AI-3 was determined as 465. AI-3 seemed to have a similar structure to that of skin tumor promoter, Olivoretin A. The compound AI-3 inhibited ACAT activity in vitro assay system using at liver microsomes with IC_(50) value of 3. 1μM.

There hasn't been a study searching for various methods of instruction and orientation in the use of a college library. By searching through the western literature in this field from 1961, I was amazed by the vast literature and enormous study done on it. I tried to introduce the most important articles in this field which can help mold our methods of library instruction. There has been quite a few cries for the use of new methods, of newer media, for increased number of students. Promising future lies in the area of programmed instruction and audiovisual aids to teaching, which should be developed into course-related library instruction.
