        Chloromethylisothiazolinone induces ER stress-induced stress granule formation in human keratinocytes

        Chloromethylisothiazolinone (CMIT), a humidifier disinfectant, is known to be toxic to therespiratory system. While the toxic effect of CMIT on the lungs has been widely investigated, itseffect on the skin is well unknown. In this study, we examined stress granule (SG) formation toinvestigate the cytotoxic effects of CMIT on human keratinocytes. We assessed the viability ofthe cells following CMIT exposure and performed immunofluorescence microscopy andimmunoblot analyses to determine SG formation and downstream pathways. The IC50 values inhuman keratinocyte HaCaT cells after CMIT exposure for 1 and 24 h were 11 and 8 μg/mL,respectively, showing no significant difference. As determined using immunofluorescencemicroscopy, SG formation was effectively induced after CMIT exposure. Moreover, thephosphorylation of eukaryotic initiation factor-2α (eIF2α), a translation initiation factor, andprotein kinase R-like endoplasmic reticulum (ER) kinase, which plays a role in the ER stressmediatedeIF2α phosphorylation, was confirmed by CMIT exposure. These results suggest thatexposure to CMIT can have detrimental effects on the skin, even briefly, by inducing SGformation through ER stress in keratinocytes.

        Use of Amplatz® canine duct occluder for closing a patent ductus arteriosus in a small-sized dog

        A 2-year-old intact female pomeranian dog presented dys- pnea, labored breathing, cough, exercise intolerance, ma- chinery heart murmur, and precordial thrill. A left-to-right patent ductus arteriosus (PDA) was diagnosed based on two-dimensional echocardiography, thoracic radiography, electrocardiography, and blood work. An angiography was performed to accurately evaluate the size and shape of the duct. An interventional approach for transcatheterial oc- clusion of the PDA was achieved using an Amplatz ® Canine Duct Occluder (ACDO), which is a commercially available ductal occluding device. Due to the limited size of the dog’s femoral artery, a device smaller [125% of minimal ductal diameter (MDD); recommended size: 150~200% of MDD] than recommended was mounted. After placement of the ACDO, precordial thrill and continuous heart murmur dis- appeared, and the patient was discharged the next day after stabilization with O 2 supply. Upon follow up examination, dyspnea, labored breathing, cough, exercise intolerance, and cardiomegaly were improved with no complications after the procedure. The ACDO was well maintained in po- sition. This case represents successful clinical application of the Amplatz ® Canine Duct Occluder to achieve closure of a PDA using a slightly smaller device than the recommended size.

        Mangifera Indica leaf extracts promote hair growth via activation of Wnt signaling pathway in human dermal papilla cells

        The crosstalk between androgens and Wnt signaling pathways is critical in the hair growth cycle. Therefore, natural products that target these two pathways for the inhibition of hair loss are sought after. In this study, we investigated the effect of water extracts of Mangifera indica leaves (WEML) on hair growth. WEML treatment significantly reduced the expression levels of both dickkopf-1 (DKK1) and type 2 5α-reductase (SRD5A2) involved in Wnt signal suppression activity and dihydrotestosterone (DHT) synthesis, respectively, in human follicle dermal papilla cells (HFDP). In addition, WEML treatment effectively upregulated Wnt target genes and downregulated DKK1 expression that was increased by DHT treatment. Degranulation analysis in rat basophilic leukemia mast cell line (RBL-2H3) using β-hexosaminidase release assay confirmed that WEML did not exhibit allergenic activity. Furthermore, hair growth was significantly enhanced in in vivo mice model treated with WEML. These results suggest that M. indica leave extract contains bioactive materials that can be used to treat hair loss.

        Antimicrobial activity of 405 nm light-emitting diode (LED) in the presence of riboflavin against Listeria monocytogenes on the surface of smoked salmon

        This study investigated the antimicrobial activityof 405 nm light-emitting diode (LED) with and withoutriboflavin against Listeria monocytogenes in phosphatebuffered saline (PBS) and on smoked salmon at differentstorage temperatures and evaluated its impact on foodquality. The results show that riboflavin-mediated LEDillumination in PBS 25 C significantly inactivated L. monocytogenes cells by 6.2 log CFU/mL at 19.2 J/cm2,while illumination alone reduced 1.9 log CFU/mL of L. monocytogenes populations at 57.6 J/cm2. L. monocytogenespopulations on illuminated smoked salmon decreasedby 1.0–2.2 log CFU/cm2 at 1.27–2.76 kJ/cm2 at 4, 12, and25 C, regardless of the presence of riboflavin. Althoughillumination with and without riboflavin caused the lipidperoxidation and color change in smoked salmon, thisstudy demonstrates the potential of a 405 nm LED topreserve the smoked salmon products, reducing the risk oflisteriosis.

        Bioactive peptides in the pancreatin-hydrolysates of whey protein support cell proliferation and scavenge reactive oxygen species

        Whey protein (WP) in milk shows physiologically active functions such as cholesterol control and immune system strengthening. In this study, we performed hydrolysis and peptide polarity fractionation to enhance the efficacy and diversity of its physiological activities, using the digesting enzyme, pancreatin. Our results indicate that hydrolysis significantly increased the cell proliferation of the WP fractions, with the lower-polarity fractions showing greater efficacy in this regard. Our results indicate that hydrolysis significantly increases cell proliferation of the WP fractions. Additionally, we confirmed differences in the antioxidant activity of the WP fractions as a function of polarity was confirmed via scavenging 2,2’-azino-bis (3-ethylbenzothiazoline-6- sulfonic acid) (ABTS) assay in vitro. WP itself did not show anti-inflammatory efficacy. However, all the hydrolyzed fractions downregulated the mRNA expression levels of inflammatory cytokines in all treated cell lines and, based on a senescence-associated (SA)-β-galactosidase assay, the fraction with the lowest polarity (F6) inhibited cellular senescence to the greatest extent. Furthermore, we identified the peptide sequences with various physiological activities from whey protein hydrolysates through mass spectrometry. Taken together, our results indicate that the fractionation of WP via hydrolysis generates novel functions including promoting cellular cell proliferation, anti-inflammatory effects, and enhancing antioxidant and anti-cellular senescence.

        Melittin-derived peptides exhibit variations in cytotoxicity and antioxidant, anti-inflammatory and allergenic activities

        Melittin is a major component of bee venom; it is widely used in traditional medicine because of its therapeutic effects, such as anti-inflammatory effects. However, melittin has limited medical applications owing to its adverse effects, such as high cytotoxicity. In this study, we investigated the physiological activities of various hydrolyzed melittin-derived peptides to eliminate the cytotoxicity of melittin and enhance its efficacy. The 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging assay confirmed that melittin-derived peptides showed antioxidant activity comparable to that of melittin. Moreover, unlike melittin, which showed high cytotoxicity in the 3-(4,5-dimethylthiazol-2-yl)−5-(3-carboxymethoxyphenyl)−2-(4-sulfophenyl)−2H-tetrazolium inner salt (MTS) assay, the melittin-derived peptides showed negligible cytotoxicity. Among the melittin-derived peptides, the peptide composed of sequence TTGLPALISWIKRKRQQ (P1) showed inhibitory effects on the mRNA expression of inflammatory cytokines and phosphorylation of IκBα, similar to the effects of melittin in RAW 264.7 cells. Degranulation of RBL-2H3 cells was analyzed using a β-hexosaminidase release assay to confirm the allergenic activity of melittin and P1, which showed remarkably reduced allergenicity of P1 compared to that of melittin. These results indicate that P1 maintained the anti-inflammatory effects of melittin while reducing its cytotoxicity and allergic reactions. In conclusion, the melittin-derived peptide P1 efficiently decreased the adverse effects while maintaining the beneficial effects of melittin, making it suitable for therapeutic applications.

      • 일반 사료와 동결건조 사료의 기호성 차이 분석

        본 연구에서는 동결건조 사료와 일반 사료에 대한 개의 기호성을 섭취 행동에 따라 비교 분석하였다. 실험 대상은 총 개 4마리로, 대형견(Golden Retriever) 2마리와 소형견(Shih Tzu) 2마리로 구성하였다. 실험은 스트레스에 의한 변수를 최소화하기 위해 실험견의 생활공간에서 진행되었다. 또한, 허기에 의한 변수를 최소화하기 위해 실험 시간은 아침 사료 급여 후로 선정하였다. 실험에 사용된 동결건조 사료와 일반 사료는 각각 세 종류씩 선정하였으며, 섭취 시간, 산만함의 정도, 섭취 후 행동, 잔여 사료량, 그릇 탐색 횟수 등을 척도로 삼아 기호성 차이를 분석하였다. 그 결과 평균적으로 네 마리의 개 모두 동결건조 사료에 대한 기호성이 일반 사료보다 높았으며, 특히 섭취 시간에 대한 차이가 두드러지게 나타났다. In this study, dog preferences for freeze-dried feed and general feed were compared and analyzed according to intake behavior. The subjects of the experiment consisted of a total of 4 dogs, 2 large dogs (Golden Retriever) and 2 small dogs (Shih Tzu). The experiment was conducted in the living space of the experimental dog to minimize variables caused by stress. In addition, in order to minimize the variables due to hunger, the experimental time was selected after the morning feed benefit. Three types of freeze-dried feed and general feed used in the experiment were selected, and differences in preference were analyzed using intake time, degree of distraction, behavior after intake, residual feed amount, and number of vessel searches as measures. As a result, on average, all four dogs had a higher preference for freeze-dried feed than general feed, and in particular, the difference in intake time was noticeable.

        말티즈견에서 발생한 판막상형 폐동맥판 협착증 증례

        A 8-month-old female Maltese dog was presented with a history of heart murmur. In physical examination,grade 4/6 systolic murmur heard at the left heart base. Electrocardiography showed sinus arrhythmia, right axis deviation,deep S wave and splintered QRS complex. Thoracic radiography revealed enlarged right side heart and bulging ofthe main pulmonary artery. Echocardiography showed mild hypertrophy of right ventricle, a supravalvular stenosis,marked post-stenotic dilation of the main pulmonary artery and a moderately increased pulmonary arterial velocitythrough the stenotic area (4.4 m/s, pressure gradient of 78.7 mmHg). The dog was diagnosed with supravalvularpulmonic stenosis based on the diagnostic imaging findings. Medical management using β1-blocker and ACE inhibitorwas started in this dog and this is first case report described diagnostic characteristic features of supravalvular pulmonicstenosis in korea.

        간호대학생의 건강신념, 안전동기, 안전환경이 감염관리 표준주의 수행도에 미치는 영향

        This study was conducted to investigate the effect of health beliefs, safety motivations, and safety-climate on the performance of taking standard precautions of nursing college students with clinical practice experience. Methods: The subjects of this study were 173 students in the 3rd and 4th grades of nursing college located in Daegu Metropolitan City with clinical practice experience, and data were collected from September 7th to September 25th, 2020 using a structured questionnaire. Results: As a result of the study, the health belief of nursing college students was 3.60 (±0.75) points, safety motivation 22.40 (±5.33) points, safety-climate 4.79 (±1.90) points, and the performance of taking standard precautions was 3.21 (±0.63) points. Hierarchical regression analysis was performed with the presence or absence of infection management education, health belief, safety motivation, and safety-climate as independent variables to identify the factors that affect the performance of taking standard precautions. Factors affecting the performance of taking standard precautions are health belief (β=0.33, p=0.003), safety motivation (β=0.28, p=0.003), safety-climate (β=0.20, p=0.009). Conclusions: According to this study, health belief, safety motivation, and safety-climate influenced the performance of taking standard precautions. These results suggest that it is necessary to emphasize the importance of practicing standardism, motivate nursing students, and provide a safer hospital practice environment in order to improve the performance of taking standard precaution when nursing college students are in hospital practice.
