Hobo Enhancer Trapping Mutagenesis to find genes involved dpp signal transduction pathway

decapentaplegic (dpp) is a zygotically expressed gene encoding a member of transforming growth factor-beta (TGF-beta) superfamily, which is most similar to the bone morphogenetic proteins (BMPs). dpp is required for several events including embryonic dorsal-ventral pattern formation, embryonic midgut development and proliferation of imaginal disk cells. We have used hobo enhancer trapping to identify other genes involved in this process. Genetic characterization of this enhancer trapping lines will be presented.
pleiohomeotic 유전자 돌연변이의 모계 영향이 초파리의 초기발생에 미치는 영향과 부계의 정상 인자에 의한 모계영향의 회복

Progenies from the homozygous pleiohomeotic parents showed various abnormal developmental aspects due to maternal effects. They caused embryonic death before larvae hatching, cell death and homeotic transformations. Head was severely disrupted and its involution was abnormal. Unlike other Polycomb group genes, pleiohomeotic mutants showed weak homeotic transformation. Thoracic segments were transformed just to the first abdominal one, which is unique among Polycomb group mutants. Sixth and seventh abdominal segments were transformed to the eighth one. As pleiohomeotic mutants often caused segment defect, it is supposed that pleiohomeotic gene may be involved in segmentation as well as regulation of the homeotic gene expression. These maternal effects were recovered in proportion as the number of paternal pho+ allele increased.

The Mound of Gaya found so far in eastern North Jeolla Province is 240 here in the Jinan Plateau (Janggye and Janggye Basin), and 150 in Unbong Plateau. Of these, the excavation team examined the remains of a 140 stone lined Tomb in Gaya. Based on this, it is assumed that the eastern region of North Jeolla Province had a national organization developed based on Gaya culture before Baekje and Silla entered the country. The location of Gaya tombs is generally a geographical feature in which streams and ancestral sites are developed. Gaya Tomb in North Jeolla Province has an oval bowl and no stone statues are found. The way to construct the Bongto and its burial facilities is to dig up the first stage, prepare a tombstone, and construct a burial facility. Some tools are being identified. It is understandable that Gaya’s culture is now being combined into a re-designated painting. The unique features of the excavated items are that the head of the Wisinjae Temple, which has few examples from Gaya, and the various relics from different regions are found inside the same ancient tomb. The pottery includes Gaya, Baekje, Silla, Mahan, and Japanese pottery. In addition to the ancient tombs, acid, beacons, and iron ore provide important information on understanding the reality and development process in North Jeolla Kaya Province.
초파리의 말초신경계와 근육 세포의 발생에 관여하는 유전자의 특성

Embryos of the A37 enhancer detector strain of Drosophila express the lacZ gene in the sensory mother cells and the developing and mature cells of the peripheral nervous system (PNS). The lacZ gene is also expressed in all muscle cells of larvae and adults, as well as in most imaginal disk sensory mother cells. However, it is confined to only a subset of the PNS cells in the adult. We characterized the developmental role of the gene, neuromusculin (nrm) which is located adjacent to the A37 insertion. The gene was cloned by screening genomic libraries of the enhancer detector strain using lacZ as a probe. Sequencing of cloned nrm cDNAs revealed that it encodes nine immunoglobulin (lg)-like domains. Whole mount in situ hybridizations using nrm cDNAs as probes exhibited an expression pattern similar to that of lacZ observed in A37 embryos. However, nrm transcripts are also present in embryonic muscles and few CNS neurons. Since many lg-like proteins have been implicated in cell adhesion, the adhesion properties of Nrm was demonstrated in Schneider 2 cells transfected with the complete nrm open reading frame. Our results suggest that Nrm mediates clustering of cells or fasciculation in the developing PNS. In addition, it may play a role in motoneuron growth cone guidance by muscles or PNS neurons.
extra sex combs 돌연변이가 초파리 중배엽에서 Ultrabithorax 유전자의 발현에 미치는 영향

초파리에서 호메오틱 유전자의 발현 유지에 관여하는 것으로 알려진 extra sex combs (esc) 유전자 돌연변이가 중배엽에서 Ultrabithorax (Ubx) 유전자의 발현에 미치는 영향을 조사하였다. 동형의 esc 돌연변이는 배아에서 치사적이며 초파리의 초기 발현에 영향을 주지 않기 때문에 모계 영향을 나타내는 돌연변이을 선택하여 Ubx reporter 초파리와 재조합시켰다. Ubx는 정상의 초파리에서는 parasegment 7에서 발현되지만, esc 모계영향 돌연변이에서는 매우 적은 수의 배아에서 전위적 (ectopic)인 발현이 관찰되었으며, 대부분의 배아에서는 발현이 억제되는 것이 관찰되었다. 이러한 억제 현상은 Ubx보다 뒤쪽에서 발현되는 abdominal-A (abd-A)와 Abdominal-B (Abd-B) 유전자가 Ubx가 발현되는 지역에서 또한 발현되어 Ubx의 발현을 억제하는 것으로 생각되며, 전위적인 발현을 보이는 것은 개체마다 모계 영향이 차이가 있어 이들 두 유전자의 영향으로부터 벗어난 결과라고 생각된다. We examined the effect of extra sex combs mutation on the expression of Ultrabithorax in the visceral mesoderm during Drosophila development. As esc zygotic mutant embryos are lethal before hatching, but do not affect the early development, we recombined Ubx reporter strain with esc alleles showing the maternal effects. Ubx is normally expressed in the visceral mesoderm of the PS7 of Ubx reporter strain. However, in the esc mutation, Ubx is ectopically expressed in the small number of embryos, and weakly or not expressed in most of embryos. The repression of Ubx in esc maternal effect mutant embryos may be caused by abdominal-A and Abdominal-B ectopically expressed in esc mutation as well. The ectopic expression shown in a small number of embryos are thought as leaking of Ubx expression by a different level of esc maternal effects among individuals.

Polycomb group proteins (PcG) maintain the transcriptional silence of target genes in cells where these genes must be silenced during Drosophila development. This repression depends on Polycomb group response elements (PREs). Two PcG protein complexes have been isolated from Drosophila embryos: PRC1 and Esc-E(z) complex. In this study, we attempted to determine whether extra sex combs (esc) and Polycomb (Pc) mutations disrupt the functions of GAGA and Zeste, using an Ultrabithorax (Ubx) transgene containing the GAGA or Zeste binding sites. GAGA and Zeste are sequence-specific DNA-binding proteins, which have been previously demonstrated to be trithorax group activators of the homeotic gene Ubx. The Ubx transgene was weakly derepressed in Pc and esc mutants, indicating that Pc and esc interacts with GAGA and Zeste in the promoter of the Ubx transgene.
