        겨우살이 Lectin의 항당뇨 효과

        겨우살이 (Viscum album)는 민간요법에서 여러 질환에 치료제로 쓰이고 있다. 이는 겨우살이가 가지고 있는 lectin의 약리작용에 의한 것으로 그 가능성을 알아보고자 겨우살이 lectin의 항당뇨 효과를 연구하였다. 이를 위해 streptozotocin으로 당뇨를 유발시킨 흰쥐에 국내 참나무류에 기생하는 겨우살이의 추출물과 정제된 lectin을 경구와 복강내에 투여하여 조사하였다. 추출물을 29일 동안 경구 투여시 혈당치가 36%까지 현저히 감소하였으나, 정제된 lectin을 경구 투여시 항당뇨 효과가 현저히 나타나지 않았다. 또한 정제된 lectin 0.4 ㎎/㎏을 복강내에 주사시 당뇨군에서 2시간과 4시간에서 saline에 비해 혈당치가 각각 32%와 28% 감소하였으므로 lectin의 항당뇨 효과는 뚜렷하였다. We investigated anti-diabetic effect of the crude extracts and the lectin purified from the leaves of Viscum album collected in Mt. Duk Yu. The induction of diabetes mellitus was confirmed by the presence of statistically significant greater serum glucose levels in streptozotocin-treated rats compared with the untreated non-diabetic group. There are no significant differences in the serum glucose levels compared extracts and purified lectin to tap water in the non-diabetic group after administration through the oral cavity in 29 day. But for streptozotocin-induced diabetic rats the serum glucose level in extracts after administration through the oral cavity was strikely decreased to 36% than tap water in 29 day. Significantly lower glucose level was obtained at 2 hr and 4 hr after administration of purified lectin (0.4 ㎎/㎏) through the peritoneal cavity. These results provide an evidence that Viscum album lectin has an anti-diabetic effect pharmacologically.

        This study was carried out to investigate effect of lipid metabolism in Viscum album lectin on rats. The lectin was purified by sepharose 4B affinity chromatography and gel filtration using sephadex G-150 with plant material from Viscum album collected in Mt. Duk Yui. After 72 h of CCl4 injection (in olive oil, 1:1, 2 ㎎/㎏) there was a significant increase in serum total cholesterol and triglycerige levels relative to the control group. However, treatment of both Viscum album and purified lectin were significantly decreased lipid parameters against the CCl4-induced. Histological observation basically supported the result obtained from serum lipid assay. The livers of rats challenged with CCl4 produced a marked increase of cytoplasmic vacuoles in number, while the number of necrotic cells and swollen hepatocytes did not change significnatly. Rats administered olive oil alone did not alter the normal hepatic architecture. Histological observation of the liver section in rats treated 72 h with either Viscum album purified lectin or CCl4-induced liver lipogenesis showed decreased numbers of cytoplasmic vaculoes and necrotic cells. The normal hepatic architectural pattern was observed in Hematoxylin-eosin stain. These results suggest that Viscum album lectin has a possible protective effect of lipid metabolim in rats.

        In the present study, the author investigated changes in liver function caused by Viscum album lectin. πle lectin was purified by 0.15 M NaCl extraction, amnonium sulfate precipitation, sepharose 4B affmity chromatography and gel filtration using sephadex 0-150 with plant material from Viscum album collected in Mt. Duk Yui. After 72 h of CC14 iniection (in olive oil, 1:1, 2 mg/kg) there was a significant increase in serum AST and AL T levels relative to the control group. However, treatment. of either Viscum album extracts and purified lectin offered no protection against CC14-induced hepatotoxicity. Histological observation basically supported the resu1t obtained from serum enzyme assay. The livers of rats challenged with CC14 produced a marked increase in cytoplasmic vacuoles and inflammatory cells in number, while the number of necrotic cells and swollen hepatocytes did not change significnatly. Rats administered olive oil alone did not have a change in the normal hepatic architecture. Histological observation of a liver section in rats treated 72 h with either Viscum album extract or purified lectin CC14-induced liver damage showed numbers of cytoplasmic vaculoe and necrotic cells. The numbers of inflammatory cells were, however, increased markedly. πle nurmal hepatic architectural pattem was lost in a Hematoxylin-eosin stain. These results suggest the conc1usion that Viscum album lectin has a physiological effect of hepatotoxicity activator.

        돼지 난소내 여포의 폐쇄과정중 과립세포의 결합능의 변화

        In order to study the mechanism of follicular atresia, follicles were classified into the normal groups and the atretic ones, according to the criteria with or without corpus luteum, size of follicles, vascularization, status of granulose cells and the hypertrophy of theca layers in the porcine ovary. To estimate the binding capacity of human chorionic gonadotropin (hCG) receptor on the granulosa cells during atresia, hCG were iodinated by the chloramine-T method and then purified through the column chromatography. The concentration" of hCG receptor in each group were measured by the hCG receptor binding assay. Binding capacity in large normal follicles were 1.16%, but atretic ones were 0.45%. But in medium and small follicles (below 6mm in diameter), the binding capacity in normal follicles were 0.09%, but atretic ones were 0.05%, which was lower than those of large follicles. The present ( ) that the concentrations of hCG receptors on granulosa cells is decreased when the follicles become atretic and be used as a sort of creteria for the identification of follicles atresia.

        In the present study, the author investigated to the cytotoxocity in cultured rat hepatocytes of Viscum album lectin. The cytotoxcity effect in Viscum album lectin on the activity of LDH was also investigated. Viscum album lectin significantly increased LDH leakage into medium of hepatocytes treated or untreated with CCl4 (p<0.001). However, Viscum album lectin significantly increased LDH leakage from CCl4-induced hepatocyte (p<0.001). There was a significant increase in LDH levels relative to the control group. Histological observation basically supported the result obtained from LDH assay. The livers of rats challenged with CCl4 produced a marked increased cytoplasmic vacuoles and inflammatory cells in number, while the number of necrotic cells and swollen hepatocytes did not change significnatly. Rats administered DMSO alone did not alter the normal hepatic architecture. Histological observation of liver section in rat treated 72 hrs with either Viscum album lectin CCl4-induced liver damage showed number of cytoplasmic vaculoe and necrotic cell. The number of inflammatory cell increased markedly. This results suggest to the conclusion that Viscum album lectin has a effect of hepatotoxicity activator.

      • 임상병리과 학생을 대상으로 한 임상 실습에 관한 연구

        The aim of this study was to investigate the problem areas in the current clinical training of biomedical laboratory science education which may guide the development of an effective strategy for managing clinical training for biomedical laboratory science students. A self-reporting questionnaire was developed by the researchers. This survey was conducted between May 1st and May 15st in 2009 and 92 questionnaires werecollected from senior biomedical laboratory science students. In Analyzing data, chi-square test was used for categorical variables. 82.6% of respondents was satisfied with conditions of the training institutes. 30~40% of the respondents reported that they were not satisfied with the programs and supervision systems for training students even though the staff members of other disciplines were supportive in general. Although83.7% of the respondents reported that they were obtaining information and knowledge for their professional carrier through the training, 70.7% of the respondents reported that they had negative experiences during the training. According to the result of the survey, it can be inferred that the structure of the training education needs to be more systematically organized in order to train the well prepared entry-level biomedical laboratory scientists.
