        Ischemic brain injury decreases dynamin-like protein 1 expression in a middle cerebral artery occlusion animal model and glutamate-exposed HT22 cells

        Dynamin-like protein I (DLP-1) is an important mitochondrial fission and fusion protein that is associated with apoptotic cell death in neurodegenerative diseases. In this study, we investigated DLP-1 expression in a focal cerebral ischemia animal model and glutamate-exposed hippocampal-derived cell line. Middle cerebral artery occlusion (MCAO) was surgically induced in adult male rats to induce focal cerebral ischemic injury. Brain tissues were collected 24 hours after the onset of MCAO. MCAO induces an increase in infarct volume and histopathological changes in the cerebral cortex. We identified a decrease in DLP-1 in the cerebral cortices of MCAO-injured animals using a proteomic approach and Western blot analysis. Moreover, glutamate treatment significantly decreased DLP-1 expression in a hippocampalderived cell line. The decrease in DLP-1 indicates mitochondrial dysfunction. Thus, these results suggest that neuronal cell injury induces a decrease in DLP-1 levels and consequently leads to neuronal cell death.

        Lac 색소 추출물의 유리아미노산과 페놀성 화합물의 조성 및 피부미용 화장품 생리활성 검색

        The composition and content of some free amino acids and non-protein amino acid derivatives of the hot water and methanol extract of the Lac pigment, and the composition and content of phenolic compounds were analyzed, and the physiological activity of some skin beauty cosmetics were as follows. Free amino acids were not detected in hot water and methanol extracts, respectively, 2 kinds of protein non-constituent amino acid derivatives were detected in hot water extract and 3 kinds were detected in methanol extract. In the composition and content analysis of phenolic acid and flavonoids, 2 types of phenolic acid were detected in hot water extract and 5 types were detected in methanol extract. In addition, flavonoids were not detected in hot water extract, but rutin and taxifolin were slightly present in methanol extract. DPPH radical scavenging activity was slightly higher in methanol extract than in hot water extract at 40.43 ± 0.21%, and in tyrosinase inhibitory activity, methanol extract was approximately twice as high as in hot water extract. Inaddition, cytotoxicity to the HaCaT cell line was not observed at all treatment concentrations, and cell growth tended to increase as the treatment concentration increased. In the treatment concentration of these extracts of 100㎍/ml, the cell activity of about 30-60% was higher than that of the control. Therefore, according to the above results, the hot water and methanol extract of Lac pigment contains some non-protein derivatives, phenolic acid and flavonoid components, and it appears to have some physiological activity for skin beauty. It is believed that it is valuable for use as an ingredient, but it is considered that additional research is needed continuously in order to actually use it.

        Radiotherapy in prostate cancer treatment: results of the patterns of care study in Korea

        Purpose: The purpose of this study was to describe treatment patterns of radiotherapy (RT) for prostate cancer in Korea. Materials and Methods: A questionnaire about radiation treatment technique and principles in 2013 was sent to 83 radiation oncologists and data from 57 hospitals were collected analyzed to find patterns of RT for prostate cancer patients in Korea. Results: The number of patients with prostate cancer treated with definitive RT ranged from 1 to 72 per hospital in 2013. RT doses and target volumes increased according to risk groups but the range of radiation doses was wide (60 to 81.4 Gy) and the fraction size was diverse (1.8 to 5 Gy). Intensity-modulated radiation therapy was used for definitive treatment in 93.8% of hospitals. Hormonal therapy was integrated with radiation for intermediate (63.2%) and high risk patients (77.2%). Adjuvant RT after radical prostatectomy was performed in 46 hospitals (80.7%). Indications of adjuvant RT included positive resection margin, seminal vesicle invasion, and capsular invasion. The total dose for adjuvant RT ranged from 50 to 72 Gy in 24–39 fractions. Salvage RT was delivered with findings of consecutive elevations in prostate-specific antigen (PSA), PSA level over 0.2 ng/mL, or clinical recurrence. The total radiation doses ranged from 50 to 80 Gy with a range of 1.8 to 2.5 Gy per fraction for salvage RT. Conclusion: This nationwide patterns of care study suggests that variable radiation techniques and a diverse range of dose fractionation schemes are applied for prostate cancer treatment in Korea. Standard guidelines for RT in prostate cancer need to be developed.

        연복자 추출물의 Phenolic acids와 Flavonoids의 조성 및 화장품 항산화 활성

        이 연구는 연복자의 열수와 80% 메탄올 추출물에 대한 총 polyphenol과 flavonoid의 함량과phenolic acid와 flavonoid의 조성 및 함량을 분석하고, 이 추출물에 대한 일부 항산화 활성을 조사하므로 스킨케어 기능성 화장품으로의 활용 가능성을 확인하는 것이 연구의 목적이다. 총 polyphenol과 flavonoid 함량은 열수와 80% 메탄올 추출물 모두에서 과피 추출물이 씨앗 추출물 보다 높은 것으로 나타났다. Phenolic acid는 열수 추출물에서 2종, 80% 메탄올 추출물에서 6종이 확인되었다. Flavonoids는 열수 추출물에서는 확인되지 않았으나, 80% 메탄올 추출물에서는 1종이 확인되었다. 또한 연복자의 열수와 80% 메탄올 추출물에 대한 항산화 활성을 조사한 결과, DPPH와 ABTS의 라디칼 소거능은 처리농도에 비례하여 증가하는 경향을 나타내어 항산화 활성이 높은 것으로 나타났다. 따라서 이와 같은 결과를 종합하여 보았을 때 연복자의 열수와 80% 메탄올 추출물은 항산화 활성을 나타내는 물질을 함유하고 있고, 항산화 활성이 확인되어 피부관리를 위한 기능성 화장품 성분원료 및 천연 항산화제로서의 가능성이 있을 것으로 사료되어진다. In order to verify the effect of using functional cosmetic ingredients for skin beauty, the composition and content of some phenolic acid and flavonoids in the hot water and 80% methanol extract of Akebia quinata fruit pericarp and seeds were analyzed, and the skin care antioxidant activity was investigated. The results are as follows. Total polyphenol and flavonoid contents were found to be higher in pericarp extract than seed extract in both hot water and 80% methanol extract. In the analysis of the composition and content of phenolic acid and flavonoids, two kinds of phenolic acids were identified in the hot water extract of pericarp, 6 kinds of phenolic acids were identified in the 80% methanol extract, and one kind of flavonoid. Two types of phenolic acids were identified in the hot water and 80% methanol extract of seeds, respectively. DPPH and ABTS radical scavenging activities tended to increase in proportion to the treatment concentration in both hot water and 80% methanol extracts, and antioxidant activity was found to be high. Therefore, from the above results, it was found that the hot water and 80% methanol extract of Akebia quinata fruit pericarp and seeds contained various kinds of phenolic acid and flavonoids, and the antioxidant activity was also high. It is believed to be of value as a natural antioxidant in skin care cosmetic ingredients.

        Atypical cystic hepatocellular carcinoma in a Himalayan cat

        A liver mass was incidentally revealed in a 10-year-old male neutered Himalayan cat on radiographic examination conducted in a local animal hospital. The abdominal distention had worsened over the past five months, along with intermittent vomiting and nausea for two weeks. Serum chemistry was unremarkable, except for elevated alkaline phosphatase and gamma-glutamyl transferase levels. Radiography revealed a round-shaped soft mass in the cranial abdomen. Abdominal ultrasonography showed a well defined, massive, fluid-filled cyst with multiple lobes, and heterogeneous echogenicity in the pancreas. Computed tomography examination revealed hypoplasia of the left lobes of the liver, but the vascular changes and enhancement of the mass were not observed on contrast computed tomography images. The herniation of the abdominal fat through the diaphragm was incidentally observed on sonography and computed tomography scan. The patient was tentatively diagnosed with pancreatic cyst. The laparotomy revealed that the origin was not related to the pancreas, but was suspended from the liver. The mass was multilobular and filled with approximately 120 ml of fluid containing a few white blood cells. The patient recovered from anesthesia, but showed postoperative respiratory depression and died of cardiac arrest 15 hours later. Histopathologically, the mass was definitively diagnosed to be hepatic cellular carcinoma.

        쑥 추출물의 유리아미노산, 유리당 및 phenolic acid와 flavonoids 조성 및 함량

        This study analyzed the composition and contents of a portion of hot water and methanol extract created from Artemisia princeps var. orientalis leaves, stems, and roots. The results were as follows. The composition and content of free amino acids in the water extract were found to be 16 species of leaves and stems, respectively, and 11 species of roots. The methanol extract was confirmed to contain 13 leaves, 11 stems, and two roots. In the water extract, the non-constituent free amino acid compounds of the protein were confirmed as the leaves of seven species, six stems, and two roots. Methanol extracts were confirmed to contain two leaves and stems, and one root. In addition, composition and contents of free sugar were confirmed to be two species of leaves and roots, and three species of stems. In the analysis of phenolic acid and flavonoids, phenolic acid was found to be contained in seven species of leaves, six species of stems, and seven species of roots in the hot water extract. As a result, it was found that the content of free amino acids, free sugar, total polyphenol, and total flavonoid compounds were high in the A. princeps var. orientalis extracts, and that there were several types of phenolic acid and flavonoids. It has been confirmed that there is a high possibility for utilization as a natural antiseptic or cell growth activating factor as well as a natural antioxidant agent in the production of functional cosmetics.

        피부미용 화장품 소재로서 연복자 추출물의 유리아미노산 및 유리당의 조성과 함량분석

        This study analyzed pericarp of Akebia quinata fruit and hot water extract of seeds and 80% methanol extract to verify the effectiveness of utilizing functional cosmetic materials for skin care. 17 components of free amino acids were identified in pericarp and seeds. Both pericarp and seeds had the highest lysine content in hot water extracts and asparagine in 80% methanol extracts. Protein-free amino acid derivatives were identified in pericarp and seeds, each with 5 components. In the hot water extract, phosphoserine was the highest in pericarp and citrulline was the highest in seeds. In 80% methanol extract, carnosine was the most common in pericarp and citrulline was the highest in seeds. Analysis of the composition of free sugar identified frutose and glucose in pericarp and sucrose and maltose in seeds. As a result, when pericarp and seeds of Akebia quinata fruit are extracted with hot water and 80% methanol, free sugar and free amino acid derivatives are present, which is expected to be valuable as functional foods and nutritional cosmetics ingredients for skin beauty.

        참마의 괴경과 영여자 추출물의 Phenolic Acids와 Flavonoids의 조성 및 피부미용 화장품 생리활성 검색

        The purpose of this study was to investigate the phenolic compound compositions and skin care cosmetic physiological activities of Dioscorea japonica freeze drying yam‘s tuber, freeze drying and hot-air drying aerial bulbil extracts with MeOH solution. Total polyphenol content was higher in the yam tuber extract than aerial bulbil extract. Total flavonoid content of freeze-drying and hot airdrying aerial bulbil extract indicating higher than about 6.3 and 5.2 times content in the yam tuber extract, respectively. The major phenolic compounds of the yam tuber, freeze-drying and hot air-drying aerial bulbil extracts was p-hydroxybenzoic acid, sinapic acid and t-cinnamic acid respectively. Similarly, the flavonoids of the yam tuber, freeze-drying and hot air-drying aerial bulbil extracts were myricetin(6.02~7.43 mg%) and isorhamnetin(3.07~5.41 mg%), respectively. DPPH radical scavenging activity of freeze-drying and hot air-drying aerial bulbil extract was higher than yam tuber extracts. Tyrosinase inhibition activity of yam tuber extract indicating higher than about 32% and 22.35% freeze-drying and hot air-drying aerial bulbil extracts, respectively. The antimicrobial activities of freeze-drying yam tuber extract and hot-air drying aerial bulbil extracts was found that; E. coli KCCM 11234 and S. aureus KCCM 11593 cell growth was some inhibited at 1000 μg/ml concentration. Whereas, S. aureus KCCM 11593 cell growth was some inhibited by freeze-drying and hot-air drying aerial bulbil extracts. In addition, cytotoxicity for NIH3T3 cell line was not appeared by yam tuber, freeze drying and hot-air drying aerial bulbil extracts. These results indicated that D. japonica yam tuber and aerial bulbil extracts are functional and effective. In particular, they would be very useful as a health food and whitening cosmetic ingredients.

        아카시아 꽃 추출물의 피부미용 화장품 소재 생리활성 연구

        This study analyzed the composition and contents of acacia(Robinia pseudoacacia) flower extracts, free amino acid of water and methanol extracts and polyphenol compound. Then, the physiological activities of some skin care cosmetic examined. The study results found the followings: The free amino acid of acacia flower hot water extracts was 36.96 mg/g in total, and a total 16 types of free amino acids were isolated. In methanol extract, however, a total of 13.41 mg/g were found in 13 different types. In terms of polyphenol contents, methanol extract was slightly higher than hot-water extract. Regarding the composition of polyphenol compound, 8 different types of phenolic acids and two types of flavonoids were found in hot-water and methanol extracts. The DPPH radical scavenging activity was increased in proportion to the concentration, and the inhibitory activity of tyrosinase in the hot-water and methanol extracts of acacia flower was increased in proportion to the concentration. The ell growth according to human skin cell line, HaCaT was very low cell growth inhibited in an concentration-dependent. The inhibitory activity of elastase and collagenase in the hot-water extract and methanol extracts increased in a concentration-dependent manner. The said results show that because free amino acid and polyphenol contents are rich in the hot-water extract of acacia and methanol extract, they could be very useful as cell growth activating factors or natural antioxidants. To use them as cosmetic ingredients, however, there should be further studies on correlations between the enzyme activity in the cell and various cosmetic components.

        무궁화나무(Hibiscus syriacus) 수피 추출물의 페놀산 조성 및 기능성화장품 소재 가능성 평가

        The purpose of this study was to investigate the phenolic acid contents and compositions, and possibility of functional cosmetic ingredients of Hibiscus syriacus bark extracts with hot water and 70% ethanol solution. The results were as follows: Total polyphenol and total flavonoid content, hot water extract was grater than 70% ethanol extract. Regarding the composition of polyphenol compound, 8 different types of phenolic acids and 4 different types of flavonoids were found in hot-water and 70% ethanol extracts. In terms of DPPH radical scavenging activity, water extract was about 2 times grater than 70% ethanol extract. Nitrite-scavenging activity against hot water extract was 52.30 ± 0.42% and for the 70% ethanol extract was 46.00 ± 0.52% of stem bark in pH 1.2, respectively. In the tyrosinase inhibition activity of IC50, hot water extract was about 1.6 times grater than 70% ethanol extract. Bacterium cell growth was inhibited the proportional to the concentration for both the hot water and 70% ethanol extracts of bark for the Gram-negative E. coli KCCM 11234 bacterium. Cytotoxicity for SK-MEL-2 cell line was not appeared for hot water, and appeared about 16% in the 100 μg for 70% ethanol extract.
