        자궁경부암 세포주들과 Nude mouse 에 이종이식한 자궁경부암 세포에서 아데노바이러스와 리포펙틴을 이용한 종양억제 유전자 ( p53 ) 의 이입율

        N/A Background: The classical treatment of the cervical cancer is surgery, radiotherapy, chemotherapy. Even though the improvement of treatment successful rate, conventional therapy has some limitations. Recent cutting edge of cancer therapy has been developed in gene level including understand the biological characteristics of the cancer cells, enhance the human immune response, suppress the cancer cell proliferation. Therefore, the gene therapy is proposed to new treatment strategy. Purpose: The transfection efficiency of cervical cancer cell lines and cervical cancer cell line xerografted nude mouse was investigated by transfection of liposome and infection of adenovirus mediated suppressor(p53) and reportor(LacZ) gene. Method: The cervical cancer cell lines was used in this study were CaSki, SiHa (HPV16 positive, wild type p53 gene), HeLa, He]aS3(HPV18 positive, wild type p53 gene) and C33A, HT3(HPV negative, mutant p53). Direct plasmide and AdCMVp53 gene transfection was performed by using liposome system (pRcCMVLacZ / lipofectin, FuGene 6, Ca-phosphate). LacZ gene was used as the reportor gene for the transfection efficiency evaluation. Expression of p53 in cell lines and tumor tissue was confirmed by western blot and immunohistochemical staining. Xenografted nude mouse of SiHa cell line was infected by AdCMVp53 and AdCMVLacZ. Transfection efficiency was observed by same as above. Results: In cervical cancer cell lines, gene transfection using liposome system(pRcCMVLacZ/lipofectin, FuGene 6, Ca-phosphate)revealed different transfection efficiency, especially pRcCMVLacZ in Fugene 6 showed 18-40% of high transfection efficiency in 6 cervical cancer cell lines by X-gal staining and AdCMVp53 showed 95-98% of the high transfection efficiency in HeLa, C33A. AdCMVp53 was significantly expressed at 2-5days after injection xenografted nude mouse on the western blot and transfection efficiency was 19.79±5.36, 26.26 ± 11.69, 14.77±3.98,15.99 ±6.43%(day1-5). AdCMVLacZ were found to immunohistochemistry analysis, in vivo transFection efficiency was 61.26±4.66, 59.63±9.12, 29.46±14.33, 31.73±22.64%(day 1-5) at ×200 and 88.68.65, 70.85±20.94, 40.75±25.44, 48.21±10.97% (day 1-5) at ×400. Conclusion: As a results , adenovirus-mediated transfection efficiency was higher in vivo experiment compared to cell lines. These high efficiency of adenovirus-mediated suppressor gene(p53) could become a significant meaningful data gene therapy strategy both transgenic mice and cervical cancer cell lines.

        스키 참가자의 여가만족 영향요인 연구

        The purpose of this study was to investigate the relationships between factor that affect leisure satisfaction of the skiers. It used an intensive survey questionnaire as the research tool in this study. they were composed of 401 people who had been participating in ski from three ski resort in Gang-won. The resulting data were processed by means of frequency analysis and exploratory factor analysis using SPSS 21.0 program. Structural equation modeling techniques were used to test the relationships among the variable using AMOS 21.0 program. The results of analysis can be acquired as follows. First, in the relations between the participation motivation and the leisure constrains, the self-development factor influenced negative to leisure constrains. Second, in the relations between the participation motivation and the leisure constrains negotiation, the self-development factor and the sociality factor influenced positive but deviation factor influenced negative to leisure constraints negotiation. Third, in the relations between the participation motivation and the leisure satisfaction, the deviation factor influenced positive to the leisure satisfaction. Fourth, the leisure constraints influenced positive the leisure constrains negotiation. Fifth, the leisure constraints influenced negative the leisure satisfaction. Sixth, the leisure constraints negotiation influenced positive the leisure satisfaction. this study showed that although a leisure constraints has negative effects on the leisure satisfaction, the leisure constraints can be relaxed though a leisure constraints negotiation.

        Wheel 형 분무건조기에서 반응표면법에 의한 치즈 분말의 특성 해석

        Wheel형 분무건조기에서 식품첨가제로 사용되는 치즈분말의 건조공정을 반응표면법에 의하여 원료의 고형분함량, wheel 회전속도 및 출구온도를 독립변수로 하고 분말제품의 수분함량, 겉보기밀도, 분말 입자크기 및 현탁액의 점도를 종속변수로 하여 중심합성계획법에 따라 분산분석한 후 유의성과 상관관계을 검증하였다. 독립변수와 종속변수들의 상관관계를 검증한 결과 입자크기와 수분함량은 wheel의 회전속도가 커질수록 감소하였고, 겉보기밀도는 출구온도가 높아짐에 따라 감소하였다. 현탁액의 점도는 wheel의 회전속도와 고형분함량이 커짐에 따라 증가하였다. 분말제품의 종속변수들의 상관관계에서 수분함량과 겉보기밀도는 비례 관계, 입자크기는 수분함량과 걸보기밀도와 반비례의 관계를 나타내었다. In the spray drying with rotating wheel atomizer of cheese powder, the relationships among variables were analyzed with Response Surface Methodology in which several independent variables such as total solid content, wheel rotation speed, and outlet temperature influenced dependent variables such as particle diameter, moisture content, bulk density, and viscosity of suspended liquid. Significance and correlation were tested according to central composite design. As a results of analyzing the correlations between independent and dependent variables, particle diameter and moisture content of cheese powder were decreased with increasing wheel rotation speed, and bulk density was decreased with increasing outlet temperature. Viscosity of suspended liquid were increased with increasing wheel rotation speed and total moisture content. In correlation among dependent variables, moisture content was proportional to bulk density, and particle diameter was inversly proportional to moisture content and bulk density.

        난치성 다발성근염/피부근염 환자에서 정맥내 면역글로불린의 효과

        목적: 부신피질 호르몬 및 면역억제제에 반응이 없거나 사용이 제한되는 난치성 다발성근염/피부근염에서 정맥내 면역글로불린의 치료 효과를 알아보고자 하였다. 대상 및 방법: 1992년부터 1995년 현재까지가 톨력의대 부속 강남성모병원 내과에서 정맥내 면역글로불린 치료를 시행하였던 6명의 환자를 대상으로 면역글로불린을 정맥내로 매달 2g/kg의 용량을 6개 월간 투여하였다. 임상적 평가는 British Medical Research Council (BRC) muscle power score, timed stand test, functional grading scale, 생화학 검사로 혈청 CPK, aldolase를 조사하였다. 치료 효과의 판정은 입상적 개선이 치료전에 비해 20%이상 나타난 경우와 근효소치의 감소가 30% 이상인 경우로 하였다. 결과: BRC muscle power score는 대상 환자 6명중 5명에서 임상적 개선이 관찰되었고, timed stand test functional grading score는 6명 모두에서 유의한 임상적 호전을 보였다. 치료전 CPK가 정상범위이었던 2예를 제외한 4예 모두에서 근효소치의 유의한 감소가 있었는데 이중 3예에서는 50% 이상의 현저 한 감소를 보였다. aldolase를 측정하였던 5명 모두에서 치료전에 비해 50% 이상의 뚜렷한 감소가 나타났다. 치료전 prednisolone 평균 1일 투여 량은 39.2±9.2mg에서 치료 6개월 후 7.03±2mg으로 뚜렷한 스테로이드 감량(81.3%) 효과를 보였다. 부작용은 1예에서 경미한 호흡곤란 및 발한이 일시적으로 나타났으나 분할 투여하면서 사라졌다. 결론: 정맥내 면역글로불린 주사요법을 기존 약제에 반응이 없거나 사용이 제한될 경우, 유용하고 안전한 치료 방법으로 생각된다. Objectives: Polymyositis (PM) and dermatomyositis (DM) are inflammatory muscle diseases of presumed autoimmune origin. Many interventions including corticosteroids, immunosuppressive drug, and plasmapheresis to treat patients with PM/DM are not always effective, and may be associated with certain serious side effects. Intravenous immunoglobulin (IVIG) has been useful in a number of autoimmune diseases. An attempt was made to evaluate the efficacy of IVIG in refractory PM/DM. Methods: Six patients with conventional treatment-refractory PM/DM [1 man and 5 women; mean age of 27(7-49)years; PM(2), DM(2), juvenile DM(2)] received high doses of IVIG (2 mg/kg) dividing over consecutive 2 or 5 days. The IVIG infusion was conducted each month for 6 months. Clinical evaluations, including proximal muscle power, functional grading, and biochemical studies (creatine kinase (CK), aldolase) were performed before each or every IVIG infusion. Clinical evaluations were considered successful if more than 20% improvement of initial score was obtained. Biochemical results were considered good if muscle enzymes decreased more than 30% of initial values. Results: Clinical improvement of proximal muscle power was noted in 5 patients after 6 courses of IVIG infusion. Timed stand test and functional grading score were also improved in all patients. All patients showed good response of biochemical results, except two cases with normal initial CK levels. Mean daily prednisolone dosage was significantly reduced (before IVIG: 39.2±9.2 mg/day vs after 6th IVIG: 7.3±2 mg/day, p<0.0l). One patient experienced mild dyspnea and sweating during the course of IVIG infusion, which were disappeared with the reduction of daily IVIG dosage. Conclusions: These results suggest that IVIG is a safe and effective therapy in patients with PM/DM, who are resistant to traditional therapies or have limitations for their use.

        전외측 도달법을 이용한 소절개 회전근 개 봉합술

        목적: 회전근 개 파열에서의 전외측 도달법을 이용한 소절개 봉합술을 소개하고 임상적 결과 및 그 효용성을 알아보고자 하였다. 대상 및 방법: 전외측 도달법을 이용한 소절개 봉합술을 시행하고 1년 이상 추시 관찰이 가능하였던 59예를 대상으로 하였다. 평균 연령은 56.6 (39~71)세였고, 남자가 39예, 여자가 20예였으며, 평균 추시 기간은 26 (12~41)개월이었다. VAS 점수 및 ADL 지수를 이용하여 동통 및 기능적 평가를 시행하였고, 임상적 결과의 판정은 ASES 점수를 이용하였다. 결과: VAS 점수는 술 전 7.04점에서 최종 추시시 1.02점으로, ADL 지수는 12.37점에서27.20점으로 호전되었으며, ASES 점수는 술 전 35.32점에서 최종 추시시 90.08점으로 향상되었다(p=0.000). 임상적 결과는 우수 41예, 양호 11예, 보통 2예, 불량 5예로 52예 (88.1%)에서 만족할 만한 임상적 결과를 나타내었다. 나이, 성별, 증상 기간, 파열의 크기, 술 전 견관절 강직과 최종 추시 ASES 점수와의 통계학적 유의성은 없었다 (p>0.05). 결론: 전외측 도달법을 이용한 소절개 봉합술은 만족할 만한 임상적 결과를 보였으며 시야 확보에 유리한 술식으로 사료된다. Purpose: To introduce mini-open rotator cuff repair using the anterolateral approach and evaluate its clinical outcomes and effectiveness. Materials and Methods: 59 consecutive cases of rotator cuff tearing which were treated with mini-open repair utilizing the anterolateral approach were evaluated. The population comprised 39 men and 20 women, with an average age of 56.6 years. An average follow-up time period was 26 months. Clinical outcomes were analyzed based on VAS, ADL, and ASES scores. Results: The average respective VAS, ADL, and ASES scores improved from 7.04, 12.37, and 35.32 preoperatively to 1.02, 27.20, and 90.08 postoperatively (p=0.000). There were 41 excellent, 11 good, 2 fair, and 5 poor results. There were satisfactory results in 52 cases (88.1%). There were no statistically significant differences between the final ASES scores and age, sex, duration of symptoms, tear size, and preoperative stiffness (p>0.05). Conclusion: Mini-open rotator cuff repair using the anterolateral approach effective in providing better visualization.

        MCF-7 세포주에서 Staurosporine 투여에 의한 세포주기 분기 변화와 p53단백 발현

        N/A Objective : We investigated the effects on the cell cycle and p53 expression by the treatment of various concentrations of staurosporine to elucidate the molecular mechanism of staurosporine induced cell cycle arrest in MCF-7 cell line. Methods : Various concentrations of staurosporine were treated in MCF-7 cells cultured with RPMI 1640 media. The harvested cells were fixed and permeabilized with 1% paraformaldehyde and absolute methanol. Then the cells were stained indirectly with anti-p53 primary antibody and FITC conjugated goat anti-mouse(GAM)-IgG secondary antibody. Sequentially DNA were stained with 0.1% RNase and PI solution. These stained cells were analyzed by the standard FACScan flow cytometer. The obtained results were analyzed further with WinList 3.0, and ModiFit LT software program. Results : MCF-7 cells were arrested mostly in G1 phase of cell cycle at 5-10 nM of staurosporine, however, the cells were arrested in G2 phase at 20-100 nM of staurosporine. The expressions of p53 protein were higher in the MCF-7 cells treated with both concentrations of 10 nM and 100 nM of staurosporine compaired with the control cells. This suggests that the p53 may be involved in the mechanism of G1 and G2M arrest of cell cycle in MCF-7 cell. Conclusion : The points of arrest in cell cycle differred depending on the concentrations of staurosporine and these cell cycle arrests at G0G1 and G2M pahse were related with p53 protein expression. It suggested that these results could be extended to study for staurosporine to be usefull as a potential anti-tumor agent.
