        TIP30 Directly Binds p53 Tumor Suppressor Protein In Vitro

        TIP30 (30 kDa HIV-1 TAT-interacting protein), also called HTATIP2 or CC3, is a tumor suppressor protein that acts as an angiogenesis inhibitor. TIP30 blocks nuclear import of the mRNA-binding protein HuR, and thereby promotes the cytoplasmic accumulation of HuR by binding to im-portin-β, which is known to facilitate the cytoplasm-to-nuclear transport of HuR. Accumulation of HuR in the cytoplasm, in turn, enhances the expression of the transcription factor p53, a tumor suppressor that plays an essential role in preserving genome stability and inhibiting cancer growth. In addition to such a post-transcriptional mechanism via which TIP30 increases the p53 level, it has been proposed that TIP30 may regulate p53 protein at the protein level by directly binding to it. In order to investigate the possibility of direct interaction between p53 and TIP30, we have used on three functional regions in p53 and examined their interactions with TIP30 using GST pull-down assay and surface plasmon resonance technique. The results show that that TIP30 binds to the DNA-binding domain and the C-terminal domain of p53.

        육회 중에 분포하는 미생물과 주요 식중독 세균의 조사

        육회는 가열처리 없이 섭취하기 때문에 다양한 미생물에 노출될 수 있다. 본 연구에서는 대구지역의 육회 전문점 5곳을 선정하여 미생물 분포를 조사하였다. 식당에서 판매되고 있는 조리된 육회를 식품공전 방법에 따라 중온성균 수와 대장균군 수를 측정하였고 식품 중에 검출되어서는 아니되는 병원성 세균인 Salmonella spp., Staph. aureus, E. coli O157:H7, L. monocytogenes의 검출 유무를 조사하였다. 조사 결과, 육회에는 중온성균이 $6.6{\times}10^3-2.7{\times}10^5\;CFU/g$, 대장균군이 $8.9{\times}10^1-2.1{\times}10^5CFU/g$으로 분포되었다. 모든 육회 시료에서 Salmonella spp., E. coli O157:H7, L. monocytogenes은 검출되지 않았으나 한 시료에서 Staph. aureus가 검출되었다. 이외 임상학적으로 중요한 미생물들이 동정되었다. 가열과정 없이 바로 섭취하는 육회는 원료의 생산 및 조리과정에서 엄격한 위생 감독이 요구된다. People can be exposed to various microorganisms when they eat yukae(seasoned raw meat) because yukae is eaten raw. The purpose of this study was to find out microbial distribution in yukae. In this experiment, 5 restaurants serving yukae were chosen in Daegu area. Mesophilic microorganisms and coliforms were measured by Korean Food Standards Codex. Analyses of Salmonella spp., Listeria monocytogenes, Staphylococcus aureus and Escherichia coli O157:H7 were conducted. As results, mesophilic microorganisms ranged $6.6{\times}10^3-2.7{\times}10^5\;CFU/g$ and coliforms ranged $8.9{\times}10^1-2.1{\times}10^5CFU/g$. Salmonella spp., Listeria monocytogenes and Escherichia coli O157:H7 were not detected in all 5 samples. However, Staphylococcus aureus was detected in 1 sample out of 5 samples. Hygiene practice during production, processing and cooking process is required.

        내경계막 제거술 후 발생한 활꼴모양의 망막신경섬유층의 부종

        Purpose: To evaluate the cause of dark arcuate striae observed in infrared photographs in idiopathic epiretinal membrane (ERM) and macular hole patients after internal limiting membrane (ILM) peeling. Methods: Forty patients (41 eyes) of idiopathic ERM and macular hole who underwent trans pars plana vitrectomy with ILM peeling and gas tamponade were included in the present study. The best corrected visual acuity (BCVA) was recorded at preoperative and postoperative 6 months. Infrared (IR) photography with spectral domain optical coherence tomography (SD-OCT) were obtained at preoperative and postoperative 1 week, 1 month, 3 months, and 6 months. When abnormal findings were visible on IR photographs, additional SD-OCT was performed at the corresponding sites. Results: Of 40 patients, 4 patients demonstrated dark striae extending from the optic nerve to near macular area in IR photographs at 1 week postoperatively. SD-OCT images of the dark striae region revealed the swelling of retinal nerve fiber layer (RNFL). At postoperative 6 months, however, RNFL swelling previously observed subsided in all 4 cases, while temporal retinal thinning and dimples were observed in 3 cases. The preoperative and postoperative BCVA did not show any statistical difference between the patients with the RNFL swelling and the patients without the swelling. Conclusions: Swelling of RNFL may occur in idiopathic ERM and macular hole patients after pars plana vitrectomy and ILM peeling, which may present as dark striae in IR photographs.       • 사회공포증 경과중 타 정신질환이 발병한 환자들에 대한 연구

        We have studied 16 patients diagnosed of social phobia among the hospitalized patients for 1 year, from September 1994 to August 1995, to evaluate whether serial phobic symptoms are the prodromal symptoms of the other psychiatric disorder or concomitant disorder. The results were as follows; 1) In the subject of this study, the mean age of onset of social phobic symptoms was 17.3, and the mean age of inital hospital visit was 27.4. 2) The proportion of offensive type was higher than the simple type : 7 cases of the subjects classified as simple type, 9 cases as offensive type. 3) According to the type of social phobia, they developed different type of psychiatric disorder. Majority of patients with simple type had final diagnosis of mood disorder. Patients with offensive type had schizophrenia spectrum disorder, such as schizophrenia, delusional disorder, and schizophrenia, personality disorder. 4) Social phobic symptoms have taken various courses according to the final diagnosis. In the patients who developed major depression and delusional disorder, social phobic symptoms have not been changed during the course. Patients with the final diagnosis of schizophrenia and schizotypal personality disorder have showed various social phobic symptoms. In the patients with the final diagnosis of bipolar disorder, their social phobic symptoms disappeared with improvement of depression.

        오요탕(五拗湯) 및 신출산(神朮散)이 GUINEA PIG의 기관지(氣管支) 평활근(平滑筋)에 미치는 영향(影響)

        This study was carried out to investigate the effect of Oyootang and Shinchulsan extract on the contractile force of the isolated guinea pig trachea smooth muscle and elucidate its mechanism The result were obtained as follow: 1. The isolated trachea smooth muscle was suspended in the organ bath with oxygenated kerb's Henseleit bicarbonate buffer solution at $37^{\circ}C$, and the developed tension by the drug was recorded with isometric tranducer(Nacro F-60). The resting tension was approximately 0.5g 2. The contractile response of the trachea smooth muscle of the isolated guinea pig by histamine $10^{-4}M$ was significantly inhibited by Oyootang and Shinchulsan extract. 3. The contractile response of the trachea smooth muscle of the isolated guinea pig by acetylcholine $10^{-4}M$ was considerably inhibited by Oyootang and Shinchulsan extract. 4. The contractile response of the trachea smooth muscle of the isolated guinea pig by histamine $10^{-4}M$ was significantly inhibited by Oyootanghabshinchulsan extract. 5. The contractile response of the trachea smooth muscle of the isolated guinea pig by acetylcholine $10^{-4}M$ was significantly inhibited by Oyootanghabshinchulsan extract. 6. The contractile response of the trachea smooth muscle of the iso-lated guinea pig by histamine $10^{-4}M$ was significantly inhibited by Oyootang, Shinchulsan and its constituent herbs extract. 7. The contractile response of the trachea smooth muscle of the iso-lated guinea pig by acetylcholine $10^{-4}M$ was significantly inhibited by Oyootang, Shinchulsan and its constituent herbs extract.

        노니(Morinda citrifolia)분말을 첨가한 분쇄돈육의 이화학적 특성 및 항산화 활성

        The purpose of this study was to examine the effect of noni (Morinda citrifolia) powder on the physicochemical properties and antioxidant activities of ground pork meat. Four samples of ground pork meat were prepared as follow: CON without noni powder, NP0.5 with 0.5% noni powder, NP1.0 with 1.0% noni powder, and NP1.5 with 1.5% noni powder. The moisture content of CON was the highest (p<0.05). The ash, fiber, fat retention, hardness, cohesiveness, gumminess, chewiness, lightness, and yellowness increased with the addition of the noni powder (p<0.05). The protein, fat, water holding capacity, cooking yield, moisture retention, springiness,and redness of the cooked meat were not significantly different among the samples. The pH of CON was the highest among the samples (p<0.05). The TBARS of CON, NP0.5, NP1.0, and NP1.5 were 0.48, 0.41, 0.36 and 0.34 mg/kg, respectively, and the NP1.5 was the lowest (p<0.05). The DPPH radical scavenging activity of CON, NP0.5, NP1.0, and NP1.5 were 10.4%, 20.8%, 34.6% and 45.3%, respectively, and the NP1.5 was the highest (p<0.05). Consequently, these results support the possible use of noni powder for meat product industry, as addition of noni powder enhances the antioxidant activities of ground pork meat.

        선학초(짚신나물) 경구투여시 항암효과 탐색 및 약물 대사효소의 변화

        Objective : This research was aimed to investigate the anti-tumor effect, safety, mechanism and metabolizing enzyme of Agrimonia pilosa LEDEB(APL) in female C57B/L mouse. Methods : At first, to evaluate the anti-tumor activity of APL, we divided into four groups, normal, control, APL100(100mg/kg), APL150(150mg/kg). LLC obtained American Type Culture Collection was used. LLC had been inoculated to induce tumor. To measure the anti-tumor effect of APL, we calibrate tumor size and weight. To study for mechanism of anti-tumor in APL, we used western blotting and to know metabolizing enzyme in APL we used to real-time PCR. Results : APL100, APL150 inhibited tumor growth after medicine injected. APL did not only induced caspase-dependent apoptosis in LLC-bearing mouse tumor. In APL100, it were decreased 72% in CYP3A11. In APL150, it were decreased 62%, 75% in CYP3A11 and MRP1a respectively. Conclusion : These results suggests that APL has some anti-tumor effects in female C57B/L mouse tumor. APL should be careful use with other drugs related with CYP3A11 or MRP1a.
