        염스트레스가 담배식물의 광합성 , proline 및 이온함량에 미치는 영향

        본 실험은 담배의 염스트레스에 대한 생장특성 및 생리적반응을 구명하기 위하여 NaCl을 농도별로 처리하여 시험한 결과를 요약하면 다음과 같다. NaCl농도가 높아질수록 생장은 크게 억제되었으며, 90mM의 지상부/지하부율이 약 2.0이하에서 생육형질이 크게 감소하여 생리적인 제한농도로 나타났다. 광합성량, 증산량 및 수분이용효율은 NaCl농도가 높아질수록 감소하였으며, 광합성량은 60mM에서 대조구의 50%의 감소로 염해에 민감하게 반응하였으며, 증산량과 수분이용효율은 처리농도간에 큰 차이는 없었다. 잎수분포텐셀은 염농도의 증가에 따라 낮아졌으며 30mM에서 대조구의 2배로 낮아졌으며 처리농도간에는 120mM에서 크게 감소하였다. Proline함량은 NaCl농도가 높아짐에 따라 120mM가지는 일정하게 증가하다가 150mM에서 감소하였다. NaCl농도에 따라 Ca^(2+), Mg^(2+), K^+의 함량은 90mM까지는 증가하였고, 120mM이상에서는 감소하였다. Na^+은 염농도의 증가에 따라 완만히 증가하다가 150mM이상에서 급격히 증가하였다. Cl^-은 염농도의 증가에 따라 30mM의 일시적 증가외는 처리간의 차이는 없이 감소하는 경향이었다. K^+/Na^+율은 염농도가 높아질수록 감소하였으며 90mM의 1.0이하에서 세포내의 항상성이 유지되는 농도로 나타났다. K^+와 Na^+함량과는 부의 상관을, K^+/Na^+율과 단백질함량과는 정의 상관을 나타냈다. This experiment was conducted to investigate effects of NaCl concentration on photosynthetic rate, free proline content and ion content in tobacco. As NaCl concentration was increased growth was retarded. The decrease growth characteristics(shoot/root ratio was 2.0) at 90mM NaCl indicated that this concentration could be a limiting level. As NaCl concentration was increased photosynthetic rate, transpiration rate, and water use efficiency were decreased. Photosynthetic rate was highly decreased at 60mM NaCl. There was no significant difference between transpiration rate and water use efficiency. Leaf water potential was decreased as NaCl concentration was increased, in that twice lower at 30mM than that of control and drop largely at 120mM NaCl. Free proline content was increased as NaCl increased until 120mM NaCl and drop at 150mM NaCl. The Ca^(2+), Mg^(2+), and K^+ contents were increased until NaCl concentration was 120mM. The Na^+ content was slowly increased as NaCl concentration increased until 120mM NaCl, and largely increased at 150mM NaCl. There was no significant difference between Cl^- and NaCl treatments except 30 mM NaCl in which Cl^- content was higher than that of control. As NaCl concentration was increased K^+/Na^+ ratio was decreased. The negative correlation between K^+ and Na^+, and positive correlation between K^+/Na^+ and protein content were found.

        This study was carried out to obtain the basic data which include the change of the photosynthetic rate and protein content according to growth stage in the process of senescence of tobacco plant The photosynthetic rate was the maximum with 26.31$\mu$mol.CO2/m2.sec and stomatal resistance was the minimum with 0.2552cm/sec at 15th days after leaf emergence. However, after 50 days the photosynthesis was very little occurred. During leaf developments the number of chloroplast was increased and reached at the maximum at 25th days after emergence of leaf, thereafter, it was decreased gradually. The content of protein increased continuously and showed the highest value at 15th days after leaf emergence. The degradation rate of soluble protein was more rapid than that of insoluble protein at early stage of senescence. The range of decrement in the insoluble protein was low at late stage of senescence. The content of Rubisco, the key enzyme of photoamthesis, corresponded to about 50% of soluble protein and reached to the maximum at 150 days after leaf emergence. As the senescence progressed, the content of large subunit(UV) of Rubisco showed a tendency to decrease more rapidly than that of small subunit(SSU). The total amount of amino acids was the highest at 15th days after leaf emergence.

      • 육묘포트 깊이가 담배의 뿌리발달 및 분포에 미치는 영향

        The study was carried out to clarify reasonable production of healthy seedling, optimal Pot depth, and appropriate transplanting time, which can be deduced from understanding of seedling quality. Seedling quality results from growth of root and shoot, morphology and distribution of root system under influence of Pot depth during seedling growing period. Stem height, shoot dry weight, leaf area and leaf number were increased in proportion to depth of pots. Growth of shoot and root during seedling growing period showed the most dramatic development between 20th and 25th day after temporary planting. Root number increased as pot depth decrease and total root length and dry weight increased as pot depth increase. In 5cm pot, relative multiplication rate was higher and mean extension rate was lower than other depth of Pot. The limitation of pot volume in which rhizosphere was located enhance the development of roots of second and third order. At 20th days after temporary Planting root distribution was relatively uniform in length and development of adventitious root on stem base was poor as Pot depth decreased.

        염스트레스가 담배식물의 Protein , ATPase 및 Peroxidase 활성에 미치는 영향

        본 실험은 담배에서 염스트레스에 따른 생리적 반응의 연구결과(제1보)를 기초로, NaCl를 농도별로 처리하여 생체내에서 일어나는 생화학적인 변화의 구명을 통해 염해기작의 기초자료로 얻고자 수행한 결과를 요약하면 다음과 같다. 총단백질과 가용성단백질은 염농도가 높아질수록 감소하였으며 처리간에는 120mM까지는 완만히 감소하였고, 150mM에서는 급격한 감소를 나타냈다. 전기영동 패턴은 염농도의 증가에 따라 새로운 polypeptide band의 생성과 소멸은 없었으며 약 74Kd의 polypeptide band에서 30mM과 60mM까지는 뚜렷한 양이 증가하였고 90mM부터는 감소하였다. 엽록소함량은 염농도의 증가에 따라 감소하였으며 특히 염해에 의한 반응은 엽록소b보다는 엽록소a가 민감하였다. ATPase활성과 peroxidase의 활성은 염농도가 높아질수록 120mM까지는 일정하게 증가하였으나 150mM에서 급격히 증가하여 담배의 염해의 생화학적인 제한범위는 120mM로 나타났다. The analysis of biochemical changes in tobacco plant as increase of NaCl concentraion was conducted. Total protein content and soluble protein content were decreased as NaCl concentration was increased, in that steady decreased until 120mM NaCl and largely decreased at 150mM NaCl. The expression of 74Kd subunit was increased until 60mM NaCl. However, the amount of 74Kd protein was decreased from 90mM NaCl. There was no difference for expression of other protein subunits. Chlorophyll a content was significantly decrease as NaCl concentration was increased, but chlorophyll b content was not much decreased. The slow increase up to 120mM NaCl and large increase at 150mM NaCl for ATPase and peroxidase activities indicated that 120mM NaCl could be a limiting concentration for salt injury.

        수분 스트레스가 담배의 잎 수분 포텐셜, 광합성 및 뿌리발달에 미치는 영향

        Development of shoot and root, leaf water potential and photosynthetic rate affected by water stress in early growing stage of tobacco were surveyed to interpret stress response in terms of plant physiological and agricultural aspects. The growth of shoot and root was highly suppressed by water stress and the difference in dry weight by rewatering was smaller in root than in shoot. The total root length was highly decreased by water stress and the lengths of root for water stress and non-stress were 74m and 84m, respectively, after rewatering. The root growth treated by water stress was increased between 2nd and 3rd day after treatment indicating that temporary water stress at early growing stage might have increased of root zone activity for early growth stage. The leaf water potentials were decreased to -7.63MPa, -9.47MPa, -11.89MPa, -13MPa at the 2nd, 3rd, 4th and 5th day by water stress. The relative water contents were 75%, 62% and 57% at the 3rd, 4th and 5th day after treatment. Photosynthesis was reduced largely by water stress. The photosynthetic rate after treatment at 2nd day and 3rd day was dropped to 18.15~mu mol. CO2 /m2·sec-1 and 9.35~mu mol. CO2 /m2·sec-1 . It was never recovered to the normal, even after rewatering. Stomatal conductance had been reduced since 2nd day after treatment and increased after rewatering.

      • 담배의 성숙에 따른 수확엽의 생화학적 활성변화

        In order to investigate biochemical activities of harvested tobacco leaves, photosynthetic rate, soluble protein contents, and peroxidale activities were analysed during different maturing period. Physiological activities of harvested leaves during maturing period were higher in topped than those of non-topped plants. Chlorophyll content and photosynthetic rate in both topped and non-topped plants decreased at 4 days and 3 days before harvest, respectively. The chloroplast numbers in topped and non-topped plants decreased at 3 days and 5 days before harvest, respectively. Changes of soluble protein and total RNA contents showed similar patterns during maturing period. Soluble protein contents were slightly decreased from 5 days before harvest in topped plants, but decreased drastically from 3 days before harvest in non-topped plants. Not much changes were found in total RNA contents in topped plants until 2 days before harvest, and it was largely decreased after 5 days before harvest in non-topped plants. The peroxidase activities drastically decreased in topped plants and increased in non-topped plants after 3 days before harvest during maturing period. The largest change of biochemical activities in tobacco leaves during maturing periods were observed at 3 days before harvest.
