Recently many efforts were focused to understand the mechanical insights of melanogenesis to develop the agents for hyper-pigmentation and hypo-pigmentation. In the melanin biosynthetic pathway, tyrosinase is the rate limiting enzyme, and ${\alpha}$-melanocyte stimulating hormone(MSH) or cAMP-elevating agents stimulate melanogenesis and enhance the melanin synthesis and the tyrosinase activity. The author has analyzed the effects of Rhizoma Bletillae on the basal melanogenic activities of B16 mouse melanoma cells. Rhizoma Bletillae alone markedly suppressed melanin content and tyrosinase activity in a dose-dependent manner. Pretreatment of the cells with Rhizoma Bletillae. The decrease in the tyrosinase activity was paralled by a decrease in the abundance of tyrosinase protein and tyrosinase promoter activity. These results suggest that Rhizoma Bletillae inhibits melanogenesis of B16 melanoma cells via suppression of tyrosinase activity.

Rhizoma Bletillae alone markedly suppressed melanin content and tyrosinase activity in a dose-dependent manner. Pretreatment of the cells with Rhizoma Bletillae also suppressed the increase of ${\alpha}$-MSH (100 nM) or forskolin (20 ${\mu}M$)-induced melanin content and tyrosinase activity. The decrease in the tyrosinase activity was paralled by a decrease in the abundance of tyrosinase protein and tyrosinase promoter activity. Pretreatment of the cells with Rhizoma Bletillae also inhibited the increase of forskolin(20${\mu}M$) induced the amount of tyrosinase protein and tyrosinase promoter activity. The results of DOPA staining revealed that pretreatment of the cells with Rhizoma Bletillae showed less intensity than B16 melanoma cells stimulated with ${\alpha}$-MSH or forskolin. These results suggest that Rhizoma Bletillae inhibits melanogenesis and abrogates ${\alpha}$-MSH and cAMP-induced melanogenesis in B16 melanoma cells.
양격산화탕(凉膈散火湯)의 항염증(抗炎症) 효과에 대한 연구

Objective: Yang Geouk San Hwa - Tang (YGSHT) has been widely used in Sasang Constitutional Medicine of Korea for treatment of acute inflammatory symptom, such as palatine tonsillitis, polydipsia, headache, papule, pimple however, the mechanism of its anti-inflammatory activity has not been clarified. In this study, therefore, we investigated the mechanism of the inhibitory effect of YGSHT on LPS-induced inflammation. Materials and methods: The effect of YGSHT was analyzed by ELISA, RT-PCR and Western blotting in LPS-stimulated RAW264.7 cells. Results: We found that YGSHT suppressed not only the production of pre-inflammatory cytokines (IL-$1{\beta}$ and TNF-$\alpha$), the generation of nitric oxide (NO) and prostaglandin E (PGE)2, but also the mRNA expression of pre-inflammatory cytokines, inducible nitric oxide synthase (iNOS), and cyclooxygenase (COX)-2. Furthermore, YGSHT was shown to inhibit the phosphorylation of ERK1/2 and JNK1/2 and the activation and translocation of NF-kB from cytosol to nuclear in LPS-stimulated RAW264.7 cells. Conclusions: These results suggest that YGSHT exerts an anti-inflammatory effect through the regulation of the ERK1/2 and JNK1/2 pathway and NF-kB pathway, thereby decreasing production of pre-inflammatory cytokines, NO, and PGE2.

Objective : The purpose of this study is to report the effect of Pharmacopuncture Therapy and Needle-embedding Therapy on Androgenetic Alopecia in woman. Methods : The patient was treated by acupucture, pharmacopuncture, needle-embedding. The improvement of the patient was judged by phtograpies and VAS(Visual Analogue Scale) score. Results & Conclusions : Her hair loss and physical condition were improved remarkably, and an author consider that continuous clinical study will be needed in other needle-embedding therapy and korean medical treatment.

Objective : The purpose of this report is to know the effect of Korean medical treatments on exfloliative dermatitis. Methods : We treated a 25-years-old man patient with exfloliative dermatitis on the limbs and back with Korean medicine. After Korean medical teatment for 26 days, we measured the decrease of dermatitis and recovery of skin exfloliate. We recorded pictures of changes on symptoms. Result & Conclusion : The symptoms of exfloliative dermatitis were significantly improved. The dry skin and itching were disappeared and skin damages were almost recovered. Thus Korean medical treatments are effective on exfloliative dermatitis.

Objective: Previously, the methanol extracts of the semen of Xanthium strumsrium could involved anti-inflammatory effects in lipopolysaccharide (LPS)-stimulated Raw 264,7 cells, We evaluated the anti-allergic effects of X. strumarium on rat basophilic leukemia (RBL-2H3) cells, Methodes : To investigate the effect of X. strumarium on the phorbol 12-myristate 13-acetate (PMA) and calcium ionophore A23187-induced RBL-2H3 cells. The effects of X. strumarium on the degranulation and the pro-inflammatory cytokines secretion and expression from RBL-2H3 cells were evaluated with $\beta$-hexosaminidase assay, ELISA, and RT-PCR analysis, In addition, we examined the effects of X. strumarium on nuclear factor (NF)-${\kappa}B$ activation and $I{\kappa}B-\alpha$ degradation using Western blot analysis. Results : X. strumarium inhibited degranulation and secretions and expressions of pro-inflammatory cytokines, such as tumor necrosis factor-alpha ($TNF-\alpha$), interleukin (IL)-4 and cyclooxygenase (COX)-2, on stimulated RBL-2H3 cells, however, X. strumarium not affect cell viability. In stimulated RBL-2H3 cells, the protein expression level of nuclear factor-kappa B (NF-${\kappa}B$) was decreased in the nucleus by X. strumarium. In addition, X. strumarium suppressed the degradation of inhibitory protein $I{\kappa}B-{\alpha}$ protein in RBL-2H3 cells. Conclusion : These results suggest that X. strumarium inhibits the degranulation and secretion of pro-inflammatory cytokines through blockade of NF-${\kappa}B$ activation and I $I{\kappa}B-{\alpha}$ degradation.
한방치료를 통해 개선된 화폐상 습진 치험 1례 : 황련아교탕(黃蓮阿膠湯)과 마황부자세신탕(麻黃附子細辛湯)을 중심으로

Objectives : The purpose of this study is to report the effect of Hwangryeonagyo-tang and Mahwangbujasesin-tang(《Shanghanlun》 prescription) on whole-body nummular eczema. Methods : We treated a female patient in her early twenties who has whole-body nummular eczema with Korean medicine. She was treated from 2020.10.8 to 2020.12.11. In this period, she received admission treatments twice because of the recurrence of the symptoms after the first admission. During the treatment, the two herbal medicines were changed depending on the patient's condition. 'Hwangryeonagyo-tang' and 'Mahwangbujasesin-tang' were used as herbal medicine. In addition, herbal external preparations and acupuncture treatment were also performed. The evaluation was conducted by photo comparison, VAS, and DLQI. Blood tests were also conducted for safety assessments. Results : After two months of treatment, the patient's condition improved. The itching and heat sensation decreased, and scabs were placed on the lesion of the eczema, reducing the oozing. VAS of itching decreased from 8 to 0. DLQI was also improved, and the stability of the herbal medicines for liver and kidney was also confirmed through blood tests. Conclusions : Hwangryeonagyo-tang and Mahwangbujasesin-tang helped relieve the itching and heat of eczema in currency. Therefore, it is believed that the proper use of thoes herbal medicine can be expected to have a useful effect on nummular eczema.
아토피 피부염에 운용 가능한 경혈(經穴) 탐구 - 상용(常用) 본초(本草)를 중심으로

Objectives : The purpose of this study is to investigate acupoints for atopic dermatitis which have similar impact with frequent herbs. Methods : We collected research materials in "The Journal of Korean Medicine Ophthalmology & Otolaryngology & Dermatology" published from Feburary, 2000 to September, 2017. The search terms are 'atopic Dermatitis', 'antiallergic', 'anti-inflammatory', 'antianaphylaxis', 'dermatitis' and we elected experimental researches about herbal medicine. Among the herbal medicine, we extracted frequent herbs using more than 5 times, and categorized through "Herbal Medicine". By Consulting "Principles of Meridians & Acupoints; A Guidebook for College Students", we matched frequent herbs and acupoints. Results : 1. There are 38 frequent herbs for study except Glycyrrhiza uralensis FISCH. because it was used for balance in herbal medicine. Arrange meridians in order of frequency, the most is 'Lung meridian'. Among the organs, the most related one with skin is 'Lung' and 'Lung meridian' can treat skin diseases. 2. We researched acupoints called 'specific acupoints' under elbow, knee because of clinical usage. Exceptively, SP6 and GV20 are not included in 'specific acupoints' but they can be effective for atopic dermatitis treatment. 3. Researching on MEDLINE about frequent herbs and acupoints, there are many studies that acupuncture is effective for reducing pruritus. Therefore, the acupoints which were mentioned in this study can be choosed for atopic dermatitis. Conclusions : We found several applicable acupoints for Atopic Dermatitis. We hope it will be useful in experiment such as randomized controllized trial on Atopic Dermatitis and acupuncture and futhermore in clinic.
