        신경역동법과 역학적 인터페이스의 중재가 요통과 연관된 하지통 환자에게 미치는 영향

        Objective: The objective of this research is to conduct a 4 week study of low back related leg pain. Management of neurodynamic and mechanical interface, the results on the patients' pain and function, centralization of phenomenon. Method: This research was conducted on 3 low back related leg pain patients undergoing physiotherapy at W hospital from January, 2013 until March, 2013. In the evaluation, pain level with Numeric Pain Rating Scale(NPRS), pain location scale(PLS), korean oswestry disability index(KODI). All measurement of each subject were measure at pre-treatment and post-treatment(after 4weeks). The physical therapy program designed of SPSS 18.0 program was used to compile result. The difference between was compared pre-test and post-test using paired t test. Results: The pain and function, centralization of phenomenon were significantly difference between pre-treatment and post treatment(after 4weeks)(p<.05) Conclusion: This study suggest that management of mechanical interface with neural mobilization technique improved ROM, Disability and decrease pain score.

        재조합 lysozyme-HJP34 단백질의 다양한 병원성 세균에 대한 항 균 효능 및 자돈 사료첨가제의 가능성 평가

        The recombinant lysozyme-HJL34 proteins were expressed and purified using commercial Escherichia (E.) coli expression system. Stx2e<sup>+</sup> F18<sup>+</sup> E. coli, Actinobacillus pleuropneumoniae (APP), Streptococcus (S.) suis, and Clostridium (C.) perfringens strains were isolated from pigs. The minimum inhibitory concentrations (MICs) of the recombinant lysozyme-HJP34 proteins were examined by means of the microtiter plate method, according to the NCCLS recommendations. The possibility of its as the alternatives to antibiotics was tested in piglets. The MICs were determined as 75 μg/mL, 300 μg/mL, 75 μg/mL, 35.5 μg/m against Stx2e<sup>+</sup> F18<sup>+</sup> E. coli, APP, S. suis, C. perfringens, respectively. A total of 25 piglets were divied 5 groups. The piglets in group A∼C were fed with commercial feed and those in groups D, E were fed with commercial feedstuff. All piglets in groups B∼E were challenged with virulent Stx2e<sup>+</sup> F18<sup>+</sup> E. coli, APP, S. suis strains. Groups C and D were treated with antimicrobial from 24 h after challenge. All piglets in group B died within 3 days after challenge. Among 5 piglets in groups C and D piglets, 80% survived after challenge. Among group E piglets, 60% were alive until the end of this study. Therefore, this study indicates that recombinant lysozyme-HJP34 proteins is a suitable possibility as a feed additive for reduction of diseases by bacterial pathogens in piglet feed.

        Artificial Mutation for Silkworm Molecular Breeding Using Gene Scissors

        Clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR associated protein (Cas)9을 이용하는 유전자 가위 기술은 미래 육종 기술로서 주목받고 있다. 본 연구에서는 3세대 유전자 가위 CRISPR/Cas9을 이용한 누에 kynurenine 3-monooxygenase (KMO) 유전자 편집을 통한 돌연변이 유발 및 선택 교배를 통하여 유전자의 세대간 전달을 분석하고자 하였다. 유전자 편집을 위하여 누에의 KMO 유전자에 대한 3종의 가이드 RNA를 제작하고, 제작된 gRNA는 Cas9 단백질과 복합체를 형성시켜 누에 세포주(BM-N)에 도입 후 T7 endonucleaseI 분석을 수행하여 최적의 gRNA를 선발하였다. 선발된 K1N gRNA는 누에 유전자를 편집하기 위하여 Cas9 단백질과 복합체를 형성시킨 후 누에 초가 배아에 미세주사하고 사육하였다. 미세주사 후 부화율은 18% 가량으로 낮게 나타났으나 생존한 개체 중 돌연변이 발생율은 60% 이상으로 비교적 높게 나타났다. 돌연변이가 발생된 G0세대의 KMO 유전자는 이형접합자 형태로 나타났으며, 표현형의 변화는 관찰되지 않았다. 하지만 형접합자들 사이의 근친 교배에 의해 탄생한 G1세대 돌연변이에서는 일부에서 알과 눈의 색 변화가 확인되었으며, 변이가 확인된 개체들사이의 근친교배를 통해 생산된 G2세대에서는 모든 개체에서 표현형의 변화가 나타났다. 이러한 결과에 비추어 볼 때, 유전자 가위를 이용한 돌연변이 육종에는 한계가 있으나 전통 교배 육종과 융합을 통하여 육종 기간을 비약적으로 단축시킬 수 있는 곤충 육종 기술로 발전가능성이 높다고 판단된다. Gene editing technology using the clustered regularly interspaced short palindromic repeat (CRISPR) and the CRISPR associated protein (Cas)9 has been highly anticipated in developing breeding techniques. In this study, we discuss gene scissors as a tool for silkworm molecular breeding through analysis of Bombyx mori Kynurenine 3-Monooxygenase (BmKMO) gene editing using the CRISPR/Cas9 system and analysis of generational transmission through mutagenesis and selective crossing. The nucleotide sequence of the BmKMO gene was analyzed, and three guide RNAs (gRNAs) were prepared. Each synthesized gRNA was combined with Cas9 protein and then analyzed by T7 endonuclease I after introduction into the BM-N silkworm cell line. To edit the silkworm gene, K1P gRNA and Cas9 complexes were subsequently microinjected into the silkworm embryos; the hatching rate was 18% and the incidence of mutation was 60%. The gene mutation was verified in the heterozygous G0 generation, but no phenotypic change was observed. In homozygotes generated by self-crossing, a mutant phenotype was observed. These results suggest that silkworm molecular breeding using the CRISPR/ Cas9 system is possible and could be an effective way of shortening the time required.

        적백하오관중탕(赤白何烏寬中湯)의 기원(基源), 변천과정(變遷過程) 및 구성원리(構成原理)

        1. Objectives: This paper was written to understand the origin, changes and the constructive principles of Jeokbaekhaogwanjung-tang(Chibaihewu-tang; 赤白何烏寬中湯). 2. Methods: Jeokbaekhaogwanjung-tang and other related prescriptions were analyzed in terms of pathology, based on "Donguibogam(東醫寶鑑)", "Donguisusebowon Chobongwon(東醫壽世保元 草本卷)", "Donguisusebowon Gabobon(東醫壽世保元 甲午本)", "Donguisusebowon Sinchukbon(東醫壽世保元 辛丑本)" and "Donguisasangsinpyeon(東醫四象新編)" 3. Results: and Conclusions: 1) The origin of Jeokbaekhaogwanjung-tang, which inherited the spirit of Zhang, Zhongjing(張仲景)'s Sasim-tang(Xiexin-tang; 瀉心湯), is discovered in the prescriptions for Sun-qi(順氣), that is, Gwanjung-hwan(Kuanzhong-wan; 貫衆丸) and Mokhyangsungi-san(Muxiangshunqi-san; 木香順氣散). 2) The Jeokbaekhaogwanjung-tang was derived from Gangchulpajeok-tang(Jiangzhupoji-tang; 薑朮破積湯) of "Dongyisusebowon Gabobon", where the herbal medicines, Panax ginseng(人蔘) of Sasim-tang was replaced with Cynanchum wilfordii(白何首烏) and Allium sativum(獨頭蒜) was newly used too. Thereafter, Polygonum multiflorum(赤何首烏) and Alpinia oxyphylla(益智仁) were first added in Jeokbaekhaogwanjung-tang in "Sinchukbon". 3) The Jeokbaekhaogwanjung-tang, composed of 8 herbs except for Zizyphus jujuba(大棗), treats Taeumjeung(太陰證) of Soeumin(少陰人) through warming the Stomach(溫胃) of Cynanchum wilfordii, Polygonum multiflorum, Zingiber officinale(乾薑) and Alpinia officinarum(良薑) and downbearing the Yin(降陰) of Citrus reticulata(靑皮), Citrus unshiu(陳皮), Cyperus rotundus(香附子) and Alpinia oxyphylla.

        Cas9 단백질/ 가이드 RNA 복합체를 이용한 누에 BmBLOS 유전자 편집

        유전자 가위를 이용한 게놈편집 기술의 등장은 다양한 분야에서 분자육종에 대한 관심을 유발하였으며, 3세대 유전자가위 CRISPR 기술의 개발은 게놈편집을 통한 분자육종 시대를 가속화하고 있다. 본 연구에서는 최근 개발된 3세대 유전자 가위 CRISPR/Cas9을 이용하여 국내 보급품종인 백옥잠의 BmBLOS 유전자를 편집하여 돌연변이를 유도하고 유전형 및 표현형 검사를 통하여 유전자가위를 이용한 누에 분자육종가능성을 분석하고 이용기술을 확보하고자 하였다. 유전자 편집을 위하여 백옥잠의 BmBLOS 유전자의 염기서열을 구명하고, 이를 바탕으로 3종의 가이드 RNA를 합성하였다. 합성된 gRNA는 Cas9 단백질과 복합체를 형성시킨 후 BM-N 누에 세포주에 도입 후 T7 endonuclease I 분석을 통하여 편집효율이 가장 높은 B4N gRNA를 선발하였다. 누에 유전자를 편집하기 위하여 Cas9/B4N gRNA를 누에 초가 배아에 미세주사하고 사육하였다. 미세주사 후 부화율은 18% 가량으로 낮게 나타났으나 생존한 개체 중 돌연변이 발생율은 40% 이상으로 비교적 높게 나타났다. 또한 유전자 편집 G0세대누에 중 70% 가량에서 표현형의 변화가 관찰되었고, 염기서열 분석결과 대부분의 개체에서 BmBLOS 유전자가 정상과 돌연변이가 같이 존재하는 이형접합자 형태로 나타났으며, 그 유전형 또한 모든 개체에서 다르게 나타났다. 이러한 결과에 비추어 볼 때 CRISPR/Cas9 시스템을 이용한 누에 분자육종의 가능성은 매우 높을 것으로 예상되나, 유전자 편집효율을 개선하고 동형접합자를 얻기 위한 교배 및 선발방법에 대한 지속적인 연구가 필요하다고 판단된다. Genome editing technology employing gene scissors has generated interest in molecular breeding in various fields, and the development of the third-generation gene scissors of the clustered, regularly interspaced short palindromic repeat (CRISPR) system has accelerated the field of molecular breeding through genome editing. In this study, we analyzed the possibility of silkworm molecular breeding using gene scissors by genomic and phenotypic analysis after editing the biogenesis of lysosome-related organelles (BmBLOS) gene of Bakokjam using the CRISPR/Cas9 system. Three types of guide RNAs (gRNA) were synthesized based on the BmBLOS gene sequence of Bakokjam. Complexes of the prepared gRNA and Cas9 protein were formed and introduced into Bombyx mori BM-N cells by electroporation. Analysis of the gene editing efficiency by T7 endonuclease I analysis revealed that the B4N gRNA showed the best efficiency. The silkworm genome was edited by microinjecting the Cas9/B4N gRNA complex into silkworm early embryos and raising the silkworms after hatching. The hatching rate was as low as 18%, but the incidence of mutation was over 40%. In addition, phenotypic changes were observed in about 70% of the G0 generation silkworms. Sequence analysis showed that the BmBLOS gene appeared to be a heterozygote carrying the wild-type and mutation in most individuals, and the genotype of the BmBLOS gene was also different in all individuals. These results suggest that although the possibility of silkworm molecular breeding using the CRISPR/Cas9 system would be very high, continued research on breeding and screening methods will be necessary to improve gene editing efficiency and to obtain homozygotes.

        서사적 지식에 초점을 둔 초등학교 국어교과서 학습활동 지도 방안 - 5-6학년군을 중심으로 -

        이 글은 초등 서사교육에서 교육내용으로 작용하고 있는 학습활동들이 서사적 지식의 학습을 위해 정렬성 있게 설정되어 있는가를 분석하고, 그에 대한 대안활동과 지도방안을 제시하였다. 이를 위해 서사적 지식 관점에서 문학 영역의 성취기준 및 학습활동을 [5-6학년군]을 중심으로 분석하고, 서사적 지식의 균형적 학습을 위한 학습활동을 새롭게 구안하였다. 그런 다음 새롭게 구안된 학습활동의 지도를 위한 방안을 제시하였다. 이 글의 논의는 서사교육의 맥락과 실제, 서사교육의 지향점과 교육내용, 실제적 활동의 수행, 실제적 활동에서의 태도나 윤리, 맥락 등이 서사교육에서 학습활동으로 구현되어야 함을 강조하는 방향에서 이루어졌다. 아울러 서사교육에서의 학습활동이 교과서에 제시된 학습활동, 즉 기본학습 및 실천학습과 분리될 수 없음을 강조하는 차원에서 이루어졌다. 이를 통해 이 연구는 학교 현장의 서사교육의 지향을 새롭게 마련할 수 있었다. This article analyzes whether the learning activities that act as educational content in elementary narrative education are reasonably established for the learning of narrative knowledge, and suggests alternative activities and guidance measures. To this end, achievement standards and learning activities in the literary domain were analyzed from an epic knowledge perspective, and learning activities were newly devised for balanced learning of narrative knowledge. It then presented a way to guide the newly devised learning activities. The discussion in this article took into account the context and practice of narrative education, emphasizing that content related to academic understanding, the performance of practical activities, attitudes, ethics and contexts in narrative education should be implemented as learning activities. In addition, it was made to emphasize that learning activities in narrative education cannot be far from the learning activities presented in the textbook, namely basic learning and practical learning. Through this, the study was able to create a new orientation for narrative education at school sites.

        부자(附子)의 활용(活用)에 대한 고찰(考察)

        1. Objectives: After arranging the contents of the usages of Aconite mentioned in the literature - "Dongmu", a power trend me to further study the details of the usages of Aconite. 2 Methods: This study mainly refers to; "Dongmu-YuGo(東武遺稿)"(DYG), "Donguisusebowon-SaSangchobongyun(東醫壽世保元四象草本卷)"(DSS), "Donguisusebowon-GabObon(東醫壽世保元甲午本)"(DGO), "Donguisusebowon-ShinChukbon(東醫壽世保元辛丑本)"(DSC), and the research was written in order to find out the physiology and pathology of Soeumin in SCM. 3 Results and Conclusions 1) Taking aconite in a scale of 3 to 15 gram as common dosage, especially, when a person was critically ill, the dosage of 45 gram was used. Different dosages of aconite were chosen in accordance with some other different conditions, such as the purpose of reducing fever or mid-energizer warming, urinary volume, presence or absence of edema, stages of growth of diseases. In addition, we found a remedy using Remedy Single head garlic(獨頭蒜) instead of aconite. 2) Adverse toxic effects of aconite can be serious, it is dangerous to patients who are administered with aconite in massive amounts or take it in different concocting methods, therefore, further evidence about the laboratory experiments studied in animals and clinical dataare needed to confirm the scope for more widespread and accurate usages of Aconite.

        산란계에서 Chlorhexidine-inactivated Salmonella Enteritidis, S. Typhimurium 및 S. Gallinarum 3가 백신의 효능평가

        Protective efficacy of trivalent Salmonella inactivated vaccine containing Chlorhexidine-inactivated S. Enterltidis (SE), S. Typhimurium (ST), and S. Gallinarum (SG) strains, was evaluated in this study. A total of 70 brown nick layers were divided into 7 groups, A to G, containing 10 hens per group. All hens in groups B to D were intramuscularly immunized with approximately 7×10<sup>8</sup> cells (3×10<sup>8</sup> cells of SE+1×10<sup>8</sup> cells of ST+3×10<sup>8</sup> cells of SG) of the trivalent vaccine in 0.5 mL of PBS. All chickens in groups E to G were injected with sterile PBS. All hens of groups B and E, groups C and F, and groups D and G were orally challenged with approximately 2 ×10<sup>9</sup> CFU of wild-type SE, ST, and SG, respectively. Serum IgG titers and CD3<sup>+</sup>CD4<sup>+</sup> T-cells, and CD3<sup>+</sup>CD8<sup>+</sup> T-cells levels of groups B to D significantly higher than those of group A. In addition, all animals in groups A to C, E and F showed no clinical symptoms and survived after the virulent challenges, whereas one chicken in group D died and all chickens in group G died following the challenge. The protection against wild-type SE and ST in liver, spleen, cecum, and cloaca of groups B and C chickens was significant effective as compared with those in groups E and F. These indicate that the trivalent inactivated vaccine can be an effective tool for prevention of Salmonella infections by inducing robustly protective immune responses and cellular immune response in chickens.
