한국의 소동물 임상에서도 애완동물(개와 고앙이)의 수명이 길어짐에 따라 노인병(geriatric:노령성 질환)의 비중이 증가하고 있다. 이러한 노인병 중에 종앙에 대한 비중도 점차 증가하고 있다. 그러다 보니 종앙의 치료에 수의사뿐만 아니라 보호자의 관심도 집중되고 있다. 현재까지 가장 많이 사용하고 있는 악성 종앙의 치료방법은 수술적으로 완전 절제하는 것이다. WHO의 기준에 근거한 Stage가 높은 종양들은 완전절제가 불가능한 경우가 많다. 이런 경우, 화학요법제 또는 방사선치료를 병행하고 있으나, 완전 관해율(remission) 또는 부분 관해율이 낮은 편이다. 이외에 다양한 치료방법이 연구되고 있다. 유전자 치료도 그 중 하나이다.

BACKGROUND: Arsenic trioxide (As2O3) has been identified as an effective drug for the treatment of acute promyelocytic leukemia (APL). However, the role of As2O3 during the erythroid differentiation of human leukemic cells remains unknown. In this study, we investigated the in vitro effects of As2O3 on the erythroid differentiation of the K562 cell line and also on the expression and regulation of the apoptotic modulators of this process. 0aMETHODS: The K562 cells were cultured in the presence of 0.1, 0.5 and 1.0micrometer As2O3, or they were cultured in the presence of 1.0 and 10micrometer all trans retinoic acid (ATRA). The expression of glycophorin A before and after treatment with As2O3 or with ATRA in the K562 cells was assessed by flow cytometry and western blotting. The expressions of Bcl-2 and caspase-3 were determined by western blotting.0aRESULTS: The viability of the K562 cells was not decreased after treating with 0.1 and 0.5micrometer of As2O3, but the viability was significantly reduced at a dose of 1.0micrometer Caspase 3 activation was not observed at 0.1 and 0.5micrometer of As2O3 until 12 days, but Caspase 3 was activated by 1.0micrometer of As2O3 from day 3. The expression of glycophorin A was increased in dose dependent manner by As2O3 treatment, but this was not changed in the ATRA treated K562 cells. The expression of Bcl-2 was increased by 0.1 and 0.5micrometer of As2O3, but it was abruptly reduced by 1.0micrometer of As2O3. 0aCONCLUSION: These results suggest that As2O3 induces the erythroid differentiation of K562 cells and that 1.0micrometer of As2O3 induces apoptosis through the down-regulation of Bcl-2.
