This study examined the antioxidant and anti-inflammatory activity of Notoginseng Root (NGR) hot water extracts to develop a cosmetics material using natural plant resources. Antioxidant effects were evaluated based on DPPH and ABTS radical scavenging activity and total polyphenol contents. In addition, anti-inflammatory effects were measured using MTT assay, nitric oxide (NO) colorimetric assay, western blot analysis, enzyme-linked immunosorbent assay (ELISA) and real-time polymerase chain reaction (real-time PCR). The DPPH electron donating and ABTS radical scavenging activities increased significantly in a dose-dependent manner. The measured total polyphenol content of NGR hot water extracts was 2.8±0.1 mg/g. MTT assay showed no cytotoxicity at a low concentration. The inhibition rate of LPS(Lipopolysaccharide)-induced NO production was increased by NGR hot water extracts. NGR hot water extracts significantly inhibited the expression level of iNOS and TNF-α mRNA as measured by real-time PCR. Furthermore, western blot analysis revealed that the expression level of iNOS and TNF-α protein was reduced in LPS-induced RAW 264.7 cells by NGR treatment. Collectively, these results indicate that NGR hot water extracts has the high potential as a cosmetic ingredient of natural product.
