        pCAMBIA 1300 벡터를 이용한 일미벼의 형질전환 및 잎의 전자현미경적 관찰

        - The argEgene of E.coliwas introduced into ‘Ilmibyeo’cultivar of rice by Agrobacterium tumefaciensand alarge number of transgenic plants were produced. Embryogenic calli were co-cultivated with A. tumefaciensstrain AGL1carrying the plasmid pCAMBIA1300 containing hygromycin resistance (HygR). Transgenic plants showing in vitroresistance to 50 mg/L hygromycin were obtained using a selection procedure. Stable integration of argEand HPTgenesinto chromosomal DNA was proven by southern blot analysis and PCR analysis of genomic isolated from T0 progenies.The fragments of 650 bp (HPT) were detected in transgenic rice lines. The 230 bp (argE) fragments were showed inagarose gel, and detected fragments were matched with size of argEspecific primer. The microscopic feature of leaf onscanning electron microscope (SEM) revealed differences between clear and chalky in shape and arrangement of stomabut did not discriminate.Key words - argE, Hygromycin, Ilmibyeo, SEM

        큰방가지똥 추출물의 항당뇨 및 항고혈압효과

        In this study, we evaluated the bioactivities of methanol extract and its solvent fractions of Sonchus asper (L.) Hill. The EtOAc fraction of S. asper exhibited more strong antioxidant activity than other extracts as evidenced by the strongest 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity with a EC_50 value at 33.55 µg/mL and reducing power, the total polyphenol (180.71 mg GAE/g) and flavonoid contents (145.86 mg QE/g) of S. asper extract were higher than other extracts. The EtOAc fraction of the S. asper also showed 47.38% mushroom tyrosinase inhibition activity, 56.22% α-glucosidase inhibition and 46.58% α-amylase inhibition ratio at 1 mg/mL. Both methylene chloride and EtOAc fractions of methanol extract of S. asper effectively reduced of the 86.34% and 62.03% angiotensin I converting enzyme (ACE) activity at 2 mg/mL, respectively. These findings suggest that the EtOAc fraction of the S. asper could be a potential antioxidant in food additive, medicinal,and industry product.

        (-)-Catechin 및 배당체의 환원력 및 α-glucosidase저해 활성

        the EtOAc fraction of the MeOH extract of Ulmus davidiana, (-)-catechin (1), (-)-catechin-7-O-β-D-api-ofuranoside (2), and (-)-catechin-7-O-β-D- xylopyranoside (3) were isolated and characterized on the basis of 1H-and 13C-NMR,and FABMS spectral data.         민들레 추출물의 항산화 활성 및 세포독성 효과

        This study was investigated antioxidant and anticancer activity of water, methanol extract from upper and root part of Taraxacum mongolicum H. Total phenolic compound contents of methanol and water extracts from upper part were 51.95±0.18 mg/g and 48.16±0.89 mg/g respectively, and total flavonoid compound contents were estimated as 20.57±1.12 mg/ g in methanol extract and 6.55±1.20 mg/g in water extract. EC50 values for DPPH radical scavenging activity of methanol and water extract from upper part were 138.47±3.78 μg/mL and 204.38±5.32 μg/mL, and methanol and water extracts from root part were as 512±8.11 μg/mL in methanol extract and 1315.05±11.98μg/mL in water extract. Reducing power and hydroxyl radical (·OH) scavenging activity estimated that methanol extract of each part were higher than water extracts. The cell viability showed that the methanol extract from upper part had a cytotoxicity in the growth of colon carcinoma cell (44.58%). Both water extract (51.97±11.43%) from upper part and methanol (53.46±19.77%), water (52.79±13.53%) extracts from root part had quite higher cytotoxicity than that of methanol extract (88.25±2.02%) from upper part. Based on the results, It was suggested that the methanol extract of Taraxacum mongolicum H. were potential materials for use as functional food and medicine.

        추출용매에 따른 산초 (Zanthoxylum schnifolium)열매 추출물의 라디칼소거능과 항염증 효과

        This study was presented to the physiological activities such as metal chelating capacity, superoxide dismutase-like activity, anti-inflammation, cytotoxicity from Zanthoxylum schnifolium fruit. The values for metal chelating capacity of 1 mg/mL of 70% EtOH and 70% MeOH extracts were 68% and 67%, respectively, it was shown that metal chelating capacity in a dose-dependent manner. The 100% EtOH and 100% MeOH extract exhibited strong superoxide dismutase-like activity. The highest SOD-like activity obtained from 100% EtOH was 51% at a concentration of 4 mg/mL. The all extracts of Zanthoxylum schnifolium fruit except H2O extract significantly decreased the concentration of LPS-induced NO in Raw 264.7 cells. The 100% EtOH, 100% MeOH and 70% EtOH extract represented the highest activity in the anti-inflammation properties in vitro. However, the 100% EtOH and 100% MeOH extracts has cytotoxicity in Raw 264.7 cells and it may affect cell viability. Conclude that 70% EtOH extract was most suitable for anti-inflammation. Our results revealed that the Zanthoxylum schnifolium fruit expected to physiological activities.

        Different Solvent Fractions of Acanthopanax senticosus Harms Exert Antioxidant and Anti-Inflammatory Activities and Inhibit the Human Kv1.3 Channel

        The potential antioxidant and anti-inflammatory activities of Acanthopanax senticosus Harms were evaluated and its effect on the human Kv1.3 potassium channel was detected. The ethyl acetate fraction possessed the highest phenolic (289.19 ± 7.43 mg tannic acid equivalents/g) and flavonoid (10.80 ± 0.67 mg quercetin equivalents/g) contents and exhibited stronger antioxidant effects than other fractions in most of the antioxidant assays. On the other hand, the dichloromethane (CH2Cl2) fraction showed the strongest anti-inflammatory activity. The CH2Cl2 fraction inhibited the expression of inducible nitric oxide synthase, cyclooxygenase-2, tumor necrosis factor-a and interleukin 1β mRNAs, and the generation of reactive oxygen species in lipopolysaccharide-induced RAW 264.7 cells. Also, the peak current was inhibited 54.8% ± 17% by the CH2Cl2 fraction in voltage-clamp recording from Xenopus laevis oocytes. Our research demonstrated that fractions of A. senticosus have great potential to be a source of edible antioxidant and anti-inflammatory agents.

        광환경조절에 따른 멸종위기식물 섬시호의 생리적 반응

        This study was conducted to investigate the physiological responses of Bupleurum latissimum, endangeredplants by light condition. We investigated photosynthetic parameters, chlorophyll contents and chlorophyll fluorescenceunder different shading treatments (Shaded 50%, 70%, 90% and non-treated). Results showed that netapparent quantum yield (AQY) and chlorophyll contents were significantly increased with elevating shading level. However, light compensation point (LCP) and dark respiration (Rd) were shown the opposite trend. Especially, nontreatedexhibited photoinhibition such as reduction of chlorophyll contents and maximum photosynthesis rate (Pnmax)also variation trend of stomatal conductance (gs), and transpiration rate (E) were decreased to prevent water loss. Photosynthetic rate (PNmax) and photochemical efficiency (Fv/Fm) of 90% treatment showed a drastic reduction inJuly. This implies that photosynthetic activity will be sharply decreased with a long period of low light intensity. The50% treatment showed relatively higher photosynthetic activity than other treated. This result suggested that growthand physiology of B. latissimum adapted to 50% of full sunlight. 차광처리에 의한 광환경 변화가 섬시호의 생리적 반응에 미치는 영향을 조사하기 위해, 차광막을 이용하여 무차광처리구(0%), 50%, 70% 및 90% 차광처리구를 설치하고 광합성 관련 인자, 수분이용효율, 엽육세포내 CO2농도, 엽록소 형광반응 등을 조사하였다. 차광수준이 높아짐에 따라 낮은 광도에서 광합성을 수행하기 위해 암호흡의 저하로 인한 광보상점이 감소가나타났으며, 광합성의 효율을 높이기 위해 순양자 수율과 엽록소 함량의 증가가 나타났다. 무차광처리구에서는강한 광에 의한 수분손실을 막기 위해 기공전도도와 기공증산속도가 감소되었고, 엽육내의 CO2를 효율적으로광합성에 활용하지 못하는 것으로 나타났다. 또한 최대광합성속도가 감소하는 광저해현상이 나타났으며, 여기에너지의 전자전달이 원활하지 못하여 광으로 인한 피해가 예상된다. 90% 차광처리구 역시 점진적으로 광합성속도가 감소하여 7월에 가장 낮은 최대광합성속도를 보였는데, 이는 광선요구도보다 매우 적은 광 환경에서 계속 생장함으로서 광합성 능력이 저하되는 것으로 생각할수 있으며, 50% 차광처리구의 경우 광합성에 효율적인광환경이 제공되어 기공전도도와 기공증산속도가 증가하였고, 광화학반응 효율이 증가하는 등 광합성 활성이 높아진 것을 알 수 있어 생육에 보다 유리한 광 조건임을알 수 있었다.
