        Synovial sarcomas are malignant soft tissue neoplasms that develop from tendon and bursa near the large joints and frequently occur in the lower extremities of young male adults. They usually recur within 2 years after surgical treatment. The lung is a common metastatic site and rarely reported as the primary site of synovial sarcoma. The stomach is a very rare primary site of synovial sarcoma and recently only two cases of primary gastric synovial sarcoma were reported. We report a case of metastatic gastric synovial sarcoma in a 53-year-old male 4 years after surgical treatment of primary lung synovial sarcoma. The tumor had histologic, immunohistochemical and ultrastructural features of biphasic synovial sarcoma of the soft tissue. (Korean J Gastrointest Endosc 2003;27:536-540) 활막육종은 관절주위의 인대나 활액낭에서 발생하는 악성종양으로 젊은 남자의 사지에서 주로 발견되며 수술적 치료 후 2년 이내에 폐로 전이되거나 국소적으로 재발하는 것으로 알려져 있다. 드물게 두경부, 폐, 복벽, 소화관 등에서 발생하는 경우도 있으나 위에서 발생한 경우는 최근에 외국에서 2예가 보고되었을 뿐 국내에서는 아직 보고된 예가 없다. 저자들은 원발성 폐활막육종으로 수술 받은 53세 남자에서 4년 뒤에 전이성 위활막육종으로 재발한 경험을 하였기에 문헌 고찰과 함께 보고하는 바이다.

        연꽃 추출물이 활성산소종인 Glucose Oxidase로 손상된 배양 C6 Glioma 세포에 미치는 항산화 효과

        활성산소종(reactive oxygen species, ROS)인 glucose oxidase(GO)의 산화적 손상이 배양 C6glioma 세포에 미치는 영향 및 이에 대한 연꽃(Nelumbo nucifera GAERTN, NNG) 추출물의 항산화 효과를 세포생존율(cell viability)을 비롯한 DPPH-radical scavenging activity, lipid peroxidation에 의하여 분석하였다. 본 연구에서 H2O2는 배양 C6 glioma세포에 처리한 농도에 비례하여 유의한 세포생존율의 감소를 나타냈다. 한편, GO의 산화적 손상에 대한 NNG추출물의 방어효과를 조사하기 위하여 세포생존율을 조사한 결과 NNG 추출물은 GO의 산화적 손상으로 감소된 세포생존율을 유의하게 증가시켰다. 또한, NNG 추출물은 DPPH-radical scavenging activity의 분석에서 유의한 자유라디칼 소거능을 보였으며, 동시에 지질과산화에 대한 억제능을 보였다. 위의 결과로 부터 GO는 배양 C6 glioma세포에 고독성인 것으로 나타났으며 NNG추출물은 GO와 같은 ROS의 산화적 손상을 방어하는데 효과적인 것으로 나타났다. This study was aimed to clarify the cytotoxic effect of reactive oxygen species (ROS), glucose oxidase(GO) on the cultured C6 glioma cells, cell viability by XTT assay was determined by colorimetric assay and also, the protective effect of Nelumbo nucifera GAERTN (NNG) extract on cultured C6 glioma cells. C6 glioma cells were grown in the media containning 10∼40mU/mL concentrations of GO for 6 hours, and cell viability, 1,1-diphenyl-2-picrylhydrazyl(DPPH)-radical scavenging activity and also lipid peroxidation by ferric thiocyanate (FTC) assay was assessed on cultured in these cultures. In this study, GO remarkably decreased cell viability in a dose dependent manner. The XTT90 and XTT50 values were determined at 10mU/mL and 40mU/mL of GO by the treatment of C6 glioma cells with GO. In the protective effect of NNG extract on the cytotoxicity induced by GO, NNG extract significantly increased cell viability decreased by the cytotoxicity of GO. NNG extract also showed DPPH-radical scaveng activity and the diminutive activity of lipid peroxidation. From these results, it is suggested that GO showed highly toxic effect on cultured C6 glioma cells and NNG extract such as flavonoids showed the protection on the cytotoxicity induced by GO in these cultures.

        배양 SK-MEL-3세포에 있어서 로즈마리 추출물의 항산화능 및 멜라닌화에 대한 영향

        To evaluate the effect of Rosemary(Rosemarinus officinalis) extract on antioxidative activity and melanogenesis on cultured SK-MEL-3, the cell viability, DPPH-radical scavenging activity, superoxide dismutase(SOD)-like activity, tyrosinase activity and a total amount of melanin were measured. In this study, SK-MEL-3 cells were treated with the media containing 1∼20μM, hydrogen peroxide(H2O2) for 4 hours. H2O2 significantly decreased cell viability compared with control and XTT50 value was determined at 20μM of H2O2. In the antioxidative effect of Rosemary extract, it showed DPPH-radical scavenging activity and SOD-like activity. While, in the melanogenic effect of Rosemary extract, it significantly decreased the tyrosinase activity and a total amount of melanin which were increased by H2O2. From above the results, it is suggested that Rosemary extract was effective in the protection of H2O2-induced cytotoxicity by antioxidative activity and inhibition of melanogenesis.

        배양 인체피부섬유모세포에 있어서 알러지성 접촉피부염 유발제인 구리에 대한 쇠비름 추출물의 항산화 효과

        To elucidate the cytotoxicity and antioxidative effect of Portulaca oleracea L. (PO) extract on copper (Cu), an allergic contact dermatitis(ACD)-induced agent, cell viability were analysed by XTT assay after the treatment of cupric sulfate (CuSO4) to cultured human skin fibroblasts (Detroit 551). And also, the effects of vitamin E and PO extract on CuSO4- induced cytotoxicity were measured after Detroit 551 cells were pretreated with vitamin E or PO extract for 2 hours. In the antioxidative effects of PO extract were measured by superoxide dismutase (SOD)-like activity, the inhibitory activity of lipid peroxidation (LP) and lactate dehydrogenase (LDH) activity. In this study, CuSO4 significantly decreased cell viability in dose-dependently compared with control, and the XTT50 value was calculated at 59.8μM of CuSO4. While, vitamin E effectively prevented CuSO4-induced cytotoxicity and also, PO extract significantly increased cell viability compared with CuSO4-treated group. In the antioxidative effect, PO extract showed the inhibitory activity of LP and the decrease of LDH activity as well as SOD-like activity. From these results, it is suggested that the PO extract effectively prevented the cytotoxicity induced by an ACD-induced agent, CuSO4 via antioxidative effect. Conclusively, plant extract such as PO may be a putative resources as an alternative antioxidant for the prevention or diminution of ACD induced by allergen of heavy metals correlated with oxidative stress.

        환경오염원인 크롬의 세포독성에 대한 청미래덩굴추출물의 보호효과

        The purpose of this study is to evaluate the cytotoxicity of chromium and protective effect of Smilax china L.(SC) extract on glucose oxidase(GO)-induced cytotoxicity in the cultured C6 glioma cells. For this study, C6 glioma cells were treated in the media containing 15~30uM concentrations of chromium trioxide(CrO₃) for 72 hours, and cell viability and lactate dehydrogenase(LDH) activity with 1,1-diphenyl-2-picrylhydrazyl(DPPH)-radical scavenging activity were assessed on these cultures. In this study, CrO₃ decreased cell viability according to the treated dosage compared with control, and than the XTT₅₀ value was determined at 30uM CrO₃ in these cultures. In the protective effect of SC extract on CrO₃-induced cytotoxicity, both of an antioxidant, catalase and SC extract significantly increased cell viability which was decreased by CrO₃-induced cytotoxicity. SC extract also showed the antioxidative effect such as DPPH-radical scaveng activity and the decreased LDH activity. From these results, it is suggested that the cytotoxicity of chromium like CrO₃ is correlated with oxidative stress of reactive oxygen species(ROS), and also SC belong to Liliaceae showed the protective effect on the chromium-induced cytotoxicity by antioxidative effect in these cultures.

        Lavender 추출물이 t-BHP로 손상된 배양 인체피부 흑색종 세포의 세포증식과 멜라닌합성 및 tyrosinase 활성에 미치는 영향

        꿀풀과(Labiatae)의 일종인 잉글리쉬 라벤더(Lavendula angustifolia L.)추출물이 tert-butylhydro- peroxide (t-BHP)에 미치는 영향을 조사하기 위하여 인체피부흑색종세포(SK-MEL-3)를 배양한 후 t- BHP의 산화적 손상에 대한 영향을 세포증식율에 의하여 조사하였다. 또한 t-BHP의 멜라닌화(melanogenesis)에 대한 라벤더 추출물의 영향을 티로시나제 활성(tyrosinase activity) 및 멜라닌합성(melanin synthesis)에 대하여 분석하였다. 본 연구에서 t-BHP는 배양 SK-MEL-3세포에 처리한 농도에 따라 세포증식을 대조군에 비하여 유의하게 감소시켰다. 이 과정에서 t-BHP의 독성이 세포증식에 영향을 미치는 초기독성값(initial-cytotoxicity value )와 중간독성값(mid-cytotoxicity value)가 각각 4 uM과 30uM에서 나타났다. 한편, t-BHP에 대한 라벤더추출물은 t-BHP에 의해 감소된 세포증식을 유의하게 증가시켰으며, 동시에 티로시나제의 활성과 멜라닌합성을 유의하게 감소시켰다. 이상의 결과로 부터 라벤더추출물은 t-BHP에 대한 항산화효과와 멜라닌화를 방어하는데 효과적인 것으로 나타났다. To examine the cytotoxicity of tert-butylhydroperoxide(t-BHP) and the protective effect of english lavender (Lavendula angustifolia L.) extract on the cytotoxicity induced by t-BHP in cultured human skin melanoma cells(SK-MEL-3), the cell proliferation rate(CPR) and the melanogenesis such as tyrosinase activity and melanin synthesis were assessed using colorimetric assay. For this study, SK-MEL-3 cells were treated with the media containing 4∼30uM, gallic acid for 4 hours. t-BHP significantly inhibited cell proliferation dose-dependently. The initial and mid-cytotoxicity(MCT) values were determined at 4uM and 30uM of t-BHP, respectively. In the protective effect of Lavender extract on t-BHP-mediated cytotoxicity, lavender extract increased cell proliferation damaged by t-BHP-mediated cytotoxicity. In the melanogenesis, lavender decreased remarkably tyrosinase activity and melanin synthesis compared with t-BHP-treated group. From these results, it is suggested that t-BHP was highly toxic on cultured SK-MEL-3 cells and lavender extract was effective in the cell proliferation and the inhibition of melanogenesis induced by t-BHP.
