건설산업에서 안전사고 예방을 위하여 많은 연구들이 진행되어 왔으나 안전사고의 숫자는 줄지 않고 있다. 이에 최근 많은 연구에서 관리자나 작업자의 개인적인 특성과 행동에서 기인하는 원인들을 분석하고, 이를 제거하고자 하였다. 그러나 기존 연구의 노력에도 불구하고 관리자나 작업자는 안전사고의 발생이 서로에게 책임이 있다고 전가하는 인식이 있어 지속적으로 안전사고가 발생하고 있다. 이에 본 연구에서는 안전사고 예방을 위하여 관리자와 작업자의 책임전가의식을 분석하고, 책임전가의식에 영향을 미치는 영향요인들과의 상관성을 분석하였다. 본 연구에서는 실제 현장에 근무하고 있는 관리자와 작업자에게 설문조사를 실시하여 책임전가의식 수준을 정량적으로 제안하였다. 특히, 관리자는 작업자에게, 작업자는 관리자에게 책임을 전가하고자 하는 영향이 있음을 입증하였다. 추가적으로 책임전가의식과 이에 영향을 미치는 요인들(Job stress, Accident experience, Safe knowledge, Optimistic bias)과의 상관성이 있음을 입증하였다. 본 연구의 결과는 관리자와 작업자의 안전의식 향상과 건설안전분야의 연구적, 실무적인 측면에서 기여를 할 것으로 기대한다.
DNA 손상요인에 의한 CHO 세포의 apoptosis 와 DNA 회복

N/A The present study has been performed to elucidate apoptosis and DNA repair to DNA damaging agents in Chines hamster ovary (CHO) cells. Four assays were employed in this study ; DNA fragmentation by agarose gel electrophoresis, quantitative assay of fragmented DNA, morphological assessment of apoptotic cells and expression of heat shock protein (Hsp70) visualized by western blot analysis. DNA fragmentation in cells treated with ultraviolet radiation (UV), heat or actinomycin D (AMD) was induced at twelve, six or nine hour, respectively. The level of Hsp70 in cells pretreated with low dose of UV or heat and subsequently treated with high dos of UV of heat was higher than that treated with high dose of UV of heat. The level of Hsp70 in cells preheated for five minutes at 43℃ and subsequently treated with 5 ug/ml AMD was higher than that only treated with 5 ug/ml AMD. DNA fragmentation was delayed in cells pretreated with low dose of UV or heat. These results suggest that the increment of Hsp70 relates with a delay of DNA fragmentation associated with apoptosis and DNA repair such as adaptive response.
Effects of Inhibitors on the Expression of Poly(ADP-ribose)Polymerase in Apoptotic Mammalian Cells

The present study has been performed to elucidate the effects of inhibitors on the expression level of poly(ADP-ribose) polymerase (PARP) in apoptotic mammalian cells induced by ultraviolet radiation (UV). After treatment with 50 J/㎡ UV to HeLa S_3 and Chinese hamster ovary (CHO) cells, cells were incubated with two kinds of inhibitors, 3-aminobenzamide (3AB) and cycloheximide (CHX) for various times. The incidence of apoptosis was much lower in CHO than HeLa S_3 cells based on the percentages of apoptotic cells in terms of the morphological change of nucleus or direct counting of viable cells and the qualitative or quantitative DNA fragmentation. The percentages of apoptotic cells treated with UV and incubated with 3AB were lower than that without 3AB in HeLa S_3 cells. The levels of 116 kDa PARP in both HeLa S_3 and CHO cells treated with UV and then incubated with 3AB for 6 or 12 hrs were higher than that without 3AB. Thus, our data indicate that CHX does not affect UV-induced expression of PARP and DNA fragmentation. And 3AB affects UV-induced expression of intact PARP in HeLa S_3 as well as in CHO cells.
Chinese hamster 난소세포에서 DNA - 단백질 교차결합과 glutathione S - 전이효소의 발현에 대한 돌연변이원에 의해 유도된 교차적응반응

The present study has been undertaken to elucidate whether glutathione S-transferase (GST) is related to cross-adaptive response to DNA-protein crosslink (DPC) induced by ethyl methanesulfonate (EMS) or ultraviolet radiation (UV)-C in Chinese hamster ovary (CHO) cells. DPC was determined by SDS-K^+ precipitation method, and GST was measured by western blot analysis. 2,4-Dinitrophenol (DNP) that depletes cellular ATP and cycloheximide (CHM) that inhibits protein synthesis were used to find whether DNP and CHM affected the expression level of GST and cross-adaptive response to EMS or UV. There is a cross-adaptive response to DPC in cells pretreated with EMS or UV and subsequently treated with UV or EMS, respectively. In cells pretreated with 2 mM EMS and subsequently treated with 5J/㎡ UV, DNP and CHM did not affect cross-adaptive response to DPC and GST. And also, in cells pretreated with 1J/㎡ UV and subsequently treated with 8 mM EMS, CHM did not affect crass-adaptive response to DPC and the expression level of GST. But DNP increased the amount of DPC, and decreased the expression level of GST in UV-pretreated cells. These results suggest that GST are involved in the induction of cross-adaptive response to DPC in CHO cells.
출아효모에서 S기 checkpoint에 관여하는 DPB11에 관한 연구

The DPB11 gene, which genetically interacts with DNA polymerase II ($\varepsilon$) encoded by POL2 and DPB2, is required for DNA replication and the S phase checkpoint in Saccharomyces cerevusiae. The transcripts of DPB11 did not fluctuated as evidently as DPB2 and POL2 genes during cell cycle. To identify the physical interaction between Dpb2 and Dpb11, we examined the interaction by two-hybrid system. The interaction between Dpb2 and Dpb11 was detected in a two-hybrid assay. These results suggest that the amount of the Dpb2-Dpb11 complex may peak at the G1/S boundary. Therefore, we strongly suggest the involvement of the Dpb2-Dpb11 complex in a progression of DNA replication and S-phase checkpoint.

Purpose: Oral squamous carcinoma (OSCC) cells exhibit resistance to chemotherapeutic agent-mediated apoptosis in the late stage of malignancy. Increased levels of heat shock proteins 70 (HSP70) in cancer cells are known to confer resistance to apoptosis. Since recent advances in the understanding of bacterial toxins have produced new strategies for the treatment of cancers, we investigated the effect of Pseudomonas aeruginosa exotoxin A (PEA) on HSP70 expression and induction of apoptosis in chemoresistant OSCC cell line (YD-9). Materials and Methods:The apoptotic effect of PEA on chemoresistant YD-9 cells was confirmed by MTT,Hoechst and TUNEL stains, DNA electrophoresis, and Western blot analysis. Results: While YD-9 cells showed high resistance to chemotherapeutic agents such as etoposide and 5-fluorouraci (5-FU), HSP70 antisense oligonucelotides sensitized chemoresistant YD-9 cells to etoposide and 5-FU. On the other hand, PEA significantly decreased the viability of YD-9 cells by deteriorating the HSP70-relating protecting system through inhibition of HSP70 expression and inducing apoptosis in YD-9 cells. Apoptotic manifestations were evidenced by changes in nuclear morphology, generation of DNA fragmentation, and activation of caspases. While p53, p21, and E2F-1 were upregulated, cdk2 and cyclin B were downregulated by PEA treatment, suggesting that PEA caused cell cycle arrest at the G2/M checkpoint. Conclusion: Therefore, these results indicate that PEA reduced the chemoresistance through inhibition of HSP70 expression and also induced apoptosis in chemoresistant YD-9 cells.
