흰다리새우(Litopenaeus vannamei)에서 cephalexin의 잔류농도 연구를 위한 LC-MS/MS 분석법 연구

Cephalexin, a semi-synthetic cephalosporin antibiotic, has long been used in fish aquaculture in various countries under legal authorization. The drug is thus widely available for use in other aquatic species except fishes like the crustacean whiteleg shrimp. This study aims to develop a sensitive method for laboratory residue studies to adopt in withdrawal period determinations. Through repeated trials from the existing methods developed for other food animal tissues, it was possible to achieve a sensitive high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) method. The results showed that at a concentration of 0.1 mg/kg, the recovery rate was 81.79%, and C.V. value was 8.2%, which meet the recovery rate and C.V. recommended by Codex guideline. After satisfactory validation of analytical procedures, applicability to the shrimp tissue was confirmed in experimentally cephalexin-treated whiteleg shrimp. As a result, most muscle samples were detected below the limit of quantification (0.05 mg/kg) after day 3, and most hepatopancreas samples were detected below the limit of quantification after day 14. In particular, the limit of quantification 0.05 ppm with the presently developed method suggests sufficient sensitive over the current legal maximum residue limit of 0.2 mg/kg set for fishes.

Orthopedic rapid maxillary expansion(RME) has been a common treatment modality used to widen narrow maxillae in young children. However, since more skeletally matured adolescents or adults has closed midpalatal suture, the result of RME was undesirable because of dental tipping with little or no basal skeletal movement and resulted to many other complications. After such treatment, complications often occurred such as alveolar bending, compression of periodontal ligament, extrusion, buccal tipping, and severe relapse. Thus, surgically assisted rapid maxillary expansion(SA-RME) is required, especially for patients over 14 years old, to skeletally release maxillary expansion. We used two methods of maxillary expansion surgery. Surgically assisted rapid maxillary expansion(SA-RME) & surgically assisted posterior segmental expansion(SA-PSE) were used for narrow maxilla. The study was divided into two groups(SA-RME group and SA-PSE group). SA-RME group was consisted of 2 males and 4 females, and the ages of materials ranged from 15 years to 25 years with a mean of 20.2 years. SA-PSE group was consisted of 1 male and 5 females, and the ages ranged from 13 years to 23 years with a mean of 18.7 years. Dental study models were fabricated before starting the expansion and immediately after the expansion was completed. It was fabricated again 1 month later, 3 months later when the expansion device was removed, and 6 months later after the expansion was completed. A repeated measures analysis of variance(ANOVA) test was applied to assess changes between each groups over time. The amount of expansion and the amount of tipping movement each in both groups were compared by using paired t-test and it was also compared between each subjects within the group by using independent t-test. Both SA-RME and SA-PSE group showed stable results, but SA-PSE group showed statical significance in tipping movement of second premolar. We compared 6 patients who recieved SA-RME with 6 patients who received SA-PSE, and appraised the clinical usefulness.
김(Porphyra sp.)에서 추출한 PDRN (polydeoxyribonucleotide)의 나일틸라피아(Oreochromis niloticus)에서의 상처 치유 효과

Polydeoxyribonucleotide (PDRN)은 특정 adenosine 수용체에 작용하여 상처 치유 과정을 촉진하는 것으로 알려져 있다. 지금까지 어류의 조직에서 얻어진 PDRN과는 다르게, 최근 해초류인 Porphyra로부터 추출된 PDRN이 개발되었다. 본 연구는 나일틸라피아(Oreochromis niloticus)에서 인공적으로 피부의 상처를 유발한 뒤 Porphyra에서 추출한 PDRN을 이용하여 상처 치유 효과를 평가하였다. 상처의 회복은 시험 시작 후 1주내에는 일부만 회복되었지만, 2주내에 거의 완전히 회복되었다. 상처를 유발하기 전 PDRN을 10 및 20 mg/kg의 용량으로 5일간 사료로 투여한 후 상처 치유 효과를 관찰하였다. 상처 치유 효과는 육안적, 면적 및 색도 측정으로 평가하였다. 본 연구에서 관찰된 Porphyra 유래의 PDRN에 의한 상처 치유 효과는 다양한 원인에 의해 발생하는 어류의 상처 치유 목적으로의 활용 가능성을 보여준다. Polydeoxyribonucleotide (PDRN) is known to accelerate wound healing process by acting on specific adenosine receptors. Differently from the PDRN substances obtained so far from piscine tissues, unique PDRN was prepared recently from Porphyra, a seaweed species. In this study, effects of the Porphyra PDRN was evaluated by inducing artificial skin wounds in Nile tilapia Oreochromis niloticus. The wound recovered almost completely in 2 weeks without any treatment, however only a partial recovery was done after one week. Facilitated healing was observed by PDRN fed for 5 days before wound induction at 10 or 20 mg/kg, against the partially recovered 1-week old wound. The healing effects were identified not only by visual observation, wound area measurement, but also by colorimetric estimations. These novel wound healing effects suggest beneficial therapeutic utility of Porphyra-derived PDRN for wound treatments in fishes occurring from various causes.

Microchannels machining can be used to make micro molds for microfluidic chips. The fluid flow in the channel can be controlled, by changing the cross sectional shape of the channel. V-shaped channels with a specific angle are not easily made with the etching process. This study presents the mechanical machining of microchannels of V-shaped cross section, on cemented carbide (WC-Co). In this study, to reduce tool wear in the process of machining, the micro conical tool was fabricated using polycrystalline diamond (PCD). The tool wear of the conical tool and form accuracy of channels, were investigated during V-shaped microchannel machining.
디스크 공구를 이용한 초경합금의 미세 홈 가공에 대한 연구

Recently, the demand for micromachining of hard materials has been increasing. Machining microholes, grooves, and structures in hard materials such as tungsten carbide is very difficult. In this study, the machining characteristics of amicrodisk tool for microgroove machining of tungsten carbide were studied. Microtools made of polycrystalline diamond (PCD) were fabricated using wire electrical discharge grinding (WEDG) to machine high-hardness tungsten carbide. Rectangular and V-shaped disk tools were fabricated by WEDG with controlled wire paths. In the micro grooving of tungsten carbide, the effects of capacitance and feedrate on the surface roughness of microgrooves and the wear of disk tools were studied. As the capacitance and feed rate decreased, the surface roughness decreased and no significant wear was observed in the PCD tool. However, an increase in tool edge radius of several micrometers was observed.
