        pH 변화와 인산 제한이 Closoidium acetobutylicum의 연속발효에 미치는 영향

        Clostridium acetobutyloicum KCTC 1037의 용매 생성조건을 알기 위하여 pH변화와 인산제한의 조건 하에서 회분 배양과 연속 배양을 실시하였다. C. acetobut-ylicum KCTC 1037은 pH5.5나 6.5에서 보다 pH 4.5에서 잘 자라는 호산성이었으며, pH4.5의 진리되지 않은 부티르산이 약 20mM인 조건에서 용매 발효로 전환되었으나, pH5.5의 전리되지 않은 부티트산이 17mM인 조건에서는 용매 발효가 일어나지 않았다. 인산 제한 조건에서 배지에 인산이 KH$_2PO_4$나 $K_2HPO_4$0.1g/l이하인 경우 용매 생성량이 증가하였으나, 생육 속도에는 영향을 미치지 않았다. 인산제한 이단 연속배양에서 pH5.5인 stage1에서는 산생성기가 유지 되었고 한편 pH4.5인 stage2에서는 초기에는 용매가 생성 되었으나 150시간 이후에는 용매의 양이 감소되었으며 산생성기로 전환되는 등 불규칙한 발효 양상을 보였다. 또 pH5.0, 희석율 $0.05h^{-1}$인 stage2에서 전리되지 않은 유기산이 36mM인 조건에서도 용매 발효가 일어나지 않았다. 이러한 결과로서 C.acelobutylicumd의 용매 생성 조건은 산성 pH나 인산 제한 조건외에 균의 최대 에너지 생성을 유지하려는 대사와 관련 되었을 것으로 추측하였다. The growth and fermentation profiles of Clostridium acetobutylicum KCTC 1037 were examined in batch and continuous modes with pH variation and phosphate limitation. Clostridium acetobutylicum KCTC 10 37 grew better at pH 4.5 than at pH 5.5 or 6.5. Acetate and butyrate were produced at pH 5.5, whereas culture at pH 4.5 produced acetone and butanol. Solvent production was increased by the phosphate limitation in a batch culture, but in a phosphate-limited continuous culture for 400 hours steady-state solventogenesis was not observed. The induction and maintenance of solventogenesis presumably require not only acidic condition or phosphate limitation but also favourable bioenergetic condition.

        Effect of Euphorbia humifusa Willd extract on the amelioration of innate immune responses

        Euphorbia humifusa Willd is an annual plant widely used as a vegetable and traditional medicinal herb. E. humifusa Willd displays a variety of pharmacological actions; however, the effect of E. humifusa Willd on the innate immune response has not been well characterized. The aim of this study is to investigate the effect of an E. humifusa Willd methanolic extract (EuH) on the activation of innate immune cells. Here, we found that the EuH increased the expression of the innate cytokines, TNFa and IL-1b, in THP-1 monocytes and THP-1-derived macrophages. In addition, the EuH enhanced the phagocytosis of THP-1-derived macrophages. These findings suggest that E. humifusa Willd may be helpful for preventing bacterial infection and could be developed as a dietary supplement for ameliorating innate immune response.

        3-Hydroxyflavanone Induces Apoptosis in HeLa Cells

        Flavonoids and their derivatives exhibit many biological properties, including anti-inflammatory and antitumor activities. However, the antitumor action of 3'-hydroxyflavanone (3'-HF) is largely unknown. Antitumor efficacy of 3'-HF was assessed using cervical cancer (HeLa) cells. 3'-HF treatment resulted in a reduction in cell proliferation. A flow cytometric analysis demonstrated that 3'-HF deregulated cell cycle progression and triggered apoptosis. 3'-HF also increased the levels of p53 and p21, but decreased the level of cyclin D1. 3'-HF-induced apoptosis was accompanied by poly(ADP-ribose)polymerase cleavage. Together, these data indicate that 3'-HF possesses antitumor activity, which is mediated via the deregulation of cell cycle progression and induction of apoptosis.

        산성 발효 산물과 배양 pH가 Clostridium acetobutylicum의 유지 에너지에 미치는 영향

        In order to elucidate the acid tolerance mechanism of Clostridium acetobutylicum against organic acid, the maintenance energy with added butyrate at different pH was determined. Maintenance coeffecient in acidogenic chemostat was higher at pH 6.5 than at pH 5.5 showing that this organism is an acidophile. The addition of butyrate at pH 5.5 and different dilution rate caused linear decrease of the cell concentration though $Y_{ATP}$ did not decrease with increasing undissociated organic acid. $Y_{ATP}$ decreased by increasing the concentration of undissociated organic acid at pH 5.0 by the addition of butyrate. From these results it is hypothesized that the ATP conaumption for pH stat of acidophile C. acetobutylicum is increased at the circumstance with over 30mM of undissociated organic acid.cid.

        Clostridium acetobutylicum 무포자 변이주와 야생주의 Protonmotive force 와 용매 생산의 상관 관계

        Wild strain and asporogenous mutant of Clostridium acetobutylicum were used in a study to investigate the effect of organic acids on the maintenance of protonmotive force and on solventogenesis. In the presence of 50 mM acetate both of the strains maintained the protonmotive force of-174--232 mV and the internal pH of 5.46-6.52. The protonmotive force determination in the presence of 25 mM acetate and 25 mM butyrate showed that normal magnitude of protonmotive force was maintained by the acidogenic culture of the mutant and by the wild strain. On the other hand the solventogenic culture of mutant strain maintained the protonmotive force of -135--148 mV in the presence of 25 mM acetate and 25 mM butyrate. From these results it is concluded that the higher solventogenic ability of the mutant strain is due to the increased permeability of butyrate into the cell.

        일차산물 부티르산이 Clostridim acetobutylicum 의 Protonmotive force 에 미치는 영향

        In order to understand the acid tolerance mechanism in Clostridium acetobutylicum against the undissociated weak acids, a two stage continuous culture of C. acetobutylicum KCTC 1037 was made to supply acidogenic and solventogenic phase culture for a study on the protonmotive force and fermentation profile in the presence of butyrate, the acidic fermentation product. Undissociated butyric acid was permeable to the membrane of acidogenic cell, while the influx of the acid was minimal into the solventogenic cell. Acidogenic cell had higher pH gradient and protonmotive force than solventogenic cell. The protonmotive forces decreased linearly with the increase in butyrate from 25 to 75 mM. Acidogenic culture maintained the internal pH of 5.6-6.2 in the presence of 75 mM butyrate, and the internal pH of the solventogenic cultures was 5.3-5.7 in the similar conditions. Though the acidogenic culture was found to maintain the protonmotive force in the presence of 75 mM butyrate in the short term experiment, a long term incubation showed that the acidogenic culture was unable to ferment sugar in the similar conditons. On the other hand the solventogenic culture maintained active protonmotive force and fermented sugar in the presence of 75 mM butyrate. The protonmotive force of acidogenic culture was dissipated by 75 mM butyrate in the presence of ATPase inhibitor, dicyclohexylcarbodiimide, whilst the protonmotive force of solventogenic culture was maintained a similar condition. From these results, it is concluded that the acidification of cytoplasm by butyric acid is prevented by proton pumping action of the membrane bound ATPase which maintains the protonmotive force for a short-term period in the acidogenic cultures, and that solventogenic culture possess the ability to reduce the influx of organic acid in long term fermentation. It is suggested that C. acetobutylicum maintains the protonmotive force by the investment of ATP during the fast-fermenting acidogenesis; on the other hand, the solventogenic culture can reduce the influx of the acidic fermentation products to maintain the protonmotive force.

        Implication of Egr-1 in trifluoperazine-induced growth inhibition in human U87MG glioma cells

        The early growth response gene-1 (Egr-1) is a tumor suppressor which plays an important role in cel growth, diferentiation and apoptosis. Egr-1 has been shown to be down-regulated in many types of tumor tisues. Trifluoperazine (TFP), a phenothiazine class of antipsychotics, restored se-rum-induced Egr-1 expression in several cancer cell lines. We investigated the effect of Egr-1 expression on the TFP-induced inhibition of cell growth. Ectopic expression of Egr-1 enhanced the TFP-induced antiproliferative activity and down-regulated cyclin D1 level in U87MG glioma cells. Our results sugest that antipsychotics TFP exhi-bits antiproliferative activity through up-regulation of Egr-1.

        심한 구역반사를 가진 성인환자에서 프로포폴 목표농도 주입법으로 시행한 깊은 진정

        The gag reflex is a physiological reaction, but, an exaggerated gag reflex can be a severe limitation not only to treat dental caries but also to do oral exam. Procedures such as surface anesthesia of the palate and pharyngeral area, sedation, or general anesthesia can be options as behavioral management. But, there are no golden rule for the sever gag reflex patients. We present a case report of propofol intravenous sedation using TCI pump for simple dental treatment. A 44-year-old man, who had past history of general anesthesia for dental treatment because of severe gag reflex, was scheduled intravenous sedation for simple dental treatment. After 8 hour fasting he entered the clinic for persons with disabilities. We explained about intravenous deep sedation and got informed consent. First, we kept intravenous catheter (22G) in the arm and started monitoring ECG, non-invasive blood pressure, pulse oximetry and end-tidal $CO_2$ through nasal cannula. We started propofol infusion with TCI pump at the target concentration of 3 mcg/ml. The patient became sedated, but he showed involuntary movement during dental treatment, so we increased the target concentration to 4 mcg/ml. We finished the dental treatment without complications during 30 min. And after 40 min recovery room stay he was discharged without any complications.
