        Bacillus subtilis subsp. krictiensis로부터 항진균물질 KRF-001의 생산을 위한 발효조건 및 돌연변이 연구

        토양세균 B. subtilis subsp. krictiensis로부터 항진균활성물질 KRF-001 복합체를 생산하기 위한 발효조건을 조사하였다. Whey와 mannitol 등이 탄소원으로, CSL, corn gluten meal 그리고 polupeptone이 복합질소원으로 좋았다. 중온균인 생산균은 중성 pH 조건에서 P. oryzae에 대한 bioactivity가 가장 높았으며, 0∼10% DO의 낮은 용존산소 조건에서도 bioactivity가 유지되었다. 인산염의 농도가 K_2HPO_4, 0.047%에서 0.097%로 높아짐에 따라 1/2의 bioactivity 감소가 관찰되었다. 한편 UV 및 MNNG에 의한 돌연변이에 의해 우수균주 3개를 선별하였다. Antifungal compound, KRF-001, was produced by Bacillus subtilis subsp. krictiensis isolated from soil. Physico-chemical factors affecting cell growth and bioactivity were examined to improve the production yield. Nutrient composition, temperature, pH and phosphate ion concentration were proved to be important factors for the production of KRF-001. Mutation was performed to select high yielding strains. First, mutation was performed with ultra-violet light, and the second mutation process was conducted by MNNG(N-Methyl-N'-nitro-N-nitroso-guanidine) resulting in three high yielding strains.

        변수 분석을 통한 아토카푸스 니티두스 추출물과 분획물의 항산화, 타이로시나제 및 콜라제나제 In Vitro 저해활성 연구

        본 연구에서는 아토카푸스 니티두스의 용매추출물 및 실리카 컬럼 분획물의 항산화 활성, 콜라제나제 저해활성 및 미백활성을 평가하였다. In vitro 실험의 활성 분석은 sigmoid 곡선형 반응에 적합한 4 매개변수회귀곡선 분석(4 parameter logistic, 4PL)을 활용하여 정량화 하였다. HPLC와 LC/MS 분석에 따르면 아토카푸스 분획물은 폴리페놀류가 주성분이었으며, 추출물의 총 폴리페놀의 함량은 갈릭산 기준 48.1 ± 2.6 mg GAE/g 이었다. 1,1-Diphenyl-2-picrylhydrazyl (DPPH) 라디칼 소거활성을 평가한 결과, 용매추출물, 분획물-1과분획물-2는 각각 16.7, 42.0 및 10.1 μg/mL의 농도에서 50%의 라디칼 소거활성(SC<sub>50</sub>)을 보였고, 분획물-2는양성 대조구 ascorbic acid (1.5 μg/mL)에 가장 근접한 활성을 보였다. Tyrosinase 저해활성은 추출물과 분획물 2종이 각각 64.9, 0.9 및 1.2 μg/mL의 IC<sub>50</sub>를 보여 대조구인 코직산(7.4 μg/mL) 및 알부틴(119.0 μg/mL) 대비 전반적으로 높은 활성을 보였다. 제브라피쉬 배아를 이용한 색소침착도 억제활성에서는 분획물-2(27.5%)가 대조구인 코직산(18.6%) 대비 유의하게 미백활성 결과를 보였으며 동시에 500 μg/mL까지 배아 생장에 영향이 없음을 확인하였다. 아토카푸스 추출물과 분획물-1 및 분획물-2는 콜라제나제 저해활성에서 각각 139.8, 20.6, 및 16.8 μg/mL의 IC<sub>50</sub>를 보여, 대조구인 1, 10-Phenanthroline (55.4 μg/mL) 대비 우수한 활성을 보였다. 추출물과 분획물-2에 대한 엘라스타제 저해활성은 각각 61.8과 67.1 μg/mL으로 암라 및 사포타 추출물과 유사한 활성 범위를 보여주었다. 이상의 결과는 아토카푸스 추출물이 항산화, 미백 및 피부노화 방지에 유용한 폴리페놀계 화장품소재로 활용이 가능하며, 추출물-2는 고농도에서도 안전함을 시사한다. In this study, the antioxidative and inhibitory activity of tyrosinase and collagenase for the solvent extract and silica column fractions of Artocarpus nitidus were evaluated. The activities were quantified using the 4 parameter logistic. LC/MS analysis showed that the major component of the fractions was polyphenol and the total polyphenol content of the extract was 48.1 ± 2.6 mg GAE/g. The radical scavenging activities (SC<sub>50</sub>) for 1,1-diphenyl-2-picrylhydrazyl of the extract, fraction-1 and fraction-2 were 16.7, 42.0 and 10.1 μg/mL, respectively. The value for fraction-2 was the closest to ascorbic acid (1.5 μg/mL). The tyrosinase inhibitory activity of the extracts and the fractions showed IC<sub>50</sub> of 64.9, 0.9 and 1.2 μg/mL, respectively, and overall activity was higher than that of kojic acid (7.4 μg/mL) and arbutin (119.0 μg/mL). In the experiment by zebrafish embryo, the whitening activity of fraction-2 (27.5%) was higher than that of kojic acid (18.6%), and there was no adverse effect up to 500 μg/mL of fraction-2. For the collagenase inhibitory activity, the samples showed IC<sub>50</sub> of 139.8, 20.6, and 16.8 μg/mL, respectively, which were competitive to 1, 10-Phenanthroline (55.4 μg/mL). The extract and fraction-2 showed IC<sub>50</sub> of 61.8 and 67.1 μg/mL for elastase. These results suggest that A. nitidus extract can be used as a cosmetic material useful for antioxidant, whitening, and prevention of skin aging without adverse effects.

        A skill is defined as the special ability to do something well, especially as acquired by learning and practice. To learn a skill, a Bayesian network model for representing the skill is first learned. We will regard the Bayesian network for a skill as an affordance. We propose a soft Behavior Motivation (BM)switch as a method for ordering affordances to accomplish a task. Then, a skill is constructed as a combination of an affordance and a soft BM switch. To demonstrate the validity of our proposed method, some experiments were performed with GENIBO (Pet robot) performing a task using skills of Search-a-target-object, Approach-a-target-object, Push-up-in front of - a-target-object.

        고지탈묵용 Cellulase 및 Xylanase 생산

        효소탈묵용 cellulase와 xylanase 생산을 위한 조건을 조사한 결과 배지초기 pH, Tween 80, 포자접종량, 질소원의 종류 및 탄소원의 종류에 따라 효소 생산량이 크게 변하였다. Cellulase 생산을 위한 최적조건으로는 pH 5.O∼6.5, Tween 80 0.02%, 포자현탄액(1×10^7㎖) 접종량 0.5∼l.0%, 질소원으로 면실박, 탄소원으로 옥분을 사용하였을 때 가장 효과적이었다. 또한, xylanase 생산을 위한 최적조건으로는 pH 6.5, Tween 80 0.Ol%, 질소원으로 corn steep liquor, 탄소원으로는 신문고지를 사용하였을 때 효과적이었으며, 이때 포자현탁액의 접종량은 cellulase 생산의 경우와 동일하였다. 한편, cellulase와 xylanase의 동시생산을 위해서는 플라스크 규모에서 질소원으로 면실박 0.5%, 탄소원으로 신문고지 1.0%와 옥분 2.0%를 첨가하였을 때 효과적이었으며, 이때 생산된 배양액의 cellulase의 활성은 6.11∼7.22IU/㎖, xylanase의 활성은 97.7IU/㎖이었다. 그러나 fermentor 규모에서는 동일 배지조성에서 효소생산이 다소 감소하였으며, 특히 xylanase 경우 배양액의 효소활성이 낮아서 이에 대한 보완이 요구된다. The optimal conditions for cellulase and xylanase production by Trichoderma reesei 28217 were studied for enzymatic deinking of old newspaper. The amounts of cellulase and xylanase from the strain was varied by initial medium pH, Tween 80, inoculum size of spore suspension, and carbon and nitrogen sources. The optimal conditions for cellulase production were pH 5.O∼6.5, 0.02% of Tween 80, 0.5∼1.O% of inoculum size of spore suspension (1×10^7/㎖), cotton-seed meal as nitrogen source, and corn flour as carbon source. On the other hand, the optimal conditions for xylanase production were pH 6.5, 0.01% of Tween 80, corn steep liquor as nitrogen source, and disintegrated old newspaper as carbon source. The inoculum size for xylanase production was the same as for cellulase production. The concomitant production of cellulase and xylanase in shake flask culture was efficiently induced in the medium containing 0.5% cottonseed meal as nitrogen source and 1.O% old newspaper and 2.O% corn flour as carbon sources. In this case the activities of cellulase and xylanase produced were 6.11∼7.22 IU/㎖ and 97.7 IU/㎖, respectively. However, the cellulase production in 5ℓ fermentor scale was slightly decreased compared with that in flask scale. Moreover, xylanase production was severely reduced in a fermentor scale. The study for the reason of decreased enzyme production in fermentor is further needed.

        Statistical optimization of culture medium for improved production of antimicrobial compound by Streptomyces rimosus AG-P1441

        The nutritional requirements for antimicrobial activity of Streptomyces rimosus AG-P1441 were optimized using statistically-based experimental designs at a flask level. Based on a one-factor-at-a-time (OFAT) approach, glucose, corn starch and soybean meal were identified as the carbon and nitrogen sources having a significant effect on antimicrobial productivity. As a result of investigating the effect of glucose concentration, the highest antimicrobial activity was observed at 3% concentration. Response surface methodology (RSM) was then applied to optimize the growth medium components (corn starch, soybean meal, MgCl2 and glutamate). Antimicrobial productivity increased sharply when the medium consisted of 3% glucose, 3.5% corn starch, 2.5% soybean meal, 1.2 mM MgCl2 and 5.9 mM glutamate. The fermentation using optimized culture medium in a 5-L bioreactor allowed a significant increase in antimicrobial activity, evaluated by the paper disc assay, revealed a 29 mm inhibition zone diameter against Phytophthora capsici.

        Purpose: The purpose of this study was functional leg length inequality effect on COP(Center Of Pressure) and LOS(limits Of Stability) and EMG activation. Methods: The participants were consisted of fourteen. Subjects were distributed 2 groups; control group, leg length inequality ${\leq}3mm$, n=8), experimental group(leg length inequality${\geq}10mm$, n=8). The participants were measured leg length wearing comfortable clothes through tape measure method(TMM). All subjects was measured COP(Center Of Pressure), LOS(limits of stability) using by Balance Trainer BT4(HUR, Finland). Results: The results COP was not exist statistical significant differences(p>0.05). LOS was not exist statistical significant difference(p>0.05). Conclusion: The results was not statistical significant differences COP and LOS depending on Leg Length Inequality. But between experimental group and comparison group was exist mean differences on COP, LOS(COP:Ex.>Com, LOS:Ex. <Com/excepted some data). Somewhat COP and LOS was affected on by Leg Length Inequality. Balance is affected on Leg Length Inequality partly.

        Cultivation Optimization of Insect-Pathogenic Fungi Paecilomyces lilacinus HY-4 to Soil-pest Adoretus tenuimaculatus

        A new insect pathogenic fungi, Paecilomyces licacinus HY-4 that was isolated from soil sample, showed biological activity to Chestnut Brown Chafer, pest of golf course. The culture condition for the fungi P. lilacinus HY-4 was optimized through statistical approach for the mass production. According th the bioassay on the pest, the insecticidal activity reached up to 30%, and positive factor for the production of the fungi was zeolite with 97.5% of signi.cance.

        상당수의 중소기업은 자체 마케팅 역량이 부족하여 제품 및 기술 개발에 있어서 초기부터 마케팅 활동을 충분히 수행하고 있지 못한 경우가 많다. 본 논문은 이러한 문제를 해결하기 위해 원료 공급에서부터 가공, 아웃소싱, 판매, 협업에 이르는 제품 생산 활동의 각 단계마다 적절한 마케팅 지향 정보가 제공되도록 공공 서비스 제공 전략을 제시한다. 또한 제품 정보, 시장 정보, 기술 정보로 구성된 마케팅 지향형 정보 제공 시스템의 프로토타입을 제시한다. Many SME have difficulties in marketing activities from early product and technology development because of lack of internal marketing abilities. This paper presents the strategy for the public information service providing so that the marketing oriented information system should provide appropriate information in every step of production from raw material procurement, processing, outsourcing, sales to collaboration. A prototype of marketing information system is implemented in terms of product information, market information, technology information and enterprise information.
