        Agrobacterium을 이용한 고추의 Transient Expression 시스템

        Agrobacterium을 이용한 GUS 유전자를 효과적으로 발현시키기 위하여 수행되어진 실험 결과를 요약하면 다음과 같다. 박테이라 농도별 실험을 수행한 결과 균의 전처리 배양 농도 O $D_{600nm}$ 0.3일때 원심분리한 후 얻어진 균을 희석한 후의 최종 농도는 O $D_{600nm}$ 0.8로 맞춘 실험 처리구에서 GUS 유전자 발현율이55%로 가장 높게 나타났다. 병원성 유도 배지 내에 Acetosyringone (AS)이 첨가되지 않은 경우 GUS 유전자가 발현된 고추 잎을 얻을 수 없었으나, 200$\mu$M을 첨가했을 때 90%의 가장 높은 GUS 유전자 발현율을 나타내어 많은 수의 GUS spots을 관찰할 수 있었다. Agrobacterium에 의한 고추 잎의 감염 정도를 조사한 바 Agrobacterium으로 감염시킨지 3일째부터는 박테리아 에한 감염 정도가 심해져서 GUS 유전자 발현 정도가 약해지므로 Agrobacterium으로 감염시킨지 2일째 되었을 때 GUS 유전자 발현이 가장 강하게 나타난 것을 확인하였다. 이 같은 결과는 박테리아에 의한 감염이 일어난지 3일째 부터는 식물체의 감염부위 고사를 일으키는 것과 관련된 것으로 보인다. We established a transient gene expression system in chili pepper leaves based on Agrobacterium-mediated transformation of GUS gene. For the best GUS transient expression, two step culture system was adopted. When the Agrobacterium tumefaciens cell density of pre-culture was $A_{600nm}$ 0.3, the cells were harvested and diluted to $A_{600nm}$ 0.8 with virulence induction medium after cell harvested. The addition of acetosyringone (200 $\mu$M) in virulence induction step was a key factor for successful transient expression. Additionally, Younger leaves showed more effective transient expression than older leaves. Temporally, the strongest intensity of GUS expression was detected at 2 days after infiltration. These results demonstrate that Agrobacterium-mediated transient expression can be used for a simple in vivo assays of plant promoters, transcription factors and furthermore provide efficient protocol for chili pepper transformation.

        중국과 일본 들깨 수집 자원의 생육 특성 및 페놀 성분 분석

        This study was conducted to find out the variation in agronomic trait and chemical composition in the collected Perilla frutescens from China and Japan. From the results of growth investigation, the maximum number if branches was 26.7ea in Japan 134 line, followed by 25 nodes number in China 119 line. Among the different lines investigated, maximum number of panicle number (108.8) were observed in China 114 line. 1000 seed weight was maximum (4.12 g) in China 118 line. Flowering time of different collected lines varied significantly with average value of 175.5 days and the average line required for maturation of seedlings was 205.1 days. Plant height was the highest (248.9 ㎝) in China 107 line. Highest number of total picking leaves was 965ea, and the average picked period was 54 days. The major phenol compounds contained in Perilla frutescens showed wide variation for Syringic acid, Benzoic acid, Naringin, o-Coumaric acid, Myricetin, Naringenin and Hesperetin. Japan 139 line showed the highest level of total phenol contents (8254.0 ㎍/g, dry weight).

        Analysis of genetic diversity in the bioenergy crop Miscanthus via ISSR and RAPD markers

        Analysis of the genetic diversity of Miscanthus, a representative crop associated with bioenergy, is necessary for the enhancement of Miscanthus biomass production, an important and challenging research prospect. This study was conducted to determine the genetic diversity of Miscanthus accessions, with regard to geographical location. Inter-simple sequence repeat (ISSR) and random amplifed polymorphic DNA (RAPD) markers were used to assess the relationships among, and genetic diversity of, 24 Miscanthus accessions collected from the USA and China. These 24 Miscanthus accessions were all shown to be diploid by fow cytometry. Using 30 selected ISSR primers, a total of 96 amplifed products were produced, with a mean of 11 products per primer, and the polymorphism percentage was 70%. Using 11 primers, the RAPD method produced 194 amplifed products, and the polymorphism percentage was 83% in all accessions. The genetic distance was 0.36–0.71, according to Jaccard’s coefcient for combined ISSR and RAPD data. The 24 Miscanthus accessions were classifed into two groups (USA and China) through UPGMA clustering analysis. This classifcation revealed signifcant genetic diversity among Miscanthus accessions according to geographical location. Future eforts to analyze genetic diversity will facilitate the improvement of biomass characteristics in other grasses undergoing development as energy crops.

      • 허브 하이드로졸 용액의 식물 생장 효과

        Background : Natural substance extracted from plant is going increased to apply for agricultural industry to improve crop product. Thus, hydrosols of lavender used to cosmetic materials was treated in plant to investigate growth effect. Methods and Results : The young plants grown for 7 days from seed were transferred to pot. They have foliage dressed with lavender hydrosols diluted to 500 × at intervals of 2 days. The results of N. benthamiana grown by foliage dressing for 2 weeks were 11.37 ± 0.31 ㎝ of leaves length, 11.07 ± 0.47 ㎝ of leaves width, 14.43 ± 0.90 ㎝ of plant length. These values were investigated to more increase than control. In the results measuring plant length at 4 weeks, there had no difference in two treatments. The activities of DPPH scavenging and SOD were checked to examine the antioxidant activity in two treatments. The DPPH scavenging activity was not shown a difference in two treatments. However, SOD activity displayed at high to level of 2 times in extracts from 100% ethanol of N. benthamiana treated with lavender hydrosols in compared to control. The RT-PCR preceeded to investigate the transcriptional changes of genes related to antioxidant activity using the respective primers for 5 genes such as NbNOA1, NbNR, NbPAL, NbAPX, NbSOD. These 5 genes were known to detect as a high level in N. benthamiana treated with lavender hydrosols in transcriptionl expression. Conclusion : The influence on plant growth through lavender hydrosol treatment was represented as a promoting effect in early growth stage.

        바이오에너지 자원으로서 강원도 지역에서 수집된 수수의 작물학적 특성 평가

        1. 수수 성숙기에 간장 길이를 조사한 결과 6개체(12.2%) 가 300 cm 이상의 길이로 바이오매스가 우량한 것으로 파악되었으며, 10개체(20.4%)정도는 200~300 cm 사이의 평균 길이를 나타내었다. 2. 성숙기의 수수 줄기 직경에는 큰 차이를 보여준 개체는 없었으며, 대부분의 수수 수집종들은 2.2±0.1 cm 정도의 직경을 갖는 것으로 드러났다. 3. 본 연구에 시험한 수수 수집종들은 엽장이 60~70 cm 정도의 길이에 속하는 유전자원이 22 개체가 되는 것으로 나타났으며, 7개체는 70 cm 이상되는 것으로 나타났다. 4. 엽형은 49% 정도의 개체가 협단으로 분류되었고, 대부분의 엽맥색은 백색으로 나타났다. 5. 본 연구에 사용된 대부분의 수수의 수형은 반밀수 타원형이 많은 것으로 나타났으며, 수장은 평균적으로 20~30 cm에 달하는 것으로 나타났다. Forty nine accessions of the Sorghum bicolor (L) Moench collected from different agronomical zone of Gangwondo province, Korea were characterized morphologically on field trial. Plant length, stem diameter, tiller number, ear type, ear length, ear width, leaves length, leaves width and leaf color differed significantly in the cultivated accessions. Out of forty nine test accessions, six accessions (12.2%) showed comparatively taller height, measuring more than 300 cm, while, other ten test accessions (20.4%) were measured from 200~300 cm. There were no significant differences in the diameter of stem in the collected test accessions (2~3 cm). Of the total test accession, seven accessions showed leaf length of over 70 cm, and twenty two test accession revealed leaf length between 60-70 cm. Forty nine of the collected test accessions showed narrow and short leaves with white mid vain. These results provided additional information for the breeding programs and conservation of Sorghum bicolor as a bio-energy crop.

        The Mediating Effect of Job Esteem on The Relationship Between Customer Misbehavior Experienced by Beauty Workers And Their Job Continuity

        This study aims to verify the mediating effect of job esteem on the relationship between customer delinquent behavior experienced by beauty workers, who are subjected to longer and more frequent face-to-face interactions with clients compared to workers in other industries, and their job continuity. Correspondingly, this study surveyed 496 individuals between the ages of 20 and 50, who are currently working in the beauty service industry where building and maintaining customer relationships is crucial in an attempt to confirm whether cultivating job satisfaction and a sense of self-worth has a direct influence on job continuity. As a result of the analysis, customer misbehavior was found to have significant and negative impact on job esteem. The frequency of such experiences is inversely related to the level of job esteem; the more incidents of customer misbehavior, the lower the job esteem of the workers. On the other hand, job esteem was found to have significant and positive effects on job continuity, indicating that the job continuity of beauty workers would decrease due to the negative impact of customer delinquent behavior. However, this study revealed that experiencing customer misbehavior does not have directly impact on the job continuity of beauty workers. Instead, customer delinquent behavior was observed to have a less direct effect on job continuity compared to the influence of job esteem on job continuity, implying that the decrease in job esteem resulting from customer misbehavior affects job continuity. Thus, in order to improve job continuity among beauty workers, measures must be taken to enhance their job esteem. This can be achieved through several means, including fostering job value consciousness, implementing an effective organizational reward system, providing education on emotion control, maintaining a positive organizational culture that values emotional and psychological well-being, and operating human service systems while taking into account the individual's self-efficacy and self-esteem.

        들깨 Limonene 유전자의 담배식물(Nicotiana benthamiana)내 Agroinfiltration에 의한 분자적 특성

        The full-length cDNA encoding Perilla frutescens limonene synthase (PFLS) (603 amino acids, GenBank accession no. D49368) was cloned. To elucidate the role of PFLS in gene regulation, we transiently transformed full-length PFLS into tobacco plants. PFLS mRNA was first detected in the intact leaves of the plants at 6 h, and the LS transcript level increased after 12 h in leaves treated with oxidative stress-related chemicals. The transient overexpression of PFLS resulted in increased transcription of NbPR1 and NbSIP in Nicotiana benthamiana leaves. Thus, our result confirmed that the infiltration of PFLS gene act as a transcriptional regulator of NbPR1 or NbSIP genes in the tobacco.
