한열변증 설문지와 일반적 건강 검진 결과와의 상호 연관성에 관한 비교 연구

Background and purpose: The cold-heat patternization is one of the most frequently use for diagnostic method in oriental medicine, But it is still an unclear scientific mechanism and the objective index. the aim of this study is to search the objective index of the cold-heat patternization, comparing with ordinary health test. Method: The study group comprised 101 Korean, male:female ratio 51:50, with a mean age of 49.74 years, The results of the questionnaires for cold-heat patternization were compared with the western health test that it consist of height, weight, muscle mass, body fat, WBC, Hb, ALT, AST, TG, Total cholesterol, glucose, HDL-cholesterol, free-T4 and TSH, measured in the health promotion center of the hospital of Chungnam university Result: The cold patternization were significantly higher in the women than the man, The somothing of the questionmaires for the heat patternization were positive correlated with TG, TSH, glucose, Hb, free T4, weight, musle mass(p<0,05), But there were no correlation in the gross. The somothing of the questionnaires for the cold patternization were negative correlated with the weight, musle mass, body fat, glucose, Hb and ALT(p<0.05), Especially there were strong negative correlation with the weight and muscle mass(p<0.01). Conclusion: The present study shows there is no definitive index for the cold-heat patternization in the ordinary health test. But the weight and muscle mass can be useful index of the cold patternization.
Thyroglobulin에 대한 단일클론항체의 제작 및 특성

Thyroglobulin과 반응하는 monoclonal antibody를 3클론(TN-1, TN-2 및 TN-3)을 확립하였다. 이들 중 1클론 (TN-2)은 thyroglobulin의 3차원적인 구조를 인식하면서 음전하를 띠는 자가항원의 일종인 phosphatidylserine과도 교차반응성을 나타내었다. 따라서 TN-2를 이용한 자가면역성 갑상선염의 진단 및 치료에의 적용효과가 기대된다. Twelve clones of monoclonal antibodies (mAbs) against thyroglobulin were produced and characterized. Among them, three mAbs (TN-1, TN-2 and TN-3) showed high binding affinity to thyroglobulin. from ELISA inhibition assay, TN-2 showed considerable reactivity with soluble thyroglobulin. TN-2 also reacted with phosphatidylserine which has a negative charge in aqueous condition. These results suggest that TN-2 has characteristics of autoantibody concerned with thyroiditis.

본 연구에서는 해파리로부터 콜라겐 추출물을 제조하였고, 이에 대한 보습 효과를 측정하였다. 보습 효과는 caspase 14 발현, filaggrin, hyaluronan synthase-3 (HAS- 3), aquaporin-3 (AQP-3) 및 desmocollin (DSC)을 측정하였다. 그 결과, caspase 14 mRNA 발현은 비교물질인 retinoic acid (RA)와 유사한 효과를 나타냈다. 그리고 콜라겐 추출물은 20 mg/mL의 농도에서 각각 211.7%, 139.9%, 212.5% 및 116.8%의 filaggrin, HAS-3, AQP-3 및 DSC 발현 증가를 나타냈다. 따라서, 본 연구에서 해파리 콜라겐 추출물은 보습 효과를 갖는 화장품 소재로서의 개발 가능성이 클 것으로 기대할 수 있다. In this study, we investigated the moisturizing effects of collagen extract from jellyfish. The mositurizing effects were measured by the caspase 14 expression, filaggrin, hyaluronan synthase-3 (HAS-3), aquaporin-3 (AQP-3) and desmocollin (DSC). As a results, effect of caspase 14 mRNA expression of collagen extract are similar to that of retinoic acid (RA) which is a reference control. And the collagen extract showed inhibitory of filaggrin, HAS-3, AQP-3 and DSC mRNA expression of 211.7%, 139.9%, 212.5% and 116.8% respectively, at the concentration of 2%. Therefore, our study suggested that jellyfish collagen extract has considerable potential as a cosmetics ingredient with moisturizing effect.
In vitro에서 발암물질에 의한 발암진행에 미치는 천궁약침액의 영향

1. 천궁약침액은 1×에서 15.1%의 cytochrome P4501A1 저해효과를 나타냈으며 그 억제효과는 약침액의 농도가 높을수록 증가하였다. 2. 천궁약침액은 benzo[a]pyrene과 세포의 DNA 결합을 유의성있게 억제시켰다. 이상의 결과를 종합해 볼 때 천궁약침액은 효과적으로 cytochrome P4501A1 효소를 저해했으며, 발암물질과 DNA의 결합을 억제시켜 외부 물질 또는 대사물에 의해 일어날 수 있는 돌연변이와 암 발생을 억제할 것으로 사료된다. The effects of Cnidium officinale Makino aqua-acupuncture solution (COMAS) and Cnidium officinale Makino water-extraced solution (COMWS) on the CYP1A1 activity and benzo[a]pyrene(B[a]P)-DNA adduct formation were examined. There were 6.8%, 12.1%, 15.1%, 18.3% and 22.6% inhibition in the activity of cytochrome P4501A1 enzyme with the treatment of 0.1×, 0.5×, 1×, 3× and 5× COMAS, respectively. At concentration of 0.1× COMAS, the binding of [3H]B[a]P metabolites to DNA of NCTC-clone 1469 cell was significantly inhibited by 56.9%. These results suggest that COMAS has chemopreventive potential by inhibiting cytochrome P4501A1 activity and benzo[a]pyrene-DNA adduct formation.
