        Functional Conservation and Divergence of FVE Genes that Control Flowering Time and Cold Response in Rice and Arabidopsis

        Recent molecular and genetic studies in rice, a short-day plant, have elucidated both conservation and divergence of photoperiod pathway genes and their regulators. However, the biological roles of rice genes that act within the autonomous pathway are still largely unknown. In order to better understand the function of the autonomous pathway genes in rice, we conducted molecular genetic analyses of OsFVE, a rice gene homologous to Arabidopsis FVE. OsFVE was found to be ubiquitously expressed in vegetative and reproductive organs. Overexpression of OsFVE could rescue the flowering time phenotype of the Arabidopsis fve mutants by up-regulating expression of the SUPPRESSOR OF OVEREXPRESSION OF CO1 (SOC1) and down-regulating FLOWERING LOCUS C (FLC) expression. These results suggest that there may be a conserved function between OsFVE and FVE in the control of flowering time. However, OsFVE overexpression in the fve mutants did not rescue the flowering time phenotype in in relation to the response to intermittent cold treatment.

        질소원이 증진된 수확후배지를 이용한 느타리버섯 수량 특성

        The aim of this study was to re-use spent mushroom substrate (SMS) with increased total nitrogen (T-N) and amino acid content and reduce the amount of cottonseed meal used as nutrient supplement in Pleurotus ostreatus cultivation. Bacteria used for improvement of the T-N content were GM20-4(Bacillus sp.) and Rhodobacter sphaeroides (RS). GM20-4 was isolated from the SMS of P. ostreatus and RS was obtained from Gwangjusi agricultural technology center. SMS in T1, T2, and T3 was reused as substrate after drying and the T-N content of dried SMS (D-SMS) was increased by 0.34% by treatment with the bacteria. T1 with 8% D-SMS and T2 with 18% D-SMS had higher rates of primordia formation compared with T3 and the control. The biological efficiency of the control and of treatment with 8%, 18%, and 26% D-SMS was 110%, 114%, 112%, and 79%, respectively. Considering the economic cost, yield, and biological efficiency, T2 with 18% D-SMS as the culture substrate for P. ostreatus was shown to be the most effective for cultivation.

      • 느타리버섯 병재배 농가 오염발생 실태조사

        최근 대규모화가 증가추세인 병재배농가는 배지제조부터 수확까지 환경조절에 의해 이루어지므로 청결유지가 가장 안정생산에 주요 요인으로 시설형태와 각각의 재배단계에 따른 오염발생 조사를 통해 오염율을 저감할 수 있는 방안모색이 시급한 실정이다. 이에, 본 연구는 재배 농가를 대상으로 냉각실 접종실의 분리 여부, 탈병장과 배양실의 연결 여부에 따라 3타입으로 농가를 구분하여 오염 이력조사를 실시하였다. 시설별 낙하균밀도, 오염율, 멸균시간 및 멸균시 온도변화측정 등을 통해 오염요인을 분석하였다. 각 조사는 계절별 2회 조사를 실시했으며 춘계조사의 결과 냉각실, 접종실 이 분리되어 있고 탈병장이 배양실과 떨어져 있는 B타입농가의 낙하균 밀도가 가장 낮았으며 오염 률 또한 가장 낮았다. 냉각실 접종실이 분리되지 않고 탈병장이 배양실과 복도로 연결되어 있는 C 타입농가의 경우 배양실의 세균류와 곰팡이류의 오염률이 각각 B농가의 228배, 36배 높았다. 냉각 실과 접종실은 분리되어 있지만 탈병장이 배양실과 연결되어 있는 A타입의 농가는 배양실에 있어 서 세균류는 B농가보다 6.4배 곰팡이류는 4.7배 높았다. 계절별로는 봄보다 여름에 오염률이 더 높 은 결과를 보여주였다. 오염발생 병으로부터 오염균을 분리한 결과 푸른곰팡이병을 일으키는 Trichoderma속이 가장 많았으며 세균으로는 Chryseobacterium속이 많았다. 시설별 낙하균에서 분리한 오염균으로는 곰팡이류의 경우 Trichoderma속, Penicillium속, Aspergillus속 순으로 높은 오 염율을 보였다. 세균의 경우 Bacillus속과 Pseudomonas속의 오염율이 높았다. 병재배농가 오염에 가장 큰 영향을 미치는 크린부스, 냉각실, 접종실, 배양실의 청결관리가 오염율을 낮추는 주요 요인 으로 판단된다.

        표고 톱밥배지 재배시 녹색LED 광량이 자실체 생육에 미치는 영향

        In previous studies, we selected the green LED(light emitting diodes) for suitable wavelength of the light by higher commercial yields and lower ratio of the abnormal fruit body in Lentinula edodes. In this study, we aimed to select efficient irradiation intensity of the green LED. Stronger irradiation intensity of the green LED resulted in more polyphenol content of fruit body. And Polyphenol content of fluorescent lamp was similar to that of the green LED 20 umol·m-2·s-1.. Ergosterol content of the green LED 5 umol·m-2·s-1 was showed higher 2.1 times than that of the fluorescent lamp. In four level of irradiation intensity, 5, 10, 15, 20 umol·m-2·s-1 there was no big difference in characteristics of the fruit body. However the yield of fruit body in the green LED 5 umol·m-2·s-1 is higher than the others. In addition, The amount of electricity used of the green LED 5 umol·m-2·s-1 was reduced 15.9% than that of the fluorescent lamp. In conclusion, we selected 5 umol·m-2·s-1 for suitable irradiation intensity in Lentinula edodes with sawdust substrate cultivation.

        살균조건 및 UV-C 조사가 느타리버섯 병재배 오염율 감소에 미치는 영향

        This study was conducted to reduce contamination ratio of oyster mushroom bottle cultivation. The optimal conditions of substrate sterilization for reducing of contamination ratio were at 121oC for 90 min. In addition, UV-C irradiation is good for lower contamination ratio to continue over 6 hours at cooling and inoculation room after sterilization. The contamination ratio and density of microorganisms of substrate were showed 0% after sterilization at 121oC for 90 min. Trichoderma sp., main pathogen of mushrooms, was detected from substrate after sterilized during 2 or 4 hours at 101oC and 105oC, respectively. The amount of electricity used was the lowest at 121oC for 90 min than that of other sterilization conditions. The UV-C irradiation treatment was used UV-C lamp(40 watts) in the inoculation room(56 m 3 ). The density of bacteria did not detected after UVC irradiation for 6 hours. And the death ratio of bacteria and Trichoderma sp. was 99.9% after UV-C irradiation for 6 hours. However, in the same UV-C irradiation time, the death ration of Cladosporium sp. was 90.9%. Therefore, the death ratio of fungi was lower than that of bacteria at the same UV-C irradiation treatment.

        세균을 이용한 수확후배지의 총질소 및 아미노산 증진 효과

        본 연구는 느타리버섯 수확후배지를 배지자원으로 재사용하기 위하여 미생물처리에 의한 수확후 배지의 총질소함량 증진효과를 조사하였다. Bacillus sp. GM20-4 와 Rhodobacter sphaeroides(RS) 배양액 30%를 함께 접종하여 5일간 상온에서 배양 시 총질소 함량이 가장 크게 증가하였다. 농가의 수확후배지 조성에 따른 GM20-4와 RS 처리에 의한 T-N함량 변화를 조사하기 위하여 경기 지역 버섯재배농가 세 곳의 수확후배지(spent mushroom substrates, SMS A, B, C)를 수집하여 GM20-4 와 RS 배양액 30%를 처리한 결과, 무처리구에 비해 건조 SMS B 에서 T-N함량이 20%p로 가장 크게 증가하였고, 건조 SMS A는 17%, C는 12% 증가하였다. 반면에 생수확후 배지에서는 미생물 처리에 의한 T-N 함량 증가 효과가 크지 않았다. 또한 아미노산 조성별 함량 변화는 건조된 SMS A, B, C 모두 GM20-4와 RS 처리로 aspatic acid와 glutamic acid가 상대적으로 높게 증가한 결과를 보였고, 특히 건조 SMS B의 처리구에서 두 아미노산의 함량이 무처리구보다 2.9배 증가를 보임으로서 미생물처리가 수확후배지의 총질소 및 아미노산 함량의 증진효과를 확인하였다. This study was conducted to reuse spent mushroom substrates (SMS) of Pleurotus ostreatus and improve their nitrogen content by bacterial treatment. Two kinds of bacteria were used to investigate the increase in total nitrogen (T-N) content. Bacillus sp. (GM20-4) was isolated from SMS of oyster mushroom, and Rhodobacter sphaeroides (RS) was obtained from Gwangju Si Agricultural Technology Center. SMS samples were collected from three oyster mushroom cultivation farms located in Gyeonggi-do province, Korea. When dried SMS was inoculated with 30% culture broth of GM20-4 and RS and incubated at room temperature (25±2℃) for 5 days, T-N content increased. To investigate the T-N content of other SMS, three dried SMS samples (A, B, and C) were treated by the same method using GM20-4 and RS. As a result, the T-N content of sample B was 20% higher than that of the control, whereas the T-N content of samples A and C increased to 17% and 12%, respectively. The change in T-N content by bacterial treatment of wet SMS was slightly higher than that of the control. The changes in amino acid content were also found to be higher than those in the control in all SMS samples by GM20-4 and RS treatment. Aspartic acid and glutamic acid contents were the highest among all amino acid compositions. Especially, the aspartic and glutamic acid contents of sample B increased by 2.9 folds higher than the control.

        표고 톱밥배지 재배시 LED 광파장이 자실체 생육에 미치는 영향

        This study was carried out to elucidate suitable wavelength of light during development of fruit body in Lentinula edodes. The four colors of LED(Light Emitting Diode), blue, green, red and yellow, were irradiated for formation of fruit-body. The effect of color of LED at all growth stage, the lightness of cap showed darker in blue and green LED irradiation than that of red and yellow LED. The longer stipe were resulted in longer wavelength. And the activity of anti-oxidant did not showed big differences according to LED wavelength. We obtained higher commercial yields and lower ratio of abnormal fruit body in green LED than those in control(fluorescent lamp).

        느타리버섯 후배양 기간과 온도처리가 발이 균일도에미치는 영향

        The study was conducted to investigate the effects of post-incubation period and temperature treatment conditionsduring incubation on the uniform primordia formation and cultural characteristics of oyster mushroom (Pleurotus ostreatus). Threekinds post-incubation period; 25, 30, 35 days and control were applied for 30 days while two kinds incubation roomtemperature 23oC and 26oC and control were used 20oC. The substrate temperature during pre-incubation was of ‘Suhan No. 1’and ‘Gonji No. 7’. Oyster mushroom varieties tended to increase between 24oC to 26oC at 11 to 15 days after inoculation andthen they were maintained in treatment temperature during post -incubation period. The CO2 occurrence was at the highest at6,500ppm for ‘Suhan No. 1’ and 5,800ppm for ‘Gonji No. 7’ at the time of the highest temperature increase. The ratio of un-uniformal primordia formation and the ratio of non-commercial fruit body were reduced by 40%, 10.5%, respectively comparedto control for ‘Suhan No. 1’ when in the post-incubation temperature was 26oC, and incubated for 10days and 15daystreatment. Also, ‘Gonji No. 7’ was reduced by 19%, 9.5%, respectively when in the post-incubation temperature was 26oC, andincubated for 10 days treatment. Therefore, the higher post-incubation temperature of room and longer post-incubation periodresulted in the higher percentage of primordia formation of two cultivars.
