        분열효모에서 THO 복합체의 구성요소인 Tex1/THOC3가 생장 및 mRNA 방출에 미치는 영향

        진핵생물에서 THO/TREX 복합체는 전사 신장, pre-mRNA 가공 및 mRNA의 핵에서 방출에 중요한 역할을 담당한다. 이 복합체는 진화적으로 잘 보존되어 있지만, 생명체에 따라 구성성분과 기능에 차이가 존재한다. 이 논문에서는 출아효모보다는 고등생물과 더 유사한 분열효모 Schizosaccharomyces pombe에서 THO/TREX 복합체의 한 구성요소인 spTex1가 생장과 mRNA의 방출에 필수적이지 않다는 것을 보였다. spTex1 유전자의 결실과 과발현 어느 것도 생장의 결함과 $poly(A)^+$ RNA가 핵 안에 축적되는 현상을 거의 보이지 않았다. 또한 spTex1-GFP 단백질은 주로 핵 안에 위치하였다. Yeast two-hybrid와 Co-immunoprecipitation 분석에서 S. pombe Tex1은 THO/TREX 복합체의 주요 구성인자인 spHpr1 (THOC1), spTho2 (THOC2)과 상호작용을 하였다. 이와 같은 결과들은 S. pombe의 Tex1도 THO/TREX 복합체의 구성인자이지만, 생장과 mRNA 방출에는 중요한 역할을 하지 않음을 의미한다. In eukaryote, THO/TREX complex plays a critical role in transcriptional elongation, pre-mRNA processing, and nuclear mRNA export. This complex is evolutionally well- conserved, but there are some differences in composition and function according to organisms. Here we showed that spTex1, a component of THO/TREX complex, is not essential for growth and mRNA export in a fission yeast, Schizosaccharomyces pombe, which is more similar to higher eukaryote than budding yeast. Deletion and overexpression of the spTex1 gene do not lead to any detectable growth phenotype and accumulation of poly(A)+ RNA in the nucleus. And the spTex1-GFP protein is localized mainly in the nucleus. Yeast two-hybrid and Co-immunoprecipitation analysis showed that the spTex1 protein interacted with spHpr1 (THOC1) and spTho2 (THOC2), main subunits of THO complex. We conclude that the S. pombe Tex1 is a component of THO/TREX complex, but does not plays important roles in growth and bulk mRNA export from the nucleus.

        분열효모에서 TREX-2 복합체의 구성요소인 Sus1이 생장 및 mRNA 방출에 미치는 영향

        Sus1 / ENY2는 진핵생물에 잘 보존된 작은 단백질로 염색사 리모델링과 mRNA 생성과정에 관여한다. Sus1 단백질은 전사 보조활성자인 SAGA 복합체와 핵공과 연관된 TREX2 복합체 등, 핵에 존재하는 2개 복합체의 구성 요소이다. 분열효모 Schizosaccharomyces pombe의 유전체 데이터에서 Sus1의 이종상동체를 찾아 기능을 분석하였다. 4분체 분석 결과 이 유전자는 생장에 필수적이 아니었으나, Sus1 유전자를 결실시키면 낮은 온도에서 생장이 느렸으며, $poly(A)^+$ RNA도 핵 안에 약간 축적되는 표현형을 보였다. 또한 Sus1-GFP 단백질은 주로 핵에 존재하였다. Yeast two-hybrid 분석과 공동면역침전 실험에서 Sus1 단백질은 TREX-2 복합체의 또 다른 구성인자인 Sac3와 상호작용을 하였다. 이와 같은 결과들은 S. pombe의 Sus1 단백질도 역시 TREX-2 복합체의 구성인자로 mRNA 방출에 관여하고 있음을 시사한다. Sus1 / ENY2 is a tiny conserved protein that is involved in chromatin remodeling and mRNA biogenesis. Sus1 is associated to two nuclear complexes, the transcriptional coactivator SAGA and the nuclear pore associated TREX2. In fission yeast, Schizosaccharomyces pombe, ortholog of Sus1 / ENY2 was identified from the genome database. Tetrad analysis showed that the S. pombe sus1 is not essential for growth. However, deletion of the sus1 gene caused cold-sensitive growth retardation with slight accumulation of $poly(A)^+$ RNA in the nucleus. And the Sus1-GFP protein is localized mainly in the nucleus. Yeast two-hybrid analysis and co-immunoprecipitation experiment showed that Sus1 interacts with Sac3, another subunit of TREX2 complex. These results suggest that S. pombe Sus1 is also involved in mRNA export from the nucleus as a component of TREX-2 complex.

        서양복(西洋服) 파스닝(fastening)의 변천(變遷)에 관(關)한 고찰(考察)

        The purpose of this thesis is to overview the periodical changes of the fastenings, from the pins and buttons in ancient age to the velcro in these days, and to study the relationships of these various fastenings to the costume types, and also to research how it is applied to the latest fashion. The fastening is closely related to the costume types, being various in its form, like pins, fibula, brooches, buttons, lacing, points, hooks & eyes, press studs, zipper and velcro. It is clear from the fact that the fastenings have developed remarkably since 13th century when the costume became tightly fitting to the body, while the fastenings had been very simple, like buttons or fibula, for so long time from the ancient time to the 12th century. The end of fastening`s development is to be fitting to the body and to make it easy to take on or off the costumes by use of it. In `99 S/S, `99-00 A/W pret-a-porter collections, ornamental fastening was more emphasized then practical one, probably due to the development of more stretchable fabric.

        Shakespeare 희곡작품에 나타난 복식 이미저리 연구 Ⅰ

        The purpose of this research was to figure out clothing imagery underlying Shakespeare`s King Lear and transfer it into actual stage costume. Three steps were taken in order to achive the purpoes. The first step exemined theoretical background of the Shakespeare`s imagery. The second step reviewed a wide spectrum of stage costume used in the last four centuries. The final step was to design and produce actual stage costume focusting on Lear, while considering clothing imagery drawn form the previous steps. King Lear contained rich clothing imagery which could contrast the relationship between material and spirit. In paricular, it shed light on the contrast between Lear`s divesting and Edgar`s wearing and denoted that wearing and divesting processes were closely related to spiritual and status changes. A review of stage costume used for four centuries was prepared in order to understand how clothing imagery was applied in producing stage costume. It was found that their stage costume was reflecting clothing imagery captured from the analysis of King Lear. On the basis of this review, this research presented Lear`s stage costume which applied clothing imagery drawn from King Lear. In producing stage costume, actual stage costume used Elizabethan fashion as its basic color and pattern. It also utilized medern clothing production techniques and ornaments acceptable in these days. As Lear was depicted as a character who underwent spiritual and status changes in the process of divesting, his costume was produced in line with this clothing imagery(i.e., divesting). The long journey of reviewing and analyzing King Lear from a stand point of clothing reassured us, on the one hand, the importance of stage costume. On the other hand, it established the groundwork for developing the methodology of fashion design by applying clothing imagery drawn from literature to design and production of stage costume.

        요하네스 베르메르의 작품을 통해 본 17세기 네덜란드 여성 시민복과 시민문화

        This thesis aims to investigate the characteristics of the civil costume in Netherlands women and the way how the civil culture was reflected on these by analyzing the women`s costume depicted in the works of the Johannes Vermeer regarded as the representative painter of Netherlands in 17th century. The method of study was to select 24 pieces of Vermeer`s works among the 30 pieces, and were analyzed in detail. These were approved to be common civil costumes by researching the works of other painters in that era on the other hand. The result shows that the civil costume of Netherlands women from 1653 until 1675 was in the simple form of two pieces dress, and minimal decorations with simple hair style and headdress would take the constitution of the frugality, chastity and practicality as the mainstream, along with using the red, yellow and blue as the primary three colors on to the costumes. These characteristics might be ascribed to the Netherlands civil culture influenced by the Calvinism that emphasized the frugality and chastity, denouncing the luxury with supporting the religion and morality. This trend was also noted in the men`s costume, giving evidence of the intimate relationship between the costume, religion and civil culture. This thesis might be a help to elucidate the relationship between the costume and cultural society, and be a affordable tools to study the contemporary costume.

        연구논문(硏究論文): (중국)中國 동북지역 다롄시 스트리트 패션에 나타난 컬러트렌드 (분석)分析(제2보) -2010 겨울, 중국의 전통 색채감정과의 비교를 중심으로-

        The purpose of this research is to analyze the effect of the traditional color sentiment on the contemporary clothing color by researching the street fashion of 2010 Winter season of Dalian in China, in view of the clothing color of women in their 20-30 years of age, along with traditional Chinese color. This is the 2nd paper following the paper no. 1, focusing on 2010 Summer season, so the research method is same as the paper no. 1. As a result, Black and deep tone PB color most frequently appeared. The vivid and strong tone of R, YR, Y color showed high frequency of clothing colors. This results illustrate that the clothing color choices of Chinese women was affected by Chinese traditional color sentiment rather than the international color trend. Characteristically, in winter season, frequency of R and Y color categories were higher than in summer season, specially in the top and accessories color, those colors showed higher frequency than the other colors. These were the important points in dealing with winter color trend, so that the fashion company and brands should take it an important points when they are planningwinter clothing colors. The achromatic colors show the high frequency in both seasons. The bottom color shows high incidence of Black and deep tone of PB color categories as in the previous researches. That means consistent color trend existed in Dalian city, however it does not seem to have its origin from traditional Chinese color preferences. This kinds of regional and seasonal research might contribute to finding the basic informations about the design and marketing strategies to launch into the Chinese fashion market.

        영국(英國) 패션의 원동력(原動力)에 관(關)한 연구(硏究)2 -후세인 샬라얀을 중심(中心)으로-

        The purpose of this thesis is how to find the way of activating Korean fashion industry, by reviewing the accomplishments of British young designers, Hussein Chalayan, who are contributors to the present peak activities of British fashion. Some results come over this depressive state of Korean fashion might be proposed. Firstly, Korean government should try to direct the Korean fashion associated group to make the unified and effective results in an organized way. Secondly, Korean fashion industries and university-level fashion school should have more intimate relationship and be closely communicated with each other. Also the fashion school should watch their curriculum. and change it gradually to the up-todate one. Thirdly, the Korean new generation designers should do effort to have the highest tailoring technique and artistic good sense. Finally, the designer should have broad range of knowledge for their design. In conclusion, the government, university-level fashion schools, fashion industries and fashion designers should be positively changed and cooperated to do the best and activate Korean fashion.
