        메티실린 내성 황색 포도상 구균에서 mecA, femA 유전자의 임상적 의의

        Purpose : In the treatment of MRSA infection, rapid detection of MRSA is extremely important. The mecA gene codes the new drug resistant polypeptides called PBP2' which mediates the clinically relevant resistance to all beta-lactam antibiotics. The identical mecA gene has been found in coagulase-negative staphylococcus with the methicillin-resistant phenotype. On the other hand, the femA gene was absent from coagulase negative staphylococcus strains with the methicillin resistant phenotype. This study is aimed at early detection and definite diagnosis of MRSA. Methods : A total of 24 MRSA strains were studied. All strains were tested for antimicrobial susceptibility and purified DNA. We amplified both mecA and femA genes by PCR in 24 strains. Results : In MRSA all the 16 strains (100%) carried femA gene and 11 strains (68.7%) carried mecA gene. In contrast, in methicillin sensitive staphylococcus all the 8 strains (100%) carried femA and only 3 strains (37.5%) were detected mecA. Conclusions : As results, there are difference in the phenotype and genotype of methicillin resistance by PCR of mecA and femA. Such disparities between methicillin resistance and the presence of mecA gene suggest the presence of control gene of the mecA.

        HJ01이 OP9세포에서의 지방 분화와 P-407로 유발한 고지혈증 흰쥐에 미치는 영향

        Objectives : This study was designed to investigate the effect of a herbal preparation HJ01 consisting of Salicornia herbacea, Citri Reticulatae Pericarpium, Crataegi Fructus and Glycyrrhizae Radix on adipocyte differentiation in OP9 cells and on poloxamer 407(P-407)-induced hyperlipidemia in mice. Methods : 1. MTT assay was used to evaluate the potential cytotoxicity of Salicornia herbacea, Citri Reticulatae Pericarpium, Crataegi Fructus, Glycyrrhizae Radix and HJ01, respectively. 2. Bone-marrow derived OP9 cells were treated with HJ01, and the alterations in fat storage in the cells were determined by the Oil red O assay. 3. The protein level of CAAAT/enhancer binding protein alpha($C/EBP{\alpha}$), as a adipocyte differentiation marker, was examined using western blot analysis in differentiated OP6 cells. 4. Adult male C57BL6 mice received intraperitoneal injections of P407 to induce hyperlipidemia, simultaneously, were treated with HJ01 for 4 weeks. Then the cholesterol (TC), triglyceride (TG) and high-density lipoprotein-cholesterol (HDL-c) levels in sera and liver tissues were measured. Results : 1. The MTT assay exhibited that Salicornia herbacea, Citri Reticulatae Pericarpium, Crataegi Fructus, Glycyrrhizae Radix and HJ01 showed no significant cytotoxicity in tested dosages. 2. Ten days' treatment with HJ01 markedly inhibited the increases in fat storage in differentiated OP6 cells. 3. Four weeks' treatment with HJ01 down-regulated the protein level of CAAAT/enhancer binding protein alpha($C/EBP{\alpha}$) but up-regulated the levels of adiponectin in differentiated OP9 cells. 5. HJ01 inhibited the accumulation of TC and TG in liver tissues and increased serum levels of TC in hyperlipidemic mice. Conclusions : These results suggest that HJ01 can in vitro inhibit adipocyte differentiation and fat storage in OP6 cells, in vivo improve the hyperlipidemia induced by P-407 in mice, which may be mediated by promoting glucose uptake and improving a lipid metabolite profile.

        고추냉이가 배추김치의 발효 중 이화학적 특성에 미치는 영향

        본 연구는 고추냉이 뿌리(이하 고추냉이)가 배추김치의 발효와 저장성에 미치는 영향을 알아보고, 배추김치의 저장성 향상을 위한 천연 첨가제로서의 고추냉이의 이용 가능성을 모색하고자 하였다. 배추 무게에 대하여 0, 2, 3, 4, 5%를 첨가하여 배추김치를 담그어 $10^{\circ}C$에서 35일간 발효시키면서 이화학적 특성을 살펴보았다. 발효 전반적으로 0% 처리구가 가장 낮은 pH를 보였고, 고추냉이 첨가량이 증가할수록 pH가 높았다. 또한 고추냉이 첨가량이 증가할수록 적숙기의 pH를 오랫동안 유지하였다. 총산은 발효 초기부터 말기까지 0% 처리구의 가장 높았고, 고추냉이 첨가량이 증가할수록 낮은 총산을 보여 고추냉이 첨가가 배추김치의 발효를 지연시켜 주었다. 특히 3% 이상 첨가할 경우 발효를 지연시켜 주는 효과가 있음을 알 수 있었다. 총비타민 C 함량은 발효가 진행됨에 따라 증가하다 0%와 2% 처리구는 발효 8일, 3%, 4%와 5% 처리구는 발효 10일에 최대값을 보인 후 감소하였다. 담금 직후부터 발효 말기까지 고추냉이 첨가량이 증가할수록 총비타민 C 함량이 높았고, 특히 3% 이상 첨가한 처리구에서 더욱 높았다. 3% 이상 첨가한 처리구가 0%와 2% 처리구에 비해 총비타민 C가 최대 함량을 보인 후 완만하게 감소하여 발효가 지연되었다. 고추냉이 첨가량이 증가할수록 환원당 함량은 높았고, 발효 전반적으로 0%와 2% 처리구가 급격하게 감소하여 발효가 빨리 진행됨을 알 수 있었다. 반면 3% 이상 첨가한 경우 완만하게 환원당이 감소하였고, 발효 말기에도 높은 함량을 유지하였다. 수용성 펙틴의 경우 배추김치가 익어감에 따라 모든 처리구에서 점차로 증가하였고, 고추냉이 첨가량이 증가할수록 수용성 펙틴의 함량은 낮았다. 발효 말기로 갈수록 3% 이상 첨가한 처리구가 0%와 2% 처리구에 비해 더욱 낮은 값을 보였고, 고추냉이 첨가량이 증가할수록 수용성 펙틴의 증가하는 폭이 감소하여 발효가 서서히 진행을 알 수 있었다. 이상의 결과로 볼 때 고추냉이를 첨가할 경우 배추김치의 발효를 지연시켜 저장성을 향상시켜 주었고, 특히 3% 이상 첨가할 경우 바람직한 효과를 보였다. 따라서 배추김치를 담글 때 고추냉이를 첨가하게 되면 품질과 저장성을 향상시켜줄 것으로 기대된다. In this study, we explored the application of Wasabi addition to baechu kimchi with regard to quality improvement, by reviewing the optimum Wasabi level and its effects on the product physicochemical characteristics during fermentation. The final weight percentages of Wasabi in the baechu kimchi were adjusted to 0, 2, 3, 4, and 5% per weight of baechu, respectively, and the samples were fermented for 35 days at $10^{\circ}C$. Following fermentation, the treatments over 3% Wasabi contained the highest pH and lowest total acidity values. Total vitamin C and reducing sugar content increased initially, up to certain fermentation times depending on the level of Wasabi contents within the treatments over 3% Wasabi, and decreased afterwards. The water soluble pectin content increased as the fermentation period increased; however, it decreased with increasing levels of Wasabi. In conclusion, the optimum levels of added Wasabi in the baechu kimchi were 3% and 5% per weight of baechu, to have a fermentation-retarding effect on the product.

        근로자(勤勞者)의 양생수준(養生水準)과 비만(肥滿)과의 관계(關係)

        In this study, the researcher tried to present the groundwork to prepare the oriental medical yangseng plan for the future obesity with the comparison between yangseng level and the obesity of workers. The researcher made up the questionnaire which asks the general character, health-related character and yangseng level, projected among 560 people. All collected material was analyzed by SPSS and tested by T-test and ANOVA. 1. The general yangseng level average is 3.27, morality yangseng 3.90, sleep yangseng 3.39, mind yangseng 3.31, sex life yangseng 3.30, exercise yangseng 3.15, activities and rest yangseng 3.08, diet yangseng 2.94, seasonal yangseng 2.84. The highest is morality yangseng and seasonal yangseng is the lowest. 2. In the aspect of yangseng level: Having a spouse, Non-smoking, Regular exercising, Sufficient sleeping have higher yangseng levels. 3. Obesity related index is changing into the higher level when he/she is older, more paid, more job experience, more education background but less metabolic calory. And drinking and regular exercising have relativity with body composition analysis. 4. After comparing yangseng level with body composition analysis, we can easily find that the more visceral fat, the higher WHR has a high yangseng level. The more mineral also has a high mind yangseng and a low diet yangseng. The visceral fat level has a high yangseng level when higher morality yangseng and mind yangseng are getting higher and higher. And sex life yangseng shows that the highest yangseng level is from 9-10 visceral fat and the lowest yangseng level is from below 4 visceral fat. The higher WHR, the higher morality, mind and sleep yangseng. The heavier, the lower diet yangseng. The mind yangseng was very high when body fat rate was higher. The more body fat, the higher morality yangseng. The higher yangseng, when we have more muscle. Yangseng level and obesity of laborer has a close relationship with individual character and daily habits. Also, relevance can be easily found between yangseng level and obesity. Now the researcher came into the conclusion that we need to control over laborers' health and prevention of their obesity.

        지류 중소하천의 물관리 탄력성 평가지수 개발 및 평가

        지류 중소하천의 미래 물관리 정책방향을 수립하기 위한 자료로, 물관리 탄력성 지수를 개발하고 이를 117개 중권역에 적용하였다. 물관리 탄력성 지수는 취약성, 견고성, 다양성의 부지수로 구성하였으며, 각 부지수는 다시 이수, 치수, 하천환경 분야의 지표들로 총 31개의 지표항목으로 구성하였다. 중권역은 다목적댐 하류 본류유역(범주 1), 용수공급 및 유량조절이 불가능한 지류(범주 2)와 가능한 지류(범주 3)로 분류하여 적용한 결과, 지류 중소하천에서의 물관리 탄력성, 특히 이수 및 치수분야의 다양성이 본류에 비해 낮은 것으로 나타났다. 또한 취약성과 견고성에 있어서는 본류와 지류가 큰 차이를 보이지 않았으나 다양성에서 지류가 낮은 값을 나타내었다. 따라서 지류 중소하천의 다양성을 높이는 방향으로 물관리 정책의 수립이 필요하다. 본 연구에서 개발된 물관리 탄력성 지수는 내외부 충격에 대한 안정성을 종합적으로 제시하여 수자원 정책 및 계획의 우선지역 선정 등에 활용할 수 있을 것이다. Water Management Resilience Index (WMRI) was developed as a policy measure of adaptability to withstand water stresses and to set up water management strategies mainly in mid-small scale tributaries, and then evaluated on 117 sub-basins in South Korea. The index consists of 3 sub-indices such as vulnerability, robustness and redundancy sub-indices, each including indicators of 3 sectors: water use, flood mitigation, and river environment. Total number of indicators selected for the index was 31. Taking into account the stream order and control capability of river flow discharge, sub-basins were categorized into 3: 1 for mainstreams of lower large dams, 2 and 3 for tributaries, respectively without and with flow discharge regulation. As a result of the evaluation, resilience index scores in Category 2 and 3 are much lower than that of Category 1, especially with very poor score of redundancy. Although there was no significant difference between mainstream and tributaries in vulnerability and robustness sub-indices, results of redundancy sub-index in tributaries were lower than those in mainstream. Thus, it is conceived that the variety of water management schemes should be considered to improve their resilience in the face of future uncertainty. Addressing comprehensive stability of river basin against internal and external impacts, WMRI in this study can also be used for the prioritization of water management plans.

        한국어 어두파열음의 발음교육 효과 연구

        본 연구는 중국인 학습자를 대상으로 한국어 어두파열음의 발음을 교육하고, 그 효과를 살펴보는 데 목적을 둔다. 그리고 발음교육 효과를 토대로 중국인 학습자를 대상으로 한 한국어 어두파열음의 발음교육 방안을 제시해 본다. 본 연구에서는 20명의 중국인 학습자가 발음한 한국어 어두파열음의 특성을 VOT값과 후행모음 F0값으로 분석하였다. 수도권 출신의 청장년층 한국인 화자들은 경음의 VOT는 짧게, 평음과 격음의 VOT는 길게 발음하는데, 평음은 낮은 F0으로, 격음은 높은 F0으로 발음함으로써 세 발성유형을 변별한다. 반면 중국인 학습자들은 세 발성유형을 F0값에 차이를 두지 않고 발음하기 때문에, 세 어두파열음을 평음으로 발음하였다. 그리고 수도권 청장년층의 실제 발음을 반영하여 중국인 학습자들에게 어두파열음 교육을 실시하였다. 그 결과, 교육 후에도 학습자의 어두파열음 발음은 한국인의 발음과 차이가 있는 것으로 나타났다. 그러나 일부 학습자들에게서 교육의 효과가 나타났는데, 25%의 학습자가 한국인 화자처럼 세 발성유형을 변별하여 발음할 수 있게 되었다. 그리고 또 다른 25%의 학습자들은 발성유형별 음높이의 특성을 불완전하게 적용하여 발음하기 시작하였다. 따라서 VOT와 후행모음 F0을 활용한 어두파열음 교육 방법이 일부 중국인 학습자들에게는 평음, 격음, 경음을 변별하여 발음하는 데 도움을 주었다고 볼 수 있다. 이러한 실험결과를 바탕으로 중국인 학습자를 위한 한국어 어두파열음의 교육 방안을 제시하였다. The objective of this study is to explore teaching materials that can help Chinese learners make a clear distinction among three phonation types of word initial-plosives in Korean, and test their effects. For the study, VOT and F0 of word-initial plosives in Korean produced by 20 Chinese speakers were analyzed. Chinese learners pronounced all three phonation types of word-initial plosives as native Korean speakers do lax, without differentiating F0. Then we instructed Chinese learners on how to pronounce word-initial plosives in Korean, referring to pronouncements of native Korean speakers from Seoul and Gyeonggi Province. After the instruction, Chinese speakers" pronouncement of word-initial plosives continued to be different from native Korean speakers. However, the instruction turned out effective for some learners as 25% of them could make a distinction among the three phonation types of word-initial plosives like native Korean speakers. Another 25% of learners began to apply different pitches for different types of word-initial plosives, although inaccurately. From this finding, we can say that instructions on pronouncing word-initial plosives using VOT and F0 as a cue can help some Chinese learners to make a distinction among lax, aspirated and fortis.

        This study aims to investigate on residence's space use by the environment-friendliness of the indoor common space in multiple dwelling housing. Space plan and specific use were subjected for the comparison between environmental symbiotic housing in Japan and regular apartment in Korea. The embodiment of the environment-friendliness at indoor common space was achieved by considering dwelling building form and space plan. Moreover, environment-friendliness led the independency of residence's space use.

        Chironomus riparius의 급성 및 만성영향에 의한 농약의 퇴적토 독성평가

        BACKGROUND: Pesticides is exposed in an aquatic environment and effected to benthic animals. Especially, sediment-associated pesticides is required for determination of sediment toxicity on aquatic organisms. This study was conducted to evaluate the impact of six pesticides (chlorfluazuron, difenoconazole, dithianon, flufenoxuron, flutianil, pendimethalin) onChironomus riparius in aquatic ecosystems. METHODSANDRESULTS: Chlorfluazuron, difenoconazole, dithianon, flufenoxuron, flutianil and pendimethalin were used as a model compounds, which have a sediment-associated potential (K<sub>oc</sub>>3). Acute and chronic toxicity tests on Chironomus riparius were performed at six concentrations of each pesticidewith four replicates of each based onOECDtest guideline 235 and 218. The calculated 48-h EC<sub>50</sub> values of chlorfluazuron, flutianil, pendimethalin, difenoconazole, dithianon and flufenoxuron were 6.72, 2.55, 2.27, 0.77, 0.30 and 0.11 mg/L, respectively. Flufenoxuronwas the lowest 48-h EC<sub>50</sub> value in this study. TheNoObserved EffectiveConcentration (NOEC) and the Lowest Observed Effect Concentration (LOEC) of flufenoxuron for Chironomus riparius in 28-days test were 30 and 60 μg/kg, respectively. CONCLUSION: Pesticides of the sediment-associated have the potential effect for Chironomus riparius in aquatic ecosystems. Therefore, sediment toxicity assessment of these pesticides should be further investigated to evaluate the impact to benthic organisms.

        사회통합형 공동주택 개발을 위한 공유공간의 디자인요소 연구

        Socially-integrated housing will be able to connect social severance, integrating multi-culture, different age groups, handicap barrier, as a futuristic housing model. Multiple dwelling housing will provide for residents to develop community spirit and to facilitate positive autonomy by supplying ample communal space. This work was focused on common space for the establishment of socially-integrated housing and analyzed the design elements and planning method of common space. Information about the concrete use of common space in multiple dwelling housing will facilitate the planning of socially-integrated housing preceding spontaneous gathering and communication within neighborhood. Consequently, for the forms of socially-integrated housing, the design elements of common space consist of openness, connection, enclosure, unity as the view of space use, and ordinary, approach, amenity as the view of space composition.
