Objectives: The purpose of this study was to compare SYBR Green qPCR, TaqMan, and bacterial selective medium cultures for accurate quantitative analysis of oral microorganisms. Methods: The SYBR Green method is widely used to analyze the total amount of oral microorganisms in oral saliva. However, in this study, MTR-PCR method based on TaqMan method was performed using newly developed primers and probes. In addition, it was designed to confirm the detection agreement of bacteria among bacteria detection method. Results: As a result of MRT-PCR and SYBR Green qPCR analysis, more than 40 times (0.9-362.9 times) bacterium was detected by MRT-PCR. In addition, more bacteria were detected in saliva in the order of MRT-PCR, SYBR Green qPCR, and bacterium culture, and the results of MRB-PCR and SYBR Green qPCR showed the highest agreement. The agreement between the three methods for detecting P. intermedia was similar between 71.4 and 88.6%, but the agreement between MRT-PCR and SYBR Green qPCR was 80% for S. mutans. Among them, the number of total bacteria, P. intermedia and S. mutans bacteria in saliva was higher than that of SYBR Green qPCR method, and bacterium culture method by MRT-PCR method. P. intermedia and S. mutans in saliva were detected by MRT-PCR and MRT-PCR in 88.6% of cases, followed by the SYBR Green qPCR method (80.0%). Conclusions: The SYBR Green qPCR method is the same molecular biology method, but it can not analyze the germs at the same time. Bacterial culturing takes a lot of time if there is no selective culture medium. Therefore, the MRT-PCR method using newly developed primers and probes is considered to be the best method.
Objectives: The purpose of this study is to evaluate the non-enzymatic antioxidants stabilities in dentifrices by ascorbic acid and tocopherol according to the chemical condition. Methods: For the analysis of two antioxidants, HPLC UV detector system was used. HPLC was performed using sodium sulfate, acetonitrile(ACN), methanol(MeOH) and measuring absorbance at 240-295 nm. To confirm general pH reaction of two compounds, buffer solution was prepared for the analysis. The dentifrice was titrated by pH so as to examine the change of elapsed time in dentifrice. Linearity of calibration curve of two antioxidants was measured. Results: Each compound showed good linearity at optimized wavelength as well as showing good precision. General pH reaction of two antioxidants was examined. Ascorbic acid showed the highest residue(63.23%) at pH 10 and the lowest residue(2.77%) at pH 4. Tocopherol showed the highest residue(55.70%) at pH 7 and the lowest residue(3.31%) at pH 4. As a result of changing elapsed time of antioxidants in dentifrice by pH, components were remained stably at low temperature(39.2℉) and pH 7. Conclusions: It is necessary to keep dentifrice including ascorbic acid and tocopherol, and non-enzymatic antioxidants at pH 7 and low temperature for improving chemical stability.
Objectives: The purpose of the study was to investigate the professionalism identity and related factors in dental hygienists. Methods: A self-reported questionnaire was completed by 347 dental hyfienists from March 5 to May 31, 2015. Except incomplete 39 answers, data were analyzed by frequency analysis, ANOVA, Pearson``s correlation coefficient, and multiple regression analysis using SPSS 22.0. The questionnaire comprised 25 items including professionalism scale by Hall, Snizek, and Anderson by Likert 5 point scale. Results: Professionalism scale revealed self-regulation(3.65±0.58), service conviction(3.64±0.62), skills and training(3.21±0.70), sense of professional vocation(3.20±0.65), and thought about professional group(3.08±0.78). The professionalism identity by general characteristics showed significant differences and there were positive correlations in main factors. The identity level was influenced by internal factors rather than economic compensation. Conclusions: There were positive correlation in main factors. The identity level was influenced by internal factors.

Osteoblasts mineralize bone matrix in the process of bone formation and osteoblastic differentiation is an important step of bone formation. Some drugs have useful effects for bone formation in oriental medicine. In this study, to confirm the anti-inflammation and osteoblast differentiation effect of Pharbitidis Semen extract used in oriental medicine, we investigated cell viability, Alizarin red S assay, and Alkaline phosphatase(ALP) activity with Pharbitidis Semen extract. Also, we studied the inhibition effect of nitric oxide(NO) generation with Pharbitidis Semen extract in RAW 264.7 cell. Our results showed that Pharbitidis Semen extract had almost no cytotoxicity to the cell and it significantly enhanced the induction of ALP activity and Alizarin red S staining for osteogenesis in MC3T3-E1 cell. Also, all Pharbitidis Semen extract inhibited the LPS-induced NO generation in RAW 264.7 cell. Especially, Pharbitidis Semen ethyl acetate extract and Pharbitidis Semen butanol extract showed to be powerful inhibitors of NO generation. In conclusion, Pharbitidis Semen extract increased the ALP activity, Alizarin red S assay activity for bone formation and attenuated NO generation for inflammation. These results suggest that Pharbitidis Semen extract may act as a successful drug for osteogenic differentiation in vitro and could have the potential to be use as a therapeutic agent for bone formation.
Purpose: The purpose of this study is dental technicians perception investigation about national health insurance of old persons denture in metropolitan area. Methods: A survey was performed on 103 subjects in metropolitan from January 14 to April 30 in 2009. The research tool was a structured questionnaire. The collected data was analyzed by frequency, descriptive and multiple linear regression. Results: Total dental technician who respond to questionnaire was 103(100.0%). Male respondent appeared by 66.0%. 30-year-old low respondent appeared by 60.2% and subjects lived the most in seoul(68.0%). When got married, was expose that realization about national health insurance of old person s denture was high(p<0.05). When made denture, was expose that realization about national health insurance of old person s denture was high(p<0.05). Also, When got married, was expose that necessity about national health insurance of old persons denture was high(p<0.001). Conclusion: When did national health insurance of old persons denture, there were a lot of problems. But, old people was need it. Therefore, I desire that this study is used to basic data of the national health insurance.
