        Detection of mPing mobilization in transgenic rice plants

        Background Various kinds of transposable elements (TEs) constitute high proportions of eukaryotic genomes. Although most of these TEs are not actively mobile, genome stress can induce mobilization of dormant TEs. Transgenic plants undergo tissue culture and subsequent whole-plant regeneration, which can cause genomic stress and in turn induce mobilization of inactive TEs. Objectives To investigate the activation of transposable elements on the genome wide of the GM plant. Methods Transposon activities were analyzed in three transgenic rice plants carrying the insect resistance gene Cry1Ac and an herbicide resistance gene by the transposon display technique. These three transgenic plants were derived from a leading Korean rice variety, Illmi. Results We detected seven mobile activities in the mPing element, which is a MITE family transposon. The identity of the novel fragments in the gel display was confirmed by checking TAA target site duplication via sequence analysis. The genomic integration sites were all on different chromosomes, and the integrations were specific to either one or two T1 transgenic lines, except for one common integration on chromosome 4. One integration was in the 5′-UTR of the Glycerol3-phosphate acyltransferase 8 gene, two integrations were in introns of expressed genes, and the other four integrations were in intergenic regions. Conclusion Thus, novel mobilization of dormant TEs occurs in transgenic plants, which must be considered in the generation of genetically modified crops (GM crops).

        NGS sequencing reveals that many of the genetic variations in transgenic rice plants match the variations found in natural rice population

        Background As the transformation process can induce mutations in host plants, molecular characterization of the associated genomic changes is important not only for practical food safety but also for understanding the fundamental theories of genome evolution. Objectives To investigate a population-scale comparative study of the genome-wide spectrum of sequence variants in the transgenic genome with the variations present in 3000 rice varieties. Results On average, we identified 19,273 SNPs (including Indels) per transgenic line in which 10,729 SNPs were at the identical locations in the three transgenic rice plants. We found that these variations were predominantly present in specific regions in chromosomes 8 and 10. Majority (88%) of the identified variations were detected at the same genomic locations as those in natural rice population, implying that the transgenic induced mutations had a tendency to be common alleles. Conclusion Genomic variations in transgenic rice plants frequently occurred at the same sites as the major alleles found in the natural rice population, which implies that the sequence variations occur within the limits of a biological system to ensure survival.

        Efficiency to Discovery Transgenic Loci in GM Rice Using Next Generation Sequencing Whole Genome Re-sequencing

        Molecular characterization technology in genetically modified organisms, in addition to how transgenic biotechnologies are developed now require full transparency to assess the risk to living modified and non-modified organisms. Next generation sequencing (NGS) methodology is suggested as an effective means in genome characterization and detection of transgenic insertion locations. In the present study, we applied NGS to insert transgenic loci, specifically the epidermal growth factor (EGF) in genetically modified rice cells. A total of 29.3 Gb (∼72× coverage) was sequenced with a 2 × 150 bp paired end method by Illumina HiSeq2500, which was consecutively mapped to the rice genome and T-vector sequence. The compatible pairs of reads were successfully mapped to 10 loci on the rice chromosome and vector sequences were validated to the insertion location by polymerase chain reaction (PCR) amplification. The EGF transgenic site was confirmed only on chromosome 4 by PCR. Results of this study demonstrated the success of NGS data to characterize the rice genome. Bioinformatics analyses must be developed in association with NGS data to identify highly accurate transgenic sites.

        The Effect of Leverage on Labor Cost Management

        [Purpose]The purpose of this study is to examine the effect of capital structure on managers’ labor cost management by focusing on the relations between leverage and hiring decisions. [Methodology]The research was conducted on firms that satisfy the sample selection criteria of this study in the Compustat database from 1976 to 2014. The wage model in Chemmanur, Cheng, and Zhang (2013) is used to test the hypotheses. [Findings]First, I find that firms with higher leverage hire fewer employees, consistent with the disciplining effect of debt. Second, I also find that leverage is negatively associated with total labor cost, which is the product of the number of employees and average employee wage. Finally, this effect is greater for firms with higher institutional shareholdings and higher bankruptcy risk. [Implications]My findings suggest that the effect of debt discipline and compensating wages simultaneously exist, but they affect firms’ human capital investment decisions through different channels. I show that debt disciplines manager to minimize overall labor costs, implying that the disciplining effect of debt on employee hiring outweighs the effect of debt on compensating wage documented in prior studies. The results also imply that labor cost management is crucial for firms facing greater bankruptcy risk.

        FlexPro MD, a Mixture of Krill Oil, Astaxanthin, and Hyaluronic Acid, Suppresses Lipopolysaccharide-Induced Inflammatory Cytokine Production Through Inhibition of NF-κB

        FlexPro MD® (FP-MD), a novel multi-ingredient dietary supplement formulation, has been demonstrated to relieve knee joint pain in humans. However, the mechanisms of action responsible for the activity of FP-MD have not been elucidated. In this study, we show the anti-inflammatory effects of FP-MD in RAW264.7 macrophage cells and mice challenged with lipopolysaccharide (LPS). FP-MD significantly inhibited the mRNA levels of pro-inflammatory cytokines, including interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), and IL-1β. In contrast, it elevated the mRNA levels of anti-inflammatory cytokine IL-10 in LPS-stimulated RAW264.7 cells. FP-MD markedly reduced LPS-induced phosphorylation levels of nuclear factor-κB (NF-κB) p65 and inhibitor of κB-α (IκB-α). Importantly, the anti-inflammatory effects of FP-MD were demonstrated in mice with LPS-induced inflammatory arthritis in which FP-MD significantly reduced the expression levels of pro-inflammatory cytokines and inflammatory markers. Thus, this study suggests that FP-MD has anti-inflammatory effects by inhibiting NF-κB that may offer a molecular basis for its pain relief property.

        초급 해금교육을 위한 숫자보 활용 연구

        숫자보는 18세기 유럽에서 만들어져 유럽 전역의 음악교육을 담당하였다. 19세기 일본을 거쳐 중국에 유입되어 중국 근대 음악교육에서부터 지금까지 중국과 그 이외의 여러 아시아 국가에서 민족음악 교육에 널리 사용되고 있다. 숫자보는 아라비아 숫자로만 이루어져 있으며 조표 이동에 상관없이 도가 숫자1이 되는 악보이다. 이러한 특성은 음악을 처음 접하는 자에게 보다 손쉽게 음악 교육을 할 수 있는 교재로 사용할 수 있다. 본 연구에서는 이런 숫자보를 이해하고 그 숫자보를 해금 초급 교육에 도입해 연구하였다 첫째, 숫자보의 이해에서는 숫자보는 아라비아 숫자 1~7까지로 이루어져 있으며 음정과 박자표기 방법을 이해 할 수 있다. 또 숫자보의 음악 표기방법과 표식은 거의 오선보와 동일하다는 것을 알 수 있다. 둘째, 해금 초급교육을 위한 숫자보 활용을 통해 해금의 바깥 줄(유현)기본연습, 안 줄(중현)기본 연습, 두 줄 교차연습, 기본 반음 연습을 초급자들에게 숫자보 연습곡들의 소개로 쉽고 간단하게 교육 할 수 있다는 것을 알 수 있다. 본 연구를 통하여 숫자보를 이해 와 활용 방법을 알 수 있으며 해금 초급 교육에 숫자보를 도입하여 보다 체계적인 초급 교육과 연습 방법을 제시 할 수 있다. Number notation(Sutjabo) was made in Europe in the 18th century and performed musical education over the European area, which flew in China in the 19th century via Japan, and is used as folk music education as well as Chinese modern musical education, widely in several Asian countries including China until now. Number notation(Sutjabo) is a music, composed of only Arabic numbers, of which ‘do’ is number 1, regardless of movement of key signature. This characteristic can be used as a textbook which can conduct musical education more easily for those who face music for the first time. This research comprehended this number notation(Sutjabo) and researched by introducing it to Haegeum beginners’ education. First, comprehension of number notation(Sutjabo) is composed of arabic number 1~7, tune and beat mark method can be comprehended. Also, music mark method of number notation(Sutjabo) and indication are almost same as staff notation. Second, it could be confirmed that basic practice of outer string(yuhyun) of Haegeum, inner string(junghyun) basic practice, two strings cross practice, and basic semi-tone practice can be easily and simply educated with the introduction of number notation(Sutjabo) etudes for the beginners through the utilization of number notation(Sutjabo) in Haegeum beginners’ education. The researcher considers that this research can suggest comprehension and utilization method of number notation(Sutjabo), also, suggest more systematic beginners’ education and practice method by introducing number notation(Sutjabo) in Haegeum beginners’ education.
