        Comparative Evaluation of Antioxidant Activities of Ethanol Extracts and Their Solvent Fractions Obtained from Selected Miscellaneous Cereal Grains

        잡곡류의 항산화활성을 조사하기 위해 국내산 11종의 잡곡으로부터 80% 에탄올 추출물을 얻어 DPPH- 및 ABTS-라디칼 소거활성을 측정한 결과, 황금찰수수(Sorghum bicolor L. Moench cv. Hwanggeumchalsusu), 찰수수(Sorghum bicolor L. Moench cv. Chalsusu) 및 식용피(Echinochloa esculenta)의 에탄올 추출물이 다른 잡곡류의 에탄올 추출물에 비해 높은 라디칼 소거활성을 나타내었다. 이들 황금찰수수, 찰수수 및 식용피의 에탄올 추출물을 n-hexane, methylene chloride, ethyl acetate 및 n-butanol로 분획하였을 때, 대부분의 라디칼 소거활성은 페놀성 화합물이 주로 함유되어 있는 것으로 나타난 ethyl acetate 분획과 butanol 분획에서 집중적으로 확인되었다. 특히, 황금찰수수의 ethyl acetate 분획과 butanol 분획의 라디칼 소거활성은 천연 항산화제인 α-tocopherol에 비해 더 높게 나타났다. 황금찰수수, 찰수수 및 식용피 유래의 ethyl acetate 분획과 butanol 분획은 지질 과산화를 저해하는 것으로 ferric thiocyanate (FTC)와 thiobarbituric acid (TBA) 방법에 의해 확인되었다. 황금찰수수, 찰수수 및 식용피 유래의 ethyl acetate 분획의 경우, tertiary-butyl hydroperoxide (TBHP) 처리에 의해 HL-60 세포에서 유도되는 에폽토시스 현상들 즉, sub-G1 세포 등장, Δψm 소실, caspase-9과 caspase-3의 활성화, 그리고 PARP와 lamin B의 분해 등을 저해하는 것으로 나타났다. 이러한 결과들은 황금찰수수, 찰수수 및 식용피가 효율적인 항산화 활성을 지니고 있으며 산화적 손상에 의해 매개되는 에폽토시스를 억제할 수 있음을 보여준다. 아울러 이러한 연구결과들은, 황금찰수수, 찰수수 및 식용피가 산화적 스트레스로부터 세포를 보호하는 항산화 식이소재가 될 수 있음을 시사한다. To examine the antioxidant activities of 11n selected miscellaneous cereal grains (proso millet, yellow glutinous proso millet, hwanggeumchal sorghum, glutinous sorghum, white glutinous sorghum, yellow glutinous foxtail millet, nonglutinous foxtail millet, green glutinous foxtail millet, golden foxtail millet, barnyard millet, and adlay), the free radical-scavenging activities of 80% ethanol extracts of the individual grains were investigated using 1,1-diphenyl-2-picryl-hydrazl (DPPH) and 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) methods. The ethanol extracts of hwanggeumchal sorghum, glutinous sorghum, and barnyard millet grains exhibited more potent free radical-scavenging activities as compared to the other grains. When these three ethanol extracts were sequentially fractionated with n-hexane, methylene chloride, ethyl acetate, and n-butanol, the majority of the antioxidant activities were detected in the ethyl acetate and butanol fractions in which phenolic ingredients were abundant. The ethyl acetate and butanol fractions of hwanggeumchal sorghum and the ethyl acetate fraction of glutinous sorghum showed higher antioxidant activity than that of α-tocopherol. Both ferric thiocyanate (FTC) and thiobarbituric acid (TBA) methods demonstrated that these organic solvent fractions could inhibit lipid peroxidation. The ethyl acetate fractions from hwanggeumchal sorghum, glutinous sorghum, and barnyard millet grains could suppress tertiary-butyl hydroperoxide (TBHP)-induced apoptotic events, including sub-G1 peaks, Δψm loss, activation of caspase-9 and caspase-3, and cleavage of PARP and lamin B, in human HL-60 cells. These results show that the grains of hwanggeumchal sorghum (Sorghum bicolor L. Moench cv. Hwanggeumchalsusu), glutinous sorghum (Sorghum bicolor L. Moench cv. Chalsusu), and barnyard millet (Echinochloa esculenta) possess efficient antioxidant activity, which could protect cells from oxidative stress-mediated cytotoxicity.

        혈관육종에 대한 임상 및 병리조직학적 분석

        Background: Cutaneous angiosarcoma (AS) is a rare malignant tumor of endothelial origin with a very poor prognosis, frequent recurrences, and high metastatic potential. Clinical suspicion is often raised too late, but histological findings and immunohistochemical assays have proved to be very helpful in the diagnostic process. Objective: The purpose of this study was to evaluate the clinical and histopathologic features of angiosarcoma. Methods: We retrospectively analyzed 12 patients with angiosarcoma who had visited our department of dermatology in Gyeongsang National University Hospital between 1998 and 2013. The clinical features, evolution, treatment, and outcome were examined, and biopsy specimens were reviewed by a trained dermatologist, with subsequent immunohistochemical assessment. Results: Cutaneous AS was clinically diagnosed in 3 of 12 patients. There were many clinical misdiagnoses including lupus erythematosus, metastatic cancer, and infectious granuloma. The male to female ratio was 1:0.71 and the mean age of the patients was 66.25 years. The most frequent site of angiosarcoma was the scalp, in 8 of 12 patients. Histopathologically, well-differentiated angiosarcoma was composed of vascular channels, lined by flattened atypical endothelial cells that could be distinguished from their benign counterparts. Variable differentiation may be observed even in the same tumor. Immunohistopathologic analysis showed that the tumor cells were positive for CD31, CD34, Ki 67, and Factor VIII-related antigen. Conclusion: This study provides useful data on the clinical and histopathologic characteristics of angiosarcoma in the Korean population. (Korean J Dermatol 2015;53(3):188∼195)

        메탄올 용매에서 산소 첨가된 다섯자리 Schiff Base Cobalt(Ⅲ) 착물들의 활성 촉매에 의한 Hydrazobenzene의 산화반응

        산소가 포화된 메탄올 용액에서 다섯자리 Schiff base cobalt(II) 착물인 [Co(II)(Sal-DPT)(H$_2$O)] 와 [Co(II)(Sal-DET)(H$_2$O)]들의 활성촉매에 의한 hydrazobenzene(H$_2$AB)의 산화 주생성물은 trans-azobenzene(trans-AB)이다. UV-visible분광광도법에 의해 이들 반응의 속도상수 $k=6.06{\times}10^{-3}sec^{-3}$ 및 $2.50{\times}10^{-3}sec^{-1}$로 주어짐을 알았다. 균일 산화 활성촉매에 의한 H$_2$AB의 산화반응 메카니즘은 다음과 같은 과정으로 주어진다. H$_2$AB + Co(II)(L)(H$_2$O) + O$_2$ $\rightleftharpoons^K_{methanol}Co(III)(L)O_2{\cdot}H_2AB + H_2O\longrightarrow^{k}Co(II)(L) + trans-AB + H_2O_2$ (L: Sal-DPT and Sal-DET). Homogeneous catalytic oxidation of hydrazobenzene was investigated by employing pentadentate Schiff base complexes such as [Co(II)(Sal-DPT)(H$_2$O)] and [Co(II)(Sal-DET)(H$_2$O)] in oxygen-saturated methanol solvent. The oxidation product of hydrazobenzene(H$_2$AB) was trans-azobenzene(trans-AB). The rate constants of oxidation reaction measured by UV-visible spectrophotometry were observed as $6.06{\times}10^{-3}sec^{-1}$ for [Co(II)(Sal-DPT)(H$_2$O)] and $2.50{\times}10^{-3}sec^{-1}$ for [Co(II)(Sal-DET)(H$_2$O)]. The mechanism of oxidation reaction for H$_2$AB by homogeneous activated catalysts has been proposed as following. H$_2$AB + Co(II)(L)(H$_2$O) + O$_2$ $\rightleftharpoons^K_{MeOH}Co(III)(L)O_2{\cdot}H_2AB + H_2O\longrightarrow^{k}Co(II)(L) + trans-AB + H_2O_2$ (L: Sal-DPT and Sal-DET)

        신경섬유종증과 동반되지 않은 악성 신경집종양

        Malignant peripheral nerve sheath tumor (MPNST) is a rare entity of all soft tissue sarcomas. It has four different types of glandular, melanocytic, rhabdomyoblastic, and epithelioid. Epithelioid MPNST is composed of predominantly epithelioid cells. In contrast, typical MPNST is consisted of spindle cells, which is arranged in a fascicular or whirling pattern. A 33-old man showed two skin-color to reddish brown protruding nodules on the left thigh. Microscopically, one nodule was made up of epithelioid cell, primarily. In addition, the other nodule was composed of epithelioid cells and spindle cells, which also showed Antoni A and B. Immunohistochemical study showed the cells being positive for S-100 protein (weak and focal) and neuron-specific enolase, and negative for HMB-45, smooth muscle actin. Herein, we report a case of two different forms of malignant nerve sheath tumor on the left thigh, which was not associated with neurofibromatosis-1. (Korean J Dermatol 2012;50(4):362∼366)

        북한 서해안권의 항만선택요인 평가에 관한 연구

        한반도의 서해안권은 동남아시아, 태평양, 중국의 동북3성, 러시아 등 글로벌공급사슬관리를 연결하는 중요한 지리적 위치에 있다. 북한은 서해안경제특구를 활용하여 경제발전을 진행하며 항만물류 영역에서 남북 협력 사업은 남북한과 중국동북 3성 및 러시아 등 유라시아 연계된 국제 복합물류망 구축이 목표이다. 북한의 서해안 항만개발은 남북한 항만공동개발 및 공동운영이 필요하며 중요한 전략적 요소가 된다. 본 연구는 북한 항만의 선택적 정책대안을 제시하기 위하여 측정영역과 측정요소를 설계하고 분석방법은 AHP(Analytic Hierarchy Process) 기법을 적용하였다. 본 연구 분석 결과는 항만공사 등의 민간차원에서 공공기관이 참여하는 비즈니스 모델 개발이 요구되며. 제2계층의 평가 속성은 해운·항만 기반 구축이 0.296로 가장 높은 수치를 보이고 있다. 북한 항만 선택세부 평가요소의 종합중요도는 정부의 관련기관이 참여하는 형태의 거버넌스 구축이 0.073이다. 본 연구에서 북한 서해안의 최종적으로 항만개발선택은 신의주특구로 0.345을 보여주고 있다. 신의주 특구는 북한 외에 중국 단동과 연계성이 매우 높으며 제도적 측면에서 안정적인 법적 장치의 운용 가능성이 높기 때문으로 해석된다. The west coast bay of the Korean Peninsula is located in China's three northeastern provinces and Russia. It is an important scientific position that connects Europe with Asia and the Pacific. Port development in North Korea requires joint development and operation of ports between South and North Korea. This study designed the measurement area and factors to present optional policy alternatives to North Korean ports; the analytic hierarchy process technique was applied. North Korea wants to use special economic zones to develop its economy. The goal of inter-Korean cooperation projects in the port logistics area is to establish an international complex logistics network linked to Eurasia, including the aforementioned areas. A business model involving public institutions should be developed at the private port authority level. The second tier's valuation attribute is the highest at 0.296 in the construction of shipping and port infrastructure. The overall importance of North Korea's port selection assessment factors is 0.073 regarding governance. North Korea's final port selection is 0.345 in the Sinuiju Special Zone. This zone has a strong connection with Dandong, China owing to its synthetically stable legal mechanism
