        역전사중합효소 연쇄반응-효소면역측정법을 활용한 장내바이러스 감염 진단

        Background : Enteroviruses are known as major pathogen for aseptic meningitis. Although rapid diagnosis for enteroviruses is very essential to exclude bacterial infections in patients with meningitis, classical diagnostic method based on virus isolation is not practicable for timely treatment of patients due to its laborious and time-consuming procedure. Recently molecular methodologies as alternatives are routinely used for rapid and sensitive diagnosis for enteroviruses infections. Methods : Reverse transcription (RT)-PCR ELISA kit for targeting 5’non-coding region (NCR) with highly conserved genetic identity among all genotypes of enteroviruses was introduced in this investigation. RT-PCR ELISA was evaluated about sensitivity and specificity through virus isolation using clinical specimens from patients suspected of enteroviral infections and enteroviral isolates comparing with conventional RT-PCR identifying them. Results : The detection limit of the RT-PCR ELISA was up to 10-100 folds higher than virus isolation using cell culture and conventional RT-PCR. On comparison between above two methods, the detection rate of RT-PCR ELISA for clinical specimens from patients with aseptic meningitis was 7% higher than that of conventional RT-PCR targeting 5’NCR (P=0.016). Conclusions : Our results suggest that RT-PCR ELISA developed in this study could be an alternative diagnostic method for the detection of enteroviral genome with high sensitivity and specificity. 배경 : 장바이러스(enterovirus)는 무균성 수막염의 주요 병 원체이다. 무균성 수막염이 의심되는 경우, 장바이러스에 대한 정확한 진단이 필수적이지만 기존의 세포배양을 통한 바이러스 의 분리법은 많은 시간과 노력이 필요하다. 그러므로 최근에는 장바이러스 진단을 위하여 신속하고 민감한 분자생물학적 방법 이 사용되고 있다. 방법 : 장바이러스의 유전자 중 잘 보존된 부분인 5’non-coding region에 대한 소식자를 이용하여 reverse transcription (RT)-PCR ELISA 키트를 제작하였다. 제작된 RT-PCR ELISA 키트는 RT-PCR법과 비교되었는데, 바이러스 분리주와 무균성 수막염이 의심되는 환자 검체에 대하여 민감도와 특이도를 측 정하였다. 결과 : RT-PCR ELISA법의 검출한계는 세포배양을 통한 바 이러스 분리법과 RT-PCR법보다 10-100배 높았으며, 또한 무 균성 수막염 환자의 임상 시료를 대상으로 한 RT-PCR ELISA 법의 검출률이 RT-PCR법 보다 7% 높게 나타났다(P=0.016). 결론 : 본 연구에서 활용된 RT-PCR ELISA법은 높은 민감 도와 특이도를 보여 장바이러스 진단법으로 이용할 것을 제안 한다.`

      • 한국인집단의 D14S299 유전자좌내 microsatellite의 다형현상

        The allele frequency of D14S299 microsatellite locus was analyzed in 242 normal blood from Korean population. Using the polymerase chain reaction and polyacrylamide gel electrophoresis. we examined the allele and genotype frequency. The size of 0145299 PCR fragments ranged from 299 bp to 331 bp. The most frequent allele in the Korean subjects was allele 30 (44.9%) followed by allele 31 (25.4%), allele 32 (12.4%), and minor alleles (17.3%). The heterozygosity of D14S299 was 0.710. The polymorphism information content (PIC) and power of discrimination (POE) were 0.67 and 0.880, respectively in the present study. The genotype distribution and allele frequency of D14S299 microsatellite in the Korean subjects was similar to those of Japanese and Chinese populations including in the same Asian Population.

        Evaluation of a New Immunochromatographic Assay Kit for the Rapid Detection of Norovirus in Fecal Specimens

        Rapid and accurate detection of norovirus is essential for the prevention and control of norovirus outbreaks. This study compared the effectiveness of a new immunochromatographic assay kit (SD BIOLINE Norovirus; Standard Diagnostics, Korea) and real-time reverse transcription-PCR (RT-PCR) for detecting norovirus in fecal specimens. Compared with real-time RT-PCR, the new assay had sensitivity, specificity, positive predictive value,and negative predictive value of 76.5% (52/68), 99.7% (342/343), 98.1% (52/53), and 95.5% (342/358), respectively. The sensitivity of the assay was 81.8% (18/22) for GII.3 and 75.7% (28/37) for GII.4. None of the 38 enteric virus-positive specimens (3 for astrovirus, 5 for enteric adenovirus, and 30 for rotavirus) tested positive in the cross-reactivity test performed by using this assay. The new immunochromatographic assay may be a useful screening tool for the rapid detection of norovirus in sporadic and outbreak cases; however, negative results may require confirmatory assays of greater sensitivity.

        Enterovirus infection in Korean children and antienteroviral potential candidate agents

        Although most enterovirus infections are not serious enough to be life threatening, several enteroviruses such as enterovirus 71 are responsible for severe, potentially life-threatening disease. The epidemic patterns of enteroviruses occur regularly during the year, but they may change due to environmental shifts induced by climate change due to global warming. Therefore, enterovirus epidemiological studies should be performed continuously as a basis for anti-viral studies. A great number of synthesized antiviral compounds that work against enteroviruses have been developed but only a few have demonstrated effectiveness in vivo. No proven effective antiviral agents are available for enterovirus disease therapy. The development of a new antiviral drug is a difficult task due to poor selective toxicity and cost. To overcome these limitations, one approach is to accelerate the availability of other existing antiviral drugs approved for antiviral effect against enteroviruses, and the other way is to screen traditional medicinal plants.

      • Ti. Ni. Mg 분말이 첨가된 복합재료의 전자파 차폐효과에 관한 연구

        Electromagnetic interference(EMI) shielding effectiveness of glass and carbon fiber reinforced composites with Ti. Ni. Mg powder were investigated. We fabricated glass and carbon fiber reinforced composites filled with metal powder and nano carbon black. The measurements of shielding effectiveness(SE) were carried out frequency range 300MHz – 1GHz for commercial purposes such as electric or telecommunication devices. The return loss and loss due to absorption were also measured as a function of frequency in the micro wave(300MHz‐1GHz) region. It is observed that the SE of the composites is the frequency dependent, increase with the increase in filler loading. The Mg metal powder filled composite show higher SE compared to that of carbon black. The Mg metal powder/epoxy composite were shown to exhibit up to 40dB of SE. The result indicate that the composite having higher filler loading can be used for the purpose of EMI shielding as well as for some microwave applications.

        This study investigated electromagnetic interference(EMI) shielding effectiveness(SE) of the copper mesh and aluminum film / fiber reinforced composites. We fabricated glass and carbon fiber reinforced composites filled with nano carbon black. The measurements of SE were carried out frequency range from 300MHz to 1.5GHz. It is observed that the SE of the bonded copper mesh and aluminum film. The aluminum film bonded composites showed higher SE compared to that of copper mesh. The aluminum film bonded epoxy composite was shown to exhibit up to 55~75dB of SE. The result that aluminum film bonded composite can be used for the purpose of EMI shielding as well as for some microwave applications.

      • 섬유강화 복합재료의 强度에 습기가 미치는 影響

        The purpose of this investigation is to estimate the strength with the variations of shape and the adhesive condition at the fiber/matrix micro interface of E-glass/PP laminates. To promote the degradation of the adhesive condition at the fiber/matrix micro interface without matrix dissolution loss, low-, hot-wet and spiking tests were carried out. The absorption properties and the tensile properties were compared according to the fiber orientation and the content. The results show that, firstly, saturated moister absorption was reached at 5cycle and their absorptions of RD-40, UD-42, and UD-50 are 0, 0.63%, 0.60%, respectively. Secondly, all the specimens investigated were mostly degraded at 5cycle, whereas UD-50 having ellipse shaped ellipse the least decrement of strength.

      • 점증적 최대부하 운동이 호흡순환계 및 젖산변화에 미치는 영향

        This study was to investigate a cardiorespiratory response and the chang of blood lactate concentration between untrained men and judo fellows after maximal exercise. The result of this study was as follows: All of the variables(HR, VE, and VO2max) were non significant as well as RER in a cardiorespiratory response. In addition, both between groups and interaction were non significant the change of a blood lactate concentration. However, not only VO2 was sigpificant(p<.05) in the comparison of between groups in a cardiorespiratory response but the chang blood lactate concentration was significant(p<.01) in measuring time. As a result of this study, the study of a cardiorespiratory variables and the reduction of blood lactate concentration related to recovery on fourties subjects after maximal exercise didn't get particular result but it is showed the condition of health of middle aged men indirectly as well as the difference of between untrained men and judo fellows. Therefore, it is showed that both untrained men and judo fellows need a exorcise, which is suggested a objective and scientific the method and the need of motivation for health.

        본 연구에서는 수소 연료전지의 엔드플레이트와 외부 연결 부분의 금속을 진동시험기를 이용하여 접촉저항을 측정하였다. 수소 연료전지의 집접판에서의 전류밀도의 변화와 온도 분포의 변화를 비교해 볼 수 있었으며, 접촉저항을 측정 할 수 있었다. 진동시험기는 5∼50 ㎐의 범위까지 설계하였으며 진동에 대한 접촉저항의 변화도 확인 할 수 있었다.

        Complete Sequence Analysis and Antiviral Screening of Medicinal Plants for Human Coxsackievirus A16 Isolated in Korea

        Objectives: Coxsackievirus A group 16 strain (CVA16) is one of the predominant causative agents of hand, foot, and mouth disease (HFMD). Methods: Using a specimen from a male patient with HFMD, we isolated and performed sequencing of the Korean CVA16 strain and compared it with a G10 reference strain. Also, we were investigated the effects of medicinal plant extract on the cytopathic effects (CPE) by CPE reduction assay against Korean CVA16. Results: Phylogenetic analysis showed that the Korean CVA16 isolate belonged to cluster B-1 and was closely related to the strain PM-15765-00 isolated in Malaysia in 2000. The Korean CVA16 isolate showed 73.2% nucleotide identity to the G10 prototype strain and 98.7% nucleotide identity to PM-15765-00. Next, we assessed whether the Korean CVA16 isolate could be used for in vitro screening of antiviral agents to treat HFMD infection. Vero cells infected with the Korean CVA16 isolate showed a cytopathiceffect 2 days after the infection, and the treatment of cells with Cornus officinalis, Acer triflorum, Pulsatilla koreana, and Clematis heracleifolia var. davidiana Hemsl extracts exhibited strong antiviral activity against CVA16. Conclusion: Collectively, our work provides potential candidates for the development of vaccine and novel drugs to treat the CVA16 strain isolated from a Korean patient.
