        Transcriptome Profiling and Characterization of Drought-Tolerant Potato Plant (Solanum tuberosum L.)

        Potato (Solanum tuberosum L.) is the third most important food crop, and breeding drought-tolerant varieties is vital research goal. However, detailed molecular mechanisms in response to drought stress in potatoes are not well known. In this study, we developed EMS-mutagenized potatoes that showed significant tolerance to drought stress compared to the wild-type (WT) ‘Desiree’ cultivar. In addition, changes to transcripts as a result of drought stress in WT and droughttolerant (DR) plants were investigated by de novo assembly using the Illumina platform. One-week-old WT and DR plants were treated with -1.8 Mpa polyethylene glycol-8000, and total RNA was prepared from plants harvested at 0, 6, 12, 24, and 48 h for subsequent RNA sequencing. In total, 61,100 transcripts and 5,118 differentially expressed genes (DEGs) displaying up- or down-regulation were identified in pairwise comparisons of WT and DR plants following drought conditions. Transcriptome profiling showed the number of DEGs with up-regulation and down-regulation at 909, 977, 1181, 1225 and 826 between WT and DR plants at 0, 6, 12, 24, and 48 h, respectively. Results of KEGG enrichment showed that the drought tolerance mechanism of the DR plant can mainly be explained by two aspects, the ‘photosyntheticantenna protein’ and ‘protein processing of the endoplasmic reticulum’. We also divided eight expression patterns in four pairwise comparisons of DR plants (DR0 vs DR6, DR12, DR24, DR48) under PEG treatment. Our comprehensive transcriptome data will further enhance our understanding of the mechanisms regulating drought tolerance in tetraploid potato cultivars.

        Expression of Jerusalem artichoke (Helianthus tuberosus L.) fructosyltransferases, and high fructan accumulation in potato tubers

        Fructans are polymers of fructose that are present as storage carbohydrates in various plants. Jerusalem artichoke (Helianthus tuberosus L.) contains a high amount of inulin. Two enzymes are involved in inulin biosynthesis. The sucrose:sucrose 1-fructosyltransferase (1-SST) enzyme mainly catalyzes the synthesis of 1-kestose from sucrose. In the next step, fructan:fructan 1-fructosyltransferase (1-FFT) catalyzes the synthesis of inulin from 1-kestose. In this study, the Ht1-SST and Ht1-FFT genes were isolated from Jerusalem artichoke and expressed in potato (Solanum tuberosum L.), either separately or together, via Agrobacterium-mediated transformation. Transgenic potato tubers overexpressing Ht1-SST accumulated 1-kestose to a high level (up to 3.36 mg/g), while tubers overexpressing both Ht1-SST and Ht1- FFT accumulated up to 3.14 mg/g short-chain inulin-type fructans, with the degree of polymerization (DP) ranging from 3 to 5, excluding high DP inulins. Transgenic potato plants accumulated fructo-oligosaccharides to a high level, following the fructan biosynthetic pathway of Jerusalem artichoke, and therefore present a high potential for the mass production of inulin through established potato breeding and cultivation methods.

        대용량 LMS 로그 데이터를 이용한 심층신경망 기반 대학생 학업성취 조기예측 모델

        Log data accumulated in the Learning Management System (LMS) provide high-quality information for the learning process of students. Until now, various studies have been conducted to predict students’ academic achievement using LMS log data. However, previous studies were based on relatively small sample sizes of students and courses, limiting the possibility of generalization. This study developed and validated a deep neural network model for the early prediction of academic achievement of college students using massive LMS log data. To this end, we used 78,466,385 cases of LMS log data and 165,846 cases of grade data. The proposed model predicted the excellent-grade students with a high level of accuracy from the beginning of the semester. Meanwhile, the prediction accuracy for the moderate and underachieving groups was relatively low, but the accuracy improved as the time points of the prediction were delayed. This study is meaningful in that we developed an early prediction model based on a deep neural network with sufficient accuracy for practical utilization by only using LMS log data. 학습관리 시스템(LMS)에 축적되는 로그 데이터는 학습 과정에 대한 양질의 정보를 제공한다. 지금까지 LMS 로그 데이터를 활용한 학업성취 예측 연구가 다양하게 수행되었지만, 상대적으로 적은 양의 학생 및 수업 데이터에 기반하고 있어 연구 결과 일반화 가능성에 한계가 존재한다. 본 연구는 대용량 LMS 로그 데이터를 이용해 대학생 학업성취를 조기예측하는 심층신경망 모델을 개발하고 성능을 검증했다. 이를 위해 가명화 처리된 LMS 로그 데이터 78,466,385건과 성적 데이터 165,846건을 활용했다. 그 결과, 본 연구에서 제안하는 예측모델은 우수학생 집단을 학기 초부터 높은 수준의 정확도로 예측하였다. 한편 보통 및 저성취 집단에 대한 예측 정확도는 제한적인 수준이었지만, 예측시점이 늦을수록 향상되었다. 본 연구의 결과는 순수 LMS 로그 데이터만을 이용해 실제로 활용할 수 있을 정도의 일반화 성능을 가진 심층신경망 기반 조기예측 모델을 구현했다는 의의가 있다.

        Transient Erythropoietin Overexpression with Cucumber Mosaic Virus Suppressor 2b in Nicotiana benthamiana

        Erythropoietin (EPO) is a glycoprotein hormone that regulates red blood cell formation in mammals. It also plays an important role in wound healing and the brain’s response to neuronal injury. Recombinant EPO has been produced in various hosts including Escherichia coli, insect cells, and yeast. Plant systems are also useful for the production of recombinant proteins. In this study, we used Nicotiana benthamiana for the production of recombinant human EPO (rhEPO) using an Agrobacterium-mediated transient expression system. We evaluated the effect of repeated agroinfiltration and Agrobacterium density on the rhEPO production. In addition, the rhEPO expression vector was coinfiltrated with a vector expressing cucumber mosaic virus suppressor 2b (CMV2b), which suppresses posttranscriptional gene silencing. We found that rhEPO expression was increased approximately 5.5-fold in N. benthamiana leaves coinfiltrated with CMV2b. In contrast, neither Agrobacterium density nor the number of infiltrations influenced rhEPO expression.

        한국판 청소년용 긍정적 정신건강 척도의 타당화

        The purpose of this study was to validate the Korean Version of the Adolescent Positive Health Scale. The confirmatory factor analysis was conducted with 1,668 adolescents (960 middle school students and 708 high school students) who completed the Korean Version of the Adolescent Positive Health Scale. Convergent and discriminant validity were investigated using an independent sample of 189 adolescents (99 middle school students and 90 high school students) who responded to the following questionnaires: the Korean Version of the Adolescent Positive Health Scale, the School Adjustment Scale, the Ego Strength Scale, and the Social Support Appraisal Scale. The results showed that reliability coefficients (Cronbach’s α) of the Korean Version of the Adolescent Positive Health Scale were .92 for the mental well-being factor and .82 for the psychological maladjustment factor. According to the confirmatory factor analysis, a model with the correlation between the mental well-being and psychological maladjustment factor evidenced a better fit than a model without correlation between the two factors. The Mental Well-being Scale positively correlated with the School Adjustment Scale, the Ego Strength Scale, and the Social Support Appraisal Scale while the Psychological Maladjustment Scale negatively correlated with those scales. Based on these findings, it was discussed that a longitudinal study would be necessary to investigate the effects of positive mental health type on the prevention of mental disorders and process of recovery. 본 연구의 목적은 한국판 청소년용 긍정적 정신건강 척도를 타당화하는 것이다. 본 연구에서는 확인적 요인분석을 위해 중학생 960명과 고등학생 708명, 총 1,668명에게 한국판 청소년용 긍정적 정신건강 척도를 실시하였다. 수렴 및 변별타당도 분석을 위해서 중학생 99명과 고등학생 90명, 총 189명을 대상으로 한국판 청소년용 긍정적 정신건강 척도, 학교생활적응 척도, 자아강도 척도, 사회적지지 척도를 실시하였다. 본 연구 결과, 한국판 청소년용 긍정적 정신건강 척도의 내적 일치도 계수(Cronbach’s α)는 정신적 웰빙 .92 그리고 심리적 부적응 .82였다. 확인적 요인분석 결과, 정신적 웰빙과 심리적 부적응의 두 요인 간 상관이 있는 모델이 그 두 요인 간 상관이 없는 모델보다 더 좋은 적합도를 나타냈다. 정신적 웰빙 척도는 학교생활적응 척도, 자아강도 척도, 사회적지지 척도와 정적인 상관을 보이는 반면, 심리적 부적응 척도는 부적인 상관을 나타냈다. 이러한 결과를 바탕으로, 긍정적 정신건강 유형이 정신장애의 예방과 회복 과정에 미치는 효과를 분석하는 종단 연구의 필요성에 대해 논의했다.

        일상 생활 속 자아 고갈 및 심리적 부담감이 피로도에 미치는 효과: 자기애적 성격을 중심으로

        Resisting the desire to conflict with goals in everyday life may lead to ego depletion and subsequent fatigue. However, even when goals and desires are not in conflict with each other, the psychological burden of goal achievement may also be a source of fatigue. The aim of the present study is to examine the moderating effect of narcissistic personality on the relationship between psychological burden and fatigue. A total of 104 adults participated in this study. Using the experience sampling method, we measured daily ego depletion and psychological burden seven times a day for one week. The level of fatigue was also measured at the end of each day. A series of multilevel analyses revealed that daily ego depletion and psychological burden positively predicted daily fatigue level. Covert narcissism was more related to psychological burden and fatigue than overt narcissism. Participants with overt narcissism were more severely fatigued when they experienced psychological burden than non-narcissistic participants. Lastly, we discussed the implications and limitations of the present study.

        Study Examines Difference between Communal Narcissism and Altruism in Korean College Students Using Close-Other Reports

        This study aimed to examine the difference between communal narcissism and altruism using close-other reports, especially in collectivistic cultures (e.g., Korea). There may be differences between individualistic and collectivistic cultures in the evaluation of communality. However, research on acquaintance evaluations of the difference between communal narcissism and altruism has never been conducted in a collectivistic culture. Accordingly, 179 Korean college students (115 females) completed self-report questionnaires to assess communal narcissism and altruism, selecting three close others who rated the psychological adjustment of the participants in terms of communality, altruism, and well-being. We found that self-reported communal narcissism was positively correlated with self-reported altruism but not significantly correlated with close-otherreported altruism. Additionally, the effect of self-reported communal narcissism on psychological adjustment as evaluated by close others was not significant after controlling for the effect of self-reported altruism. However, after controlling for the effect of self-reported communal narcissism, the effect of self-reported altruism on psychological adjustment as evaluated by close others was significant. Although communal narcissism and altruism are closely related in self-reports, findings based on reports of close others provide empirical evidence that they are distinguishable personality traits.

        캘러스 활용도를 향상시키기 위한 벼(Oryza sativa L.) 형질전환 시스템 구축

        Plant molecular farming has attracted a lot of attention lately in the field of mass production of industrially valuable materials by extending application of the plant as a kind of factory concept. Among them, protein expression system using rice(Oryza sativa L.) callus is a technology capable of mass culture and industrialization because of a high expression rate of a target protein. This study was carried out to develop an Agrobacterium-mediated transformation system to increase the utilization of rice callus. The transformation efficiency was improved by using the hand when seeds were de-husked for callus induction. Furthermore, we were possible induction of callus from 6 years old seed smoothly. Selection of the callus contained the target gene was required a cultivation period of at least 3 weeks, and the most efficient selection period was after 6 weeks of culture including one passage. This selection was confirmed that the gene was stably inserted into the genomic DNA of the plant cell by the southern blot analysis and progeny test. Such an efficient selection system of rice callus that can be cultured in the long term will be contribute to the industrialization of useful recombinant proteins using rice.

        정신건강 2요인 모델 타당화 연구: 젊은 성소수자(LGB) 집단을 중심으로

        본 연구에서는 국내 성소수자(LGB) 집단을 대상으로 정신건강 2요인 구조의 타당성을 검증하고자 했다. 이를 위해 퀴어 포털사이트에서 모집한 464명을 대상으로 정신적 웰빙 척도(K-MHC-SF)와 한국형 정신장애 척도(K-MDI), LGB 정체성 척도(LGBIS)를 실시했다. 그 후 모형 적합도를 비교하기 위해 연구 모형과 경쟁 모형에 대해 각각 확인적 요인분석을 실시했으며, 이를 바탕으로 Keyes(2002)의 정신건강 집단 분류에 따라 성소수자의 정체성 측면에서 심리사회적 적응도의 차이가 있는지 확인하기 위해 일원변량분석과 사후분석을 실시했다. 연구결과 정신건강 모델은 정신건강을 긍정적 정신건강과 정신장애의 단일축으로 보는 단일모델과 정신건강을 긍정적 정신건강과 정신장애의 독립적인 관계로 보는 2요인 독립모델에 비해 두 변인이 독립적인 동시에 상호상관이 있다고 가정한 2요인 상관모델이 더 적합한 것으로 나타났다. 또한 성소수자 집단 중 완전정신건강 집단은 나머지 집단에 비해 LGB 정체성 척도 하위 변인에서 심리사회적응 수준이 더 뛰어났고, 단순장애 집단이 단순쇠약 집단보다 심리사회적 적응 수준이 더 뛰어났다. 이러한 결과에 기초하여 성소수자 집단을 대상으로 한 긍정적 정신건강 연구와 개입의 필요성에 대해 논했다.
