        가미십전대보탕의 RAW 264.7 세포에서 항산화 및 항염증 효과

        가미십전대보탕(이하 KSD로 표기) 추출물은 생약 성분으로 인체 및 동물에 투여 시 세포 독성 및 장기에 중금속 등에 의한 독성 물질이 발생할 수 있어 안전성이 매우 중요하다. 따라서 본 연구에서는 KSD의 안전성 확보를 위해 중금속 검사, 간과 신장 기능 지표 검사 및 RAW 264.7 세포 생존 시험을 거친 후 RAW 264.7 세포를 이용하여 KSD 추출물의 항산화 및 항염증 효과를 알아보았다. KSD 추출물에서 중금속은 발견되지 않거나 기준치 미만으로 검출되었다. 간기능지표인 AST, ALT는 정상군에 비해 낮게 나타났으며 신기능 지표인 Cr, BUN도 정상군과 같거나 낮아 인체에 안전함을 확인하였다. RAW 264.7 세포 독성은 대조군 대비 농도별 생존율에 차이가 없었다. KSD 추출물의 항산화 효능 측정 결과 DPPH 라디칼 소거능과 ABTS 라디칼 소거능은 10 μg/mL 이상의 농도에서 의존적으로 활성이 증가하였다. 항염증 효과 측정 결과 NO 생성량은 대조군에 비해 10, 100 μg/mL에서 유의성 있는 감소를 보였으며, IL-1β와 IL-6는 농도 의존적으로 발현량이 감소하였고, TNF-α는 유의적 차이점을 발견할 수 없었다. 이상의 결과를 보아 KSD는 골절 치료를 위한 생약으로 안전성 및 항산화 및 염증 감소에 효과가 있을 것으로 생각된다. As Kamisipjeondaebotang (KSD) extract is an herbal ingredient, safety is very important due to possible cell poisoning or heavy metal toxicity to organs when administered to humans or animals. Accordingly, this study examined the antioxidant and anti-inflammatory effects of KSD extract to confirm its medicinal safety by using RAW 264.7 cells after heavy metal screening, functional index test of the liver and kidney, and cell survival rate test. Heavy metals were not found in KSD extracts or were less than standard amounts. Liver function indices such as aspartate aminotransferase and alanine aminotransferase revealed low values and kidney function indices such as creatinine and blood urea nitrogen were not significantly different from the normal group. This proved the safety to the human. RAW 264.7 cells showed no poisoning compared to the control group in terms of survival rate. Regarding the antioxidant effect of KSD extract, 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity and 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging activity increased at concentrations over 10 μg/mL. The anti-inflammatory effect of KSD extract significantly decreased based on the amount of nitric oxide at concentrations of 10 and 100 μg/mL compared to the control group. Expression of interleukin (IL)-1β and IL-6 decreased in a concentration-dependent manner. There was no significant difference in tumor necrosis factor-α level. Based on the results, KSD can be regarded as a safe antioxidant with anti-inflammatory effects for fracture treatment.

        야생당근(Daucus carota L.) 주스를 첨가한 개호식품(젤리) 제조 및 품질 특성

        This study evaluated the quality characteristics of jelly prepared with different levels (0%, 5%, 10%, 15%, 20%, and 25%) of wild carrot (WC, Daucus carota L.) juice as a care food for the elderly. The lightness, redness, yellowness, and delta (${\Delta}$) values of the jelly (Control, WCJ5, WCJ10, WCJ15, WCJ20, and WCJ25) decreased with increasing amounts of wild carrot juice added. The mechanical properties, such as hardness, springiness, chewiness, and gumminess, of the jelly were decreased with increasing amounts of wild carrot juice added. The total polyphenol and total flavonoid contents of the jelly increased with increasing amounts of wild carrot juice added. The DPPH radical scavenging activity ($IC_{50}$) also increased with increasing amounts of wild carrot juice added. The ${\alpha}$-glucosidase inhibitory effects of wild carrot (WC) and WCJ25 were 71% and 54.4%, respectively, compared to the positive control (acarbose). The lipase inhibitory effects of WC and WCJ25 were 44.2% and 14.4%, respectively, compared to the positive control group (orlistat). On the other hand, the sensory evaluation score was the best at WCJ20, which contained 20% wild carrot juice. In conclusion, WCJ20 or WCJ25 is expected to be a care food for the elderly with respect to texture as well as the antioxidant and enzymatic activity (${\alpha}$-glucosidase inhibitory and lipase inhibitory activities).
