러시아 철갑상어(Acipenser gueldenstaedtii) 발생 시료의 RT-qPCR 분석을 위한 내재 대조군 유전자의 선정

To evaluate appropriate reference genes for the normalization of quantitative reverse transcription PCR (RT-qPCR) data with embryonic and larval samples from Russian sturgeon Acipenser gueldenstaedtii, the expression stability of eight candidate housekeeping genes, including beta-actin (ACTB), elongation factor-1A (EF1A), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), histone 2A (H2A), ribosomal protein L5 (RPL5), ribosomal protein L7 (RPL7), succinate dehydrogenase (SDHA), and ubiquitin-conjugating enzyme E2 (UBE2A), were tested using embryonic samples from 12 developmental stages and larval samples from 11 ontogenic stages. Based on the stability rankings from three statistic software packages, geNorm, NormFinder, and BestKeeper, the expression stability of the embryonic subset was ranked as UBE2A>H2A>SDHA>GAPDH>RPL5>EF1A>ACTB>RPL7. On the other hand, the ranking in the larval subset was determined as UBE2A>GAPDH>SDHA>RPL5>RPL7>H2A>EF1A>AC TB. When the two subsets were combined, the overall ranking was UBE2A>SDHA>H2A>RPL5>GAPDH>EF1A> ACTB>RPL7. Taken together, our data suggest that UBE2A and SDHA are recommended as suitable references for developmental and ontogenic samples of this sturgeon species, whereas traditional housekeepers such as ACTB and GAPDH may not be suitable candidates.
쥐노래미 (Hexagrammos otakii) 성장호르몬 cDNA 유전자의 염기서열 변이 및 발현 특성

우리나라 주요 해산 어종인 쥐노래미 (Hexagrammos otakii)로부터 성장호르몬 유전자 cDNA를 클로닝하고 이의 염기서열과 발현 특성을 분석하였다. 뇌하수체 조직으로부터 cDNA library를 제작하였으며, membrane filter hybridization 및 expressed sequence tag기술을 이용하여 성장호르몬 cDNA transcript 들을 대량 발굴하였다. 총 확보된 full-length clone 39개 중 31개가 동일한 형태로 나타났으나, 너머지 클론 들에서는 5`쪽의 염기서열 변이, ORF 내의 염기서열 삽입, 3`쪽의 염기서열의 변이 등이 검출되었다. RT-PCR과 RNA dot blot 분석을 수행한 결과 본 연구에서 얻어진 쥐노래미 성장호르몬 transcript 들은 뇌하수체 특이적인 전형적인 어류 성장호르몬 발현 특성을 나타내었다. Different types of transcripts encoding growth hormone (GH) were identified from cDNA libraries constructed with pituitaries of a marine fish species, greenling (Hexagrammos otakii). GH-homologous cDNA clones were isolated using the high-density filter hybridization and the expressed sequence tag techniques. Of 39 full-length positive cDNA clones, 31 clones (79%) displayed an identical sequence, however, remaining 8 clones exhibited several polymorphisms in their sequences including (1) the length and sequence variability in the 5` upstream region, (2) insertional sequences in open reading frame, and (3) deletion and/or single nucleotide polymorphism in the untranslated 3` region. Based on RT-PCR and RNA dot blot analyses, these transcripts were proven to be ecpressed in a pituitary-specific manner.

Female Siberian sturgeon Acipenser baerii were crossed with male Russian sturgeon A. gueldenstaedtii and the development, ontogenetic behavior, and phototactic characteristics of the resultant hybrid prolarvae were examined. The fertilization rate of the hybrid group was similar to that of Siberian sturgeon (the maternal species), and the overall developmental characteristics of hybrid embryos were not significantly different from those of either parental species. The time window from first hatch to the completion of hatching was wider in Russian sturgeon than in Siberian and hybrid sturgeon groups. The prolarval viability of the hybrid group was similar to that of the maternal species, and significantly higher than that of the paternal species. Hybrid prolarvae displayed ontogenetic behavioral patterns that were quite similar to those of Siberian sturgeon. In an illumination preference test, hybrid prolarvae displayed significant positive phototaxis, similar to the maternal species, and clearly distinct from the negative phototactic character of the paternal species. Taken together, our data indicate that the prolarval traits of hybrid sturgeon from this cross more closely resemble those of the maternal than the paternal species.
