        신장 상피세포주 A498을 이용한 대황(Rheum undulatum L.) 추출물의 세포독성

        본 연구에서는 전통적으로 담석증, 신장치료 등의 한약제로 많이 사용하는 대왕을 여러 용매를 사용하여 얻은 대왕추출물 분액에 대한 세포독성을 여부를 MTT 정량법, NR 정량법, SRB 정량법을 이용하여 조사하였다. 1. 추출 용매 methylene chloride, ethyl acetate, butanol, water로부터 얻은 대왕추출물 모두 처리농도에 따라 세포에 미치는 영향이 증가하였다. 2. Butanol을 용매로 사용하여 얻은 대왕추출물 분액이 다른 3가지 용매로부터 얻은 대왕추출물보다 세포에 미치는 영향이 크게 나타났고 water를 용매로 사용하여 얻은 추출물이 A498세포주에 미치는 영향이 가장 낮게 나타났다. 3. Butanol을 추출 용매로 하여 얻은 대왕추출물이 A498세포주에 미치는 영향이 가장 컸는데 그 추출물에 대한 MTT_50, NR_50, SRB_50값은 각각 0.63㎎/m1, 0.65㎎/m1, 0.68㎎/m1이었고, 가장 영향이 적은 water의 경우 MTT_50, NR_50, SRB_50값은 각각 0.84㎎/m1, 0.82㎎/m1, 0.80㎎/ml이었다. 4. 정량방법 간의 대왕추출물에 대한 반응은 MTT 정량법이 가장 민감하게 나타났다. We have evaluated cytotoxic effects of four crude extracts of methylene chloride, ethyl acetate, butanol, water layer isolated Rheum undulatum in A498 cell line, human kidney epithelial cells. The cytotoxic evalutation was measured by colorimetric assay using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT), neutral red (NR) and sulforhodamine protein B (SRB) . These results obtained are as follows : MTT, NR and SRB quantities were significantly decreased in cultured A498 cells treated four crude extracts by increased concentrations. The cell cytotoxic effect of crude extracts of butanol layer was more stronger than others layer. The values of MTT_50, NR_50, SRB_50 of crude extract of butanol layer and were measured both 0.63 ㎎/ml, 0.65 ㎎/ml, and 0.68 ㎎/ml, respectively and the values of water layer were 0.84 ㎎/ml, 0.82 ㎎/ml, and 0.80 ㎎/ml, respectively in cultured A498 cell line.

        미용 칼라렌즈 관리방법과 렌즈탈색의 관계

        미용칼라렌즈는 미용상의 효과와 채색효과 뿐 만 아니라 임상적으로도 웅용되어 착용자가 점점 증가하는 추세이다. 본 연구는 일반적으로 미용 칼라렌즈의 관리에 이 용되는 다목적 콘돼트렌즈 용액 (MPS) , 열이나 화학 소독방법, 단백질 제거제, 용액의 pH, 알코올 성분이 함유된 세척액 (Miraflow™) 및 에탄올이 칼라렌즈의 탈색에 미치 는 영향을 조사하기 위해 국내에서 생산된 Blue 색상의 렌즈(미광, 뉴 바이오)를 대 상으로 시행하였다. 각 요인별 탈색 정도는 ELISA Reader로 486, 587, 656 nm에서 홉광도를 측정하여 비교하였다. 국내에 유통되는 소프트렌즈 다목적 관리용액으로 언 한 미용칼라렌즈의 탈색은 없는 것으로 판명되었으며 일부 미용칼라 렌즈에서 렌즈 팽윤제인 에탄올에서 심한 팽윤과 탈색이 나타났다. 또한 열, 과산화수소, 단백질 제 거 제 및 pH변화에 의해 칼라렌즈가 탈색되지 않았디. 미용 칼라렌즈의 제조과정과 제품에 대한 규제가 있어야 착용자들이 안심하고 착용할 수 있올 것으로 사료된다. Colored cosmetic contact lenses have become increasingly popular. The purpose of this study was to investigate which factors bleach the cosmetic lenses. Absorbance for bleaching effects in several factors was measured by ELISA reader and microscope. The factors studied included: multipurpose soft contact lens solution(MPS), heat and H2<:)z(AOSept) disinfection methods, protein remover, solution pH(4.0/1O,O), Isopropylalcohol(Miraflow™), and ethanol. Ethanol among several factors only bleached colored cosmetic contact lenses manufactured by Mikwang Co. We suggest that strict regulation of color contact lens should be required for manufacturing excellent lenses.

        RGP 콘택트렌즈 다목적 용액이 L929 세포에 미치는 저해효과

        L929 세포를 배양하여 현재 시중에서 유통되고 있는 8 개 회사의 Alcon Optiso와, Ciba SOLO care hard, Allergan Total Care, Boston Simplicity, B&L Wetting and Soaking Solution, Menicon pharma 02 Care, 새 한 Aquas - multi, 중외 제 약 앵 피 에스액 둥 8종류의 가스투과성 경성 콘택트렌즈를 위한 MPS를 25% 농도로 배양세 포에 처리하고 48시간 동안 배양한 후 MTT, SRB assay 그리고 NR assay로 세포 수준의 독성을 검정하였다. L929세포에 MPS를 처리하여 MTT assay한 결과 4종류 의 MPS 에서 세포 중식 저해율이 30% 이상으로 나타났고, 3종류의 MPS는 대조군과 유사하였다. SRB assay의 경우는 2종류를 제외한 모든 제품에서 30% 이하의 세포 중식 저해율이 나타났다. NR assay는 6종류의 MPS 에서 세포 증식 저해용이 30% 이상으로 나타났고 2종류의 MPS는 대조군과 유사하였다. 이러한 결과로 볼 때 일부 제품의 RGP 렌즈용 MPS가 L929세포 중식올 저해하는 것으로 사료된다. To investigate the i며1ibitoη effect of multipurpose solutions, L929 cells were cultured and treated with eight multi purpose solutions(MPSs) for RGP of eight companies(Alcon Opti - soak, Ciba SOLO care hard, Allergan Total Care, Boston Simplicity, B&L Wetting and S때선ng Solution, Menicon ph없ma 02 C따e, Saehan Aquas - multi, Junwae MPS solution). The cytotoxicity was measured by MTT, SRB and NR assay. 1n MTT assay at 48 hrs, four of them had over 30% of growth inhibitioin on L929 cells. 1n SRB assay at 48 hrs, six of them had less 30% of growth inhibitioin and in NR assay at 48 hrs, six of them had over 30% of growth inhibition on L929 cells.

        Numerical simulation have been conducted to obtain the basic information for the prevention of explosion in a rectangular enclosure with n-Butane(n-C_(4)H_(10)) gas leak. A numerical Model was established to predict the characteristics of flammable mixture formation with the variation of variables: leak location, leak time. leak velocity and aspect ratio. When the gas leaks from the bottom of the rectangular enclosure, the closer the leak location is to the side wall, the faster mass transfer towards the horizontal direction than the vertical direction is as the leak velocity increases, the fuel reaches higher and in the case of the same leak velocity and leak time, mass transfer in the leak direction is less than that in the horizontal direction because of the gravitational force. Aspect ratio of the rectangular enclosure also affects the formation of the flammable mixture proportional to the leak time and the leak velocity.

        하늘타리 형질전환근의 생장 및 Trichosanthin의 생합성을 위한 최적화

        하늘타리 (Trichosanthes kirilowii)로부터 조직배양 기법을 이용하여 약리작용을 나타내는 생리활성물질인 trichosantin을 생산하기 위한 연구로 Agrobacterium rhizogenes ATCC15834 의 감염을 통하여 기내 배양된 유묘의 잎절편으로부터 형질전환 모상근을 유도하였다. 유도된 모상근 clones으로부터 성장속도와 분지화 정도가 빠르며 단백질 합성능이 가장 우수한 TR-03 clones을 선발하였으며, 이를 사용하여 생체량과 배지내 가용성 단백질의 생산을 최대로 높일 수 있는 최적 배양조건을 조사하였다. TR-03 clone을 4% sucrose가 첨가된 MS 배지에 초기 접종농도 2 g (생중량)을 접종하여 100 rpm으로 배양시 생중량과 배지내 가용성 총단백질의 함량이 플라스크당 약 2.4 g (건중량)과 28.3ug/l으로 최대치를 나타내었다. 한편, 배지내 가용성 단백질에 대한 RIP활성 검정에서 배양 4주 후 21.3 unit로 최대치를 보였다. Transformed hairy roots were induced from in vitro grown plantlets of Trichosanthes kirilowii by infection with Agrobacterium rhizogenes strain ATCC15834. Transformed hairy roots exhibited active growth with high branching of roots on plant growth regulators-free medium. Cloned line (TR-03) of hairy root was tested for its growth and extracellular protein accumulation in medium under various culture conditions. Among the culture media tested, a full-strength MS medium had a pronounced effect on root biomass and extracelluar protein accumulation in medium. The maximum root biomass (2.4 g DRW/flask) and extracellular total protein contents (28.3ug/ml) in medium was obtained at inoculum size of 2 g (FRW) and in MS medium supplemented with 4% sucrose. In addition, the optimal shaking speed for root growth and extracellular protein accumulation in medium were 100 rpm. The total extracellualr protein concentration reached a maximum of 28.3ug/ml at 4 weeks and decreased thereafter. Protein translation inhibitory activity was observed in culture broths and reached levels of 21.3 unit. These studies demonstrate that the transformed hairy roots can be utilized for the in vitro production of ribosome-inactivating proteins.

        미니멀리즘 특성과 비달사순 커트스타일의 상관성 분석

        This study attempted to improve the value of the hair industry considering the factthat trend in art and cosmetology now complement each other instead of coexistingseparately. The connection of these two areas with other genres sponsors a creative styleand expands the scope of design expression. This study takes another look at howminimalism, which plays a pivotal role in modern design, correlates with Vidal Sassoonhaircut styles. For this, books, literature documents and previous studies were examined. As a result, it was found that the art trend of minimalism was very similar to VidalSassoon haircut styles in terms of simplicity, geometric aspect, universality and function. Although they are different genres, they pursue the same goal. Therefore, it is veryimportant to connect these areas with other genres in terms of hairdressing in order tofoster the development of the cosmetology industry.

        말린눈썹지의(Parmotrema reticulatum) 추출물을 이용한 모발 염색성

        This study was performed on decolorized hair under various conditions – dye concentration, temperature, time, pH - using pigment extract obtained from Parmotrema reticulatum. The dyeing properties of dyed hair were measured by Lightness(L*), redness(a*), yellowness(b*), color difference and K/S value. As the result, the highest dyeing property was observed at 75%, 40oC temperature, 60 minutes and pH 12. In the case of using mordant, Fe showed higher dyestuff than Al, Fe and Mn. Hunter color of dyed hair showed yellow and red dye in non-mordant but Cu showed yellow and green yellow. These results suggest that the pigment extracts obtained from Parmotrema reticulatum may be useful for the development of natural hair dyeing products.

        헤어크리에이티브 디자인 선의 흐름영역 및 컬의 형태에 관한 연구 - 디자인 영역별 포인트를 기준으로 -

        This study is to use a questionnaire made up with the extracts from books published from 2003 to 2013. The copies of it were distributed to 105 hair experts and finally 87 out of them were collected. As for the contents of the analysis flow of curls at the start point, middle point, and the end point of S, C, & J curl the were researched and analyzed. The analysis of frequency was performed on the collected data by means of SPSS windows 20.0. As a result, it showed that in the analysis of the results suggest: From the analysis of the 11 years of hair creative style work from 2003 to 2013, the following results were obtained. First, the starting point of the curl was the ㉓front center medium 1128(31.6%), the midpoint flow was the ㉔left side part 657(18.4%), and the end point flow was the ㉖right crown side 413 (11.6%). The C-curved flow region is an area suitable for emphasizing the volume of bangs in horizontal or slightly oblique flow with respect to the eyelashes inside the face region. In the second S-Curl, the flow of the starting point of the ㉒right side was 752(21.1%), the ㉓midpoint flow was 1033(28.9%) and the flow of the ㉘end point was 446(12.5%). The flow of S curl is expressed by the characteristic of the S line which is balanced with the flow of curves according to the ratio of up, down, left and right based on the center inside the face area. Third, the flow of the starting point of the ㉓J curl was 1036(29.0%) in the ⑰front center medium, 19 (900) (25.2%) on the right eyebrow in the middle point flow, and 12.8% in ⑫the right cheek bone 564(15.8%). The flow area of the jerk indicates the asymmetry of the left and right sides with respect to the center of the face and the interval between the starting point and the ending point connecting the outer side and the inner side of the face is expressed in a lengthwise direction. Respectively. Thus, the trend of the design line has been expressed that the C, S, and J curls have been removed from the existing stereotypes and become more creative and diverse. The C, S, and J curl styling methods used in hair dry techniques can be seen to be practically used in the beauty industry scene in the contemporary women's outer style. It can be seen that the flow of lines, which emphasize the volume and the indentation, is represented by the asymmetrical harmony of C, C, S curl & J curl, which is a design component influencing the change of the size of the work.
