금속펩타이드를 이용한 Pseudomonas alcaligenes의 5S rRNA의 구조 연구

$Ni(II){\cdot}Gly$-Gly-His(Arg)COOH와 $Cu(II){\cdot}Gly$-Gly-His(Arg)COOH 형태의 금속펩타이드를 이용하여 P. alcaligenes에서 얻은 5S rRNA의 구조를 조사하였다. 그 결과 금속 펩타이드들은 5S rRNA의 줄기-고리 구조에서 염기쌍을 이루지 않거나 불안정하게 이루는 부분을 선택적으로 변형시켰다. 금속펩타이드의 선택성은 중심 금속이 Ni(II)인 경우와 Cu(II)인 경우에 차이가 거의 없었다. 금속펩타이드를 이용한 절단 결과를 금속 착물 M(II)CR을 이용한 결과와 비교하면 금속펩타이드에 의한 선택성이 더 크게 나타났다. 금속펩타이드와 금속착물을 이용한 절단 결과로부터 P. alcaligenes에서 얻은 5S rRNA의 이차구조를 살펴보았다. The recognition and cleavage of 5S rRNA from P. alcaligenes by metallopeptides to the form $Ni(II){\cdot}Gly$-Gly-His(Arg)COOH and $Cu(II){\cdot}Gly$-Gly-His(Arg)COOH were investigated. The results of RNA cleavage analyses suggest that metallopeptides selectively target the unpaired or unstably paired bases of stem-loop structure of 5S rRNA. The selectivity of metallopeptides was little affected by the species of metal ion, Ni(II) or Cu(II). When the result of cleavage by metallopeptides was compared with that of by metal complexes M(II)CR, the recognition by metallopeptides was more selective and structure specific. The cleavage data by metallopeptides and other metal complexes were used to probe the secondary structure of 5S rRNA from P. alcaligenes.
감정코칭을 활용한 교사교육이 교사의 회복탄력성과 교사-유아 간 의사소통에 미치는 영향

본 연구의 목적은 감정코칭을 활용한 교사교육이 교사의 회복탄력성과 교사-유아 간 의사소통에 미치는 영향을 밝히는데 있다. 연구대상은 전라북도 J시에 소재한 2곳의 어린이집 교사 7명이다. 연구 도구는 교사의 회복탄력성을 Reibeach와 Shatt’e(2003)가 개발한 회복탄력성 지수 검사(Resilience Quotient Test:RQT)를 한국문화에 맞게 수정 보완한 김주환(2011)의 한국형 회복탄력성 지수 검사(KRQ-53)와 교사-유아 간 의사소통을 박지은(2012)이 개발한 검사도구를 교사-유아 간 의사소통에 맞게 재구성 한 교사-유아 간 의사소통 척도이다. 실험처치는 감정코칭 전문 강사가 5주 동안 총 15시간의 감정코칭 교사교육이 실시되었다. 자료분석은 교사교육을 통해 수집된 유아교사의 회복탄력성과 교사-유아 의사소통의 점수를 비모수 통계인 부호 순위 검증(Wilcoxon signed rank test)으로 분석되었다. 또한 감정코칭 교사교육 프로그램의 효과를 심층적으로 이해하기 위해 프로그램 실시 전과 후에 유아에 대한 교사들의 감정의 변화에 대해 기록한 저널이 분석되었다. 연구결과 감정코칭을 활용한 교사교육은 교사의 회복탄력성과 교사-유아 간 의사소통에 긍정적 영향을 미치는 것으로 나타났다. 이는 유아교사를 위한 감정코칭을 활용한 교사교육의 가능성을 시사하고 있다. The objective of this study is to identify the effect that teacher training using emotion coaching has on the resilience of teachers and their communication with children. Subjects were seven teachers at two daycare centers located in city J of Jeollabuk Province. The tools used were the Resilience Quotient Test developed by Reibeach and Shatt’e(2003) which was revised to fit the Korean culture by Juhwan Kim (2011) (KRQ-53) and the scale for measuring communication between teacher and child that was reconfigured from the test tools developed by Ji Eun Park (2012) for communication between teacher and child. For the experiment, a total of 15 hours of emotion coaching teacher training was conducted by an expert lecturer in emotion coaching. For data analysis, the scores on the resilience of daycare center teachers collected through teacher training and the scores for communication between the teacher and child were analyzed by the Wilcoxon signed rank test. In addition, to further understand the effects of the teacher training program using emotion coaching, journals that recorded changes in the teachers’ emotions were analyzed. The findings show that teacher training using emotion coaching has a positive effect on the resilience of teachers and communication between teacher and child. This implies the potential for teacher training using emotion coaching.

EGFR 돌연변이 검출에 있어 PNA-Mediated Real-Time PCR Clamping과 직접 염기서열 분석법의 비교 분석

Background: Although the gold standard method for research trials on epidermal growth factor receptor (EGFR) mutations has been direct sequencing, this approach has the limitations of low sensitivity and of being time-consuming. Peptide nucleic acid (PNA)-mediated polymerase chain reaction (PCR) clamping is known to be a more sensitive detection tool. The aim of this study was to compare the detection rate of EGFR mutation and EGFR-tyrosine kinase inhibitor (TKI) responsiveness according to EGFR mutation status using both methodologies. Methods: Clinical specimens from 112 NSCLC patients were analyzed for EGFR mutations in exons 18, 19, 20, and 21. All clinical data and tumor specimens were obtained from 3 university hospitals in Korea. After genomic DNA was extracted from paraffin-embedded tissue specimens, both PNA-mediated PCR clamping and direct-sequencing were performed. The results and clinical response to EGFR-TKIs were compared. Results: Sequencing revealed a total of 35 (22.9%) mutations: 8 missense mutations in exon 21 and 26 deletion mutations in exon 19. PNA-mediated PCR clamping showed the presence of genomic alterations in 45 (28.3%) samples, including the 32 identified by sequencing plus 13 additional samples (6 in exon 19 and 7 in exon 21). Conclusion: PNA-mediated PCR clamping is simple and rapid, as well as a more sensitive method for screening of genomic alterations in EGFR gene compared to direct sequencing. This data suggests that PNA-mediated PCR clamping should be implemented as a useful screening tool for detection of EGFR mutations in clinical setting.
