Vol.11 No.3

Extensive characterization of feline intra-abdominal adipose-derived mesenchymal stem cells

2017 Journal of Veterinary Science Vol.18 No.3

Mesenchymal stem cells (MSCs) isolated from various tissues have been well characterized for therapeutic application to clinical diseases. However, in contrast to MSCs from other animal species, the characteristics of feline MSCs have not been fully documented. In this study, we conducted extensive characterization of feline adipose tissue-derived MSCs (fAD-MSCs). Study fAD-MSCs were individually isolated from the intra-abdominal adipose tissues of six felines. The expression levels of cell surface markers and pluripotent markers were evaluated. Next, proliferation capacity was analyzed by performing cumulative population doubling level (CPDL) and doubling time (DT) calculation assays. Differentiation potentials of fAD-MSCs into mesodermal cell lineages were analyzed by examining specific staining and molecular markers. All fAD-MSCs positively expressed cell surface markers such as CD29, CD44, CD90, CD105, CD166, and MHC-I, while CD14, CD34, CD45, and CD73 were negatively expressed. The CPDL of the fAD-MSCs was maintained until passage 5 to 6 (P5 to P6), whereas DT increased after P3 to P4. Also, stem cell-specific pluripotent markers (Oct3/4, Nanog, and SSEA-4) were detected. Importantly, all fAD-MSCs demonstrated mesodermal differentiation capacity. These results suggest that fully characterized fAD-MSCs could be beneficial when considering the use of these cells in feline disease research.

2001 Restorative Dentistry & Endodontics Vol.26 No.5

When cavity floor is near the pulp, polymerization of light-activated restorations results in temperature increase. this temperature increase cause by both the exothermic reaction process and the energy absorbed during irradiation. Therefore instating base is reqired. Most frequently used insulating base is glass ionmer. The purpose of this study was to evaluate intrapulpal temperature changes of glass ionomer according to various curing intensity and curing time. Caries and restoration-free mandibular molars extracted within three months were prepared Class I cavity of 3×6mm with high speed handpiece. 1mm depth of dentin was evaluated with micrometer in mesial and distal pulp horns. Pulp chambers were filled with 37.0±0.1℃ water to CEJ. Chromium-alumina thermocouple was placed in pulp horn for evaluating of temperature changes. Glass ionomer material was placed in 2mm. total curing time was 40s ; continuous 40s, intermittent 20s, intermittent 10s. Glass ionomer material was cured with 300mW/㎠, 550mW/㎠ light curing unit. The results were as follows : 1.Temperature in pulp increased as curing unit power is increased. 2.Temperature in pulp more increased continuous emission than intermittent emission. Key words : Sealing ability, Retrograde filling material

Effect of moisture on sealing ability of root canal filling with different types of sealer through the glucose penetration model

        Jang, Jin-Ah,Kim, Hee-Lyang,Her, Mi-Ja,Lee, Kwang-Won,Yu, Mi-Kyung 2010 Restorative Dentistry & Endodontics Vol.35 No.5

Objectives: To compared the effect of different levels of moisture of root canal on the sealing ability after filling with four different types of sealer. Materials and Methods: Single-rooted teeth (n = 90) instrumented to and apical size of 0.06 / 45 were randomly assigned to 12 experimental groups (n = 7 per group), positive/negative control groups (n = 3 per group). The teeth of the experimental groups (a. DRY; b. PAPER POINT DRY; c. WET) were obturated with sealer (Group 1-3: Sealapex; Group 4-6: AH plus; Group 7-9: Tubuli-seal; Group 10-12: EndoRez) and warm vertical compaction method. After 7 days in 37˚C, 100% humidity, the coronal-to-apical microleakage was evaluated quantitatively using a glucose leakage model. The leaked glucose concentration was measured with spectrophotometer at 1, 3, 7, 14, 21, and 30 days. Data were recorded ad mmol/L and statistically analysed with the two-way ANOVA and Duncan test (p = 0.05). Results: Throughout the experimental period Tubuli-seal/WET (Group 9) showed the highest mean cumulative glucose penetration (178.75 mmol/L), whereas AH plus/DRY (Group 4) had the least (20.78 mmol/L). Conclusions: The results of this study demonstrated that the moisture condition of root canals at the time of obturation and the type of sealer that was used had a significant effect on leakage and sealing ability. Thus drying procedure according to sealer types is a critical step and should not be missed in endodontic treatment. 실험군을 3가지 습윤 상태에서 (a, DRY; b, PAPER POINT DRY; c, WET) 4종류의 sealer로 (Group 1-3: Sealapex; Group 4-6: AH plus; Group 7-9: Tubuli-seal; Group 10-12: EndoRez), warm vertical compaction method를 이용해 충전하였다.

Vol.30 No.2

Purpose: The purpose of this study is to examine effects of a multifactorial program for preventing the frailty of older adults and effects of a follow-up program applying a capacity building strategy. Methods: A quasi-experimental pretest-posttest design was used for the nonequivalent control group. The follow-up group (n=75) and non-follow-up group (n=68) received the same multifactorial program comprising muscle strength exercise, cognitive training, and psychosocial programs for 12 weeks. After completion of multifactorial program, the follow-up group took follow-up programs applying the capacity building strategy for following 12 weeks. The data of physical function, cognitive function, and psychological function, and self-rated health were collected from both groups three times: before intervention, after intervention, and 12 weeks after intervention. The data were analyzed using x2 test and t-test. Results: In comparison with the non-follow-up group, the scores of Timed Up & Go Test, and physical activities energy expenditure were significantly improved in the follow-up group. Conclusion: These results indicate that a multifactorial program with follow-up adapting the strategies of capacity building for the older adults group is feasible to prevent the physical frailty in community.

Canine mesenchymal stem cells immunomodulate atopic dermatitis through the induction of regulatory T cells in an ex vivo experimental study

Vol.40 No.1

Mesenchymal stem cells (MSCs) are multipotent stem cells, which can be induced to differentiate into several cells. MSCs are also reported to possess immunomodulatory properties through secretion of inflammatory cytokines and generation of regulatory T (Treg) cells. Treg cells play an important role in allergic disorders, including atopic dermatitis. We examined the immunomodulatory effects of canine adipose tissue derived-MSCs (cAD-MSCs) in 3 groups: Group 1, untreated normal dog; Group 2, dogs with Dermatophagoides farinae ointment-induced atopic dermatitis; and Group 3, dogs with atopic dermatitis. Canine peripheral blood mononuclear cells (PBMCs) were collected from each group and co-cultured with cAD-MSCs. After co-culturing, PBMCs were separated and the expression of Treg cells was examined by flow cytometry. According to the results, the populations of Treg cells were increased in 3 ex vivo experimental groups, co-cultured with cAD-MSCs. These results would be important for the application of MSCs in clinical trials.
