NADH oxidase(nox) 유선자의 PCR- RFLP를 이용한 돼지 intestinal spirochetes 국내분리주의 동정

In this study, we performed a PCR-RFLP analysis of NADH oxidase gene(nox) for the characterization of porcine intestinal spirochetes isolated from Korea by the comparison with Brachyspira hyodysenteriae and B. pilosicoli reference strains. Eleven strains including four reference strains, B. hyodysenteriae B204, B234, B169, B. pilosicoli P43/6/78 and seven Korean isolates were used. PCR products of 939 bp were amplified using nox-specific primers and digested with two restriction enzymes, Bfm I and Dpn II. In study using Bfm I, both strains showed no difference in fragmented size(197 and 741 bp). When use Dpn II, B. hyodysenteriae showed two bands(209 and 684 bp), however B. pilosicoli showed a single band of 896 bp. Our results indicate that nox-specific PCR-RFLP could be used as a typing method of Brachyspira species and as an epidemiological method for identifying spirochetes isolated from swine.

MC1R 유전자의 PCR-RFLP를 이용한 한우육과 젖소육/black Angus 수입육의 구분

The melanocortin 1 receptor (MC1R) plays an important role in regulation of melanin pigment synthesis within mammalian melanocytes. Mutations within the gene encoding MC1R have been shown to explain coat color variations within several mammalian species including cattle. To develope a rapid and accurate method for the identification of Hanwoo, we performed a modified PCR-RFLP analysis of MC1R gene using single nucleotide polymorphism (SNP) within MC1R as a target. A size of 538 bp (537 bp for Hanwoo) was amplified by PCR, digested with Hpa II, and electrophoresed on a 1.5% agarose gel. A PCR product from Hanwoo showed a single band of 537 bp, whereas two fragments of 328 bp and 210 bp were detected in both Holstein and Black angus. The current result suggests that the PCR-RFLP using our primers and enzyme digestion system would be very accurate, easy and reproducible method to discriminate between Hanwoo and Holstein/Black angus meat.
국내의 개에서 Brachyspira pilosicoli의 분리 및 동정

This study was performed to report the outbreaks of canine intestinal spirochetosis and to characterize the canine isolates. Three canine isolates were weakly beta-hemolytic and had sharp end shape with 5 flagella. Isolates didn't produce indole but fermented fructose. In $API-ZYM^{(R)}$ study, isolates were alpha-glucosidase negative and alpha-galactosidase positive, which is the typical characteristics of B. pilosicoli. In multilocus enzyme electrophoresis(MEE) study, isolates were divided into 2 electrophoretic types. Isolates showed different properties with B. hyodysenteriae but closely related with pathogenic canine intestinal spirochete(B. pilosicoli). This is the first report of the isolation and characterization of canine intestinal spirochete in Korean dogs.
유세포 분석기를 이용한 galectin-3에 의해 유도된 흉선세포의 apoptosis 분석

Galectin-3 plays an important role in cell development, differentiation and cancer metastasis, including cell-cell/extracellular matrix (ECM) interactions and is supposed to have an effect of apoptosis on T-cells in thymic clonal selection. In this study, to know the effect of galectin-3 on thymocyte development, we used recombinant human galectin-3 (rHgal-3) from Escherichia coli, JM105, which was inserted with human gal-3 gene-transformed plasmid vector (prGal-3) to express human galectin-3. Expressed rHgal-3 was confirmed by western blot using the culture supernant of hybridoma (M3/38) producing monoclonal antibody to human galectin-3. Sepharose gel affinity chromatography was used to purify the expressed rHgal-3. Thymocytes and hepatocytes from 6-week-old male BALB/c mice were incubated with rHgal-3 and showed marked increase of apoptotic population on analysis using flow cytometry with 7-AAD in a dosedependent manner. However, rHgal-3 failed to induce apoptosis on hepatocytes. Interestingly, this apoptotic effect was not inhibited by lactose, a specific lectin domain inhibitor. From these results, we concluded that extrinsic -3 induces apoptosis on mouse thymocytes, and galectin-3 may have an apoptotic effect on T-cells in thymic clonal selection.
대장균에서 흉막폐렴균 독소 Apx I과 Apx II의 대량발현

Actinobacillus pleuropneumoniae is the causative agent of a porcine contagious pleuropneumonia. Among several virulence factors including exotoxin (Apx toxins), LPS, transferrin-binding proteins, OMPs, and some proteases, Apx toxins have been major targets for the protection study. In this study, cloning and expression of A. pleuropneumoniae Apx I and Apx II toxin, which are produced by all highly virulent strains, were performed by Escherichia coli expression system. Genes coding Apx I and II toxin were amplified from the A. pleuropneumoniae serotype 5 genomic DNA using polymerase chain reaction and cloned to a prokaryotic expression vector, pRSET. Expression of the Apx I and Apx II coding sequences in E. coli resulted in the formation of insoluble inclusion bodies purified according to a denaturing purification protocol, which employs the use of guanidium. Recombinant proteins were purified using $Ni^{2+}$-charged resin affinity purification. This expression and purification system made it possible to produce Apx I and Apx II in large amounts for further immunologic studies.
