Profiling of Differentially Expressed Genes in Human Stem Cells by cDNA Microarray

Stem cells are unique cell populations with the ability to undergo both self-renewal and differentiation, although a wide variety of adult stem cells as well as embryonic stem cells have been identified and stem cell plasticity has recently been reported. To identify genes implicated in the control of the stem cell state as well as the characteristics of each stem cell line, we analyzed the expression profiles of genes in human embryonic, hematopoietic (CD34+ and CD133+), and mesenchymal stem cells using cDNA microarrays, and identified genes that were differentially expressed in specific stem cell populations. In particular we were able to identify potential hESC signature-like genes that encode transcription factors (TFAP2C and MYCN), an RNA binding protein (IMP-3), and a functionally uncharacterized protein (MAGEA4). The overlapping sets of 22 up-regulated and 141 downregulated genes identified in this study of three human stem cell types may also provide insight into the developmental mechanisms common to all human stem cells. Furthermore, our comprehensive analyses of gene expression profiles in various adult stem cells may help to identify the genetic pathways involved in self-renewal as well as in multi-lineage specific differentiation.

Murine erythroleukemia (MEL) cells are widely used to study erythroid differentiation thanks to their ability to terminally differentiate in vitro in response to chemical induction. At the molecular level, not much is known of their terminal differentiation apart from activation of adult-type globin gene expression. We examined changes in gene expression during the terminal differentiation of these cells using microarray-based technology. We identified 180 genes whose expression changed significantly during differentiation. The microarray data were analyzed by hierarchical and k-means clustering and confirmed by semi-quantitative RT-PCR. We identified several genes including H1f0, Bnip3, Mgl2, ST7L, and Cbll1 that could be useful markers for erythropoiesis. These genetic markers should be a valuable resource both as potential regulators in functional studies of erythroid differentiation, and as straightforward cell type markers.
Follicular Unit Transplantation: Comparison of Video Microscopic vs. Combination Methods

Background In 2004, Sharon Keene introduced a video microscope to the hair transplantfield and discussed about the benefits such as ergonomics, quality assurance, and easy teaching. In 2009, our clinic set up digital video microscope systems. CCD-chip-loaded hand-held digital video microscope was connected to a 19-inch HD (high definition) LCD monitor. We compared the transection rates and graft-cutting time of two graft-dissecting methods to decide which one is more efficient. Methods Two technicians performed follicular unit graft dissection via two different methods of video microscopic and a combination of video microscope for slivering and loupe magnification for graft-cutting. All the procedures were recorded on high-definition digital video camera and cross-checked on the video clips. Results The transection rate of digital video microscope use was 2.2% while the combinationmethod with 1.6%. For comparison of graft-producing time, the combination method could produce about equal amount of grafts in 186% faster the time. Conclusions The work efficiency was greater with the combination method. The total graft productivity was nearly 2 times larger while the transection rates of two methodsshowed very close results.

CHO 세포에 있어서 강력한 poly(ADP-ribose) polymerase 저해제인 benzamide와 3-aminobenzmide는 MMS와 MNNG에 의한 DNA 단사절단을 증가시켰으며 단사절단의 재결합을 유의성 있게 억제하였다. 이들 저해제는 또한 MNNG에 의한 비주기성 DNA 합성(UDS)과 자매염색분체 교환(SCE)에 상승 효과를 나타내었다. MMS와 MNNG의 복합처리에 의한 UDS와 단사절단은 상가효과를 보이나 저해제 처리에 의해 이의 효과는 상승되었다. 이들 결과로 보아 poly(ADP-ribose)는 DNA에 단사절단이 일어날때마다 염색사구초를 안정화 시킴으로서 DNA 손상에 대한 회복에 조절작용을 나타낸다고 사료된다. Amounts of UDS and strand breaks induced by the combined treatment with MMS and MNNG were found to be additive but these amount were enhanced in the presence of inhibitors. These results suggest that poly (ADP-ribose) functions a regulatory role in the repair of DNA damage by virtue of stabilizing chromatin structure whenever strand breaks occur in DNA.

아동기의 경험과 부모로서의 양육효능감이 보육교사의 교수효능감에 미치는 영향

본 연구의 목적은 보육교사를 대상으로 아동기 때 자신의 부모로부터 받은 긍정적 양육경험과 현재 자녀를 키우는 부모로서의 양육효능감 간에 관계가 있는지 살펴보고, 두 변인이 보육교사로서의 교수효능감에 어떻게 영향을 미치고 있는지 분석해 보는 것이다. 연구목적을 달성하기 위해 자녀가 있는 보육교사 301명을 대상으로 아동기 양육경험, 부모로서의 양육효능감 및 교사로서의 교수효능감을 조사하였다. 자료는 빈도분석, t-검증, 상관관계분석, 위계적 중다회귀분석을 사용하여 분석하였으며, 주요 결과는 다음과 같다. 첫째, 아동기의 긍정적 양육경험, 부모로서의 양육효능감, 보육교사로서의 교수효능감 간에는 유의한 정적 관계가 있었다. 둘째, 보육교사의 연령이 높을수록, 부모로서의 양육효능감 중 훈육능력이 높다고 인식할수록, 보육교사로서의 교수효능감이 높은 것으로 나타났다. 셋째, 아동기의 긍정적 양육경험은 부모로서의 양육효능감을 거쳐 보육교사로서의 교수효능감에 간접적인 영향을 미치고 있는 것으로 나타났다. This study examined the relation between positive childhood experience and parenting efficacy as current parents of childcare teachers and analyzed the effect of the two variables upon teaching efficacy as childcare teachers. To achieve such objective, childhood experiences, parenting efficacy and teaching efficacy were analyzed to 301 childcare teachers who had a child. Frequency analysis, t-test, correlation analysis and hierarchical multiple regression analysis were carried out to analyze the data. The main results are as follows. First, there was significantly positive relation between positive childhood experience, parenting efficacy as parents and teaching efficacy as childcare teachers. Second, the teaching efficacy was getting higher when the age of childcare teachers was higher and disciplining ability among parenting efficacy was higher. Third, positive childhood experience demonstrated that it had influenced on teaching efficacy as childcare teachers indirectly through parenting efficacy of parents.
