        약용식물과 커피 혼합물로부터 기능성 건강음료의 항균 및 항산화 효과

        본 연구에서 기존의 생리활성이 알려진 차가 버섯(Inonatus obliquus), 모링가(Moringa oleifera), 그라비올라(Anona muricata), 단풍마(Dioscorea quinquelaba), 가시오가피(Acanhopanax senticosus), 음양곽(Epimedii herba), 인진쑥(Artemisia capillaris), 그리고 오디(Morus alba) 추출물과 원두커피를 혼합하여 기능성 음료를 제조하였으며, 각각의 음료를 C1에서 C7으로 명명하였다. 각 음료의 항균, 항곰팡이, 항산화 활성 및 총 폴리페놀 함량을 조사하였다. 항균 및 항곰팡이에 대한 활성은 Gram positive bacteria인 Staphylococcus aureus에 대하여 C4, C5, C6 및 C7이 활성을 나타내었고, 특히 C7인 경우 항생제 내성균주인 MRSA에 대하여 강한 활성을 나타내었다. Gram negative bacteria에 대한 결과는 E. coli D31에 대하여 1.0 mg/ml의 농도에서 C1에서 C7 모든 음료가 활성을 나타내었고, 반면에 곰팡이인 Candida albicans에 대하여는 활성을 나타내지 않았다. 총 폴리페놀 함량은 C6의 경우가 7.37 mg/g으로 가장 높은 결과를 나타내었고, C5의 경우는 3.78 mg/g으로 가장 낮은 함량을 나타내었다. DPPH로 항산화 능력을 측정한 결과, C1에서 C7 모두 강한 활성을 나타내었고, 특히 C7인 경우 0.5 mg/ml의 농도에서 88%라는 높은 free radical 소거능력을 나타내었다. 이러한 결과를 바탕으로 각 기능성 음료의 생리활성의 기초자료를 제공하고 다양한 기능성 건강음료뿐 만 아니라 고부가가치 기능성 제품을 창출 할 수 있는 근거를 마련한 것으로 생각된다. In this study, our investigated the antioxidant activities and compared other bioassay including antimicrobial, antifungal activities and total polyphenol contents of functional healthy drinks from some medicinal herbs and coffee mixture. The kind of medicinal herbs, chaga mushroom (Inonatus obliqurs), moringa (Moringa Oleifera), gravila (Anona muricata), mulberry (Morus alba), Dioscoreaceae (Dioscorea quinquelaba), Berberidaceae (Epimedii Herba), Asteraceae (Artemisia capillaries) and siberian ginseng (Acanthopanax senticous,). The functional healthy drinks, named C1, C2, C3, C4, C5, C6 and C7 were summered in Table 1. The in vitro antimicrobial activity was examined against Gram positive bacteria, Gram negative bacteria and a fungus. The functional healthy drinks were broad spectrum of antimicrobial activity without antifungal activity against Candida albicans KCTC7965. In particularly, the C7 showed strong activity against Methicillin Resistant Staphylococcus aureus CCARM3089, CCARM 3115 and CCARM3561. And, the C7 showed 88% of free radical scavenging effect on 0.5 mg/ml using 1,1-diphenyl-2-picrylhydrazyl (DPPH) method. Functional healthy drink C7 was mulberry extracts from Morus alba, chaga mushroom from Inonatus obliqurs and moringa from Moringa olifera in additionally coffee extracts. Its results confirm that the potential use of mulberry extracts as a good source of antibacterial compounds or as a health promoting food and health drinks.

        모링가 잎 (Moringa oleifera, Leaf) 에탄올 추출물의 화장품 소재 특성

        To investigate the cosmetic effects of drumstictree (Moringa oleifera, leaf) extract on skin care, we measured antioxidant and antimicrobial activities, tyrosinase and elastic activity inhibition. B16F10 melanoma cell has been used to measure cell viability. It was 42±2.66 mg/g GAE base and 16.5±1.7 mg/g Naringin base as a measurement result of total polyphenol and total flavonoid contents. DPPH free radical scavenging activity was 89% in a concentration at 500 μg/mL. Drumstic-tree (Moringa oleifera, leaf) extract exhibited in vitro antimicrobial activity against MRSA CCARM3115. EtOH extract of Moringa oleifera showed 25.1% (L-Tyrosine) and 20.1% (L-DOPA) of tyrosinase inhibition, respectively. In elastase inhibition assay, EtOH extract of Moringa oleifera showed 29.6% of inhibition. At a concentration of 200 μg/mL, tyrosinase and elastase activation were found to inhibited dose-dependent. These results suggest EtOH extract from Moringa oleifera has excellent activities in the antiwrinkle and whitening. Therefore, it expected to be used effectively for development of functional cosmetic materials.

        대왕버섯 (Pleurotus nebrodensis) 추출물의 미백 및 주름 개선 효과

        Pleurotus nebrodensis have been used as a traditional remedy and a food source. Our study focuses on figuring out the possibility of Pleurotus nebrodensis as cosmetic materials. We examined anti-oxidant, anti-wrinkle, anti-inflammation and whitening effects. The SOD-like activity was 69.7 ± 2.31 at 125 ㎍/mL, compared to the standard sample L-ascorbic acid 1 mM 34.7 ± 0.79. It was 11.5 ± 0.27 ㎎/g Naringin base and 64 ± 1.45 GAE (Gallic acid) base as a measurement result of total flavonoid and total polyphenol contents. GME showed 24.5% (L-tyrosine), 16.6% (L-DOPA) of tyrosinase inhibition assay, respectively. In elastase inhibition assay showed 36.2% of inhibition at a concentrations 200 ㎍/mL, tyrosinase and elastase activation were found to inhibited dose-dependent. As a result of measuring the collagen precursor pro-collagen type 1, the amount of collagen synthesis was 434 ng/mL at a concentration of 500 ㎍/mL. The higher the concentration of GME the greater the wrinkle reduction and whitening effects. Anti-inflammatory activites measured nitric oxide (NO) and cell viability in LPS-induced macrophage RAW264.7 cell, it was found to inhibit NO production in a concentration-dependent manner. These results suggest methanol extracted Pleurotus nebrodensis has excellent activities in the anti-wrinkle, anti-inflammation and whitening. Therefore, Giant mushroom Pleurotus nebrodensis may have value as the potential cosmetic formulations.

        홍합 (Mytilus coruscus) 추출물 RM 60을 사용하여 E. coli D31을 대상으로 항균활성 펩타이드를 분리 정제하였다. 정제한 펩타이드는 MALDI-TOF Mass spectrometry를 사용하여 분자량을 측정한 결과, 1464.92 Da으로 나타났으며 Edman 분해법을 이용한 peptide sequencer를 사용하여 일차구조를 분석 중에 있다. Antimicrobial peptides (AMPs) play an important role in this response by rapidly killing invading microorganisms. In this study antimicrobial peptide has been isolated from acidified whole body extract of a bivalve mollusk, the marine mussel (Mytilus coruscus). This peptide purified to homogeneity by gel-filtration and reversed-phase high performance liquid chromatography. The molecular weight was 1464.92 Da, determined by MALDI-TOF Mass spectrometry. In addition to growth inhibition of Escherichia coli D31.

        한국산 어류 8종 추출물의 생리활성

        N/A A search for contraction and relaxation responses on the smooth muscles, antimicrobial and antioxidant activity in different body parts of 4 freshwater fish and 4 marine fish were conducted. The four freshwater fish studied were Sarotherodon niloticus (tilapia), Channa arus (snake head), Cyprinus carpio (Israel carp) and Silures osotus (catfish), and the four marine fish were Scyliorhinus torazame (tiger shark), Atelepus japonicus (tadpole fish), Mugil cephalus (gray mullet) and Thamnaconus modestus (file fish). Frozen samples were extracted with distilled water containing 1% acetic acid. Antimicrobial activity against Bacillus subtilis and Escherichia coil was detected in extracts from several tissues in all species tested. Relatively high antimicrobial activity could also be detected in the bile extracts from C. carpio, M. cephalus, and T. modestus. Contraction and relaxation responses on smooth muscles could be detected in all species tested, especially in the intestine extracts. Antioxidant activity was also detected in extracts from several tissues in all species tested, while in the extracts from S. osotus, excluding livers and spleens, no antioxidant activity was detected. Results from this study suggest that fish are a potential source for the discovery of novel bioactive materials.

        커피 에탄올 추출물의 항균 및 미백활성

        본 연구에서는 커피(Coffea arabica)의 에탄올 추출물을 이용하여 기능성 화장품소재로서의 가능성을 검토하였다. 미백활성으로는 tyrosinase 저해 활성과 멜라닌 함량을 측정하였으며, 항산화 활성으로는 SOD-like activity를 측정하였고, 그 결과 500 μg/mL의 농도에서 94.8 ± 6.2%의 활성을 나타내었다. CAE는 3개의 그람양성균과 6개의 그람음성균을 대상으로 항균활성을 조사하였으며, 그 결과 그람음성균에 대해서는 폭넓은 활성을 나타내었지만, 그람양성균에 대한 활성은 나타내지 않았다. 하지만 항생제내성균주인 MRSA CCARM3561에는 강한 활성을 나타내었다. 반면에 Candida albicanse에 대한 항곰팡이 활성은 나타내지 않았다. Tyrosinase 저해활성과 DOPA 산화능은 대조군인 arbutin 보다는 낮게 나타내었고, B16-F10 melanoma cell에 대한 멜라닌 생합성 저해능은 농도 의존적으로 감소하는 경향을 보였으며, 100 μg/mL의 농도에서 89.2%를 나타내었다. 이러한 결과들은 커피 추출물이 천연물, 무독성 항균제 또한 천연 화장품 소재로써의 가치가 있음을 나타내는 것이다. In this study, Coffea arabica ethanol extract (CAE) was tested for possible functional cosmetic agent. Whitening effect was measured by tyrosinase inhibition assay, and anti-oxidant activity was checked by SOD-like activity. SOD-like activity of CAE showed 94.8 ± 6.2% at 500 μg/mL. The anti-bacterial activities CAE was evaluated against three different gram-positive bacteria and six gram-negative bacteria including MRSA strains. CAE exhibited in vitro broad spectrum antimicrobial activities of gram-negative bacteria without antifungal activity. CAE was strong exhibited against MRSA CCARM3561. The tyrosinase and L-DOPA inhibitory activities of the CAE lower than those positive control arbutin. CAE reduced melanin contents of B16-F10 melanoma cell in a dose dependent manner and decrease about 89.2% at a concentration 100 μg/mL. These result highlight the potential of coffee extract as a naturally active and non-toxic antibacterial suitable for cosmetic applications.

        식용 달팽이 (Achatina fulica)의 추출물 RM 60을 사용하여 E. coli D31을 대상으로 순수한 항균성 물질을 분리정제하였다. 정제한 항균성 물질은 MALDI-TOF Mass spectrometra를 사용하여 분자량을 측정한 결과, 1392.64 Da의 단일 peak를 얻을 수 있었으며, 이 후 Edman 분해법을 이용한 peptide sequencer를 사용하여 일차구조 분석을 조사하고 있다. An antimicrobial substance was purified from the giant snail body extract (Achatina fulica) using solid-phase extraction and separation on HPLC reversed-phase chromatography. The primary structure were determined by a combination of an automated amino acid sequence and MALDI-TOF Mass. Its molecular mass was found to be 1392.64 Da. This result was in excellent agrement with the theoretical molecular mass calculated from the amino acid sequence. purified peptide showed antimicrobial activity in vitro against Escherichia coli D31. This result indicate that giant snail whole body was potentially antimicrobial.

        큰실말(Cladosiphon novae-caledoniae Kylin) 부산물의 화장품소재 특성

        큰실말(Cladosiphon novae-caledoniae kylin, C. novae-caledoniae kylin) 부산물을 이용하여 화장품 소재로써 가능성을 조사하고자, 항산화 효과(DPPH 자유 라디칼 소거 활성), 항균(anti-microbial activity), 미백(tyrosinase inhibition assay), 주름개선(elastase inhibition assay) 및 B16F10 mouse melanoma cell (MTT assay)을 이용한 세포 생존률을 측정하였다. 큰실말 추출물 원물(MC)과 불용성(MI)은 Staphyloccus aureus (S. aureus)와 항생제내성균주인 MRSA에 대하여 활성을 나타내었지만 항곰팡이 활성은 나타내지 않았다. 큰실말 추출물 수용성(MS)은 tyrosine에 대한 tyrosinase 저해효과가 49%로 표준물질인 arbutin 보다 미백효과가 우수하였지만 주름개선효과는 비교적 낮은 활성을 나타내었다. DPPH 라디칼 소거능력은 500 μ g/mL에서 89%로 나타내었다. 따라서 본 연구인 큰실말 추출물은 화장품 미백소재로써 좋은 후보군임을 확인 할 수 있었다. To investigate the effect of dietary fiber from mozuku, Cladosiphon novae-caledoniae kylin (C. novae-caledoniae kylin) on the skin care, we measured anti-oxidant activity, anti-microbial activities, tyrosinase activity inhibition and elastic activity. B16F10 melanoma cell (MTT assay) were used to measure cell viability. MC and MI exhibited in vitro antibacterial activity against Staphyloccus aureus (S. aureus) and MRSA without antifungal activity. Mozuku extract (MS) showed excellent tyrosinase inhibition effect compared to arbutin as a positive control (to 49% for tyrosine). The wrinkle-improving effect was relatively low. However, wrinkle-improving effect was relatively low. DPPH free radical scavenging activity was 89% in a concentrations at 500 μg/mL. These results indicate that the mozuku extracts may be an effective cosmetic ingredient for skin whitening.

        Our investigation of the seaweed extracts, Ulva lactuca. The biological activities antioxidant, antimicrobial activity, antifungal and haemolytic activity of ethy-ether and ethyl-acetate extracts from the seaweed, Ulva lactuca were investigated. They were separately extracted using ethyl-ether and ethyl-acetate from dried samples at room temperature and freeze dried. Seaweed extracts were found to cause significant free radical scavenging effects on DPPH (1,1-diphenyl-2-picrylhydrazyl). Seaweed extracts had not significant haemolytic activity against human erythrocyte. This extracts exhibited in vitro broad-spectrum antimicrobial activity of gram-negative, gram-positive bacteria and without antifungal activity.
