수종생약의 Brine Shrimp 독성에 관한 연구(I)

Brine shrimp toxicity of water extracts of twenty three crude drugs was tested by the brine shrimp bioassay. Eight crude drugs were shown to have 100% mortality in $1000{\;}{\mu}g/ml$. They were fractionated by Amberlite XAD-2 column chromatography and were tested by the brine shrimp bioassay. Methanol fraction of Rhei Rhizoma $(LC_{50}=49.60{\;}{\mu}g/ml)$ and Cantharis $(LC_{50}=54.08{\;}{\mu}g/ml)$ were shown to have potent brine shrimp toxicity in this test.

Effects of the aqueous extract of Aquilariae Lignum (Thymelaeaceae) on the allergic reactions were investigated. Oral administration of this extract (50, 250, and 500mg/kg) exhibited a dose-dependent inhibition on passive cutaneous anaphylactic reactions in rats. Administrations of this extract (500mg/kg, i.p.) at 60 min before and 5, 10 min after the compound 48/80 treatment (8mg/kg, i.p.) decreased the mortality rates to 0, 0, and 14.2%, respectively. The aqueous extract of Aquilariae Lignum (0.05 ~ 1.6mg/ml) showed a dose-related inhibition on histamine release from rat peritoneal mast cells. The morphological examination also clearly showed that the aqueous extract of Aquilariae Lignum prevented the degranulation of mast cells in rats.

One hundred and seventeen crude drugs were screened for DNA-binding activity in vitro. The DNA-methyl green assay is a useful biochemical screen for the detection or development of biologically active compounds which bind to DNA. This assay is based upon the fact that free methyl green undergoes rapid spontaneous molecular rearrangement to its colorless carbinol base so that the liberation of the dye from DNA by displacement can be followed spectrophotometrically as a decrease in absorbance at 65Onm. Seven methanolic extracts of crude drugs including Cinnamomi Cortex spissus, Coicis Semen, Coptidis Rhizoma, Perillae Semen, Plantaginis Semen, Polygalae Radix and Zanthoxyli Fructus showed less than 2,000mcg/ml as their IC50 values. The active principles of Plantaginis Semen and Zanthoxyli Fructus were transferred into organic solvents, which showed the IC50 values with 588 and 574mcg/ml, respectively. These fractions have been selected for isolation of biologically active constituents.

The current study evaluates the capacity of eugenol to regulate immediate allergic reaction by control of histamine release. Administrations of eugenol (1M/kg, i.p.) at 60 min before and 5, 10 min after the compound 48/80 treatment (8mg/kg, i.p.) were shown the mortality rates as 0, 44.4, and 77.8%, respectively. A 60 min before administered group revealed a significant inhibition of serum histamine release compared with those of 5 and 10 min after the compound 48/80 injection. Eugenol (6-48mM) was also showed a dose-dependent activity on the compound 48/80-induced histamine release from the highly purified population of Alcian Blue-positive peritonea] mast cells. These results indicate that in vitro treatment with exogenous eugenol inhibited the active response of mast cell populations and modulated its characteristics.

In order to find the new hepatoprotective agents preliminary screening of twelve prescriptions which have been used for the treatment of hepatic disease in Oriental traditional medicine has been carried out. Both DPPH free radical scavenging and hepatoprotective effect on tacrine-induced cytotoxicity in Hep G2 cells are performed. Osumokkya-tang which is one of the prescription belongs to interior-warming properties showed the DPPH free radical scavenging effect as well as hepatoprotective activity significantly. It is also suggested that its hepatoprotective effect is derived from Chaenomelis Fructus.

From the 60% aqueous methanolic fraction of Moutan Cortex Radicis two hyaluronidase inhibitors were isolated and their structures were elucidated by spectroscopic methods. Their structures were identified as paeoniflorin (com-pound I) and oxypaeoniflorin (compound II). Compound I and II exhibited hyaluronidase inhibitory activities with $IC_{50}$ of 1.71 and 1.73mM, respectively.

Hyaluronidase is one of the mucopolysaccharide-splitting enzyme and is related to the permeability of the vascular system and inflammation. An anti-hyaluronidase assay guided fractionation of the methanolic extract of Uncariae Ramulus et Uncus has furnished a pentacyclic triterpene, ursolic acid (compound I). Compound I exhibited hyaluronidase inhibitory activity with $IC_{50}$ value of 0.15 mM, and disodium cromoglycate showed the inhibitory activity with $IC_{50}$ value of 1.78 mM as a positive control.
