We separately cultured follicular oocytes collected from individual ovaries of slaughtered Korean native cows and examined both the embryonic development rate and pregnancy rate after embryo transplantation according to the meat yield and quality grades of the source beef carcass. Oocytes from meat yield grade B cows exhibited a higher fertilization rate and embryonic developmental rate to the eight-cell stage than oocytes from grade A or C animals (p<0.05), but there was no significant difference in rate of development to the blastocyst stage among meat yield grades A, B and C. The oocyte cleavage rate and development rate to the eight-cell stage from meat quality grade 3 cattle was higher than grades 1++, 1+, 1 and 2 (p<0.05). Embryos derived from grade 3 animals displayed a development rate to the blastocyst stage of 19.4%, which was also higher than all other meat quality grades (p<0.05). Transplantation of in vitro-cultured oocytes from meat yield grade A ovaries led to a higher pregnancy rate (64.2%) than in vitro-cultured oocytes from meat yield grade B ovaries (56.5%), but there was no significant difference between the two groups in pregnancy or abortion rates. In conclusion, embryonic development rate and pregnancy rate has a close relation to meat quality grades of the source beef carcass, this results is to give information for the Korean native cows improvement of breed.
한우 체외수정란이 이식된 수란우의 임신과 유산에 영향을 미치는 시술자 측 요인들

본 연구는 임상 조건하에서 수정란이식의 효율성 증대를 위하여 이식 시술자 측면의 숙련도, 난이도, 소요시간, 주입 위치, 자궁의 출혈 및 위치 조절이 체외수정란이 이식된 수란우의 임신과 유산에 미치는 효과를 검토하였다. 임신율은 숙련된 시술자, 자궁각 2/3 지점에 이식 및 위치 조절군이 유의하게 높은 경향이었으나, 이식 난이도, 소요시간 및 출혈에 따른 차이는 인정되지 않았다. 한편 시술자 측의 요인에 따른 유산율은 유사한 경향이었다. This study was conducted to investigate the effects of various factors of practitioner's techniques such as skill, difficulty, consumed time, implantation depth, bleeding and altering of implant location, on pregnancy and abortion after transfer of in vitro produced Korean Native Cow embryos. The pregnancy rate of skilled transfer was significantly higher than that of unskilled transfer , of 2/3 location in uterus was significantly higher than in 1/2 or 4/5 uterus, and of altering implantation location was significantly increased. There were no difference in pregnancy among the groups of difficulty of transfer, consumed time and bleeding of uterus. The abortion rates from skill, difficulty of transfer, implantation depth, consumed time, bleeding of uterus and altering of location were not differ.
소에서 Interferon이 동결-융해 정자의 운동성과 인공 수정 임신율에 미치는 효과

The increase in the meat quality and milk production of cows, which breed Korean Native Cow (KNC) and Holstein cow, is not improving reproductive efficiency. In this study, we examined the effect of interferon (IFN supplementation on motility of frozen-thawed semen and pregnancy rate after artificial insemination of KNC and Holstein cow. In experiment 1, we investigated the effect of IFN-tau concentration (10,000 IU and 20,000 IU) on the percentage of total motility (TM) and progressive motility (PM) of frozen-thawed spermatozoa. In experiment 2, KNC were infused 20,000 IU IFN-tau at insemination or after insemination. In experiment 3, KNC or Holstein cow were inseminated with frozen-thawed semen and infused 20,000 IU IFN-gamma or -tau after insemination. In experiment 1, the average of TM (23.9% to 30.9%) and PM (18.5% to 21.9%) were similar between control and treatments. In experiment 2, the pregnancy rates of IFN infusing times were not different from 45.8% to 53.6%. In experiment 3, the pregnancy rates of Holstein cow infused different kinds of IFN were similar (control, IFN-gamma, IFN-tau; 42.9%, 40.5%,48.0%). In the case of KNC, however, the pregnancy rate of control was 55.6%, which was significantly lower than that of IFN-gamma (68.9%; p<0.05). Thus, IFN is effective on the improvement of reproductive efficiency, but further study is needed.

본 연구에서는 소에 있어서 수정란 이식을 다각적으로 분석하고 개선하기 위해 이식한 결과를 비교 검토하였고 그 결과는 다음과 같다. 1. 채란된 한우 난포란을 이용하여 생산된 체외수정란을 서로 다른 지역별로 이식한 바 B지역(경주)에서는 106의 수란우에 이식하여 51두(48.1%)가 수태하였고, A지역(김천) 수태율(33.8%)과 D 지역(경산) 수태율(35.3%)보다 유의하게 높았지만, C 지역(탑리)에서는 유의차가 존재하지 않았다. 따라서, 수정란이 These studies were carried out to establish an effective in vitro embryo transfer methods by analyzing several factors. The base media were TCM-199 solution for in vitro maturation(IVM) of bovine follicular oocytes and Fer-TALP solution for in vitro fertilization(IVF) and CRlaa medium for in vitro culture(IVC). IVC used the fertilized oocytes of 24-hr culture (day 1)after IVF. Embryos were cultured in drop-culture that contained 25 embryos per 10. Blastocysts cultured for 7 to 9 in vitro were transferred to recipients. The results obtained were as follows: 1. The pregnancy rate according to different region of embryo transfer were 33.8%, 48.1%, 45.0% and 35.3% respectively. 2. The pregnancy rate according to the parity of recipient when embryos were transferred to nulliparous (42.9%) was higher than that of 1∼3nd parlous(36.9%), however there were not show significant difference each other. 3. According to the stage of blastocyst, the pregnancy rate when middle blastocysts (MB) (45.5%) were transferred to recipients were higher than that of late blastocysts (LB) (41.0%). 4. When IVF-derived blastocysts cultured for 7 to 9 day were transferred to recipients, the pregnancy rate was higher 7 day of blastocyst than that of 8 day or 9 day of blastocyst. The results of embryo transfer according to the regions, the parity of recipient and the development stage showed that blastocyst formed for 7day transferred to nulliparou were higher pregnancy rate than others.

본 연구는 임상 조건하에서 수정란이식의 효율성 증대를 위하여 수란우 측면의 산차, 체점수(BCS), 발정 발현과 유도, 황체 형태, 자궁 크기 및 자궁각 위치가 체외수정란이 이식된 수란우의 임신과 유산에 미치는 효과를 검토하였다. 미경산우의 임신율이 경산우에 비하여 유의하게 높았으나, 유산율은 유사한 경향이었다. BCS에 따른 임신율은 차이가 없었으나 유산율은 BCS 3.0 미만군이 군 에 비하여 유의하게 높았다. 수란우의 발정 발현과 유도 방법, 황체 This study was conducted to investigate the effects of various factors of recipients such as recipient parity, body condition score(BCS), estrus type, corpus luteum, uterine size and uterine horn, on pregnancy and abortion after transfer of in vitro produced Korean Native Cow embryos. The pregnancy rate of nulliparous was significantly higher than that of multiparous , but there were no difference in the abortion rates . There were no difference in pregnancy among the groups of BCS treatment, but the abortion rate was significantly higher in the group of below BCS than the group of BCS . The pregnancy and abortion rates from the estrus type, corpus luteum, uterine size and uterine hem were not differ among treatment groups.
한우 육질등급이 난포란의 배반포 체외생산에 미치는 영향

These studies were conducted to establish the practical Hanwoo (Korean native cattle) improvement system through the combining of embryo transfer technology and confirming individual Hanwoo oocyte culture system and to investigate that correlation of Hanwoo carcass classification (intramuscular marbling) and oocyte donor for blastocyst production in vitro. In case of Hanwoo, the carcass meat quality grades were divided to grade 1⁺⁺, 1⁺, 1, 2, and 3 depends on fat distribution of longest muscle cross-sectional surface. As results, the numbers of follicular oocytes collected from individual fundamentally-registered Hanwoo yielded 1⁺⁺, 1⁺, 1, 2 and 3 meat quality were 9.5, 9.4, 8.5, 8.8 and 8.8 per ovary, respectively. The numbers of retrieval oocyte from follicles were significantly higher in the cattle yield 1⁺⁺, 1⁺ meat quality than in the cattle yield 1, 2 and 3 meat quality (p<0.05). The rates of blastocyst formation were 18.2, 21.3, 29.4, 30.9, and 31.5% in the cattle yield 1⁺⁺, 1⁺, 1, 2 and 3 meat quality of after in vitro maturation, respectively. It was significantly lower in the cattle yield 1⁺⁺ and 1⁺ meat quality than in the cattle yield 1, 2 and 3 meat quality (p<0.05). In order to evaluate embryos quality, TUNNEL assay was conducted for each meat quality grade using blastocyst stage embryo on day 8. The results showed that apoptosis cell number was higher tendency in the cattle yield 1⁺⁺and 1⁺ meat quality (81 and 79, respectively) than in the cattle yield 1, 2 and 3 meat quality (51, 48 and 50, respectively) but there was no statistical significance in each group. After embryo transfer, the conception rate of recipient was 53.5 (23 out of 43), 52.1 (38 out of 73) and 58.0% (58 out of 100) in the meat quality of 1, 1+ and 1++, respectively. These results showed that the conception rate was significantly higher in the cattle yield 1 meat quality than in the cattle yield 1⁺⁺, 1⁺ , 2, and 3 meat quality (p<0.05). In summary, these results indicate that the application of confirming Hanwoo individual oocyte culture system and embryo transfer technology can make good use of the genetic resources conservation and improvement of Hanwoo. Relevance of the meat quality grade and reproductive ability of carcasses of Hanwoo will be considered to be one of the effective means for the associated research with obesity and reproduction.

The Transgenic livestock can be useful for the production of disease-resistant animals, pigs for xenotranplantation, animal bioreactor for therapeutic recombinant proteins and disease model animals. Previously, conventional methods without using artificial nuclease-dependent DNA cleavage system were used to produce such transgenic livestock, but their efficiency is known to be low. In the last decade, the development of artificial nucleases such as zinc-finger necleases (ZFNs), transcription activator-like effector nucleases (TALENs) and clustered regulatory interspaced short palindromic repeat (CRISPR)/Cas has led to more efficient production of knock-out and knock-in transgenic livestock. However, production of knock-in livestock is poor. In mouse, genetically modified mice are produced by co-injecting a pair of knock-in vector, which is a donor DNA, with a artificial nuclease in a pronuclear fertilized egg, but not in livestock. Gene targeting efficiency has been increased with the use of artificial nucleases, but the knock-in efficiency is still low in livestock. In many research now, somatic cell nuclear transfer (SCNT) methods used after selection of cell transfected with artificial nuclease for production of transgenic livestock. In particular, it is necessary to develop a system capable of producing transgenic livestock more efficiently by co-injection of artificial nuclease and knock-in vectors into fertilized eggs.
한우 체외수정란이 이식된 수란우의 임신과 유산에 영향을 미치는 수정란측 요인

본 연구에서는 한우 체외수정란의 이식에 있어서 수정란 측 요인들이 수란우의 임신과 유산에 미치는 영향을 검토하였다. 이식에 제공하는 배반포의 수량에 따른 임신율은 1개, 2개 및 3개 이식 군에서 각각 32, 44 및 였고, 유산율은 로서 2개 이식군의 임신율과 유산율이 가장 높았으나 유의차는 인정되지 않았다. 배반포 등급에 따른 임신율은 , 유산율은 로서 유사한 경향이었다. 배반포의 발생 단계에 따른 임신율은 로서 비슷하였으나, 유산율은 HB군이 LB In this study, we investigated the effects of number, grade, stage and day of embryos on pregnancy and abortion rates of Holstein heifer recipients after transfer of in-vitro produced Korean Native Cattle (Hanwoo) embryos. When one, two or three blastocysts were transferred to recipients, pregnancy (32.0, 44.0 and ) and abortion (12.5, 13.6 and ) rates did not differ among treatments. The pregnancy and abortion rates of different grades of blastocysts were , respectively. However, they were not significantly different. According to the stage of blastocyst, the pregnancy rates were similar between . However, the abortion rate in hatched blastocyst stage was significantly higher than that in late blastocyst stage . The pregnancy rates in day and day blastocyst (47.1 and , respectively) was significantly higher than that in the day blastocyst (29.8 and , respectively). The abortion rate was the highest in the day blastocyst among treatments, and it was significantly different compared to that in the day blastocyst . Therefore, this study suggested that the pregnancy rate was affected by only day of embryos, while the abortion rate was affected by stage and day of embryos.
한우 체외 수정란이 이식된 수란우에 백신 투여 시기가 임신과 송아지의 생존에 미치는 영향

본 연구에서는 수정란 이식을 위해 선발된 대리모에 이식 전 백신의 투여 시기가 임신과 유산에 미치는 효과와, 임신한 대리모의 분만 예정일 전에 백신의 투여가 송아지의 질병과 생존에 미치는 효과를 검토하였다. 실험 1에서는 한우 체외 수정란의 이식에 선발된 대리모의 백신 투여 시기가 임신과 유산에 미치는 영향을 검토하였다. 이식 전 주째 백신 투여시 임신율이 31.5%로서 대조군 및 주 투여군의 42.4 및 45.9%에 비하여 유의하게 낮았다(p<0.05 The present study was conducted to investigate the effects of time of vaccination in recipients on the pregnancy rate and the viability of calves derived from embryos produced in vitro. In experiment 1, control group was non-vaccinated, group 1-1 received vaccine (Pfizer, Exton, PA, USA) during 04 weeks and group 1-2 received vaccine during 48 weeks before embryo transfer. The pregnancy rates in the control (42.6%) and group 1-2 (45.3%) were significantly higher (p<0.05) than that in the group 1-1 (32.6%). However, the abortion rates were similar among groups (4.9 to 13.5%). In experiment 2, the recipients received embryos produced in vitro were non-vaccinated (control) or vaccinated. Vaccine was injected during 04 weeks (group 2-1) and 48 weeks (group 2-2) before parturition. The incidence of a disease in calves was significantly higher in the control (22.4%) than in other vaccinated groups (2.2% and 3.1%, p<0.05). The mortality of calves in the control is 27.6%, which was significantly higher than that of group 2-1 and group 2-2 (11.1% and 7.8%).
