We examined the transcription of the hepatitis B virus surface antigen(HBsAg) gene in COS cells transfected with pSHS and pSMS containing the transcriptional termination signal of SV40 and HBV, respectively. The amount of HBsAg and its transcripts from cells transfected with pSHS were higher than in cells transfected with pSMS. These results represent that the poly(A) addition signal (UAUAAA) of HBV is leaky and weak in transcriptional process.
계배 근세포의 분화단계에 따른 cDNA library 제작과 분석

계배근세포의 분화단계에서 발현되는 mRNA들에 대한 cDAN library들을 제작하였다. 분화전과 후의 cDNA들을 서로 상쇄시켜 분화과정의 초기단계에 특이적인 한종의 cDNA를 분리하였다. 이 cDNA의 크기는 약 0.8kb이고 하나의 HindIII 제한효소 자리를 가지며 EcoR I, BamH I, Xba I, Kpn I, Sma I, 그리고 Pst I 자리는 포함하지 않았다 The cDNA libraries for mRNAs expressed on differentiation stages in chick embryonic muscle cells were constructed. A species of cDNA specific on early stage of differentiation process was isolated by subtracting ones after differentiation from cDNAs before differentiation of muscle cells. Size of the cDNA is about 0.8kb and it contains one HindIII site but no EcoR1, BamHI, XbaI, KpnI, SmaI, and PstI site.
야행성 또는 돌연변이체 B형 간염바이러스 DNA로 감염시킨 HepG2 세포로부터 RNA 스플라싱에 의한 다양한 전사체의 확인

HBV에서 RNA 스플라이싱에 의한 전사체의 합성 특성을 조사하기 위하여 야생형과 돌연변이체 HBV DNA로 HepG2 세포에 감염시켜 생산된 전사체들을 분석하였다. 두 종류의 감염된 세포로부터 mRNA를 추출한 후 RT-PCR을 한 결과 스플라이싱에 의하여 크기가 감소한 DNA 절편들이 증폭되었고 증폭된 양상은 두 경우가 유사하였다. DNA 염기서열분석을 통하여 전사체의 정확한 구조를 조사한 결과, 야생형과 돌연변이체 HBV 모두에서 5종류의 RNA 스플라이싱에 의한 전사체가 확인되었다. 이 결과는 HBV의 e 항원과 X 단백질이 HBV의 RNA 스플라이싱과는 관련이 없음을 의미하였다. 전사체 중 dsp2의 앙호틀에서 HIV의 단백질가수분해효소와 유사한 구조를 가지는 지역이 나타났다. Hepatitis B virus (HBV) transcripts produced by RNA splicing have been reported recently and we also previously detected a likely viral spliced RNA from HepG2-K8 cell line which was transfected with a mutant HBV DNA and produced the viral particles. In order to confirm the existence of HBV spliced transcripts, we carried out the reverse transcription-polymerase chain reaction (RT-PCR) in HepG2 cells transfected with the wild-type or mutant viral DNA. Five spliced species were identified in both cells and the junction region of deleted sequences showed the conserved sequences of splice junction sites, GT-AG. This result suggests that the e antigen and X protein of HBV may be not concerned with RNA splicing of HBV because the mutant can not produce both proteins. Two species were ones by singly splicing and three were by doubly splicing. Sizes of the spliced transcripts ranged approximately 1.7 to 2.1 kb. A spliced RNA of 2.1 kb was same to that reported by others but four species were newly identified. Sequence analysis of the CDNA of these spliced transcripts shows that these transcripts can potentially encode new proteins that comprise the fused core-surface antigen domain and the reverse transcriptase domain of HBV.
한국산 굴(Crassostrea gigas)과 바위굴(C.nippona)의 16S rRNA에 대한 미토콘드리아 DNA 염기서열의 비교

한국산 굴(Grossostrea gigas)과 바위굴(C. nippona) 미토콘드리아의 16S rRna 지역의 염기서열을 조사하고 비교하였다. PCR을 이용하여 16S rRNA 유전자에서 319-bp DNA 절편을 증폭시키고 그 중 277-bp의 염기서열을 분석한 결과, 굴의 경우 지역별 차이는 보이지 않았다. 즉, 한국의 두 지역(남해, 서해)과 일본산에서 100% 동일한 염기서열을 나타내었다. 굴과 바위굴과의 비교에서는 12개 뉴크 레오타이드가 서로 달랐다(4.3%). 이 결과는 앞으로 굴에 대한 계통분류학적 연구를 위해 사용될 수 있을 것이다. Mitochondrial DNA (mtDNA) sequence variation for the large subunit rRNA-coding gene (16S rRNA) was characterized from two Pacific oyster species (Crassotrea gigas and C. nippona) in Korea. As the sequences of 319-bp DNA fragments amplified from mtDNA of each species by polymerase chain reaction (PCR) were compared, there was no nucleotide difference among C, gigas from West Sea, South Sea in Korea, and Hiroshima in Japan. In comparison of 277-bp DNA sequences for 16S rRNA between two Crassostrea species, C. gigas and C. nippona, twelve nucleotide differences (4.3%) were found. These results could be applied to phylogenetic studieds for Pacific oyster species.
