        고지방식이 동물모델에서 통곡물 시리얼의 근감소성 비만 예방 효과

        Whole grain cereal (WGC)-rich diets provide macronutrients that are important for the regulation of energy metabolism. The current study evaluated whether WGCs had a preventive effect on sarcopenic obesity in high-fat diet (HFD)-induced obese mice. C57BL/6N mice were fed a normal diet (ND), ND+WGC, HFD, and HFD+WGC for 12 weeks. WGCs significantly reduced body weight gain, food efficiency ratio, fat mass, and adipocyte size in HFD-induced obese mice. WGCs attenuated HFD-induced nonalcoholic fatty liver disease by decreasing liver weight and hepatic fat accumulation. In addition, WGCs increased muscle strength and muscle mass in HFD-induced obese mice as well as in ND mice. Taken together, WGCs can be employed as functional food materials for the prevention of sarcopenic obesity by inhibiting fat accumulation and increasing muscle mass.

        Standardized Siegesbeckia orientalis L. Extract Increases Exercise Endurance Through Stimulation of Mitochondrial Biogenesis

        Siegesbeckia orientalis has been reported to exhibit anti-allergic, anti-infertility, anti-inflammatory, anti-rheumatic, and immunosuppressive activities. However, there are very few studies describing its stimulatory effects on exercise capacity. This study elucidated whether S. orientalis extract (SOE) standardized to kirenol content can enhance exercise endurance by increasing mitochondrial biogenesis. SOE significantly improved the running distance and time in mice fed normal diet (ND) and high-fat diet (HFD). SOE also enhanced mitochondrial biogenesis by stimulating the mitochondrial regulatory genes including peroxisome proliferator-activated receptor gamma co-activator 1 alpha (PGC-1α), estrogen-related receptor α (ERRα), nuclear respiratory factor 1 (NRF-1), and mitochondrial transcription factor A (TFAM) in the skeletal muscles of ND and HFD mice. Furthermore, SOE upregulated the AMP-activated protein kinase (AMPK)/sirtuin 1 (SIRT1)/PGC-1α/peroxisome proliferator-activated receptor delta (PPARδ) signaling pathway in the skeletal muscles of ND and HFD mice. Kirenol markedly increased adenosine triphosphate production and mitochondrial activity by stimulating the expression of markers of mitochondrial biogenesis and upregulating the AMPK/SIRT1/PGC-1α/PPARδ signaling pathway in L6 myotubes. These results show that SOE has the potential to be used to develop an exercise supplement capable of stimulating mitochondrial biogenesis through the AMPK/SIRT1/PGC-1α/PPARδ signaling pathway.

        안면신경기능의 평가방법에 대한 고찰;House-Brackmann scale이후의 New grade

        Background and Objective : The facial nerve grading system proposed by House and Brackmann is most widely accepted for the clinical assessment of facial nerve injury. Because of the limitations and subjectivity of the House-Brackmann scale, several new scales of varying degrees of objectivity and ease of use have been introduced. To assess methods of evaluating the function of the facial nerve that have been introduced over the past 20 years, We compared with the House-Brackmann scale. Method : We referred to the information through Entrez Pubmed and Korean studies information(KSI) from 1985 to 2006 about methods of evaluating facial nerve function. We choose 7 scales that focused on objective and easy of use. Result and conclusion : Sunnybrook scale is a weighted, subjective scale with incorporation of secondary defects into a single composite score. Sunnybrook scale can be recommended over House-Brackmann scale.

        경도근시(가성근시 포함) 환자에 있어 눈 경혈 마사지기와 침치료간의 증상개선 효과 비교를 위한 임상시험(단일기관, 무작위 배정)

        Objective : The purpose of this clinical research is the investigation of the effects between acupuncture group(control group) and NURIEYE-1 group (experimental group) in myopia Methods : Comparison clinical study was conducted with the randomly arranged 32 patients for acupuncture group(control group) and NURIEYE-1 group(experimental group). For effectiveness evaluation, eye sight test, MQOL and DITI were tested three times with visits for every 4 weeks. With SAS, Statistical analysis was performed by using paired t-test and two sample t-test. Statistical significance was achieved with less than 5% probability (p<0.05). Result : 1. There was no significant effect from the eye sight comparison between the two group. We had improved results in average with MQOL, however, there was no statistical significance. Though there was also no significant result with DITI average comparison between two group, we witnessed significant effect in some part of MQOL. 2. Nevertheless Periodical comparison test showed weak significance with eye sight and DITI, significant effect was found in some part of MQOL. 3. With periodical comparison tests, there were significant effects in eye sight, MQOL and DITI with the experimental groups and control groups, respectively. Conclusion : There was no significant eye sight improvement. However, with acupuncture point stimulation, we think there are some positive effects for the relaxation of excessive stress in eyes.

        한방 진단 시스템(DSOM)를 통한 이명(耳鳴) 환자와 건강대조군의 변증 비교분석

        Objective : The purpose of this study is to investigate the difference of pathogenesis between tinnitus patients group and healthy volunteers group by using DSOM (Diagnostic system of Oriental Medicine). Methods : Patients group is consisted of people who do not have any disorder except tinnitus, and healthy volunteers group is consisted of people who do not have any special symptom and past history. DSOM was used for pathogenesis investigation of these two groups. Results : There were significant differences between volunteers group and patients group in blood (血虛), dampness (濕), phlegm (痰) (p<0.05), In gender of patients group, deficiency of blood (血虛) was shown with female group statistically more than male group (p<0.05). On the other hand, with volunteers group, there was negligible difference between male and female. In age (over and below 50years), noticeable deficiency of blood (血虛) was shown in patients group. Especially, the old people who are over 50 years showed more deficiency of blood (血虛) than younger group (under 50 years) (p<0.05). With volunteers group, there was only negligible difference. Conclusion : This result showed definite difference in the pathogenesis between tinnitus patiensts group and healthy volunteers group.

        Standardized Kaempferia parviflora Extract Enhances Exercise Performance Through Activation of Mitochondrial Biogenesis

        Exercise enhances mitochondrial biogenesis in skeletal muscle. Increased mitochondrial function and content can contribute to the improvement in skeletal muscle function and the benefits of exercise by increasing the response to energy demands. The effect of standardized Kaempferia parviflora extract (KPE) on exercise performance was accessed in L6 myotubes and C57BL/6J mice. KPE significantly activated peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α) and increased mitochondrial density in L6 myotubes. KPE also upregulated the expression of transcription factors for mitochondrial biogenesis (estrogen-related receptor-α [ERRα], nuclear respiratory factor-1 [NRF-1], and mitochondrial transcription factor A [Tfam]) through activation of PGC-1α in L6 myotubes. In vivo models including normal diet mice and high-fat diet obese mice showed that KPE effectively enhanced running endurance and increased the skeletal muscle weight/body weight ratio. Furthermore, these observations were associated with a significant upregulation of mitochondrial biogenesis regulatory genes in skeletal muscle tissue. KPE enhanced the protein expression of the sirtuin 1 (SIRT1)/adenosine monophosphate (AMP)-activated protein kinase (AMPK)/PGC-1α/peroxisome proliferator-activated receptor-δ (PPARδ) signaling pathway components in vitro and in vivo, acting as an exercise metabolism regulator. These results suggest that KPE has the potential to enhance exercise performance through mitochondrial biogenesis and the SIRT1/AMPK/PGC-1α/PPARδ signaling pathways.

        Optimization of High Hydrostatic Pressure Process for the Extraction of Kirenol from Siegesbeckia orientalis L. Using Response Surface Methodology

        High hydrostatic pressure (HHP) extractionmethod was optimized to maximize the extraction yield ofkirenol from Siegesbeckia orientalis. Operating parameterssuch as extraction pressure (100-600 MPa), pressureholding time (3-20 min), feed-to-solvent ratio (1:10-1:90(w/v)), and solvent (ethanol) concentration (0-100%) wereinvestigated individually by mono-factor experiments. Then, the optimal extraction conditions were determinedusing response surface methodology. Box-Behnken designwas applied to evaluate the effects of three independentvariables (extraction pressure, solvent concentration, andfeed-to-solvent ratio) on the extraction yield of kirenolfrom S. orientalis. Extraction pressure, solvent concentration,and feed-to-solvent ratio affected the extraction yieldssignificantly, whereas the pressure holding time had nosignificant effect. The optimal processing conditions,which gave a maximum extraction yield of 85.9% kirenolfrom the raw material, were as follows: extracting pressure320MPa, pressure holding time 5min, ethanol concentration18%, and feed-to-solvent ratio 1:76 (w/v).

        High hydrostatic pressure extract of Siegesbeckia orientalis inhibits adipogenesis through the activation of the Wnt/β-catenin signaling pathway

        St. Paul’s Wort (Siegesbeckia orientalis L.)confers anti-oxidative, anti-inflammatory, anti-allergic,anti-infertility, and immunosuppressive properties. Here,we elucidated whether high hydrostatic pressure extract ofSt. Paul’s Wort (SPW-HHPE) had anti-adipogenic activity. SPW-HHPE inhibited adipogenesis by reducing intracellularlipid accumulation. SPW-HHPE reduced the mRNAand protein expression of adipogenic regulatory factors[peroxisome proliferator-activated receptor gamma(PPARc), CCAAT/enhancer binding protein alpha (C/EBPa), and sterol regulatory element binding protein-1c]. In addition, SPW-HHPE decreased the mRNA expressionlevels of lipogenic enzymes (fatty acid synthase and acetyl-CoA carboxylase) as well as adipocytokines (adiponectinand leptin). The inhibitory effect of SPW-HHPE on adipogenesiswas mainly attributed to the enhancement of theWnt/b-catenin signaling pathway. When b-catenin siRNAwas transfected into 3T3-L1 adipocytes, the mRNAexpression of PPARc and C/EBPa was upregulated; however,their expression was attenuated by SPW-HHPE. These results suggest that SPW-HHPE suppresses adipogenesisby stimulating Wnt/b-catenin pathway. Keywords

        제주 자생식물 고압용매 추출물의 통합적 항산화 능력

        제주 자생식물 20종을 대상으로 고압용매 추출(추출용매 100% methanol, 추출 온도 40℃, 추출 압력 13.6 ㎫, 추출시간 10분)하여 총페놀 함량과 통합적 항산화 능력을 측정하고 폴리페놀 성분을 동정하였다. 추출수율은 붉나무, 말오줌 때, 사방오리나무, 사람주나무, 팥배나무가 각각 21.8, 21.5, 21.1, 20.7, 20.1%로 가장 높았다. 총페놀 함량은 아그배가 68.3 ㎎ GAE/g로 가장 높았고, 다음으로 사람주나무, 석위, 말오줌때가 각각 57.6, 56.6, 55.1 ㎎ GAE/g을 나타내었다. 수용성 항산화 능력은 이질풀, 사람주나무, 산딸나무, 붉나무가 각각 598, 394, 293, 270 μ㏖ ascorbic acid equivalent/g로 높았고, 지용성 항산화 능력은 백량금, 새우나무, 이질풀, 붉가시나무가 611, 314, 296, 242 μmol trolox equivalent/g로 높았다. GC/MS에 의한 폴리페놀성분을 동정한 결과 15개의 주요 피크를 얻었으며, 그 중 2종의 폴리페놀류(gallic acid(체류시간 19.7분)와 quercetin(체류시간 33.5 분)), ascorbic acid(체류시간 35.3분) 그리고 다수의 지방산류(체류시간 18.6, 21.0, 21.8, 21.9, 23.6분)를 확인할 수 있었는데, 이 중 gallic acid는 다른 성분보다 peak area가 높은 것으로 나타나 사람주나무의 가장 중요한 폴리페놀 성분으로 추정되었다. Twenty natural plants from Jeju were extracted by pressurized organic solvent (100% methanol, 40℃, 13.6 ㎫, 10 min). Extraction yield, total phenolic content (TPC) and integral antioxidant capacity were measured, and each component was identified by GC/MS. Extraction yields were high as 21.8%, 21.5, 21.1, 20.7 and 20.1% in Rhus javanica, Euscaphis japonica, Alnus firma, Sapium japonicum and Sorbus alnifolia, respectively. The extracts containing high TPC (㎎ GAE/g of dry sample) were obtained from Malus sieboldii (68.3), Sapium japonicum (57.6), Pyrrosia lingua (56.6) and Euscaphis japonica (55.1). Integral antioxidant capacities of water-soluble substances were 598, 394, 293 and 270 μmol ascorbic acid equivalent/g in Geranium thunbergii, Sapium japonicum, Cornus kousa and Rhus javanica, respectively. Integral antioxidant capacities of lipidsoluble substances were 611, 314, 296 and 242 μ㏖ trolox equivalent/g in Ardisia crenata, Ostrya japonica, Geranium thunbergii and Quercus acuta, respectively. Fifteen major peaks were identified by GC/MS from the extract of pressurized organic solvent from Sapium japonicum. Two polyphenols (gallic acid (retention time (RT) 19.7 min)) and quercetin (33.5 min)), ascorbic acid (RT 35.3 min), and several fatty acids (retention time 18.6, 21.0, 21.8, 21.9 and 23.6 min) were identified, and gallic acid was the major polyphenol component due to high peak area.
