        The Effects of Crystal Lattices on Luminous Properties in Eu3+ Doped Sr9-xLa1+x(PO4)7-x(SiO4)x System

        The red-emitting phosphors of Eu3+-doped Sr9xLa1+x(PO4)7x(SiO4)x (x = 0, 1, 3, 5, 7) were prepared via a solid-state reaction method. The structural and the optical properties were investigated by using X-ray powder diffraction (XRD) and photoluminescence (PL) measurements, respectively. An analysis of the XRD patterns revealed that the crystal structure changed from a whitlockite to an apatite structure with increasing x. A Rietveld refinement of Sr9La(PO4)7 (x = 0) and Sr2La8(SiO4)7 (x = 7) was performed to obtain more detailed information on the crystal structure. In the PL excitation spectra, with increasing x, the charge-transfer-band (CTB) shifted towards longer wavelengths with enhanced excitation intensity. However, the excitation intensity from the f-f transition of Eu3+ centered at 393 nm decreased. The PL spectra showed a strong red emission (5D0 ! 7F2) and a considerable orange emission (5D0 ! 7F1). To obtain insight into the possible structural changes, the Judd-Ofelt theory are used to calculate the experimental intensity parameters Ω2 and Ω4. The red-emitting Sr9xLa1+x(PO4)7x(SiO4)x (x = 0, 1, 3, 5, 7) phosphors can play key roles for phosphor-converted white-light-emitting diodes (pc-LEDs).

      • 생식주기 동안 흰쥐 자궁에서 발현되는 c-myc 관련 RCL 유전자의 기능

        자궁은 착상 임신 유지 및 분만을 위해 증식과 분비의 주기적인 시기를 겪는다. 배아가 착상하지 못한다면 자궁내막 조직은 퇴화되지만 배아 착상 시에는 자궁내막조직은 유지가 되며 임신 유지와 분만을 위한 과정이 진행된다. 이러한 자궁의 변화는 직접적으로 난소호르몬인 프로게스테론 (progesterone)과 에스트로겐(estrogen)의 영향을 받으며 간접적으로는 다양한 성장인자(growth factor), 사이토킨(cytokines)의 영향을 받는다. 발정주기에는 세포의 분화와 증식에 관련된 많은 유전자들이 발현된다. 본 연구는 생리 주기에서 발현되는 유전자를 분석하기 위하여 cDNA microarray를 통하여 실험하였다. 실험동물을 발정전기(proestrus), 발정기(estrus), 발정후기 (metestrus), 발정말기(diestrus)로 분류하였다. 자궁 적출 후 total RNA를 추출하였다. 추출한 RNA로부터 32P로 표지된 cDNA probe를 합성하였고 Atlas Rat Toxicology 1.2 Array (Clontech)에 hybridization을 하였다. 선별된 유전자로는 growth-related c-rnyc responsive protein RCL, heat shock 47-kDa protein (HSP47), cytochrome c oxidase polypeptide VIc2 (COX6C2), calreticulin (CALR)등이 있다. RCL 유전자는 RT-PCR을 이용하여 검증하였다. RCL 유전자 발현은 발정전기에 가장 높았으며 생식주기가 진행됨에 따라 감소 하였다. 본 연구를 통해 RCL유전자는 생식주기의 조절과 임신, 불임에 관한 중요한 정보를 제공할 것이며 이러한 유전자의 기작과 기능에 대한 연구가 더 필요하다. The uterus undergoes dynamic changes during the cycle and displays many features typical of developmental process. In order to be prepared for implantation, endometrium undergoes predictable, sequential phases of proliferlation and secretory changes. In the absence of an embryo the uterus will progress through a predictable series of stages ultimately terminating in tissue regression and apotosis. In the presence of an embryo, the endometrium not only is maintained, but also progress through an additional program of events. The uterus during estrus cycle synthesize a complex of signaling molecules with specific spatial and temporal modes of expression and which are critical for cell proliferlation and differentiation. The purpose of this investigation was to use cDNA microarrays to evaluate the expression of genes in rat uterus during estrus cycle. Animals were sacrificed on pro-estrus, estrus, metestrus, diestrus. Total RNA was isolated from the uterus. 32P-labelled cDNA was generated from total RNA and hybridizied with the Atlas Rat Toxicology 1.2 Array.(Clontech). Differential gene expression profiles were revealed growth-related c-myc responsive protein RCL, heat shock 47-kDa protein, cytochrome c oxidase polypeptide VIc2 and calreticulin. Reverse transcription polymerase chain reaction (RT-PCR) was used to validate the relative expression pattern of RCL gene. Using this approach we found that RCL gene expression in rat uterus was altered with estrus cycle. The identification of differentially expressed genes may provide important information in the regulation of estrus cycle, pregnancy and infertility. Our long-term goal is to determine the role of these differnetially expressed genes during estrus cycle

        동결 건조법을 이용한 옥천산 캡슐 제형 개발에 관한 연구

        Objective : Okchun-San(OCS) is known as an effective herbal medicine on Type 2 diabetes. We performed to change OCS formulation for freeze drying capsulation. Methods The mixtures of OCS were extracted with water. finally, the filtered solution were evaporated and lyophilized to dry granules. The various ratio of excipients were studied to determine the formation for capsulizing. The samples were inspected for any difference in color, taste and appearance. Results: The prepared form of OCS were dried and weighted 260kg. The lyophilized dry powder yielding 40kg. The suitable ratio of OCS-dry powder and excipient was 10:1. The average weights of On and placebo capsules were $440{\pm}5.28mg$, $465{\pm}7.95mg$, respectively. There was no notable change in color and appearance for both capsuled samples throughout the study period. Conclusions: Therefore in can be concluded that freeze drying capsulation is appropriate form of OCS.

        Treatment with Astragali Radix and Angelicae Radix Enhances Erythropoietin Gene Expression in the Cyclophosphamide-Induced Anemic Rat

        Anemia is a common cause of morbidity and disease and reduces the quality of life. This study examined the effect of a combination treatment (AAC) using Astragali radix (AMW) and Angelicae radix (AGW) in cyclophosphamide (CYP)-induced anemic rats on erythropoietin (EPO) expression and hematological parameters. Male 4-week-old Sprague-Dawley rats were divided into five groups with or without CYP-induced anemia and individual or the combined herbal treatments according to the experimental protocol. After treatment, reverse transcription-polymerase chain reaction was used to evaluate the effects of AAC on erythropoietin expression, and blood and serological parameters were measured. The EPO mRNA levels were lower in the CYP-treated group, compared to the normal group, and higher in the AAC-treated group. In the CYP-treated group, the serum iron concentration, total iron-binding capacity, and vitamin B12 level were lower, but these were normal or almost normal in the AAC-treated group. The CYP-treated group gained less weight than the normal group, but weight gain was partially normalized in the AAC group. The feed efficiency ratio was lowest in the CYP group, but the differences were not significant. The numbers of red blood cells, white blood cells, and platelets, the hematocrit, and the hemoglobin level were measured. The results revealed a reduced number of blood cells in the CYP-treated group, whereas the AAC-, AMW-, and AGW-treated groups showed significantly enhanced blood cell numbers compared to the CYP-treated control group and the AAC-treated group. AAC enhanced EPO mRNA expression in the CYP-induced anemic rat and improved the hematological parameters and vitamin B12 status.

        Hydrogen Peroxide에 의해 유도된 남성생식세포의 세포독성에 미치는 소양(銷陽)의 효과

        The purpose of this study is to examine the protective effect of Cynomorii herba on $H_2O_2$-induced cytotoxicity in male germ cells. The effects were studied by a modified MTT assay. Hydrogen peroxide significantly induced cytotoxicity in GC-1 spg cells, and co-treatment or pre-treatment of Cynomorii herba extract has reduced the cytotoxicity in a dose dependant manner. These results suggest that Cynomorii herba extract increases the survival rate of GC-1 spg cells through the antioxidant effect against $H_2O_2$-induced cytotoxicity.

        右歸飮이 hydrogen peroxide에 의해 유도된 Leydig cell에 미치는 항산화 효과 연구

        Objectives : The purpose of this study was to investigate the antioxidant activity of water extract of Woogyuyeum (WGY) in Leydig cells. Methods : We investigated the cytoprotective effect of WGY in cultured mouse Leydig cells by MTT assay. Leydig cells treated with WGY were incubated in the presence or absence of 50 μM hydrogen peroxide at 37℃ for 24 h. The protective effects of WGY against hydrogen peroxide-induced oxidative stress, lipid peroxide (LPO), superoxide dismutase (SOD), and catalase activity assays were performed in Leydig cells. Results : As a result, WGY showed no significant cytotoxicity in Leygdig cells. WGY showed cell viability as 103.65% in 5 μg/ml concentrations. The cytotoxicity induced by hydrogen peroxide in Leygdig cells, the antioxidant effects of WGY was increased in 1, 5, 50, 100 ug/ml concentraions. 100 μg/ml concentration of WGY showed maximum antioxidant effects. Treatment of cells with 100 μg/ml WGY significantly reduced the MDA concentration to 0.23 nmoles/mg protein. SOD activity was increased at 1, 100 μg/ml concentration of WGY and catalase activity was significantly increased at 50, 100 μg/ml concentrations of WGY, respectively. Conclusions : In conclusion, WGY has antioxidant activities against hydrogen peroxide-induced oxidative stress in Leydig cells.

        DPPH 방법을 통한 토사자, 보골지(補骨脂), 사상자(蛇床子), 음양곽(淫羊藿)의 항산화 활성에 대한 연구

        The present study was conducted to compare antioxidant activity of Cuscutae Semen, Psoraleae Fructus, Cnidii Fructus and Epimedii Herba by DPPH radical scavenging activity. The extract was studied using diphenyl-picryl-hydrazyl (DPPH) for DPPH method. DPPH radical scavenging activity was measured after. .10, 20 and 30 minutes. The extract was tested by 1, 5, 10, 50, 100, 500 and $1000{\mu}g/ml$ concentrations. The results showed that the extract scavenged DPPH radical with time-dependent manner. Also, the extract showed dose-dependent DPPH radical scavenging activity. The extract of Cuscutae Semen, Psoraleae Fructus, Cnidii Fructus and Epimedii Herba scavenged DPPH radical with the IC50 being 2.7, 3.2, 2.9 and 1.1 mg/ml, respectively. In conclusion, the extract of Epimedii Herba, Cuscutae Semen, Cnidii Fructus and Psoraleae Fructus have antioxidant activity for the treatment of male sterility.

        CITES 분류에 의한 사향(麝香)의 기원 동물 분류 및 특성에 대한 고찰

        Objectives : The purpose of this study was to investigate musk deer in taxonomic status and genus species. We investigated the characteristic of musk deer for medicinal usages. Methods : To identifications of musk deer on the taxonomic status and genus species, the literary investigation were conducted on the Korean, China, and Japan pharmacopoeia and published herbal books, CITES Species Lists too. Results : Musk deer placement in a separate family status is the Moschidae. M. chrysogaster Hodgson and M. sifanicus Przewalski was the same species. So, Alpine musk deer revised M. chrysogaster Hodgson[=M. sifanicus Przewalski]. Geographic distribution of M. moschiferus L. divided Sibirica group and himalaica group. Group himalaica contains three subspecies: Korean musk deer(M. moschiferus parvipes Hol.), Chinese musk deer(M. chrysogaster Hodgson), and Himalayan musk deer(M. leucogaster Hodgson). The genetic divergence between M. moschiferus L. and other species was clearly distinguished from the others. M. berezovskii Flerove was less than the others. However, the divergence among M. chrysogaster Hodgson, M. fuscus Li, and M. leucogaster Hodgson were quite low. Musk deers are mostly distributed around the high-plateau. Moschus were from Nepal, Bhutan, Russia, China. Forest musk deer(M. berezovskii Flerove) farming was conducted in China from 1950s. In the Korean hebal pharmacopoeia, Moschus include l-muscone($C_{16}H_{30}O$ : 238.40) over 2% for quantitative test. Conclusions : There are three species of musk deer, Siberian musk deer(M. moschiferus L.), forest musk deer(M. berezovskii Flerove), and Alpine musk deer(M. chrysogaster Hodgson) for medicinal usages.

        쇄양(鎖陽)의 Diphenyl-picryl-hydrazyl (DPPH) 소거 활성 및 HepG2 세포에 대한 항산화 효과

        The purpose of this study was to investigate the anti-oxidant effect of Cynomorium songaricum. The extract of Cynomorii Herba was studied for diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity, HepG2 cell viability and $H_2O_2$-induced cytotoxicity by a modified MTT assay. DPPH radical scavenging activity was measured after 30 minutes. The extract was tested by 1. 5, 10, 50, 100 and 500 ${\mu}g/ml$ concentrations. HepG2 cell viability by a modified MTT assay was measured in the concentrations of 10, 50, 100, 250, 500 ug/ml for 24 h. The results showed that the extract scavenged DPPH radical up to 52.2% with 50 ug/ml concentration. The extract did not reduced the cell viability and $H_2O_2-induced$ cytotoxicity (69.4%) was blocked by the extract in the concentrations of 50, 100, 250 and 500 ${\mu}g/ml$. In conclusion, the extract of Cynomorii Herba has potent antioxidant activity.
