무지개송어 성어에 대량 폐사를 유발하는 IHNV의 분리와 특성

2000년 12월 충북 제천과 2001년 3월 경북 구미에 소재한 무지개 송어 양식장에서 무지개 송어가 대량 폐사되어 병어를 검사한 결과, 외부증상은 지느러미의 출혈과 아가미 빈혈을 보였으며, 해부학적 검사를 행한 결과, 부레와 복강내 지방, 근육층의 출혈 등을 수반하였다. 바이러스 sampledmf 제작하여 세포에 접종한 결과 세포의 구형화를 특징으로 하는 CPE가 관찰되었다. 감염세포를 TEM으로 관찰한 결과 탄환형의 바이러스입자를 확인할 수 있었다. 바이러스 입자의 크기는 직경 80㎚, 길이 170㎚이었고 입자의 표면은 envelope으로 둘러싸여 있는 것을 관찰할 수 있었다. 이러한 형태적 특징으로 볼 때 Rhabdovirus의 일종임을 확인할 수 있었다. 분리 바이러스주 RTK와 RTJ를 Anti-IHNV-ChAb serum, Anti-RTK serum, Anti-RTJ serum 그리고 Anti VHSV serum과 교차중화반응 시험한 결과 분리 바이러스주 RTK와 RTJ는 Anti-IHNV-ChAb serum, Anti-RTK serum 그리고 Anti-RTJ serum에 높은 중화 항체가를 나타낸 반면, Anti-VHSV serum에는 매우 낮은 항체가를 나타내었다. 단백질 구조 분석의 결과로 L,G,N,MI이 관찰되었으며, 이들의 분자량은 각각 180, 68, 41, 32 kDa에 해당하였다. Anti-IHNV serum과 Anti-VHSV serum을 이용한 교차반응에서 Anti-IHNV serum에 대해서만 반응이 나타났다. 당년생 무지개 송어를 대상으로 병원성 실험을 한 결과, 실험 기간동안 Control 어류와 IHNV-ChAb에 감염된 어류는 폐사 예가 없었으나, IHNV-RTK와 IHNV-RTJ에 감염된 어류는 (자연 감염어와 유사한 증세를 보이며) 각각 80%와 50%가 폐사되었다. High mortality with signs similar to viral haemorrhagic septicemia (VHS) such severe haemorrhages in the skin, muscle and air bladder occurred in the farmed adult rainbow trout, Oncorhynchus mykiss, in Ku-mi and Je-chun area in Korea. The isolates were neutralized by an antiserum against IHNV but not by antisera aganist VHSV. Electron micrograph of an ultrathin section showed large numbers of bulltet-shaped virus particles. The nwly isolated rhabdovirus was composed of five structural proteins. In the western blot analysis the anti-IHNV serum strongly reacted with G, N and M1 protein. The cumulative mortalities of RTK infected rainbow trout(10-12㎝, 9-12g) with 10^(3.5) and 10^(1.5)TCID_(50)/㎖ were 80% and 30%, respectively. RTJ infected fish showed 50% mortality by infection with 10^(3.5)TCID_(50)/㎖. Control group and IHNV-ChAb exhibited no mortality. From theses results, the viruses were identified IHNV although diseased fish showed similar signs with VHS and caused high mortality in large-sized fish.

In this study, PLA blends with an Acrylic elastomer as block copolymer (MMA-co-BA) and Raspberry-like PMMA-Silica hybrid nanoparticle at various compositions were prepared from the internal mixer. Hybrid nanoparticles were prepared using tetraethyl orthosilicate (TEOS) via sol-gel method in the presence of PVP-functionalized PMMA with a silica content of about 15 wt%. The flexibility of PLA increased about 150 % by adding the optimized amount, about 30 wt% of the acrylic elastomer. The addition of hybrid nanoparticles on the PLA/Acrylic elastomer blends showed increase tendency in the mechanical and thermal properties, and it was found that the silica having difficulty to be dispersed in polymer induced from the hydrogen bonding is well dispersed in PLA/ Acrylic elastomer blends. This could be attributed to not only the physical dispersion of raspberry-type PMMA-Silica, but better compatibility between PMMA and PLA, a carrier system in the blend.

Cellulose nanofibers(CNF) have attracted much attention because of their high mechanical properties, renewability and potential biocompatibility. However, CNF is agglomerated by high polarity has a limitation in nanomaterials. This study aimed to investigate synthesis and surface treatment behavior of silica nanoparticles onto CNF. The adsorption behavior and silica particle size could be controlled by adjusting the synthesis conditions. Increasing the water content reduced the packing efficiency of the silica particles. Hydrophobic CNF/silica nanocomposites were modified using hexadexyltrimethoxysilane(HDTMS). The silica contents, molecular structure, morphology and contact angle were characterized for CNF/silca nanocomposites. As a result, The nanocomposites were confirmed that the silica content was about 55~60wt% and raspberry morphology. In addition, the CNF/silica/HDTMS nanocomposites of superhydrophobic showed a contact angle of about 160°.

This study aimed to investigate oxidative biodegradation behavior in oxo-biodegradable low-density polyethylene (LDPE) films. The oxo-biodegradable LDPE films were prepared by T-die twin extrusion with 1, 2, 3 wt% additives. It does not show a significant decrease in melting and crystallization temperature, and elongation and tensile strength compared to LDPE. The degradation behavior was investigated by a change in mechanical properties and molecular structure on heat or UV/heat exposure. The oxidative biodegradation at 50 ℃ was able to observe the COO peak after about 50, 70, and 100 days, and the elongation decreased after about 130, 150, and 170 days. In accelerated weathering analysis at UV and 50 ℃, the elongation decreased after about 4 and 8 days, and the COO peak also increased significantly. Finally, the oxo-biodegradable LDPE films were decreased mechanical properties by 100%, and the molecular weight was decomposed from about 280,000 g/mol to less than 30,000 g/mol.
