        The degree of major histocompatibility complex matching between purebred Maltese and mongrel dogs using microsatellite markers

        Long-term maintenance of transplanted organs is one of the major factors that increases survival time of recipients. Although obtaining a major histocompatibility complex (MHC)-matched donor with the recipient is essential for successful organ transplantation, there have been limited reports on MHC matching between dogs. In this study, we analyzed the canine MHC matching rates using Maltese, one of the most popular purebred dogs, and mongrel dogs in Korea. Genomic DNA was extracted from blood leukocytes and DNA was amplified by polymerase chain reaction with primers specific to MHC microsatellite markers. TheMHC matching degree was confirmed by the microsatellite markers using polyacrylamide gel electrophoresis. The MHC matching rates of each donor-recipient groups including Maltese- Maltese, mongrel-mongrel and Maltese-mongrel were 4.76%, 5.13% and 6.67%, respectively. There were no significant differences in the MHC matching degree between each group. These results demonstrate that MHC-matched donors could be selected from other breeds as much as from the same breed for transplantation. Knowledge of the MHC matching degree of purebred and mongrel dogs would offer valuable information not only for improving the success rate of organ transplantation surgery in canine patients but also for transplantation research using experimental canine models.

        돼지 장기이식에서 급성거부반응 연구에 효과적인 엉덩오목이식 동물모델: 줄기세포유래 Humanized 조직의 안전성 평가모델

        To consider the iliac fossa as the vascular anastomosis site of kidney transplantation for the short-term study of acute rejection in pigs. Twelve domestic pigs weighing 39~48 kg underwent heterotopic renal allgraft transplantation. The experimental animals were divided into 2 groups in terms of renal vascular anastomosis site; the external iliac artery and vein were used in iliac fossa model (n = 6), the abdominal aorta and the caudal vena cava inferior to the kidney were used in abdominal cavity model (n = 6). Renal function was evaluated by daily measurement of plasma creatinine and BUN concentrations. The experiments’ health including postoperative complications was also assessed daily for 8 days after transplantation. After euthanazation gross and histopathologic analysis was performed. All six pigs in iliac fossa model developed neuropraxia and lameness of the ipsilateral pelvic limb. However, no necrosis was observed in any pigs. In the abdominal cavity model, durations of both the surgical operation and the vascular anastomosis were significantly longer than those in the iliac fossa model. Furthermore, ischemia injury of the transplanted kidney was increased in abdominal cavity model, which induced accelerated-acute immune response from day 4 after transplantation. Despite of pelvic limb complication, the iliac fossa model showed more advantages including not only less ischemia time related to easy vascular anastomosis, but also less immune response during the acute rejection period. The results indicate that the iliac fossa model may be appropriate to the study of acute rejection in porcine kidney transplantation.

        탈세포화 기법을 이용한 돼지 바이오 스캐폴드: 환자 맞춤형 장기재생을 위한 역분화 줄기세포 동물모델

        The shortage of transplantable kidneys has many efforts to regenerate bioartificial kidneys using transgenic animals and diverse kinds of scaffolds which are important tools for cell seeding. However, there are many limitations for clinical applications so far. Recently, decellularized bioscaffolds using animal organs come into spotlight because of its many superior advantages. In current study, we produced decellularized kidney bioscaffolds of pig which is an attractive animal as a clinical model for human. We decellularized pig kidneys with 1% SDS detergent solution using peristaltic pump systems for 12h. After decellularization process, the kidney bioscaffolds preserved intact 3D morphology including glomerular structure and almost DNA from pig was entirely removed. In addition, this process could preserve micro vascular network which is necessary for cell survival. Although, additional studies for recellularization and transplantation should be required, the decellular vascularized kidney bioscaffolds might have many potentials for kidney regeneration.

        간엽유착과 담낭염을 동반한 선천성 복막심낭 횡격막 허니아의 효과적 수술처치

        A six months old, male Shih Tzu dog was referred to the Veterinary Medical Teaching Hospital at Kangwon National University, for lethargy, respiratory embarrassment. Survey radiography revealed enlarged cardiac silhouette due to displacement of gas-filled intestine in the pericardium. Although there were no specific clinical signs, this case was diagnosed as peritoneopericardial diaphragmatic hernia (PPDH) based on the results of positive contrast radiography and thoracic ultrasonograpic findings. In addition, the thickened gall bladder wall observed in ultrasonography, increase of alkaline phosphatase and neutrophils indicated cholecystitis. Two hepatic lobes and gall bladder were severely necrotized and adhere to the diaphragm. Direct reduction of the herniated organs might cause hepatic hemorrhage and bile juice leakage which may induce very poor prognosis. To solve the problems, the adhered organs were dissected with part of diaphragm, and lobectomy with cholecystectomy was performed for prevention of some possible complications including peritonitis. This is case report describing resection of part of diaphragm adhered to herniated organ reduce the risk of possible hepatic hemorrhage during surgical correction of PPDH followed bycholecystectomy and lobectomy.

        어린 cocker spaniel 종에서 발생한 신부전증

        In the present study, we address systemically a case of renal disease developed in a 1 year-old male cockerand pathological examinations. The animal has been sufferedfrom renal dysfunction signs such as polyuria, anorexia, vomiting, diarrhea and weight loss. The dog was very weakand emaciated and had foamy contents with foul-smell in oral cavity. The animals showed notable decrease in thenumber of red blood cells and severe decreases of hemoglobin and hematocrit with or without changes of meancorpuscular volume and mean corpuscular hemoglobin concentration values, indicating microcytic or normocytichypochromatic anemia. In serum chemistry, blood urea nitrogen, creatinine, phosphorous, Na and Cl, which areassociated with renal function, were dramatically increased. In addition, alanine aminotransferase, aspartate transferase,alkaline phosphatase, cholesterol, lipase and amylase were also significantly elevated, while K concentration was notablydecreased. Urinalysis indicated prominent proteinuria with increase of bilirubin. Despite of symptomatic treatments,the dog was getting worse in healthy condition and dead in the end. At necropsy, both kidneys were brownish, pale,ts. Histologically, the kidneys indicated prominent segmentalor diffuse interstitial fibrosis in cortex and medulla as well as glomerulonephritis. The clinical signs, clinical pathologyand histopathological abnormalities of the young dog presented were consistent with chronic glomerulonephropathy,which was suspected to be a case of familial renal disease in the juvenile cocker spaniel dog.

        Necroptosis Is a Mechanism of Death in Mouse Induced Hepatocyte-Like Cells Reprogrammed from Mouse Embryonic Fibroblasts

        Liver transplantation is recommended for patients with liver failure, but liver donors are limited. This necessitates the development of artificial livers, and hepatocytes are necessary to develop such artificial livers. Although induced hepatocyte-like cells are used in artificial livers, the characteristics of mouse induced hepatocyte-like cells (miHeps) reprogrammed with embryonic fibroblasts have not yet been clarified. Therefore, this study investigated the mechanisms underlying the survival, function, and death of miHeps. miHeps showed decreased cell viability, increased cytotoxi-city, decreased hepatic function, and albumin and urea secretion at passage 14. Addition of necrostatin-1 (NEC-1) to miHeps inhibited necrosome formation and reactive oxygen species generation and increased cell survival. However, NEC-1 did not affect the hepatic function of miHeps. These results provide a basis for development of artificial livers using hepatocytes.

        늑골에서 발생한 개연골육종의 증례

        We report large tumors of the left thoracic cavity, which arose from the ribs, were diagnosed as chondrosarcoma in a 4-year-old male mixed dog. The dog was presented with swelling in the left side of the chest wall and lameness. The masses were found to be circumscribed, whitish-grey, large and firm. The cut surface revealed whitish-grey lobules of varying size with cartilaginous consistency. Those were subsequently metastasized to the lung and mandibular lymph node. Histologically, the tumors were made up of clusters of chondroid cells having pleomorphic hyperchromatic nuclei with occasional mitosis. A special and immunohistochemical staining was performed to confirm the diagnosis. Chondroid matrix in tumor showed a positive reaction for alcian blue-periodic acid-Schiff staining. Immunoreactivity to S-100 proteins was present in the cytoplasm and/or nucleus of chondrocytes in tumor. The final diagnosis was grade III chondrosarcoma in the rib, considering histological features in grading criteria.
