        뇌경새(腦梗塞) 환자(患者)와 건강인(健康人)의 맥상(脈象)과 사상체질(四象體質)에 관(關)한 연구(硏究)

        This study was performed to determine whether a pulse analyzer was useful 1) to characterize the variables of pulse wave of cerebral infarction patieno (CI), compared with those of healthy subjects, as well as 2) to determine Sasang Constitution in CI and healthy subjects. 1. Calibrated in Gwan, the amount of energy(Energy), height of main peak(H1), height of aorticvalley(H2), height of aortic peak(H3), total area of pulse wave(At), and area of main peak width(Aw) of the CI group were higher than those of the healthy group. 2. Calibrated in Cheek, Energy, H1, H2, H3, height of valve valley(H4), At, Aw, and main peak angle(MPA) of the CI group were higher than those of the healthy group. 3. Among the healthy (subjects) group, Taeumin showed the highest contact pressure(CP) and height of valve peak(H5) calibrated in Chon. The main peak width divided by whole time of pulse wave(MPW/T) calibrated in Gwan and Cheok, was highest in Soyangin and was lowest in Taeumin. The H3 divided by H1(H3/H1) and the time to valve valley minus the time to main peak and divided by T[(T4-T1)/T] calibrated in Cheek were highest in Soyangin. The time to main peak(T1) was longest in Soumin. 4. Among the CI group, At calibrated in Chon was widest in Taeumin and was narrowest in Soumin The time to aortic peak(T3) calibrated in Cheek was longest in Soumin and was shortest in Soyangin. The time to valve peak(T5) was shortest in Soyangin. 5. There were main effects of cerebral infarction in the area of systolic period(As) and area of diastolic period(Ad) calibrated in Chon, Energy calibrated in Cwan, and Energy, H1, H2, H3, (H4+H5)/Hl, and MPA calibrated in Cheek. 6. There were main effects of Sasang Constitution in (T4-T1)/T, area of systolic period(As), and Ad calibrated in Chon. 7. The interactions between the cerebral infarction and Sasang Constitution were observed in H5/H1 , T, At, As, Ad, and MPA calibrated in Chon, H4, T4, (T4-T1)/T, As, and Ad calibrated in Cwan, and 74,75, and MPW calibrated in Cheok. Therefore, we concluded that pulse analyzer was useful to determine the risk degree of cerebral infarction and Sasang Constitution.

        소양인(少陽人)에 응용(應用)되는 인동등지골피탕(認冬藤地骨皮湯)이 Streptozotocin 투여(投與) 고혈당(高血糖) 백서(白鼠)에 미치는 영향(影響)

        In order to investigate experimented effects of Indongdeung Jigolpi-Tang, which is applied to Soyangin, to diabetes, the measurement, comparison and observation made on the contents of volumes of blood glucose, total cholesterol, triglyceride, phospholipid, total protein and electrolyte($K^+$, $Na^+$) in blood serum by giving a high blood glucose, induced by streptozotocin, to hyperglycemic rats, dosing the cooked liquid of I.J.T., and following conclusion was attained. 1. The volume of glucose urine, induced by streptozotocin to hyperglycemic rats, was shown an increase in comparision to a normal flock when $12.50m{\ell}/kg$ of I.J.T. dosed to the rats and checked after two and four weeks and thereby, a sinificant reduction in the volume of blood glucose was made. 2. The volume of total cholesterol of blood serum in the diabetic rats, induced by streptozotocin, was increased in comparision to the normal flock, and in the fouth week of dosing $1.25m{\ell}/kg$ of I.J.T., a significant reduction of total cholesterol volum was shown in the blood serum. 3. The volume of triglyceride in the blood serum of the diabetic rats, induced by the streptozotocin, was increased in comparision to the normal flock, and with dosing $1.25m{\ell}/kg$ and $12.50m{\ell}/kg$ of I.J.T., a significant reduction of triglyceride volium was shwon in the blood serum after second and fourth week. 4. The volume of phospholipid in the blood serum of the diabetic rats, induced by the streptozotocin, was somewhat reduced in comparision to the normal flock, and with dosing $1.25m{\ell}/kg$ of I.J.T., a significant reduction of phospholipid volium in the blood serum was shown after first and second week. 5. The volume of total protein in the blood serum of the diabetic rats, induced by the streptozotocin, was somewhat reduced in comparison to the normal flock however, with dosing the I.J.T., no significant change on the volium of total protein was made in the blood serum. 6. The volume of potassium in the blood serum of the diabetic rats, induced by the streptozotocin, was increased in comparision to the normal flock however, with dosing the I.J.T., no significant change of potassium content was made in the blood serum. 7. The volume of sodium in the blood serum of the diabetic rats, induced by the streptozotocin, was somewhat reduced in comparision to the normal flock, and with dosing $12.50m{\ell}/kg$ of the I.J.T., a significant increase was shown in the blood serum after first week. Soyangin, accompanying with a middle and upper heat, the I.J.T. - that has a merits of cleaning up fever, solving fever, solving poison, solving thirst and restoring exhaustion - can be proved that it has a blood sugar reduction effect and it is considered that it can be meritorious for strengthening vitality, further, many other exhausting diseases, and can actively used for the cases such as thirst causing disease and a disease that causes an abnormal food hunger, thirst causing and frequent going to pass urine, in particular.

        열다한소탕(熱多寒少湯) 전탕액(煎湯液)이 Hydrogen Peroxide에 의해 손상(損傷)된 배양(培養) 해마신경세포(海馬神經細胞)에 미치는 영향

        To evaluate the effect of Yuldahansotang(YHT) water extract on cultured hippocampal cell was inhibited by hydrogen peroxide, MTT assay, NR assay, Neurofilament enzymeimmuno assay and DNA synthesis assay were carried out after the cultured hippocampal cells were preincubated with various concentrations of YHT water extract for 3 hours prior to exposure of hydrogen peroxide. The results obtained were as follows: 1. Hydrogen Peroxide decreased the survival rate of the cultured hippocampal cells on NR assay and MIT assay. 2. YHT water extract have efficacy of increasing a amount of neurofilament decreased by hydrogen peroxide in cultured hippocampal cells. 3. YHT water extract have efficacy of increasing DNA synthesis decreased by hydrogen peroxide in cultured hippocampal cells. From above the results, It is concluded that YHT has marked efficacy in preventing for the damages by hydrogen peroxide.

        조위승청탕(調胃升淸湯) 전탕액(煎湯液)이 XO/HX에 의해 손상(損傷)된 배양(培養) 해마신경세포(海馬神經細胞)에 미치는 효과(效果)

        Jowiseungcheongtang(JST) has been in Sasang constitution medicine for many years as a therapeutic agent for cerebral disease. But the effect of Jowiseungcheongtang(JST) on neurotoxicity is not known. The purpose of this study was to determine the effects of Jowiseungcheongtang(JST) on the hippocampal cell injured by Xanthine Oxidase/Hypoxanthine. The results were as follows: 1. XO/HX decreased the survival rate of the cultured hippocampal cells on NR assay and MTT assay. 2. JST water extract have efficacy of decreasing a amount of lipid peroxidation increased by XO/HX in cultured hippocampal cells. 3. JST water extract have efficacy of increasing DNA synthesis decreased by XO/HX in cultured hippocampal cells. From the above results, It is concluded that JST has marked efficacy in preventing cultured hippocampal cells from the damages by XO/HX.

        글루타티온 S-전환효소 다형성과 뇌혈관질환(腦血管疾患) 및 사상체질 사이의 연관성(聯關性)에 관한 연구

        Glutathione S-transferase polymorphisms (GST) were examined in 98 cases with cerebrovascular disease (CVD) to test the hypothesis that GST polymorphisms confer a risk to an individual to develop CVD. Tobacco smoke is a major cause of both cancer and vascular disease. We therefore were stratified the subjects with CVD for smoking status, and then examined whether polymorphisms in this detoxification enzyme gene, GST, influence risk of CVD. Neither GSTM1 nor GSTT1 genotypes in the CVD group was significantly different from the control group (n=230), even in smokers. We attempted the combined analyses for GSTM1 and GSTT1 genotypes in CVD for smoking status. No significant association observed between the combined genotypes and CVD. We also classified the subjects and control group into four types according to Sasang Constitutional Medicine, Korean Traditional Oriental Medicine, and investigated the association among GST genotypes, CVD, and Sasang constitutional classification. Our observations do not confirm the effect of the GSTM1 and GSTT1 genotypes as a risk factor for CVD, even in smokers. Furthermore, we first attempted to evaluate the efficacy of Sasang Constitutional Medicine, and to find an association with CVD.

        A Study on the association between tumor necrosis factor alpha gene polymorphism and Sasang Constitution in cerebral infarction

        Objectives : Tumor necrosis factor-α (TNF-α), a potent immuno-modulator and pro-inflammatory cytokine, has been implicated in many pathological processes. In this study, the authorexamined whether promoter region polymorphism in the TNF-α gene at position –308 affect the odds of cerebral infarction (CI) and whether genetic risk is enhanced by Sasang constitutional classification. Methods : 212 CI patients and 610 healthy controls were genotyped and determined according to Sasang constitutional classification. The amplified genotypes were analyzed on 8% polyacrylamide gel. The alleles were visualized by ethidium bromide staining. Primers for TNF-α were designed to incorporate a polymorphic site at a position –308 bp of the TNF-α gene into an NcoI restriction site. Restriction digests generated products of 87 and 20 bp for G allele and 107 bp for A allele. Results : A significant decrease was found for the TNF-α A allele in CI patients compared with controls (P=0.033, odds ratio, O.R.: 0.622). However, there was no significant association between TNF-α polymorphism and Sasang constitution in CI patients. Conclusions : My finding suggests that TNF-α promoter region polymorphism is responsible for susceptibility to CI in Koreans.

        Apoptotic Effects of Sipimikwanjung-tang of?Sasang Constitutional Medicine in Human Hepatoblastoma Cells

        Objectives : This study on the antioxidant effect of Sipimikwanjung-tang were undertaken to evaluate the antioxidant capacities and antiperoxidation activities in rat liver tissues. Sipimikwanjung-tang which has been one of the prescription in Sasang constitutional medicine is usually applied for the therapy of various liver diseases. It is elucidated that Sipimikwanjung-tang has the antioxidants on liver tissue of rat and the cytotoxic effects on human hepatoblastoma Hep G2 cells. Methods : Sipimikwanjung-tang extract in antioxidant effects of Hep G2 cells is evalated by MTT assay, DAPI staining, DNA fragmentation assays and FACScan analysis. Results : Sipimikwanjung-tang induced apoptosis in Hep G2 cells, and induced G1 and G2M arrest of the cell cycle as well as a significant increase in PARP and caspase-3 activity. Sipimikwanjung-tang induced an increase in H2O2 generation and the subsequent NF-κB activation and also induced cell apoptosis through the caspase-3-dependent pathways in the low concentration of Sipimikwanjung-tang extracts. However, the high dose of Sipimikwanjung-tang extract in Hep G2 cells inhibits TGF-β1-induced apoptosis via increase in cellular H2O2 formation and NF-κB activation in human hepatoblastoma Hep G2 cells. Conclusions : From this study, The possibility that Sipimikwanjung-tang extracts apply to antioxidentic and apoptotic treatment needed disease is revealed.
