        인체 테논낭 섬유아세포에 대한 닐로티닙의 항섬유화 효과

        목적: 본 연구에서는 인체 테논낭 섬유아세포에 대한 닐로티닙의 항섬유화 효과를 확인하고자 하였다. 대상과 방법: 녹내장수술 도중 채취하여 일차배양한 인체 테논낭 섬유아세포를 1, 5, 10, 20 μM 농도의 닐로티닙에 24시간 동안 노출시켰다. 3-(4.5-dimethylthiazol-2-yl)-2.5-diphenyl tetrazolium bromide (MTT) assay를 이용하여 세포의 생존 정도를 측정하였고, annexin V/propidium iodide (PI) 이중염색 후 유세포분석을 시행하여 세포자멸사 정도를 확인하였다. 이후 세포자멸사 관련 단백질을 웨스턴블롯 분석 방법으로 정량적으로 평가하였다. 결과: MTT assay 결과, 닐로티닙은 10, 20 μM의 농도에서 인체 태논낭 섬유아세포에 대한 항증식 효과를 보였다(p<0.001, p<0.001, 각각). Annexin V/PI 이중염색 후 시행한 유세포분석상 닐로티닙은 세포자멸을 증가시켰다. 또한 닐로티닙은 caspase-3, -9, poly adenosine diphosphate ribose polymerase의 분열을 활성화시켰으며, B-cell lymphoma-extra large, Bax 발현을 억제하였다. 이는 닐로티닙에 의해 유도되는 세포 자멸사가 미토콘드리아 경로를 통해 매개된다는 것을 의미한다. 닐로티닙은 α-smooth muscle actin, transforming growth factor beta의 발현 역시 감소시켰다. 결론: 본 연구를 통해 닐로티닙은 인체 테논낭 섬유아세포의 세포 생존을 저하시키며, 미토콘드리아 경로 매개 세포자멸사를 유도하는 것을 확인하였다. 본 연구의 결과를 토대로, 녹내장수술 후 발생하는 섬유화 문제의 해결책으로서 닐로티닙이 사용될 수 있을 것이라 기대된다. Purpose: To evaluate the anti-fibrotic effects of nilotinib on the survival of cultured human Tenon s capsule fibroblasts (HTFs). Methods: HTF primary cultures were obtained from samples following glaucoma surgery. Primarily cultured HTFs were exposed to 1, 5, 10, and 20 μM nilotinib for 24 hours. The effects of nilotinib on HTF proliferation and cell viability were determined using the 3-(4,5-dimethylthiazone-2-yl)-2,5-diphenyl tetrazolium (MTT) assay, and apoptosis was determined by flow cytometry using annexin-V/propidium iodide (PI) double staining. Apoptosis-related proteins were detected by western blotting. Results: The MTT assay showed that nilotinib induced an inhibition of HTF proliferation at concentrations of 10 and 20 μM (p < 0.001 and p < 0.001, respectively). Annexin V/PI double staining showed significantly increased apoptosis in cells treated with nilotinib. Nilotinib activated caspase-3, -9, and poly adenosine diphosphate ribose polymerase cleavage, and downregulated the expression of B-cell lymphoma-extra large and Bax, which indicated that nilotinib-induced apoptosis was partly mediated through the mitochondrial pathway. In addition, treatment with nilotinib decreased the expression of α-smooth muscle actin and transforming growth factor-β. Conclusions: Nilotinib decreased cell survival of cultured HTFs and induced mitochondria-mediated apoptosis. The results suggested that nilotinib may exert antiproliferative effects on HTFs, making it a possible agent to control postoperative fibrosis in patients undergoing glaucoma surgery.

        In this study, kinematic analysis and simulation of a linear-adjustment pipe-reproduction robot were performed. A straight adjustment pipe used in ships consists of two flanges and a straight pipe that connects the pipes of the ship at a precise angle. A robot that accurately reproduces a straight adjustment pipe is referred to as a reproduction robot. To accurately control a reproduction robot, the moving distance and rotation angle of the robot must be calculated accurately. For this purpose, the kinematics of a linear adjustment pipe reproduction robot were analyzed, and a simulation program was created based on the analysis results. The simulation results confirmed that the adjustment pipe on the drawing can be accurately reproduced using a reproduction robot. Therefore, the kinematic analysis and simulation results of the linear adjustment pipe reproduction robot performed in this study can be used to operate the reproduction robot.

        The Effect of Acupuncture on the Intestinal Smooth Muscle Dysfunction Caused by Enteric Bacterial Infection in Weaning Piglet

        It is now generally accepted that acupuncture is effective in diarrhea caused by bacterial infection. However, its effect on the intestinal smooth muscle dysfunction is not clear. Therefore, we investigated the effect of acupuncture therapy at Jiao-chao (GV-1) on the intestinal muscle dysfunction in weaning piglets orally infected by Escherichia coli. The animals are divided into four groups; 1) E. coli + no-treatment, 2) E. coli + antibiotic, 3) E. coli + acupuncture, 4) normal group. In the three E. coli infected groups, low frequency electrical field stimulation (EFS, 1 Hz) provoked triphasic responses composed of initial relaxation followed by on-contraction and off-contraction. While in the normal group, EFS (1Hz) induced biphasic responses composed of relaxation during the stimulation and off-contraction. At the high frequency (16Hz) EFS, both on-contraction and off-contraction of the E. coli + antibiotic, E. coli + acupuncture and the normal group were larger than those of the E. coli + no-treatment group. In the non-adrenergic non-cholinergic (NANC) condition, only biphasic responses occurred to EFS in all experimental groups and the off-contraction of E. coli + antibiotic, E. coli + acupuncture and the normal group were larger than those of the E. coli + no-treatment group. The response to carbachol of those three groups was also significantly greater than that of the E. coli + no-treatment group. These results suggest that acupuncture is as effective as antibiotic in the dysfunction of colonic circular muscle caused by E. coli infection. The maintenance of contractile neuromuscular transmission seems to be involved in the mechanism of the acupuncture effects on diarrhea.

        고속액체크로마토그래피를 이용한 가축용 호르몬제(고나도렐린, 프로게스테론, 옥시토신, 에스트라디올) 분석방법 개발

        The aim of this study is to develop an optimal analytical method for gonadorelin, progesterone, oxytocin and estradiol, the major components of hormones. A relatively simple and reproducible method using high performance liquid chromatography was developed and as a result of the measurement of specificity, linearity, repeatability, accuracy and intermediated precision, the validity of the developed method was verified with the result of meeting the verification criteria of analytical method validation. Using this newly developed method, 12 post-market veterinary products were tested and the ingredient content were 91.9∼116.4%, which satisfied the 90∼120% condition of the administrative measure standard. Therefore, if the newly developed method is used for the collection examination of hormone in veterinary medicine, it can be useful as an approved test method.

        고속액체크로마토그래피를 이용한 항원충제 분석법 개발: 톨트라주릴 및 디클라주릴

        This study was undertaken to develop new analytical methods for assessment of anticoccidials. High-performance liquid chromatography (HPLC) was found to be a fast, reliable, and practical method. The anticoccidials used in this experiment were toltrazuril and diclazuril, and the analysis factors were specificity, linearity, accuracy, repeatability, and intermediate precision. The linearity of each anticoccidial was better than 0.99, and the accuracies were 99.5% and 99.1% with relative SD of 0.5 and 0.4, respectively. To assess whether the developed HPLC method could be effectively applied, toltrazuril and diclazuril post-market veterinary products (five products) that are currently sold were tested. The results revealed no non-compliant items and the method was applied successfully. Therefore, the newly developed HPLC method for anticoccidial assessment described in this study may be useful as a reference method in the Korean Standards of Veterinary Pharmaceuticals for the analysis of toltrazuril and diclazuril.

        2010년 국내산 및 수입 축산물의 잔류 동물용 의약품 탐색 조사

        The Korean National Residue Program consists of three sampling plans for domestic and imported foods of animal origin: monitoring, surveillance/enforcement and exploratory testing. Monitoring and surveillance/enforcement testing programs are routinely implemented by 17 Provincial Veterinary Services for domestic products and two regional offices of Animal, Plant and Fisheries Quarantine and Inspection Agency (QIA) for imported products, respectively. The exploratory testing is designed to test substances which are not included in the list of monitoring and enforcement testing programs controlled by headquarter of QIA. In 2010, the exploratory testing was carried out in domestic and imported foods of animal origin for 24 veterinary drugs including florfenicol, clavulanic acid, four quinolones (nalidixic acid, difloxacin, marbofloxacin, orbifloxacin), two anthelmintics (closantel, levamisole), two sedatives (azaperone, carazolol), six glucocorticoids (dexamethasone, betamethasone, flumethasone, prednisone, prednisolone, methylprednisolone), eight non-steroidal anti-inflammatory drugs (phenylbutazone, paracetamol, carprofen, flunixin, ketoprofen, meloxicam, tolfenamic acid, acetylsalicylic acid). In the total of 1,153 domestic samples, only florfenicol was detected from 17 pig muscles at levels of 0.2~614 ng/g. Of 17 positive pig muscles, 16 samples were non-violative and one sample was violative. In the total of 1,065 imported samples, florfenicol was detected at 0.4 ng/g in one pork. Also, flunixin was detected at 22 ng/g in one beef.

        In order to know whether in understanding concepts of the falling motion of the middle school students the learning guidance based on the history of science is the effective learning method or not, on the basis of investigation and analysis of the degree of preconception of the falling motion of students, the conceptual lesson is done through learning the process of changes of the scientific concepts appearing in the history of science and discussion. In the light of the student' motivation and interest, when lesson was over, they said that the learning guidance based on the history of science was better than the traditional ones. Students' grades were about 15% better than that of the control group.

        The physical significance of the activity constant ζ, which is needed to describe electrodynamics of an isotropic medium, is discussed in electroconductive media and non-electroconductive media. In non-electroconductive media, where the sources of the charge density ℓ and current density J , and the conductance σ vanish, the speed of propagation and the refractive index of two circularly polarized transverse modes are different, and the physical phenomena called as the optical activity take place. Also in electroconductive media, in which energy of electromagnetic wave through the medium is damped, the phase velocity and the absorption coefficient of the electromagnetic waves depend on ζ . The skin depth and the angular frequency also depend on ζ .
